It starts slowly, the worry that builds in Nick’s chest. 
First comes the absences. Ellie Bishop, dedicated federal agent, starts missing days of work. She always has an excuse—her car broke down, she had a family emergency back in Oklahoma, a friend at NSA needed a favor she’s not at liberty to discuss. Nick believes her, at first, but after a while, it becomes too common to be mere coincidence. (He knows better than to believe in coincidences, anyway.)
Then comes the exhaustion. Ellie comes in with shadows under her eyes one morning, and they don’t leave after that. She’s still her generally chipper self, she’s just… muted. Quiet. She’s clearly so tired that she can barely keep her eyes open when she’s sitting still, but she brushes Nick off every time he asks her why.  
Then comes the change in her appearance. She starts wearing her hair in ponytails every day, often with thick headbands around her face. She gets thinner, her already slim figure shrinking and her clothes getting baggier. Again, she sidesteps every question her teammates have for her.
Then—perhaps most worryingly—comes the change in her behavior. She begins to move as if her joints are stiff and the benefit of every step has to be weighed against what it’ll cost her. She stops snacking all the time and eventually Nick realizes that he hasn’t seen her eat a thing in front of him in weeks. 
Finally something happens that he can’t ignore. She can’t evade him after this.
They’re assisting with a drug bust, routine and unconcerning, when a Navy ensign high on meth takes a swipe at Ellie with his knife. She stumbles slightly trying to get out of the way and mostly succeeds, but the knife catches her in the forearm. 
Nick tackles the guy furiously, but the damage is done. 
As soon as he’s got the guy handcuffed and squared away, he goes back to check on his partner. “Ellie, are you alright!?”
She’s sitting where he left her, her jacket balled up and pressed against her skin to staunch the flow of blood. She’s grimacing, but she turns her expression into some approximation of a smile when she sees him watching her. “Yeah, I’m fine. It could have been a lot worse.”
“Let me see.”
Ellie rolls her eyes but pulls her jacket away to uncover the wound. It’s not very long and doesn’t look very deep, but there’s a lot of blood. “Oh my god, you need stitches! Go to the freakin’ hospital and stop trying to be a hero, Bishop.”
Ellie gives him a look. “I’m not ‘trying to be a hero’, okay? It’s just a scratch.” She presses her jacket to her arm again.
“‘Just a scratch’ wouldn’t bleed like that.”
“How would you know? You’re not a doctor.” She gets to her feet angrily, intending to march away from him and his overprotective nonsense, but she sways slightly once standing and realizes that moving right now isn’t a great plan. 
“I might not be a doctor, but I’m not an idiot.” It’s only then that Nick notices how pale she is, and he grabs her uninjured arm urgently to keep her upright. “Seriously,” he insists, his voice hard. “I’m not joking. You need to be looked at.”
“No, I don’t! I’m fine!”
Nick shakes his head at her, not understanding why she’s being so stubborn. “You’re obviously not! What are you, afraid of needles or something? Come on, I’ll hold your hand and you can look away. Stitches aren’t that bad.”
“I’m not afraid of stitches,” Ellie snaps, “I just know what a doctor’s going to say if I show up to the ER, that’s all!”
“Oh, yeah? And what’s that?”
She wrenches her arm out of his grasp. “They’re going to say that I shouldn’t be working in the field where I can get hurt because I bleed too easily right now. They’re going to want to bench me, Nick! I can’t do that, alright? Are you happy now!?”
Nick catches onto the most important part of her little rant. “Why do you bleed too easily right now?” he asks her softly.
Her answer rips the breath from his lungs.
“Because I have leukemia.”
Her defiant expression fades as she watches him process the bomb she just dropped, and after a moment of silence, she sighs. “Don’t freak out, okay?”
“Don’t freak—don’t freak out—what!? How could I not freak out?” Nick feels wild, maybe slightly unhinged, and he has the almost undeniable urge to kick something. 
