Immortalized cell lines are highly regarded for scientific research. Over the period of a century, we have been able to collect numerous immortalized cell lines. They are either harvested from the tumors of individuals suffering from the condition. Unlike primary cells, which have a limited lifespan, immortalized cell lines can be generated by making genetic alterations that encourage continuous growth and division. This conversion is essential for providing a stable and sustainable source of biological material, which greatly benefits areas such as drug development, cancer studies, and genetic research. Having a deeper understanding of these procedures improves our capacity to effectively harness immortalized cell lines across various scientific disciplines.

man i love fanfiction but i find i barely read any these days. as i get older, I really cannot fucking tolerate how in SO MANY fics, the Main Guy's dialogue is extremely strong and rich and interesting, and then Female Lead's voice/dialogue is reduced to just. cracker dust. from the bottom of the bag.

buck: let them fire us, who cares!
chimney, not holding the braincell: you hear that tommy THAT is the spirit of the 118!
buck, very clearly not the one holding the braincell: that should be our new motto, who cares!
eddie, not really holding the braincell either: that's not a very good motto
buck: well not if you take it out of context
tommy, currently holding the braincell: guys i dont think we should be worrying about getting fired right now

(I had double incision top surgery)
HOLY CRAP MY CHEST LOOKS SO GOOD!! IM SO HAPPY!!
I've seen ALOT of post op photos. I had watched multiple videos of the actual surgery I was getting, and also looked at a FUCK TON of pre-op, post-op, and other healing stages photos.
I have literally never seen a chest look this good at any of the first healing stages. That's insane, I'm so happy.
My actual surgeon didn't have any photos of their masculinizing top surgeries, even though they'd done it multiple times before, so i kinda felt a bit nervous about that. I wasnt sure exactly how it'd look, even though i know the initial stages can look gnarly but heal phenomenally.
IT. LOOKS. SO. GOOD.
I'm so happy and also a bit in shock about how good!! Ahhhhhhhh!!!
(I really want to share photos tbh, but i have tattoos that could be identifying so i won't here. I may post somewhere else with my tattoos blurred. Maybe transbucket?)
~~~~~~~~~~~~~~~~~☆~~~~~~~~~~~~~~~~~~
Anyways the surgery went perfectly!! My mast cell was totally under control!! My chest has healed so well!! I didnt need the ICU and have been recovering at home!!
He was able to hook up my nerves on each nipple, so i should expect sensation still! AND he was able to preserve my tattoos, which all have extreme emotional/sentimental significance to me!
All the issues I have been having, are sooooo much less than we were expecting, and all the doctors, nurses, and just medical teams for this entire thing have been so good!! Especially the anesthesiologist and pain team that worked with me for the actual surgery.
So, besides some hiccups and more mild expected issues related to my other health conditions, I'm doing so well!!!
This is such good news, and means a couple of my conditions are much more controlled than I thought!!
(the "more" section just has more detailed health info, and my experiences so far)
~~~~~~~~~~~~~~~~~☆~~~~~~~~~~~~~~~~~~
I did have some significant & expected prolonged issues from the anesthesia.
I didn't wake up for multiple hours, then woke up but was still totally paralayzed for multiple hours and couldnt communicate that so they thought I wasn't waking up for even longer, RIP.
Then once my right side started to return, my left was still almost totally paralayzed for over an hour too.
(We're hoping that was just a hemiplegic migraine, because I do get complex migraines and have had hemi-paresis during some. That doesn't follow the pattern of my paralysis episodes from my channelopathy (which is why i was paralyzed still for so long.) So the assumption is I was having paralysis episode and a hemiplegic migraine at the same time.)
I'd also had significant increased weakness for about 2 days, I still have increased weakness now, over a week post op but its probably not related to the anesthesia directly. Most likely my mitochondria taking a hit, contributing to overall fatigue, worsening of my channelopathy, and my adrenal issues.
