University of Houston Researchers Create New Treatment and Vaccine for Flu and Various Coronaviruses - Published Aug 6, 2024
Team Develops Two Nasal Sprays – An Immune Activator and a New Vaccine – To Prevent Virus Transmission
HOUSTON, Aug. 6 — A team of researchers, led by the University of Houston, has discovered two new ways of preventing and treating respiratory viruses. In back-to-back papers in Nature Communications, the team — from the lab of Navin Varadarajan, M.D. Anderson Professor of William A. Brookshire Chemical and Biomolecular Engineering - reports the development and validation of NanoSTING, a nasal spray, as a broad-spectrum immune activator for controlling infection against multiple respiratory viruses; and the development of NanoSTING-SN, a pan-coronavirus nasal vaccine, that can protect against infection and disease by all members of the coronavirus family.
NanoSTING Therapeutic’s Highlights -NanoSTING, a nasal spray, can prevent multiple respiratory viruses by activating the immune system and preventing infection from viruses. It can also protect against SARS-CoV-2 reinfection.
-A single intranasal dose of the NanoSTING has proven effective against strains of SARS-CoV-2 and the flu virus.
-NanoSTING is complementary to vaccines and enables cells to be in a heightened state of alert to prevent attack from respiratory viruses.
NanoSTING-NS Pan-coronavirus Vaccine’s Highlights -UH researchers have developed NanoSTING-SN, a nasal vaccine that prevents transmission to the unvaccinated and fights multiple COVID variants.
-NanoSTING-SN provides the exciting potential towards a universal coronavirus vaccine and may end the cycle of onward transmission and viral evolution in immunocompromised people.
-Intramuscular vaccines prevent disease but are less efficient in preventing infections. NanoSTING-SN can provide improved protection against transmission for COVID variants and related sarbecoviruses.
NanoSTING -NanoSTING is a special formula that uses tiny fat droplets to deliver an immune-boosting ingredient called cGAMP. This formula helps the body's cells stay on high alert to prevent attack from respiratory viruses.
“Using multiple models, the team demonstrated that a single treatment with NanoSTING not only protects against pathogenic strains of SARS-CoV-2 but also prevents transmission of highly transmissible variants like the Omicron variants,” reports Varadarajan. “Delivery of NanoSTING to the nose ensures that the immune system is activated in the nasal compartment and this in turn prevents infection from viruses.”
As the recent COVID19 pandemic illustrated, the development of off-the-shelf treatments that counteract respiratory viruses is a largely unsolved problem with a huge impact on human lives.
“Our results showed that intranasal delivery of NanoSTING, is capable of eliciting beneficial type I and type III interferon responses that are associated with immune protection and antiviral benefit,” reports first author and postdoctoral associate, Ankita Leekha.
The authors further show that NanoSTING can protect against both Tamiflu sensitive and resistant strains of influenza, underscoring its potential as a broad-spectrum therapeutic.
“The ability to activate the innate immune system presents an attractive route to armoring humans against multiple respiratory viruses, viral variants and also minimizing transmission to vulnerable people,” said Leekha. “The advantage of NanoSTING is that only one dose is required unlike the antivirals like Tamiflu that require 10 doses.”
The mechanism of action of NanoSTING is complementary to vaccines, monoclonal antibodies and antivirals, the authors noted.
Nano STING-SN Despite the successful implementation of multiple vaccines against SARS-CoV-2, these vaccines need constant updates due to viral evolution, plus the current generation of vaccines only offers limited protection against transmission of SARS-CoV-2.
Enter NanoSTING-SN, a multi-antigen, intranasal vaccine, that eliminates virus replication in both the lungs and the nostrils and has the ability to protect against multiple coronaviruses and variants.
“Using multiple preclinical models, the team demonstrated that the vaccine candidate protects the primary host from disease when challenged with highly pathogenic variants. Significantly, the vaccine also prevents transmission of highly transmissible variants like the Omicron variants to vaccine-naïve hosts,” reports Varadarajan.
The authors further show that the nasal vaccine was 100% effective at preventing transmission of the Omicron VOCs to unvaccinated hosts.
“The ability to protect against multiple coronaviruses and variants provides the exciting potential towards a universal coronavirus vaccine,” said Leekha. “The ability to prevent infections and transmission might finally end this cycle of onward transmission and viral evolution in immunocompromised people.”