“Nick! Nick, look at me.” She places her hands on his shoulders and faces him squarely. “It’s not as bad as it sounds. I swear.”
“Leukemia means cancer, Ellie!”
“I know. But mine is at an early stage and it’s a kind that’s treatable, okay? The prognosis is pretty good.”
“Pretty good!? Pretty good’s not good enough. Not when—” He shuts his mouth into a hard line and looks away. Not when it’s you. Not when they’re talking about someone he can’t stand to lose.
“Not when what?” Ellie prompts.
Nick shakes his head and refuses to answer.
She winces a little, imagining what’s going through his mind. “I know I should have told you a long time ago. It’s just… I don’t want word getting around. If Gibbs didn’t bench me, Vance would.”
“You really think I would tell anyone if you didn’t want me to?” 
“No, of course not, but… you know secrets are hard to keep at NCIS. It wasn’t personal.”
“So you’ve just been… going through this alone?” 
There’s a heartbreak in his voice that Ellie doesn’t know how to deal with. She was expecting anger or sympathy, not pain. “Not alone, no,” she hastens to assure him. “I mean, my family knows.”
“And all those days you’ve been missing, were those appointments?”
“I have to have chemo infusions every now and then, and scans. And meetings with an oncologist and a few other doctors.”
“Alone,” Nick repeats. “Your family is what, a thousand miles away? It doesn’t matter if they know. They can’t exactly be there for you.”
Ellie shrugs uncomfortably. “It is what it is. Look, I’m sorry for not telling you, okay?”
Nick sighs deeply, feeling like he’s aged ten years in the span of ten minutes, and pulls her into a gentle hug, careful to avoid jostling her injured arm. “That’s not what’s important here,” he murmurs, reveling in the sensation of having her in his arms; it seems right, no matter how frail she might be at the moment. 
He feels her smile against his shoulder and his heart stutters. She’s so precious to him that he can’t imagine her death without something catching in his throat. 
“I’ll be alright. I promise.”
“Of course you will,” he murmurs, and after a second of hesitation, he shuts down the part of him that’s terrified of being made vulnerable. “You have to be, and you will, because you have me. I won’t let anything happen to you.”
Despite knowing deep down that Nick is as powerless here as she is, Ellie can’t help but believe him. He clearly means it, and who is she to argue with a man ready to take on the universe and win?
“You’ll be just fine,” he whispers, and kisses the top of her head. 
Of course she will be.

Playing with Genes: Gene Therapy : Part 2
Candidate diseases
During the last 4 decades, gene therapy for many diseases has progressed from preclinical to clinical studies; these range from monogenic recessive disorders such as hemophilia and cystic fibrosis to more complex diseases such as cancer, cardiovascular disorders, human immunodeficiency virus (HIV), neurological and ocular pathologies. The prevalence of diseases that fall under the scope of gene therapy is enormous and most of them have catastrophic or fatal outcomes. Therefore, gene therapy approach for almost any of the diseases mentioned above has an obvious appeal and rationale. To date, more than 1,540 gene therapy clinical trials have been initiated, and these are continuing or have been completed in 28 countries, using more than 100 genes, including antigens, cytokines, tumor suppressors, growth factors, and deficiency genes.