There were also some issues getting a new pain medicine since i had a lot of adverse reactions to what was initially prescribed. But the doctors have been very good about helping me find alternatives.
They'd gave me a nerve block on both sides initially (which apparently is almost never available, but i was able to get!!), and a localized pain pump for the first 3 days, then a muscle relaxer and pain medicines. I've couldnt take the muscle relaxer or the pain meds due to like severe adverse reactions, so now that the localized things ran out, I've been suffering a bit while waiting for a dr to prescribe something else for me.
And I'm actually still needing increased steroids for my adrenal issues, which is considered "abnormal" to still be needing at this point. I'm hoping once I have a bit more pain control from these new meds we're trying, then I won't need as much steroids. *fingers crossed*
Then, I have an adhesive allergy, and we've been able to avoid tapes and adhesives almost entirely. I haven't had any blistering from the paper tape they've used in some areas. I did have some itching which I've been taking benadryl for and it is more controlled now.
~~~~~~~~~~~~~~~~~~~☆~~~~~~~~~~~~~~~~
I feel very very blessed. ♡♡♡♡♡♡♡
Im so grateful I was able to get this surgery, to have it covered entirely by insurance, to have such a good team of doctors helping me, for my body to have done so well with this whole process, that my chronic illnesses have been controlled as well as they have with my current treatments, that my doctors are helping me find alternatives for pain control, that my chest has already healed so well, that i have the supports in place where i can recover at home, even with prolonged issues or temporarily worsened symptoms, that the surgeon could hook up both nerves and preserve my tattoos. I am so blessed and I am so grateful, and I am so happy, I'm so excited for when its all healed up.
[Henry] had asked Lady Shelton whether Mary [was] persisting in her obstinancy. Hearing that she was, Henry became certain that she was being encouraged by secret communication from Katharine. Lady Shelton thought the only possible messenger was Mary's chamber woman. In this she was correct. The maid had been smuggling letters in and out. She was dismissed, as was Mary's confessor, to be replaced by one whom Chapuys characterised as 'Lutheran'.
The King’s Pearl: Henry VIII & His Daughter Mary, Melita Thomas
#as i'm going through this refresher in tandem with reading weir's new novel...#she actually writes shelton as being the one that managed to get her mother's letters to her into her hands#even for fiction that feels...far fetched#ostensibly someone had to be getting her letters from chapuys as well; even chapuys reports at times#that he doesn't know how it's possible she's getting letters out to him#but i doubt it was either of the boleyn aunts here#nor margaret bryan; anne's maternal aunt#even the interpretation that anne was a nonentity by this point and had no clout; basically#would not bear this out; if they didn't fear anne then they certainly would've had reason to fear henry#and i doubt they would've circumvented what he ordered#until after jan 1536 (where shelton is allowing visitors from chapuys bcus she's been sent gifts) this just does not seem to be the case#melita thomas#(also had weir been more faithful to primary sources. then this interpretation would mean shelton threw this chamber woman under#the bus...which she did; but in her rendering it would be to save her OWN skin#rather than at great personal risk which is what she#portrays; for the construction a sympathetic character in lady shelton)#i also think there's a question of agency on this unnamed maid's part that i don't really ever seen given space...#insofar as the hierarchy of privilege etc#was she actually willing to risk her income to do this? that's generally how it's portrayed#but it's just as possible that she felt constrained to do so bcus mary; despite her demoted status; was obviously her superior#even if not her employer#not to mention after being dismissed for such a reason; it's not like she was going to get a reccomendation to another household#it's fair to talk about how both coa and mary were placed in these hostile environments but the hostility and tension#those placed as their servants (not those that had chosen to be there; like elizabeth darrell for coa)#is again...not given the same space; generally#it was probably very frustrating to serve two highly privileged women that refused to answer or look at or acknowledge them#because they were addressing them as the law required.#you can imagine the eye-rolls of the servants which coa called 'gaolers'. since. yk.#a person of a servant's status was likely to have a friend or relative that spent time in an *actual* jail cell. if not themselves .
