MONDAY MATINEE MUSIC VIDEO: “Hold On” by Rupert’s People https://www.youtube.com/watch?v=EYbrxbXH5bs. Rupert’s People was a “made in the studio” band formed around members of Fleur de Lys. In 1967, Rod Lynton was a “rising star” songwriter, so managers and record companies got behind him to record his songs by using bands-for-hire—in this case it was Fleur de Lys with Chris Andrews on lead vocals. To avoid confusion over the Fleur de Lys “brand,” the band re-named themselves “Rupert’s People.” “Hold On” was the B-side to the single Rupert’s People “Reflections on Charles Brown,” which received positive response in 10 countries. However, many said “Hold On” is the stronger track, amped up with muscular organ by Pete Solley (future sideman w/Eric Clapton, Al Stewart, Whitesnake, etc.) and edgy lead guitar by Phil Sawyer. The song was co-written with bassist Gordon Haskell (later of “Harry’s Bar” and King Crimson) and guitar hero Bryn Haworth (not credited on the record label). On a cosmic jukebox “Hold On” plays next to the soulful hard-edged blues-rock fusions of Spencer Davis Group, Pretty Things, and Brian Auger & The Trinity.

Lynton went on to collaborate with John Lennon, and Rupert’s People tracks have been reissued on soundtracks and compilations celebrating the best of 60s British rock. Fleur de Lys became a launching pad for several careers, with Chris Andrews going on as a solo act and collaborator with Roger Daltrey, David Essex, Davy Jones (Monkees), and other notables. Potential of Negative Ion Mode Proteomics: MS1-Only Approach

Current proteomics approaches rely almost exclusively on using positive ionization mode, which results in inefficient ioniza-tion of many acidic peptides. With an equal quantity of acidic and basic proteins and, correspondingly, the similar number for their derived peptides in case of the human proteome, this inefficient ionization poses both a substantial challenge and a potential. In this work, we study the efficiency of protein identification in the bottom-up proteomic analysis performed in negative ionization mode, using the recently introduced MS1-only ultra-fast data acquisition method DirectMS1. This method is based on accurate peptide mass measurements and predicted retention times. Our method achieves the highest rate of protein identifications in negative ion mode to date, with over 1,000 proteins identified in a human cell line at a 1% false discovery rate using a single-shot 10-min separation gradient, which is comparable with hours-long MS/MS-based analyses. Evaluating the proteins as a function of pI indicated preferable identification of the acidic part of the proteome. Optimization of separation and mass spectrometric experimental conditions facilitated the performance of the method with the best results in terms of spray stability and signal abundance obtained using mobile buffers at 2.5 mM imidazole and 3% isopropanol. The work also highlighted the complementarity of data acquired in positive and negative modes: Combining the results for all replicates for both polarities, the number of identified proteins increased up to 1,774. Finally, we also investigated the efficiency of the method when different proteases are used for protein digestion. Among the four studied proteases (LysC, GluC, AspN, and trypsin), we found that trypsin and LysC performed best in terms of protein identification yield. Okay here’s the thing. The thing is THIS. The first thing JC does when he finds out WWX is alive and kicking again? He gets angry at him, because of course he does, because it’s what he does whenever he needs to deal with Emotions, but what does he get angry about?

That WWX didn’t think to immediately hoof it to Lotus Pier after he was resurrected. JC feels personally slighted that WWX didn’t think to come to him, and dude, I get it. That shit hurts. But please consider this from WWX’s point of view, the last thing he remembers you saying to him is to just die already, so why on earth would he feel in any way entitled to go see you?? His entire personality is a rollercoaster of everyone is better off without me, and JC is included in that, of course.

Whenever I think about their interactions in the show, I keep coming back to that scene when they talk, or rather JC talks, while WWX is backed into a corner by Fairy. This is, I remind you, the guy who got rid of his puppies as a child so that WWX could come stay with him. He knows this isn’t just some stupid little fear of WWX’s, but a genuine phobia. He can see him shivering, barely incapable of answering, but he does it anyway, it’s why he does it, because in his eyes WWX deserves it for what he’s put him through, put them all through.

