Nasal COVID-19 vaccine halts transmission - Published July 31, 2024
Study in hamsters indicates vaccines targeting nose, mouth may be key to controlling spread of respiratory infections
A nasal COVID-19 vaccine blocks transmission of the virus, according to an animal study by researchers at Washington University School of Medicine in St. Louis. The findings suggest that vaccines delivered directly to the nose or mouth could play a critical role in containing the spread of respiratory infections.
The lightning-fast development of COVID-19 vaccines just months after the virus appeared was a triumph of modern science and saved millions of lives. But for all the good they did in reducing illnesses and deaths, the shots were unable to end the pandemic because of one notable weakness: They couldn’t stop the spread of the virus.
A new study by researchers at Washington University School of Medicine in St. Louis indicates that next-generation vaccines that target the virus’s points of entry — the nose and mouth — may be able to do what traditional shots cannot: contain the spread of respiratory infections and prevent transmission. Using a nasal COVID-19 vaccine based on Washington University technology, approved for use in India and licensed to Ocugen for further development in the U.S., the researchers showed that vaccinated hamsters that developed infections did not pass the virus on to others, breaking the cycle of transmission. In contrast, an approved COVID-19 vaccine that is injected failed to prevent the spread of the virus.
The findings, published July 31 in Science Advances, provide further evidence that so-called mucosal vaccines sprayed into the nose or dropped into the mouth may be the key to controlling respiratory infections such as influenza and COVID-19 that continue to circulate and cause significant illness and death.
“To prevent transmission, you need to keep the amount of virus in the upper airways low,” said senior author Jacco Boon, PhD, a professor of medicine, of molecular microbiology and of pathology & immunology. “The less virus that is there to begin with, the less likely you are to infect someone else if you cough or sneeze or even just breathe on them. This study shows that mucosal vaccines are superior to injected vaccines in terms of limiting viral replication in the upper airways and preventing spread to the next individual. In an epidemic or pandemic situation, this is the kind of vaccine you’re going to want.”
Developing vaccines that can control virus levels in the nose has proven challenging. Viruses such as influenza virus, SARS-CoV-2 (the virus that causes COVID-19) and respiratory syncytial virus (RSV) multiply rapidly in the nose and spread from person to person within a few days of initial exposure. Traditional injectable vaccines generate immune responses that can take a week to build to full strength and are much less potent in the nose than in the bloodstream, leaving the nose relatively unprotected against a fast-multiplying, fast-spreading virus.
In principle, a vaccine sprayed or dropped directly into the nose or mouth could limit viral reproduction and thereby reduce transmission by eliciting an immune response right where it’s needed most. But gathering evidence that mucosal vaccines actually do reduce transmission has proven tricky. Animal models of transmission are not well-established, and tracking person-to-person transmission is fiendishly complicated, given the number and variety of encounters a typical person has on any given day.
For this study, Boon and colleagues developed and validated a model for community transmission using hamsters and then used it to assess the effect of mucosal vaccination on the spread of SARS-CoV-2. (Unlike mice, hamsters are naturally susceptible to infection with SARS-CoV-2, making them the ideal laboratory animals for a transmission study.)
The researchers immunized groups of hamsters with laboratory versions of approved COVID-19 vaccines: the nasal iNCOVACC used in India or the injected Pfizer vaccine. For comparison, some hamsters were not immunized. After giving the vaccinated hamsters a few weeks for their immune responses to fully mature, the researchers infected other hamsters with SARS-CoV-2 and then placed the immunized hamsters with the infected hamsters for eight hours. This first step of the experiment mimics the experience of vaccinated people who are exposed to a person with COVID-19.
After spending eight hours rubbing shoulders with infected hamsters, most of the vaccinated animals became infected. Virus was found in the noses and lungs of 12 of 14 (86%) hamsters that had received the nasal vaccine, and 15 of 16 (94%) hamsters that had received the injected vaccine. Importantly, while most animals in both groups were infected, they weren’t infected to the same degree. Hamsters that had been nasally immunized had virus levels in the airways 100 to 100,000 times lower than those that had received the shot or had not been vaccinated. The study did not assess the animals’ health, but previous studies have shown that both vaccines reduce the likelihood of severe illness and death from COVID-19.
The second step of the experiment yielded even more striking results. The researchers took vaccinated hamsters that subsequently developed infections and placed them with healthy vaccinated and unvaccinated hamsters for eight hours to model transmission of virus from a vaccinated person to others.
None of the hamsters that were exposed to nasally vaccinated hamsters became infected, regardless of whether the recipient hamster had been vaccinated or not. In contrast, roughly half of the hamsters that were exposed to hamsters vaccinated by injection became infected — again, regardless of the recipient’s immunization status. In other words, vaccination through the nose — but not by injection — broke the cycle of transmission.
These data, Boon said, could be important as the world prepares for the possibility that avian influenza, currently causing an outbreak in dairy cows, might adapt to humans and trigger a flu epidemic. An injectable vaccine for avian influenza already exists, and a team of researchers at Washington University is working toward a nasal vaccine for avian influenza. That team includes Boon and co-author Michael S. Diamond, MD, PhD, the Herbert S. Gasser Professor of Medicine and one of the inventors of the nasal vaccine technology used in this paper.
“Mucosal vaccines are the future of vaccines for respiratory infections,” Boon said. “Historically, developing such vaccines has been challenging. There’s still so much we don’t know about the kind of immune response we need and how to elicit it. I think we’re going to see a lot of very exciting research in the next few years that could lead to big improvements in vaccines for respiratory infections.”