I have a lot of thoughts about the premise(?) of Hololive -Advent-*, the newest set of Hololive VTubers, which is weird, because in the end, there's not much to it. (And that's the problem.)
*That's not me, that's how their group name is consistently formatted in, like, video descriptions and stuff. I don't know why.
All five of them are escapees from The Cell, which is...I'm not sure what it's supposed to be. On one hand, Nerissa and Bijou are in there because they're supernatural anomalies who put people in danger by being around them; on the other hand, Shiori and the Abyssgard twins are there because they angered "the gods" by discovering forbidden knowledge and being annoying. The Cell is kind of an SCP-Foundation securing-dangerous-anomalies thing, kind of a prison for people "the gods" don't like, and VTuber lore just isn't substantial enough to make those two tropes feel like a single thing instead of two tropes.
The next problem is, in my opinon, more significant. It feels like Cover is trying to make absolutely nobody in this premise come off as truly unsympathetic. (I don't know if they're actually trying to do that, but that's what it feels like.) Now obviously, they don't want the VTubers to come off as unsympathetic monsters; they range from completely innocent girls deemed threats to the world, to annoying brats.
But at the same time, plenty of details paint the Cell as not being so bad. First off, there's the SCP vibes. Bijou is a jewel whose luster (shininess) drives people into violent conflict over her, and Nerissa's singing could drive the world mad. Something needs to be done about their anomalous properties, and while locking them up is obviously wrong, it's at least a sympathetic motivation.
Second, the Cell seems pretty comfy. This may just be a consequence of needing to explain why a bunch of prisoners know about anime and Minecraft and Twitter, but it still serves to dilute the Cell. If the prisoners are treated well, it's not much of a prison.
Which leads into problem 3: The escaped-prisoners thing is barely even an aesthetic. Which isn't inherently a problem; look at HoloJP generation 3, aka Hololive Fantasy—a necromancer, an elf, a knight, a rabbit, and a pirate. Not much there, but that's not a problem, because AFAIK they didn't try to make a big deal out of it.
But HoloEN puts more effort into the lore of its generations, because Anglophone fans respond well to it or something. This usually works fine, and I think that's because Hololive Council and Tempest picked themes that don't require much support or consistency. A bickering council of divine beings and an adventuring party are both simple ideas, ones that would be suited to an episodic slice-of-life anime or fantasy vlog channel or something. Their divine council/adventuring party things are as superficial and undeveloped as Advent's prison escapee thing, but they fit the vibes and rhythm of VTubing.
A bunch of criminals who just escaped prison? Not so much. That premise has conflict baked into it. There's an antagonist, a clear goal (avoid going back to jail), all sorts of stuff. There's necessary worldbuilding; you can't escape from prison if there isn't a prison, and you can't have a prison without laws and someone to enforce them. And it's hard to make this mesh with a typical VTuber thing.
To make this work, I feel like Cover would need to go in one of two directions.
One would be to drop or de-emphasize the "prison escapee" angle. I'm thinking something along the lines of HoloX, the JP branch's evil secret society. Just as HoloX trying to take over the world is in the theoretical future, you could put this hypothetical generation's prison escape far enough in the past to be less significant. They're not on the run from whatever authorities are in charge of The Cell, they're just vaguely renegade people doing slice-of-lifey renegade things.
Two, commit to the bit. Make the Cell a place where "the gods" or whomever stick people they don't like, make it unpleasant, build VTuber personas and backstories within the constraints of that backstory. It would be different, it would be unfamiliar, but it would be unique.
I want to end this on a positive note, so I'll say this: In the abstract, the Cell is a fun idea. If nothing else, it's a fun fanfiction idea. I would read a fanfic about Holomems escaping the Cell for reasons other than the novelty of VTuber fanfiction.