The research was conducted by a collaborative team at UH including Xinli Liu, College of Pharmacy and Vallabh E. Das, College of Optometry along with Brett L. Hurst of Utah State University and consultation from AuraVax Therapeutics, a spinoff from Varadarajan’s Single Cell Lab at UH, which is developing NanoSTING.

Owing to their lipophilic nature, EOs are advocated to penetrate viral membranes easily leading to membrane disruption. Moreover, EOs contain multiple active phytochemicals that can act synergistically on multiple stages of viral replication and also induce positive effects on host respiratory system including bronchodilation and mucus lysis. At present, only computer-aided docking and few in vitro studies are available which show anti-SARC-CoV-2 activities of EOs. In this review, role of EOs in the prevention and treatment of COVID-19 is discussed. A discussion on possible side effects associated with EOs as well as anti-corona virus claims made by EOs manufacturers are also highlighted. Based on the current knowledge a chemo-herbal (EOs) combination of the drugs could be a more feasible and effective approach to combat this viral pandemic.

Various in vitro and ex vivo studies were conducted to study the effects of eucalyptus oils and eucalyptol treatments on monocytes and macrophage recruitment in response to lung inflammation and infections. Data of these studies demonstrate marked immunomodulatory properties of both eucalyptus oil and its active ingredient, i.e. eucalyptol. Both treatments reduced the release of pro-inflammatory cytokines from monocytes and macrophages, but their phagocytic properties were not halted (Juergens et al. 2020; Sadlon and Lamson 2010). Eucalyptol is also known to have mucolytic and bronchodilatory properties (Juergens et al. 2020). Interestingly, eucalyptus oil has also been shown to have disinfection properties and inhibited the growth of viruses on various utensils and filter devices (Usachev et al. 2013). Taken together, data from both preclinical and clinical trials point towards the promising therapeutic potential that resides in eucalyptus oil and its active constituent, i.e. eucalyptol in the prevention and treatment of COVID-19. Therefore, further studies are urgently warranted in this regard.

Bad News of the Week:
One in three people in Germany will need a blood donation in their lifetime, according to statistics from the German Blood Donor Service. This is only in rare cases (12%) due to blood loss in accidents or injuries, as seen on TV. Much more frequently, for example, cancer patients (19%), gastrointestinal patients (16%) or heart patients (16%) are dependent on it during operations or treatments. Even the most modern medicine cannot do without the red lifeblood. Since blood still cannot be produced artificially.
This is why the current development is so dramatic: presumably because of the Corona pandemic, many people stayed as far away from medical facilities as possible. Out of fear of becoming infected as well as out of good will not to be a burden on the already overloaded health care system. Blood donations have therefore fallen dramatically. Currently, only about half of the required blood is available. Regular willingness to donate is also important, because the blood components remain usable only up to the 42nd day after donation.
The situation is just as bleak for organ donations. According to the German Foundation for Organ Transplants (DSO), there are currently around 8,500 seriously ill people urgently waiting for a donor organ - of which there are too few. In 2022 alone, 407 kidney and 86 heart patients therefore died. Yet (to put it sarcastically) no one who dies as a healthy person, for example in an accident, still needs their organs. Others do. Leaving religious convictions aside, the reason for the lack of willingness to donate organs is a frighteningly stupid one: the lack of documentation.
According to the German Federal Center for Health Education (BZgA), 82% of the population is basically willing to donate an organ. But only 44% have documented this - the easiest way is an organ donor card, about which relatives are informed. And this even includes the refusals. The way out could be simple: the so-called "contradiction solution". In this way, everyone would be an organ donor in principle, unless he or she expressly objects. But neither the health minister of the last government - Jens Spahn of the conservative CDU - nor the current one - Karl Lauterbach of the social democratic SPD - could get their way politically. In addition, a national register would have to be set up first. This would take years just for data protection reasons. So for the foreseeable future, only one thing remains: the rare commodity of solidarity. Sadly, this is obviously not enough right now. It costs lives. In Germany alone, every single day.
Good News of the Week:
Like Xi Jinping or Vladimir Putin, Turkish President Recep Tayyip Erdogan has tinkered "his" state to maintain his personal power. The principle is called the "presidential system," which tends to weaken parliament and strengthen the position of the president. Many can't even remember a time before Erdogan; the leader of the AKP (Adalet ve Kalkınma Partisi) party has now been in power for twenty years. Elections are due again on May 14 of this year, and of course the autocrat is seeking re-election.