Never leaves his room? You take that as a challenge, and you know just the answer to Night Raven College’s biggest riddle.
Idia…I wanna play a fighting game. You down?
Which one? I can connect you.
How about the one you left at Ramshackle with me?
I can literally mod that.
But you can’t eat my freshly baked cookies from your dorm.
(I want it emoji) Fine, you win.
Idia can never resist your baked goods, no matter how hard he tries. He needs them to function on that console of yours.
When he comes over, you don’t expect him to have a bag of spare clothes with him, but you remember that when it comes to gaming, it’s going to be a long night. India immediately goes for your choco cookies.
“Power up!” he cheers after eating two of them.
Then, you two are duking it out for the virtual champion title.
The both of you spend three long hours blistering your thumbs on your controllers. The health bars on the corners of the TV screen are at the chokehold. You and Idia don’t notice how a storm has brewers outside. It’s storming so much that it brings on the thunder. One large crack is all it takes for the power in Ramshackle to drop from fifty percent to zero. India and you take a few seconds to adjust to reality, but you also realize how dark the dorm is without any source of light.
“Good Seven, this would never happen in Ignihyde,” mutters Idia.
You, on the other hand, don’t comment. You point out how late it is instead. You offer to make up Idia’s room as he takes a shower. He might as well. You’ve never had to do this in the dark though. Not that it’s impossible, but your limbs are quaking as you hold your phone as a flashlight under your chin and tighten the sheets onto the spare bed.
You are in the middle of fluffing the pillows when Idia exits the washroom. He peers into the room, watching you shaking like a leaf. He’s all too familiar with the sensation. He clears his throat, alerting you of his presence. You can’t be happier to see Idia in his unexpectedly athletic looking pajamas. The contradiction makes you snicker.
“What? If you were going to catch me in anime tees, that’s never going to happen,” he says.
“Why not? You rarely leave your room anyway. Not like anyone would care.”
“Incorrect. I learned my lesson during my first year. I’ve been wearing my STYX night gear ever since.”
You frown at this. “Sorry.”
Idia suggests that you get a candle or two to light up the place. It would help you sleep better than having your flashlight from your phone beaming in both of your faces. You explain that you tried looking for candles the other day, but Ramshackle doesn’t have any, and Sam doesn’t provide candles unless it’s for the main school building’s events. Idia remarks that he should’ve expected that.
“What? You’re not gonna say anything?” you ask, notioning that Idia knows about your fear of the dark.
Of course, he does, but…“Not really. A NEET like me calling someone like you a baby for a fear of darkness is the most “pot calling the kettle black” scenario some airhead with no creative RPG ideas could ever come up with.”
There’s no argument that you can come up with to combat that.
“As much as I wish I could fix this problem, there is no one-hit solution for that.”
“Gee, thanks.”
“But uh…”
You elevate an eyebrow.
Idia unzips the jacket to reveal a gray, Premo shirt underneath, revealing how Idia still has his interests, but he has told the truth about no anime related clothing. His face begins to pinken, and the tips of his long blue hair fade into blush as well. “I’ve seen this corny thing a ton of times, but I heard it works. If you wanna…then…I guess…”
You carefully step around your bed to prevent yourself from stubbing your toe, and when you are in the clear, you take Idia’s offer to hug into his night jacket. Having you touch him is already nerve-wracking for the introverted Housewarden, but then, to have him wrap you in the jacket folds turns the pale third-year into an overheating furnace. He suggests that you guys hide under the covers before he’d faint onto the floor.
You’re not too pleased with having to turn the flashlight of your phone off, but as you are not alone this evening, both you and Idia suppose that safety is in both of your corners tonight.

