AMPK & SirtuinsAMPK (AMP-activated protein kinase) is another critical regulator that responds to low energy levels within the cell (a high AMP ratio). Such a far cry from his beginnings... he's truly taken after the man he hates the most.
Unfortunately for Wesker, adding Uroboros in intensely massive quantities to an existing Progenitor-67 infection is a poor choice that makes him spend much of his fight with Chris alternating between pouncing him like a rabid animal and curling like the death throes of a dying spider.
And unfortunately for Chris, Wesker doesn't die when he's left at the volcano to stew in a puddle of himself. Even with the awakened lack of thermophilic qualities as a result of the genetic influence of G, there is enough local flora and fauna to keep him alive... albeit in a subdued state.
Physiological:
Uroboros, combined on the initial foundation of Progenitor 67, takes after Progenitor 67's metabolic inefficiency and drives that far higher than it was ever meant to go in an attempt to sow compatibility.
Due to this, Wesker needs a truly ridiculous amount of calories (~6,000 - ~9,000 kcal/d) to remain fully intelligent.
A lesser amount will generally retain him, though he will experience a plethora of negative symptoms related to starvation (~3,500 - ~5,500 kcal/d).
The very brink of his sanity and humanity will find him lacking if he consumes the average human male's daily caloric intake (~1,800 - ~3,000 kcal/d).
Wesker cannot digest iron bound to salt or any organic chelates except heme (or hemeprotein) as a result of initial Progenitor 67 infection.
Progenitor 67 epigenetically alters his enterocytes. The genes involved with this are the upregulation of HCP1 and the downregulation of DMT1 and, less intensely, Dcytb, and is a result of both Progenitors' own natural leaning and, primarily, the hasty introduction of leech genes when microinjection became available.
This ultimately means that, as time drags on, he begins to find his palate shifts from a lot of power smoothies to meat.
But there is something crucial here: 67% or more of heme iron is lost when you cook meat. That means...
↑ He slowly slips into preferring his meat blue or raw, otherwise he won't be able to properly supplement his need for heme iron.
↑ With the addition of Uroboros, which dethrones and replaces Progenitor, this change persists and couples roughly with his modified caloric needs.
Usually present in brown adipocytes, UCP1 and UCP2 are both overactive as a result of the improper inclusion of Uroboros' gene signalling.
As a result of this, Wesker's natural temperature is hotter than a normal human being's.
He's probably uncomfortably warm to others - almost naturally feverish.
UCP1 and UCP2 cause proton leakage across the mitochondrial membrane - UCP1 being the primary uncoupler regarded in thermogenesis.
↑ As a result of this leak, ATP use is less efficient. This exacerbates his metabolic disorder.
↑ The inclusion of UCP2 upregulation alongside UCP1 was done out of an abundance of caution.
The gluconeogenesis enzyme PEPCK-C, involved in the burning of fat to produce glucose, is overexpressed in dual infection.
While this gives Wesker higher stamina and physical endurance, it comes at the expense of contributing to insulin resistance and is a key player in his bodies' teetering metabolic imbalance.
↑ This is because Uroboros doesn't upregulate and express PEPCK-C in just skeletal muscular cells - it does so where it is already present, too; this is a personal oversight.
↑ PEPCK-M was personally avoided in its' development because it is a popular oncogene.
Uroboros; a monster that eats itself continually without end. It physically presents a sign of Wesker's malingering, endless need to consume across his skin.
Patches of necrotizing, hardening skin and flesh form across his arms and legs in uneven patterns.
Scaly, itchy lesions creep along his cheekbones and the sides of his neck like a corrupted skin flush.
These are a weaker, useless vestigial presentation of the hardening of skin he undergoes when he's being flash-fried by high temperatures.
They're positive for many of the proto-oncogenes related to melanomas and are undoubtedly at least oncogenetic, but aren't strictly cancerous; instead, they're a misguided attempt at defense that vacillates 'on' without facing heat danger.
One of the scars of Wesker's infection and subsequent mutative transformation are hypermelanotic deposits that coat previously-mutated skin in a dark fade.
They start strongest at his fingertips and weaken at the base of his shoulders.
They are also strongest in the center of his chest where his macronuclei is present and taper off approaching his shoulders and abdomen.
His face has some of the same tinting in uneven splotches descending strongest from where he was burned.
Uroboros enhances the strength of many of his bones - more potent the closer to the surface of his body - by fortifying them with calcium and double-thickening them.
As a feature of the beginning stages of Uroboros' starvation, it will begin to leach calcium out of bones and the deposits it creates.
