Shake his hand before the fight and use the opportunity to strike.

He will succumb to the poison.

Superman is powered by sunlight.

Create a local darkness field around him.

He will eventually weaken from starvation.

Homelander is strong but not invulnerable.

He can be penetrated on thin regions of skin.

Go to whisper in his ear and inject poison into his veins on the inflow to brain using the thin skin behind the ear.

They may have planned ahead and regularly dosed themselves with anti-toxin, so use the taipan's ability to repurpose toxins by consuming large quantities of extremely spicy food.

They may stop the taipan venom, but the cepacian from the spice has no antidote, and will cause an inflammation that will disable brain function and make him functionally braindead.

Pai can't hurt Saitama, but Saitama can hurt Pai.

Pai can use shadows to regenerate, which will just make her a punching bag for Saitama's normal punches.

If punched into space, by a special punch, the darkness will regenerate her.

Just like ground dragon, i can see her choosing to retire.

The species that Jamil is based on here is called the Blood Python (special thanks to @kirexa for the information!), which can reach an average of 4-6 ft in length and up to 30 lbs in weight.