Sevika's tastes
Sevika is an old lady and she just wants to be left alone. She likes to look good but when it comes to clothes, Miss thing just doesn’t care that much. She always has practicality in mind. So, no dresses, nothing flowy, has to have natural fabrics so that her skin can breathe, and she requires that things are comfortable. Her shoes are always made for hardware with a strong sole and often reinforced. In the modern world, I see her working in metal working (specifically welding), so she has to have clothes that are multipurpose. Though, if she was forced to wear anything really nice, it would be a simple well-cut blazer and a button down with jeans or slacks that conform to her legs nicely. She prefers earthy colors, nothing too flashy. I think she’d really appreciate a nice dark green, or perhaps brown. I also feel that she would enjoy a nice flannel regularly.
With food, I’m afraid her palette is as unrefined as her clothing choices. She genuinely does not care what she eats, though she really likes chicken- loves hot wings, spicy food is her love. But, her comfort food will always be the food native to what part of India her family is from. I don’t think she’s the best cook, but she has a few family recipes that she knows so well (aloo gobi, chai, samosa, tikka masala, saag paneer). And, I think that on nights where she’s feeling really sad or lonely she always craves those foods. She’d love to cook with or for her partner, it’d be the best way to get to know her honestly. Because it allows for her to show vulnerability through actions and without words. She loves to take care of people and I think in modern times she’d mother her friends just a bit, always making sure they’re eating well, drinking their water, and sleeping right (if not she’ll give them some chai). She doesn’t eat beef or dark meats in general, and she isn’t the biggest fan of seafood or turkey. So, she sticks with her chicken and her paneer. She’ll eat tofu but it needs to be in curry or something similar.
This woman would love 80s hair metal, music is something that I genuinely believe she’d love so much. She’d play drums as a teenager, dead set on becoming the drummer of the next Metallica. She’d also love the old school heavy metal bands, Iron Maiden, Metallica, Black Sabbath, Pantera. She’d love them all. I think she’d like some old school 90s rap too, but none of the new-age mumble rap that’s going on. She wouldn’t really like Taylor Swift’s music, just because it didn’t vibe with her, but she respected Taylor’s ability to get a bag. She has had a huge crush on Adele ever since she heard the album 25 when it came out. She liked some of her music, but thought Adele was drop dead gorgeous and all mature and soulful and shit, hit her in the feels and made her whipped for this woman she didn’t even know.
For movies she loves shitty 80s slasher horror, nothing that makes her think. She’d sit back in her old recliner in her pajamas and house slippers whilst watching Slumber Party Massacre for the third time, and then put on Golden Girls because she feels that Dorothy Zbornak is her spirit animal. She likes a good sitcom too and a ridiculous drama (she loves Desperate Housewives), she likes the camp, the over the top acting and dumb plots, it makes her laugh and feel care free in a way she hasn’t been in a long time. She just wants to curl up with her pets (she would have many) and watch teen-based tv shows that revolve around crime or secrets (Pretty Little Liars, Riverdale, Vampire Diaries, even Buffy etc.). She likes how bad they are, but she gets so invested it’s ridiculous.
For personal scents she’d like more woody, alluring scents that are also kind of sweet. Think Amber by Rag n’ Bone (it smells so good), she doesn’t spray much, just a spritz, it wafts around her just slightly, just enough for women to fall at her feet. Her individual smell wouldn't be overpowering but it would definitely be clear. It’s grounding and soothing. In general, Sevika is an old woman who couldn’t give less of a shit. She wants to be left alone with her life and her people and chill. Which is why, I feel like she isn’t that opinionated on much unless it’s boundaries or causes she cares about. She just doesn’t have the energy to be bothered with trivial things like which movie to choose for the night, or which restaurant to go to. She is tired and all she wants to do is eat good food with her partner and her pets in a little cottage in the middle of nowhere. She doesn’t like neighbors and she doesn’t like people in her business. She doesn’t need a perfect life, just one that’s hers.