↑ These deposits are not found in the brain or the skull.
When resources are threateningly low, the bones will be stripped for the calcium they uniformly contain, causing him to experience an effect similar to calcium gout / pseudoarthritis, malaise, and brittling.
This leaching undoes itself and becomes re-fortifying under the duress of stress hormones like cholecystokinin and epinephrine. Less specific but still counted in re-fortification is cortisol and norepinephrine.
↑ Re-fortifying can be triggered accidentally through rigorous & sustained physical exercise or copulation.
Epigenetic changes brought on by Progenitor physiologically are stacked upon inappropriately and pseudorandomly by Uroboros like re-patchwork, which leads to a general instability and accidental incorporation of Progenitor's physical defense mechanisms.
This is what ultimately enables him to survive the events of RE5.
Neurological:
Progenitor infection spiraling into his temporal lobes, brain stem, and, minorly, his frontal lobes as years progress are the cause of his irritation and aggression in RE5.
It does this because it has no antimutagenic properties unlike the more developed t-Virus, slowly mutating out of control into a dysfunctional beast as Wesker experiences more and more stress and trauma.
A bit like G in this regard...
His SERT is disrupted by its' presence in the midbrain raphe nuclei along the brain stem.
This causes elevated extracellular serotonin levels initially, followed by receptor downregulation and eventual serotonin deficiency.
The lack of available serotonin leads him to experience, at first, heightened periods of aggression. Later, this becomes mania and even delirium if he forfeits sleep or a PG/67 shot, because...
As time ticks on, his dopamine begins to become dysregulated as well.
This imbalance contributes to mood swings, aggression, and impaired decision-making.
It also leads to a loss of the ability to feel pleasure or joy in things, making him manic and paranoid, taking risks uncharacteristically to feel something, anything.
As Progenitor 67 degrades brain function, particularly areas involved in impulse control and aggression, Wesker experiences the uncontrollable urge to consume.
This leads him to cannibalize interns and people he calculates won't be missed, as well as people who piss him off enough. During this time it does change - and become true - that a misplaced boop! or headpat! might get you killed.
His ego begins to vacillate and strengthen itself in an effort to maintain itself despite his decline. If he can just finish Uroboros, everything will be okay again, he can fix himself...
His brain practically relies on his PG67-A/W dose before he gains Uroboros.
A rapid decline in cognitive and emotional stability when he’s off the dose is present.
It's not unlike sundowning...
You would not want to see his MRIs during this time. They look awful - his brain has lesions and there are parts that are grossly overgrown; there are tiny black specks where Progenitor has gnawed holes in the fabric of him. If he hadn't roped it in, he would probably end up eventually going completely feral from his brain swelling up against the sides of his skull; he'd just become another victim of Cannibal Disease and turn, simply painfully slowly.
Uroboros is not supposed to crawl into the brain. It's supposed to leave it unaffected - but Progenitor having already curdled and spread unchecked into it gives it an opening for opportunistic infection.
It attempts to knit the holes and tamp down overgrown regions, but in doing this, it feeds from a sort of 'genetic template' that restores Wesker's ability to feel and feel greatly. This includes the reactivation (and reanimation) of the greatly disordered regions of his brain responsible for empathy, sleep regulation, attachment, and guilt.
It also cannot perfectly recreate lost tissue or resurrect memories from dead, liquefied tissue, so he retains some of his symptoms, like mood swings.
Rage becomes consuming, hunger becomes voracious, and guilt becomes haunting; he is plagued with an unfortunate (to him) tendency to consider others.
He finds it a prying weakness, but he can no longer tamp it down into nonexistence as he did; he is, essentially, thermally reduced to his Arklay days in behavior. He’s still intelligent and cunning, but he must now wrestle with intense emotional responses that don’t align with his goals at all...
Guilt and empathy are so foreign to him they are nearly out-of-body; he views them clinically and tries to rationalize them away, but his inability becomes the stake that impales him and imparts a malingering sense of conflict.
...and he must take a good, hard, long look at his goals and how far they've diverted from their initial precedence.
... what is he now? What use does he serve in this world - to what master does he heel, clearly dethroned from his position? These questions and more are things he asks himself now.
Is he doomed to roam aimlessly without purpose until he perishes?