But I think eventually, JC gets, above all, tired. WWX is tired too, in different ways, but JC has nursed this grudge for sixteen/thirteen years. he’s so ready to let it go, he doesn’t even know it. I think what WWX says to him in the Guanyin temple is, in a way, exactly what he needs to hear. He hates his brother for always playing the martyr, for loving him so much he’d give the very core of himself away to save him, but fuck me, he’s lost so much.

Jiang Cheng has lost so much, and somehow, Wei Wuxian has been returned to him?? You cannot expect me to believe for a SECOND that he would spend the remainder of his living days wishing him ill, wishing to never see him again. No, they’re going to argue A LOT, there’s going to be some accusations, a lot of yelling on JC’s part and that annoying gentle emotional scoffing on WWX’s, which JC hates, why can’t you get on my level and just yell about things like a normal coping adult, Wei Wuxian, why is it so easy for you to just let things go, but they’re going to get better.

Slowly but surely, they’re going to get better. JC will cry at WWX’s wedding, and WWX will cry when he’s allowed to step foot in Lotus Pier again (what the fuck do you MEAN you thought you could never come back again Wei Wuxian are you or are you NOT YUNMENG JIANG!! / You were the one who said I wasn’t!! / You LEFT! / And now I’m here!), But they Will. Get. Better. Slowly, but surely, they will build something new, something that never could have grown if they’d stuck to who they used to be. They knew each other as children, as teenagers, they knew each other in war and in betrayal, and now they get to learn to know each other in peace, as adults, with all the time in the world to figure out how to be able to talk again.

Jiang Cheng allows himself to accept that the world sometimes isn’t an endless shithole of bad luck and grief. Wei Wuxian allows himself to accept that he is wanted, that he still has a home in Yunmeng, and slowly, ever so slowly, walking around Lotus Pier will start to feel less and less like picking at a fresh wound. Together, they will talk to their big sister, just the two of them and no one else in that quiet, slumbering ancestral hall, and both will pretend not to notice the tears in the other’s eyes. 

JC will always hurt a little bit when he looks at WWX, he will always fuss over him in that furiously overprotective you are a fucking idiot but you’re, unfortunately, MY idiot kind of way, and WWX in turn will never give up on teasing JC until he elbows him in the ribs and rolls his eyes at him and maybe threatens to electrocute him where he stands without ever actually going through with it, but he’s always stupid relieved when that happens, because it’s how his baby brother shows his love. 

It’s how he shows his love.

I don't think that anon's been to medical school, as they obviously don't know what it's like, so I really can't imagine why they think they're qualified to comment on how you react to it. I'm also in MS2 right now, and studying for step, and trying to survive a f*cking pandemic, and I guarantee there's not a single person in my class who wouldn't like a break right about now. Many of us also indulge in a good whine about it once in a while, as is only natural. Good luck with your studying <3<3

and then! and then! even though JGY's MO so far would dictate that he should now hate LXC, as he came to hate NMJ after he'd hurt him, and maybe try to do a revenge...he instead begs LXC to stay with him while he dies, and then lets his last act be to save him. Like damn JGY loved one (1) person and that person just hit him hard enough to put him on the ground.

@lysces-96 there’s that funny meme post that has the text post “you only get 782 chances with me bitch I don’t tolerate shit” put over a screencap of LXC, but honestly it applies to JGY too. He let NMJ kick him twice before he did his revenge! He let his father abuse him for years before going for murder! If he loves you he will let you do whatever to him right up until he can’t take it any more. 

And at this point he’s balancing his whole sense of self worth on top of “no matter what, at least I never hurt Er-ge.” I think the only thing that would ever get him to hurt LXC is being directly called a son of a whore, and that would completely destroy him anyway.

Well, the book will come out next month, end of January, in Québec libraries. It’s in French. There’s a negociating process with English publishers to sell the translation deal and that will come later- but I will keep everyone up to date. My main goal is to sell enough original copies so it gets translated and sold elsewhere in Canada and the US.