Candidate monogenic disorders that are considered to be good candidates for treatment by gene therapy include the hemoglobinopathies, X-linked genetic disorders, amino acid metabolism disorders, and lysosomal and other storage diseases. Currently, there are more than 4000 monogenic diseases registered in the OMIM database. The ultimate aim in treating monogenic diseases with gene therapy is the correction of the disorder by the stable transfer of the functioning gene into dividing cells (stem/progenitor cells), which will ensure the permanence of the correction. The first recognized successful clinical gene therapy trial involved the treatment of 11 children who suffered from SCID-X1 (Severe Combined Immunodeficiency), an X-linked inherited monogenic disorder caused due to a mutation in the common cytokine receptor gamma chain (γc). In these patients, immunity was not fully developed due to the blocking of T-cell and natural killer cell development as a result of mutation. Unfortunately, the trial in SCID-X1 also exemplified one of the potentially serious side effects of gene therapy. Three of the children developed uncontrolled clonal T-cell proliferation, that is, leukemia, almost 3 years after treatment. This case was associated with the integration of the retroviral vector close to the promoter of the LMO-2 proto-oncogene. As a result, LMO-2 protein expression was up-regulated in an abnormal way and resulted in leukemia. Another candidate monogenic disorder for gene therapy is cystic fibrosis (CF), in which abnormally thick mucus is produced in the lungs of the patients, causing difficulty in breathing and increasing the frequency of serious lung infections. CF is known as the most common inherited genetic disease in Europe and USA, especially within the Ashkenazi Jewish population. The average life expectancy of patients with CF is less than 40 years; hence the treatment of this disease has become one of the prime targets of gene therapy research.
In addition to its use in the treatment of monogenic diseases, gene therapy is also becoming a therapeutic alternative for the treatment of various forms of cancer. Indeed, almost 65% of ongoing clinical trials are related to cancer (more detailed information is available at www.wiley.co.uk/genmed/clinical), an area in which much more promise can be seen; this is also a reflection of the urgent need for new therapies to tackle the escalating incidence of this disease. Several different principles are used to treat cancer, including gene therapy that is targeted at tumor suppressor genes, such as p53, or central signaling molecules, as well as "suicide gene" therapy, in which the transgene is capable of converting pro-drugs (selectively less active drugs) into drugs that are toxic for tumor cells. Furthermore, various gene therapy protocols have been developed to strengthen the host’s anti-tumor immune responses by immunotherapy. Most of these studies have been early clinical trials designed primarily as studies of the safety, applicability, and toxicity of gene therapy. Several of these phase I and II studies have, however, shown partial remission of tumors and, in rare cases, complete remission. However, complete cure of the tumor has not yet been achieved. In some trials, including TP53 gene therapy trials, regression in tumor size has been observed in up to 50% of patients. China has become the first country to license gene therapy as a regular treatment for neck and head cancer; here Gendicine, a replication-defective Ad5 vector expressing p53 from a Rous sarcoma virus (RSV) promoter, is used for therapy.
In addition to its applications for cancer and monogenic diseases, gene therapy has become one of the favorite methods in cardiovascular research field. This is in step with the rise in clinical trials for cardiovascular gene therapy from 8.3% to 9.1% during the last few years, becoming the second most popular application for gene therapy. In accordance with the variety and occurrence of cardiovascular diseases, different gene therapy strategies have been developed to tackle each disease on its own terms. The expectation is that gene therapy will provide a new avenue for therapeutic applications in the growth of blood vessels, as well as the protection, regeneration, and repair of heart tissue, the prevention of the reoccurrence of constricted or narrowed arteries following cardiovascular intervention, the prevention of the rejection of a bypass, and risk-factor management. Long-term therapeutic gene expression is required in some diseases, such as hypertension research, where reversal and prevention are the key targets. On the other hand, for some other types of cardiovascular diseases, such as ischemia, atherosclerosis, and restenosis, shorter-term gene control will be sufficient to prevent further progress of the symptoms. Therefore, different gene-therapy vectors have to be considered for the treatment of each specific cardiovascular disorder.
Consequently, gene therapy offers new avenues of treatment for diseases including monogenic disorders, cancer, cardiovascular diseases, infectious pathologies and many more. As we follow the tradition, “God did not send down any illness for which He did not also send a cure (Bukhari)”, gene therapy using either viral, non-viral, or any other novel methods may pave the way for the cure of many diseases that are highly prevalent in the world and which for decades have been perceived as untreatable.

so...what's your thoughts on the ending of raw?
I feel like they pulled the trigger on everything really quick because of the leukemia relapse, but obviously they’re making it work as best as they can for what’s happening. It’ll be interesting to hear Dean cut a promo next week about his reasoning because I feel like the way that they set it up, Dean was obviously emotional about the whole night and then got offended for Seth celebrating their victory or something? It’s weird, I need to watch it again. 