Imagine lying back in an enclosed chamber where you bask for 90 minutes in a sea of pure oxygen at pressures two to three times that felt at sea level. This is the world of hyperbaric oxygen therapy (HBOT), a technology that’s been around for decades and is now being explored as a possible treatment for Long COVID.
"The silence on the inside is deafening at first,” says John M.,* who has undergone dozens of HBOT treatments for his persistent Long COVID symptoms. Fortunately, there is a television outside the chamber in view, and it is easy to communicate with the provider if needed. While the potential protocol is still being refined, patients may undergo up to 40 HBOT sessions to address some of the most problematic, lingering symptoms of this complex condition.
HBOT is a therapeutic process that has been widely used to treat such conditions as decompression sickness in scuba divers, carbon monoxide poisoning, and diabetic foot ulcers. In HBOT, the body is exposed to 100% oxygen, a significant increase from the 21% oxygen concentration we typically breathe. The therapy takes place in an enclosed chamber where the air pressure is elevated above normal levels. The combination of high-pressure and high-oxygen conditions enhances the amount of oxygen that can reach the body's tissues. The hope is that this therapy can provide the same relief and healing to people with Long COVID that it does for those with other conditions.
According to John M., HBOT was the first treatment that helped with his sleep and reduced his heart palpitations. “At one point after hospitalization, my Long COVID symptoms were so bad that I could barely walk or talk. HBOT was a great tool that really assisted with my recovery,” he said. John added that he hopes the medical community will achieve a better understanding of how HBOT can help relieve suffering for patients with Long COVID and that more research will increase access to this innovative therapy.
Does HBOT improve Long COVID symptoms? One key observation from the work of Inderjit Singh, MBChB, an assistant professor at Yale School of Medicine (YSM) specializing in pulmonary, critical care, and sleep medicine, is that Long COVID patients often experience debilitating fatigue. Based on Dr. Singh’s previous Long COVID research, the exhaustion is thought to be linked to the muscles’ inability to efficiently extract and utilize oxygen.
To picture how HBOT might work, you can think of your muscles as engines sputtering, struggling to get the fuel they need. If oxygen is the gas that fuels the muscles, it’s as if you are trying to complete your daily routine while the gas tank is running on “empty.” By aiming to directly address this oxygen utilization impairment, HBOT may be a potential solution.
A systematic review by researchers at the China Medical University Hospital noted that HBOT could tackle another major factor in the Long COVID puzzle: oxidative stress. This relates to the body's struggle to maintain balance when harmful molecules, known as free radicals, run amok, causing chronic inflammation.
Research co-authored by Sandra K. Wainwright, MD, medical director of the Center for Hyperbaric Medicine and Wound Healing at Greenwich Hospital in Connecticut, suggests that HBOT, with its high-oxygen environment, might dampen this chronic inflammation by improving mitochondrial activity and decreasing production of harmful molecules. Other potential benefits of HBOT in the treatment of Long COVID may include restoration of oxygen to oxygen-starved tissues, reduced production of inflammatory cytokines, and increased mobilization of hematopoietic stem cells—primary cells that transform into red blood cells, white blood cells, and platelets.
HBOT for Long COVID: Current and ongoing research Several small-scale reports have indicated that HBOT is safe for patients with Long COVID.
To address this question, a trial that followed the gold standard of modern medical research—a randomized, placebo-controlled, double-blind design—assigned 73 Long COVID patients to either receive 40 sessions of HBOT or a placebo of only 21% oxygen. The study observed positive changes in attention, sleep quality, pain symptoms, and energy levels among participants receiving HBOT. In a longitudinal follow-up study published in Scientific Reports, the authors at the Tel Aviv University found that clinical improvements persisted even one year after the last HBOT session was concluded. In a second study, the same authors focused on heart function, measured by an echocardiogram, and found a significant reduction in heart strain, known as global longitudinal strain, in patients who received HBOT.
In another study, 10 patients with Long COVID underwent 10 HBOT treatments over 12 consecutive days. Testing showed statistically significant improvement in fatigue and cognitive function. Meanwhile, an ongoing trial at the Karolinska Institute in Sweden has reported interim safety results wherein almost half of the Long COVID patients in the trial reported cough or chest discomfort during treatment. However, it was unclear whether HBOT exacerbated this symptom or if this adverse effect was due to the effort of participation by patients suffering from more severe Long COVID symptoms.
Is HBOT currently available as a treatment for Long COVID? For HBOT to become a mainstream treatment option for Long COVID, several critical priorities must be addressed. First, there is currently no established method for tailoring HBOT dosages to individual patients, so researchers must learn more about the specific features or symptoms that indicate potential benefits from HBOT. At the same time, we need to identify factors that may be associated with any adverse outcomes of HBOT. And finally, it’s important to determine how long these potentially beneficial effects last in a larger cohort. Will just a few HBOT trials be enough to restore patients to their baseline health, or will HBOT become a recurring component of their annual treatment regimen?
For now, HBOT remains an experimental therapy—and as such is not covered by insurance. This is a huge issue for patients because the therapy is expensive. According to Dr. Wainwright, a six-week course of therapy can run around $60,000. That’s a lot to pay for a therapy that’s still being studied. In the current completed studies, different treatment frequencies and intensities have been used, but it’s unclear how the treatment conditions affect the patient’s outcome.
“I have had some patients notice improvements after only 10 or 15 treatments, whereas some others need up to 45 treatments before they notice a difference,” notes Dr. Wainwright. “I think that HBOT is offering some promising results in many patients, but it is probably a strong adjunctive treatment to the other spectrum of things Long COVID patients should be doing, like participating in an exercise, rehab, and nutritional program.”
Dr. Singh notes that “a major challenge for research is the heterogeneity of Long COVID. It is hard to determine which symptoms to treat and enroll patients into trials based on them.”
Perhaps treatments that target multiple issues caused by Long COVID, like HBOT, may help overcome this challenge.