Researchers from Western and Brown University have made groundbreaking progress towards identifying the root cause and potential therapy for preeclampsia.
The pregnancy complication affects up to eight per cent of pregnancies globally and is the leading cause of maternal and fetal mortality due to premature delivery, complications with the placenta and lack of oxygen.
The research, led by Drs. Kun Ping Lu and Xiao Zhen Zhou at Western, and Drs. Surendra Sharma and Sukanta Jash at Brown, has identified a toxic protein, cis P-tau, in the blood and placenta of preeclampsia patients.
According to the study published in Nature Communications, cis P-tau is a central circulating driver of preeclampsia – a “troublemaker” that plays a major role in causing the deadly complication...
“The root cause of preeclampsia has (so far) remained unknown, and without a known cause there has been no cure. Preterm delivery is the only life-saving measure,” said Lu, professor of biochemistry and oncology at  Schulich School of Medicine & Dentistry...
“Our study identifies cis P-tau as a crucial culprit and biomarker for preeclampsia. It can be used for early diagnosis of the complication and is a crucial therapeutic target,” said Sharma...
Until now, cis P-tau was mainly associated with neurological disorders like Alzheimer’s disease, traumatic brain injuries (TBI) and stroke. This association was discovered by Lu and Zhou in 2015 as a result of their decades of research on the role of tau protein in cancer and Alzheimer’s.
An antibody developed by Zhou in 2012 to target only the toxic protein while leaving its healthy counterpart unscathed is currently undergoing clinical trials in human patients suffering from TBI and Alzheimer’s Disease. The antibody has shown promising results in animal models and human cell cultures in treating the brain conditions.
The researchers were curious whether the same antibody could work as a potential treatment for preeclampsia. Upon testing the antibody in mouse models they found astonishing results.
“In this study, we found the cis P-tau antibody efficiently depleted the toxic protein in the blood and placenta, and corrected all features associated with preeclampsia in mice. Clinical features of preeclampsia, like elevated blood pressure, excessive protein in urine and fetal growth restriction, among others, were eliminated and pregnancy was normal,” said Sharma.
Sharma and his team at Brown have been working on developing an assay for early detection of preeclampsia and therapies to treat the condition. He believes the findings of this study have brought them closer to their goal...
“The results have far-reaching implications. This could revolutionize how we understand and treat a range of conditions, from pregnancy-related issues to brain disorders,” said Lu.

