Multiple myeloma is a blood cancer affecting plasma cells, white blood cells that produce antibodies, causing them to multiply abnormally in the bone marrow. Right now, only half of patients can expect to survive five years after diagnosis, so better treatments are sorely needed. Promising targets include fatty acid-binding proteins (FABPs), a multi-functional family of proteins that can pick up lipids, involved in a variety of processes. A recent study found that cultured myeloma cells (pictured, their nuclei in blue) expressed high levels of one member in particular, FABP5 (in red); blocking it reduced cell proliferation, while patients with higher FABP5 levels tended to experience worse outcomes. Inhibiting FABPs more broadly appeared even more effective, improving survival in some mice with myeloma. To build on these results, further research needs to investigate why not all mice responded so well, and whether inhibiting FABPs would be safe and effective in humans.

Abstract Leptin is mainly produced from adipose tissue and released into the circulation. Circulating leptin binds to the leptin receptor (LEPR) in the brain, which activates signaling pathways that inhibit feeding and promote calorie expenditure. The leptin receptor (LEPR, also known as Ob-R) gene is located at chromosome 1p31. DNA methylation consists of addition a methyl group at position 5′ of the pyrimidine ring of the cytosines upstream of a guanine (dinucleotide CpG) catalyzed by DNA methyltransferases. Methylation of cytosine in CpG sites is an important epigenetic modification way that could suppress the gene expression. This study was conducted to reveal the role of leptin (-2548 G/A, rs7799039) and LEPR (-668 A/G, rs113711) gene polymorphisms in patients diagnosed with multiple myeloma (MM). Patients who were diagnosed with MM and followed-up in our clinic between January 2010 and January 2022 were included in the study. The genotypes of the leptin (-2548 G/A, rs7799039) and LEPR (-668 A/G, rs113711) genes were statistically compared between patients and healthy controls. Additionally, the statistically significant effects of these genotypes on survival were examined. In addition, the methylation status of the patients was compared to the healthy control group, and the effect on survival was evaluated. A total of 300 patients diagnosed with MM and 170 individuals to form a healthy control group were included in this study. In the statistical analysis performed to investigate the effect of leptin and LEPR gene polymorphisms on disease susceptibility, GA and AA genotypes of the leptin gene were found to be significantly higher in the patient group compared to healthy controls. In the statistical analysis for -31 NT and -51 NT methylation of the leptin gene, -51 NT methylation was found to be significantly higher in healthy controls (p = 0.002). In the survival analysis, progression-free survival (PFS) of patients with GG genotype of the LEPR gene was found to be significantly shorter compared to others, there was no effect on the overall survival. In the multivariate analysis, it was revealed that the PFS of patients with GG genotype of the LEPR gene was 2.02 times shorter compared to others (RR: 2.017; CI: 1.191–3.418, p = 0.009). MM and leptin polymorphisms have significant features in terms of both disease susceptibility and treatment response.

James McCaffrey, actor best known to video game fans as the voice of Max Payne, among other characters, has passed away. He was 65.
TMZ reports that McCaffrey died on Sunday after a battle with multiple myeloma, a form of cancer. A rep for McCaffrey tells the outlet that the actor died surrounded by friends and family.
McCaffrey’s extensive on-screen credits include roles in Law & Order, Suits, and Blue Bloods, and video games such as 2008’s Alone in the Dark and 2005’s Area 51. However, Remedy fans knew him best as the voice of Max Payne in all three entries. Furthermore, McCaffrey voiced Thomas Zane in Alan Wake, Zachariah Trench in Control, and, most recently, Alex Casey in Alan Wake 2.

Chemotherapy: Chemotherapy is a frequently employed approach for leukemia treatment. It entails the use of medications to either eliminate blood cancer cells or impede their growth. Chemotherapy can be administered orally, intravenously, or via injections and is often combined with other treatments like radiation therapy or stem cell transplantation. The chemotherapy regimens vary based on the type and stage of leukemia. For instance, aggressive chemotherapy may be necessary for inducing remission in cases of ALL or AML. Patients typically undergo multiple cycles of chemotherapy with breaks in between to allow their bodies to recover from side effects.
Radiation Therapy: Radiation therapy employs high-energy X-rays or other radiation forms to target and destroy cancer cells. It is especially effective in treating leukemia that has spread to specific body areas, such as the central nervous system. Often, radiation therapy is used alongside chemotherapy.