The last word from Lisa Sanders, MD: Hyperbaric oxygen therapy (HBOT) is just one of the many existing treatments that are being looked at to treat Long COVID. We see this with many new diseases—trying to use a treatment that is effective in one set of diseases to treat another. And there is reason for optimism: We know that HBOT can deliver high levels of oxygen to tissues in need of oxygen. That’s why it’s used to treat soft tissue wounds. If reduced oxygen uptake is the cause of the devastating fatigue caused by Long COVID, as is suggested by many studies, then perhaps a better delivery system will help at least some patients.

saw a bunch of libertarians citing the recent and from what i can tell very awful case of indi gregory, a small child with a rare mitochondrial illness taken off nhs life support whose parents were denied the right to transfer her to an italian hospital that agreed to see to her medical needs. which was ofc trotted out as an illustration of the evils of statist health care, death panels, etc
the obvious retort here ofc is that the main effect of privatising healthcare on cases like these is to multiply them, but what particularly interested me was the judge who issued that ruling, robert roger peel. turns out peel is currently the lead judge of the uk's financial remedies court (for determining financial disputes between divorced or separated couples), and has published an interesting couple of articles on his (very positive) assessment of the court. a lot of his focus is trained on time/cost minimising, settlement:trial ratio maximising measures for the court to implement or that the court has implemented, including (from the 2nd one)
the extensive use of "private fdr's," a sort of privatised dispute resolution in place of the court itself, in which an ex-couple hires a "private fdr judge" (could be a solicitor, barrister or retired judge) to rule on their case without recourse to the actual judicial system. ("So too the widespread use of Private FDRs. Judges need little persuasion to permit parties to attend a Private FDR and return to court thereafter for, as the case may be, a mention hearing to endorse the consent order, or a directions hearing to timetable to trial. The use of Private FDRs has in turn relieved pressure on the courts.")
the use of single lawyers simultaneously representing both members of the former couple, explicitly in order to undermine the costly adversarial nature of the legal proceedings. in short, while this is definitely not an area where i have domain knowledge, my first impression is that the judge responsible directly for this decision is sort of a miserly ghoul happy to undermine the rights of individual brits in the service of shrinking and cost-cutting that portion of the govt over which he exercises authority. (to his credit he does at least declare the lack of public funding for legal representation to be "iniquitous", tho ofc that bit is out of his hands.) many such cases!

I'm actually surprised AC Pocket Camp is still going strong, but good for them! It means we have some new stuff to cover! In particular, there are 10 new things I will cover as an extension of this series and, if ACPC continues to live on and give us new fish, then we'll keep going. Today we will begin the series humbly with the Sailfin Sandfish.

What a cute little guy. This fish first appeared in ACPC in December of 2023 and appears to be a new staple of winter time fishing at Saltwater Shores. It is currently available, most likely into March just as it was last winter. It's very common, so if you have played ACPC recently, you probably already have this.