The state of Kentucky has legalized the use of medical marijuana. The bill received final passage on Thursday. Democratic Gov. Andy Beshear signed it into law Friday morning after a decade of failed attempts in the state legislature.
The news makes Kentucky at least the 38th state in the U.S. to legalize medical marijuana.
Now Indiana is surrounded by weed states. The encirclement is complete 😂
2. The Maryland House of Delegates voted Saturday to approve the Trans Health Equity Act

The Maryland House of Delegates voted Saturday to approve the Trans Health Equity Act — a bill that just a year ago disappeared from the chamber’s agenda ahead of a floor vote.
The bill would require Maryland Medicaid, beginning on Jan. 1, 2024, to provide coverage for additional gender-affirming treatments, which are currently disallowed in the state’s plan but commonly covered by private insurance. The expanded treatments include hormone therapy, hair alteration, voice therapy, physical alterations to the body, and fertility preservation.
3. FDA approves over-the-counter Narcan. Here's what it means

The approved nasal spray is the best-known form of naloxone. It can reverse overdoses of opioids, including street drugs such as heroin and fentanyl and prescription versions including oxycodone.
Making naloxone available more widely is seen as a key strategy to control the nationwide overdose crisis. Effects begin within two minutes when given intravenously, and within five minutes when injected into a muscle. The medicine can also be administered by spraying it into a person's nose.
4. Boston expands tuition-free community college program to all residents

Boston has expanded its tuition-free community college program to include all city residents regardless of age, income or immigration status.
Starting this fall, any city resident will be eligible to pursue an associate’s degree or certificate at one of six partnering local institutions without paying to attend. The program also includes a $250 stipend for incidental expenses each semester for up to three years, and up to $2,500 of debt relief for students whose account balances are keeping them from re-enrolling.
5. First cheetah cubs born in India since extinction 70 years ago

India has welcomed the birth of four cheetah cubs - more than 70 years after the animals were declared officially extinct there.India's environment minister announced the good news, calling it a "momentous event".
The country has been trying to reintroduce the big cats for decades, and last year brought eight cheetahs over from Namibia as part of the plan. Another 12 cheetahs were brought to India from South Africa last month.
6. BBC education show in Afghanistan helps children banned from school

The BBC has launched a new education programme for children in Afghanistan who are banned from school.It is aimed at children aged 11 to 16, including girls whose secondary education has been stopped by the ruling Taliban.
The weekly programme is called Dars, which means lesson in Dari and Pashto, Afghanistan's official languages. It is hosted by BBC Afghan female journalists who were evacuated from Kabul during the 2021 Taliban takeover.
Each new weekly half-hour episode of Dars will air four times a day, Saturday to Friday, on the newly launched BBC News Afghanistan channel.
7. A Trans Creator Has Raised Over 1.5 Million for Trans Healthcare on TikTok Live

Transgender TikTokers are celebrating Trans Day of Visibility by raising over $1.5 million for gender-affirming care around the world.
Mercury Stardust — a DIY TikToker and trans advocate who calls herself the “Trans Handy Ma’am” — raised $120,000 last year in a livestream for the mutual aid nonprofit Point of Pride, which maintains funds for surgeries, hormone therapy, and free binders and gaffs. This year, Stardust and cohost Jory, a.k.a. AlluringSkull, set themselves a goal of raising $1 million in a planned 30-hour live stream…and then smashed that milestone less than six hours after starting the stream Thursday evening.