But headwinds are forming: six Turkish opposition parties forged an alliance this week to prevent just that. The alliance wants to transform the presidential system back into a parliamentary system, strengthen the rule of law and freedom of the press, and bring the president's power back to a democratic level. For example, the president should no longer be able to rule by decree, and his term of office should be limited to seven years.
The initiative comes at just the right time. Many opposition figures have been muzzled by Erdogan's power apparatus in recent years. Most recently, Istanbul Mayor Ekrem İmamoğlu, who was considered a promising candidate. He was sentenced to two years in prison and banned from politics for allegedly insulting the election commission. Perhaps for this reason, the newly formed alliance has not yet named a top candidate.
That alone will not end in one fell swoop the massive economic, administrative, monetary, and judicial crisis that Turkey has now endured for years. But the mere prospect that there might be an alternative with a realistic chance of success to the increasingly inevitable permanent president is welcome. Especially for the fragile democracy on the Bosporus. After all, democracy is the essential basis for Turkey's perspective in the EU. And the nation will not be able to achieve lasting peace, prosperity and security any other way.
Personal happy moment of the week:
Last week I got a new colleague. I was allowed to choose him myself. The good impression his application made was confirmed in an online meeting. We agreed before we had even met in person. Because that didn't happen until his first day at work. Which started chaotically right away, because we had just learned that the office would be moving in the next few days. So the actual work is left undone and we have to deal with sorting out documents, clarifying furniture, planning workstations, packing office supplies, etc. And the new colleague proves to be helpful, hands-on, uncomplicated and resilient. It's nice when the second impression even surpasses the first.

When the Corona pandemic caused x million employees worldwide to move to the home office, not even a quarter of companies took security measures to protect all their private data connections. ...I am disappointed in Brazil's President Lula da Silva. He wanted to do many things differently and better than his unspeakable predecessor Jair Bolsonaro. In environmental protection, for example. Amcor Flexibles goes for Vetaphone corona equipment
Amcor’s Gent production facility is a giant, housing gravure printing in up to nine colors, digital printing, solvent and water-based coating, lamination, coextrusion, both cast and film up to nine layers, and Vision Systems for conversion and extrusion. It all began back in 1925 with the production of cellophane, which was the first plastic material used for food packaging. In 1950, the plant became the first to extrude in Europe, and by the 1960s Amcor Flexibles was making strides in the pharmaceutical, films & packaging, and chemical sectors, with the establishment of a strong R&D culture.
Throughout the 1990s, the Group expanded with numerous acquisitions, including Danisco, A&R, Rexam, and Tobepal, with extruded volumes being consolidated at Gent and the UK plant in Ilkeston. 2008 brought a major 17m Euros investment program at Gent with four new extrusion lines and the construction of a new production hall, while the acquisition of Alcan in 2010 saw conversion and extrusion separated at Gent and in 2019, with the addition of Bemis, the group became a major global leader in package print production.
Today, the Gent plant has a broad product range including peel and barrier packaging that serves all sectors. PE, PP, PA, and PETg are extruded in up to nine layers and on the converting side, gravure and digital printing combine with lamination, coating, and slitting. Both extrusion and converting sectors are monitored by Vison Systems to assure quality.
It was into this production environment that Vetaphone set about selling its surface treatment technology, as Dirk Den Haese, sales manager of Vetaphone Benelux explained, “The challenge of working with a large and diverse group like Amcor is knowing where to start! But I knew that the company valued its reputation for quality very highly, and that the accuracy of much of the packaging it produced was critical, so there was clearly no room for any weak link in the production chain.”
Beginning in 1990, Vetaphone has supplied a diverse range of corona technology to the Belgian plant that ranges from 1400 to 2300 mm in web width and generators from 2 x 6kW for extrusion lines to 2 x 10kW for different lamination lines. For its food and pharmaceutical work, Amcor is using exclusively Vetaphone VP2 ceramic rollers to ensure no contamination from the silicone sleeves, vulcanized and aluminium backing rollers.
This has become a standard item for Amcor Flexibles in Gent, where the treaters have performed well, drawing praise for their low maintenance requirements and easy access, and reliable performance that provides the consistently even treatment essential for a quality end-product.
Speaking for Amcor, Andries Van Nieuwenhove, plant general manager at Amcor Flexibles Conversion Gent said, “It is key for us to have good partners who support us in our journey to be the reference supplier of specialty peelable and barrier laminates.”