I can watch Dean and Seth fight all day and be entertained. I dunno. I don’t follow any wrestling blogs for their opinions and I don’t track tags so I don’t know what anyone else thinks and I think I’m happier for it. I’m wondering if someone will go to smackdown at the end of the day as well, and I’m really super wondering if Crown Jewel is just going to implode on itself and be canceled. 
Hoping Roman recovers fine and we get a really dramatic WWE 24 I can use to clear out my sinuses mostly. I think he’ll at least enjoy some time with his family. I know a lot of cancer survivors and while it’s not something to be scoffed at, a lot of people get it and at least from the angle of mentally tackling it, he’s been there before so he knows what to expect. Now it might be even easier because he has buckets of money to throw at it.

BIOGRAPHY --
For the first few years of Raul Vargas’s life, nothing happened. At least nothing out of the ordinary. He was the first-born child – a son at that – and as such, the trajectory of his life had more or less been decided for him before he had even been born. He was the heir, the future protector of the family name, and the shoulder upon which all of the responsibilities would lay. This was nothing new; Raul had been socialized to accept these things as his lot in life from the moment of his birth, as had his father before him, and any son that he should have in the future. This all came to a head, though, when his younger sister, Elena, was born. Raul was five years old and already everything his parents could ever have hoped for – he was serious and articulate, the spitting image of his father, and he carried himself with an arrogance that could only be born of knowing your place in the world. Elena, though, was the opposite. She was fragile and small for her age; it seemed like she was always in and out of the hospital. Just days before her first birthday, she was diagnosed with leukemia. As unfortunate as this was, the Vargas family was prepared to tackle this problem head on, as they would any other that came their way. They even thought that they were more aptly prepared than other families. After all, there was no expense that they would not pay to help their child, and no length they would not go to. It also helped that the Vargas name was well known in medical circles as the Vargas patriarch had sat on the board of one of the most outstanding hospitals in the New York metropolitan area for decades.
Despite all of their resources and their far reach, Elena Vargas did not live passed her second birthday. The trauma of it deeply affected young Raul and he withdrew into himself. A once bright spirit had been dulled under the guilt of having failed to save his sister. Noticing this change in their son, the Vargases instituted monthly family camping trips in the hopes that getting away from the responsibilities of their life, even for just a weekend, would help their family recover from the loss. It was on one of these camping trips that the Vargas family experienced an upheaval once again. A teenage Raul had just returned from a job around the campsite when Mariana and Silas stumbled in. They were disoriented and terrified. Little did the Vargas know that what was supposed to be a routine camping trip would lead to two adopted children. For the first few months after the adoption, Raul tried his best to maintain his distance from his new siblings. There was a part of him that was terrified of the idea of becoming too close to them; after all, he had loved and failed Elena at a young age and the idea of failing another sibling was too much for him to bear, but despite his best interests, his new siblings wheedled themselves into his heart. Mariana especially. She was feisty and protective, often attempting to take care of Raul despite their great age gap, and he loved her for it.
When Raul turned 18, there was little discussion about the course that his life would take. Naturally, it was assumed that he would go to college, major in business, and one day take over his father’s role on the hospital’s board. Raul, though, had different ideas for his future. Without his parents’ knowledge or their blessing, he enlisted in the United States Army. The country was embroiled in overseas wars and Raul believed that his skills would be better served actually helping people. He served two tours overseas in the role of combat medic, and when he returned, the world was different. Raul was different. He had lost some of the spark in his eyes and grown a much harder outer shell, but there was still a part of him that was as Vargas as before he had left. Arrogant as ever and determined to stave off the PTSD that plagued other servicemen, Raul applied to medical school. Now, at thirty-six, he has since completed medical school and is serving as an ER doctor in the same hospital on whose board his father sits.