Ten years ago, musician Usman Riaz grabbed a pencil and started to sketch.
He might have hoped, but didn't know at the time, that it would start him on a path to making history.
That initial drawing became The Glassworker - Pakistan's first ever hand-drawn animated feature film.
It follows the story of young Vincent and his father Tomas, who run a glass workshop, and a war that threatens to upend their lives.
Vincent's relationship with violinist Alliz, the daughter of a military colonel, begins to test the bond between father and son.
Usman tells BBC Asian Network the characters ultimately come to learn "that life is beautiful but fragile, like glass”.
He describes The Glassworker as an "anti-war film" set in an ambiguous and fantastical world that takes inspiration from his home country.
“I wanted to tackle issues and themes that would have been difficult to tackle if it was based in Pakistan," he says.
The country doesn't have the thriving film industry of neighbouring India and there is no government support or incentive for budding creatives like Usman.
So The Glassworker was a passion project, he says.
“These 10 years for me have just been purely driven with passion and obsession.
“Since I was a child, I have loved hand-drawn animation and there's something so magical about it.
"The beauty of the lines drawn and painted by the human hand always resonated with me.”
Usman says he travelled the world looking for mentors and his search took him to Japanese animation house Studio Ghibli.
The influence of the Oscar-winning artists behind classics such as Spirited Away and Princess Mononoke can be seen in The Glassworker's own style.
Usman says the industry veterans at Ghibli were also the ones who encouraged him to start the production himself.
After raising $116,000 through a 2016 crowdfunding campaign he founded his own studio, Mano Animations.
From there it's been a painstaking process, especially since full production started in 2019.
“What you are watching is essentially a moving painting,” says Usman.
“Every single frame you see, whether it's a background or the character moving, it's all drawn by hand.”
Usman says that, so far, he hasn't made any money from the project and has been unable to pay his wife Maryam and cousin Khizer, who he recruited to help him.
But there's hope that the labour of love could be the start of something bigger.

Sharmeen Obaid Chinoy is another experienced industry figure Usman turned to for advice about getting The Glassworker off the ground.
She directed 3 Bahadur, a computer-generated tale that was Pakistan's first-ever animated feature film.
On its 2015 release it broke box office records, even surpassing US imports and dethroning previous record-holder Rio 2.
Her studio was also the country's first female-led animation studio, and she understands the challenges of getting started better than most.
“Everything in Pakistan is driven by passion” she says. “I had to run pillar to post.
“We're a country that has limited access to electricity and our industry is heavily taxed.
"We're unable to import computers and hardware needed for animation.”
But Sharmeen – who is going to direct upcoming Star Wars film New Jedi Order – says The Glassworker could be a “monumental step” for Pakistan’s animation scene.
If it finds commercial success, she believes it will “ignite” something in the country, but there are barriers to home-grown animation becoming a red-hot trend.

The Glassworker is partly about the romance between characters Vincent and Alliz
Arafat Mazhar from Lahore-based Puffball animation agrees that “the technical skills are already there” in Pakistan despite there being “no formal training or schools available”.
But “how do you not censor yourself?” he asks.
It's a question facing any Pakistani filmmaker who has to deal with its strict board of film censors.
“Every time there's a good film that comes out that's sincere, the state ends up censoring it,” says Arafat.
He doesn't believe the rules are likely to relax soon.

Sharmeen agrees the government will only encourage the domestic film industry to grow if they work to "provide opportunity to create a level playing field for us to compete with the rest of the world".
“There is a lot of scope in Pakistan for animation," she says. "We've just never been given the opportunity to create it."
She shares Arafat's pessimism about the pace of change.
"Unfortunately, it will just be a few filmmakers who have that passion, who will continue to create films," she says.
But Sharmeen says she is eager to see how the world embraces The Glassworker.
"I know that there is so much in there that will touch people's hearts," she says.

Usman says The Glassworker has an anti-war message
Usman will finally get to find out how audiences react to the work he's spent 10 years pouring his energy into as The Glassworker goes on general release.
He says he hopes to “put Pakistan on the map” and show it can stand up to the giants across the border in Bollywood.
But he admits the process has been “gruelling”.
“It is extremely difficult, but we've done something nobody has ever done in the country before," he says.
"I think we've created something special that can stand toe-to-toe with the rest of the animation produced in the world.”