Primary Cells Market Trends, Review, and Forecast 2024–2030
The Primary Cells Market was valued at USD 1.6 billion in 2023 and will surpass USD 3.1 billion by 2030; growing at a CAGR of 10.0% during 2024 - 2030. Primary cells, derived directly from living tissues, maintain the physiological relevance of human biology, making them invaluable in scientific research. Unlike immortalized cell lines, primary cells retain their unique characteristics, providing more accurate models for in vitro studies. This blog explores the key trends, growth drivers, opportunities, and challenges within the primary cells market.
Key Market Trends Driving Growth
Increasing Adoption in Drug Discovery and Development Pharmaceutical companies and research institutions are leveraging primary cells for drug screening and toxicity testing. These cells offer a more accurate prediction of drug responses compared to traditional cell lines. As personalized medicine gains momentum, primary cells enable more individualized and predictive models, allowing researchers to identify specific responses to therapeutic agents.
Advancements in 3D Cell Culture and Organoid Models One of the major trends in the primary cells market is the increasing use of 3D cell culture and organoid models. These advanced culture systems more closely mimic the structure and function of human tissues, offering an enhanced platform for studying disease progression, drug efficacy, and patient-specific therapies. The integration of primary cells into these models is expected to further accelerate research in fields such as oncology, neurology, and regenerative medicine.
Growing Demand in Cancer Research Primary cells, especially cancer-associated cells such as tumor cells or cancer-associated fibroblasts, are crucial in cancer research. With the rising incidence of cancer, there is a pressing need for more accurate in vitro models that replicate the complex tumor microenvironment. Primary cancer cells, derived directly from patient tissues, are providing researchers with the tools to develop more effective therapies and understand tumor behavior better.
Expansion of Biobanking and Cryopreservation The expansion of biobanks and cryopreservation services is another major factor contributing to the market’s growth. Primary cell biobanks offer vast repositories of cells from diverse human populations, allowing researchers to study genetic variations and disease-specific models. With the increasing emphasis on precision medicine, the demand for high-quality, well-characterized primary cells has surged, enhancing the role of biobanks in supplying these valuable resources.
Opportunities in the Primary Cells Market
Rising Interest in Regenerative Medicine Regenerative medicine is poised to transform the treatment of various degenerative diseases, and primary cells play a key role in this revolution. Stem cells, a type of primary cell, have shown tremendous potential in regenerative therapies for conditions such as heart disease, neurological disorders, and diabetes. The growing pipeline of regenerative therapies represents a lucrative opportunity for companies specializing in primary cell production and related services.
Emerging Markets and Technological Innovations Emerging markets, particularly in Asia-Pacific and Latin America, are becoming attractive for key players in the primary cells market. The increasing healthcare investments, supportive government policies, and growing focus on biotechnology research in these regions are expected to fuel demand for primary cells. Additionally, technological innovations in cell isolation, culture, and cryopreservation techniques are likely to open new avenues for growth in the market.
Partnerships and Collaborations As the complexity of cellular research increases, partnerships between academic institutions, biotech companies, and pharmaceutical firms are becoming more common. Collaborations in areas such as cell sourcing, assay development, and therapeutic applications are enhancing the capabilities of market players. These partnerships are expected to drive innovation and accelerate the adoption of primary cell-based models in various industries.
Ask for Customization Report: https://intentmarketresearch.com/ask-for-customization/primary-cells-market-3630.html
Challenges Facing the Primary Cells Market
Limited Availability and High Costs One of the primary challenges in the market is the limited availability of certain types of primary cells, particularly those from rare or difficult-to-access tissues. The cost of isolating, culturing, and maintaining these cells can be prohibitively high, which can restrict their widespread adoption, especially in resource-constrained settings. Additionally, ethical concerns surrounding the sourcing of human tissues remain a challenge that needs to be carefully managed.