In 2001 it was discovered that the sandfish most likely belong to Scorpaenaformes, the scorpion fish, like lionfish. There is so much debate around what is and isn't in Perciformes. Just know that these guys are a pretty niche selection for ACPC to pick, and it is most definitely because they are fished extensively in Japan.
But how do they know that? What did they do to find that out? There are a bunch of ways that biologists employ to find out which animals are related to other animals, or simply put, figure out their phylogeny. In the past, and sometimes still used today, organisms are put into groups together based on common traits. The idea, at least we assume, is that if two animals share a trait, it means they are more closely related to each other than they are to animals without that trait. Now, this isn't without caveats (looking at convergent evolution scenarios) but it's still helpful.
In recent times, to get to the nitty gritty of phylogeny and determine with more certainty who is related to whom, we can use DNA. Now what does that really mean? Well, it could mean a lot of things - there are a lot of ways to use DNA to find out all sorts of secrets about disease, heredity, speciation, etc. But we don't have time for that. Without getting down a deeper rabbit hole, you have seen the double helox of DNA before in textbooks and stuff. The DNA is located in the nucleus of the cell where it stores all of the instructions the rest of cell reads in order to build and maintain a living thing. But it's the same concept - the fewer differences in genes (traits) between two species, the more related they are.

But what if I told you that the nucleus isn't the only place you can find DNA? There is also a little bit in your mitochondria (yes, the powerhouse of the cell). The mitochondrial DNA (mtDNA) is only inherited maternally, meaning this DNA usually comes from the mother, the egg cell (there are always exceptions in bio...also, we're just going to focus on animals, but fun fact, plants have some DNA in their chloroplasts, too). mtDNA is really useful in phylogeny, especially when we start to parse out species that are very closely related. The mtDNA is unique from nuclear DNA because it gets less efficient DNA repair, giving it a chance to keep mutations that can build up in a species. Again, this is just comparison at the end of the day - if two species have fewer differences in their mtDNA genome, the more closely related they are. The advantage of using mtDNA is that it is even more precise than nuclear DNA because it evolves faster. (This is actually one way in which new species are found!) You can even use mtDNA to show the relatedness between populations within a species. It's just another really cool way we can find stuff out about our living world!

In this study, they included 25 patients with Long COVID and 21 age- and sex-matched controls who had fully recovered from COVID-19. They collected blood and muscle biopsies before and after exercise. Both groups were healthy and socially active. None of the participants were hospitalized due to COVID-19 infections. Fatigue questionnaires and exercise data confirmed the impact of Long COVID on daily lives. The study found that people with Long COVID have lower VO2 max, indicating decreased oxygen absorption and reduced peak power output. They also had lower gas exchange (CO2 and oxygen) levels, suggesting that their muscles were affected by physical activities. Structural changes in the muscle were observed, with a shift from type 1 fibers (aerobic) to type 2X and 2A fibers (anaerobic). This indicates a shift towards glycolytic metabolism, which is less efficient for sustained energy production. Further, mitochondrial function was compromised in patients with Long COVID, showing lower oxidative phosphorylation capacity. This means their cells were less efficient at producing energy. Magic, being so untameable a force, requires much practice, skill, and control. Therefore, in order to learn how to properly control their magic, young witches are taught using wands, which serve as conduits that focus and strengthen their magic control, much like a metal rod for electricity.
Once strong enough, witches will abandon wands and perform their magic on their own. However, there are still occasions in which they will opt to use a wand, typically when casting magic above their own caliber, as the wand will serve to amplify their own magic and lessen the burnout. If performing a particularly powerful and/or dangerous ritual or spell, they might choose to use a staff, which is far stronger than a simple wand.

Other benefits seem due to increased grey matter in 'front office' functions of the cortex; and neuronal birth, or neurogenesis, in the hippocampus, a brain region that controls aspects of memory and mood regulation. Whatever the mechanism, giving your body a workout will produce substantial benefits in terms of brain health. Remember, a body in motion tends to stay in motion, and your brain and body will be together your whole life.
Mental activity is an obvious, and critical, ingredient for optimizing and maintaining brain function. Studies have established relationships between the degree of life-time mental activity and late-life cognitive function. It's clear that those who engage in intellectually challenging endeavors on a regular basis reap the benefits of a clear mind. There is, however a need for each individual to balance sufficient variety with a proper degree of challenge. Without variety and challenge, tasks become too mundane and too easy, eventually growing stale and losing their capacity to adequately stimulate the brain. We must also realize that mental activity goes beyond 'cognitive' tasks. Mental activities also include practices like meditative focus, relaxation and stress reduction techniques, as well as social interaction.