Variability and Short Lifespan Unlike immortalized cell lines, primary cells have a finite lifespan, and their characteristics can vary between donors. This variability can introduce challenges in reproducibility and consistency of experimental results, making it difficult to standardize protocols across different labs. While efforts to improve cell culture techniques and reduce variability are ongoing, this remains a significant obstacle for researchers.
Regulatory Hurdles As primary cells are increasingly used in drug development and regenerative therapies, navigating the complex regulatory landscape is becoming a key challenge. Regulatory bodies such as the FDA and EMA require stringent validation of cell-based models, which can delay the approval and commercialization of new therapies. Ensuring compliance with ethical standards for human tissue sourcing and use further complicates the regulatory process.
Conclusion
The primary cells market is poised for robust growth in the coming years, driven by advancements in personalized medicine, drug discovery, and regenerative therapies. The increasing adoption of 3D cell culture systems, expansion of biobanking, and the growing focus on cancer research are key trends shaping the market's future. However, challenges such as high costs, cell variability, and regulatory complexities must be addressed to unlock the full potential of primary cells. As technological innovations continue to emerge and collaborations expand, the primary cells market is set to play a pivotal role in the future of biomedical research and therapeutic development.
my sister got engaged and we’re all really happy for her but my bitter rain cloud of a dad (who naturally she told last) is giving her a bit of passive aggressive grief about it despite her boyfriend being like the best man of our generation (presumably either because he’s not catholic or because my dad sees them as young dumb unemployed people who aren’t ready for marriage or because he’s mad he barely has any real love with his own wife or something). so like pray for us? i wish i knew what to do
#if my dad had any brain cells or observational skills whatsoever#he’d realize that in terms of our faith the problem is not the boyfriend. that guy is brilliant and open minded and would probably ace RCIA#the problem is my sister. who is catholic in name but it’s clear to me how hard she’s fallen away from the faith#but like my dad has created such a bitter home environment we never have meaningful conversations with him#so like he doesn’t know *anything* about our inner lives#all he sees is labels. all he judges people by is labels#literally you can still get married in the church to a non catholic it’s just a matter of expecting them to convert eventually#and promising to still live according to the principles of the church and raising your children as such#but my parents are absolute fools if they think that’s the issue. if my sister was true in her faith her bf would have converted already#i am sure of it. the guy is smart he just needs to be guided the right way#evidently my parents don’t realize that about him either#if my dad could become a decent parent for once and stop trying to drive his kids away from the faith by only cherrypicking the parts of it#that intersected with republican/conservative boomerisms#ugh. if he was a virtuous father she’d be a virtuous daughter and therefore all her friends and loved ones would be virtuous as well#should i blame my dad for all our family problems? no.. not rightfully……#but like. the impact a father has on one’s life cannot be understated#ugh i’ve had the sense for a while that God wants me to be the one to fix this family#because looking around it doesn’t look like anyone else is gonna do it#but that’s such a daunting task… especially alone… i don’t have any true friends (ie who share both my faith and life experiences)#and like. it’s really hard to try to assume the role of a teacher or counselor when someone is older than you#or uh. in a position of direct power over you for that matter. esp when clearly deeply mentally ill#the concept of trying to essentially parent my own parent while i myself am miserable and unstable#esp when he is the primary cause of that#just. ughhhhh it’s such a vicious circle#like i’ll do this if i have to i’ll undertake that daunting mission but i have to be so careful and really sort myself out first#or for that matter if i were to volunteer to like. catechize my sister’s boyfriend (heaven knows she couldn’t do it)#i’d have to really study my stuff bc i think the intellect is the only real appeal here#like i said tho his conversion can probably never really happen as long as my sister remains the way she is#what i know is that the first step is fixing myself. i have to be a pillar of virtue if i wanna stand as any sort of authority on the faith#problem is i suck and shouldn’t be regarded as a role model for anything. i have the knowledge down but that alone won’t fix me
