Is it possible to have NPD, APD, BPD AND HPD at the same time?-

It's honestly complicated. You know, personality disorders, particularly those within the same cluster, often share multiple traits at the behavioral level, such as attention-seeking, unstable personal relationships, and violent outbursts. However, similar behaviors can stem from very different motivations. For example NPD might act aggressively to dominate their partner and maintain their ego, while someone with BPD might exhibit the same behavior out of a fear of abandonment. What should technically allow for differentiation between disorders is their core mechanisms that underlie everything going wrong. However, in my opinion, this is largely a theoretical concept because the core of the psyche is interpreted differently across psychological paradigms. That's partially what makes diagnostic procedure so difficult. Also there are for example multiple studies about "BPD in men" or "APD in woman" which only showcases that classification is not carved in stone as symptoms can differ across other categories like gender. At the group therapy I encountered very few people with a single diagnosis. In the majority of cases, it was "XPD with traits of YPD" because fitting neatly into one prototype is not easy. Also DSM V is only one of available classifications, ICD lists like 5 more specific personality disorders.

What I'm saying is that the labels we use are the flawed results of statistical cluster analysis, and it's not uncommon for someone to exhibit traits that span across multiple clusters. In cases of someone showing traits from three or more personality disorders I would just describe it as mixed personality disorder or unspecified personality disorder, ICD style.

It's been ten days since anyone has seen Jhenae Cedeno and Youngstown police believe she is in danger here.

Jhenae Cedeno is a 16 year old trans girl from North Carolina that police in Youngstown, Ohio believe travelled here to meet with a stranger, possibly someone she met online. They also believe she is in danger because she has no friends, family or known associates in Ohio and nobody has seen or heard from her since July 10th.

As most people know, Ohio is a dangerous state for any LGBTQIA+ person, but especially for a black trans girl and YPD has not been putting any effort into finding Jhenae beyond simply publishing a release on facebook through their juvenile department. We have multiple digital billboards in Youngstown, Boardman, Canfield ,and Austintown that they could be putting her info on, but they haven't. We have multiple local news stations they could be sending her information to, but they aren't. We have multiple community centers in the different neighborhoods of Youngstown they could be putting her information up in, but they aren't. There are several other community sources all throughout the north side, the west side, the east side, and the south side they could be working to find out if anyone has seen her, yet they aren't.

I know posting this does very little in the grand scheme of things, and I'm currently trying to find out if there are any search efforts going on here in my city that I can volunteer with to help find her but more people also need to be aware of her case here in general because as of right now Jhenae is being let down by our cops severely when the worst possible outcomes are still on the table and I can't imagine what her family is going through.

No one asked but here's some trivia about those misfits:

Soleil and Lune have no official family names/surnames, but they do have families/relatives.

Soleil’s family is rich, but he doesn’t like to shot it down everyone’s throats (he would rather prefer to not be associated with his family – OG reason is they’re shitty; the Young Prince Detective version is that they’re a bunch of airheads);

Lune’s family is not that big, but she’s an only child with at least three cousins (she’s the baby cousin)

Étoile’s name being Noel is a reference to a LupPat character, Takao Noel.

The Space Trio has French space names BECAUSE I was in love with Cure Étoile and Cure Soleil’s names. In-universe they just grabbed it from google.

The trio are counterparts of the 02 kids in YPD. Soleil rivals with the boys’ traits, Lune with the girls. Étoile is an odd case in which he might be just a “Bridge” connecting both sides.

If you call Soleil by “Octo-head” he might snap and it won’t be good :V

Lune hates resorting to violence, but her only “violent” method to intimidate others is just giving them a deadly deep stare into your soul with the ire of 10 gods.

Their DigiTimeLines versions are actually 15 (Soleil), 14 (Lune) and 10 (Étoile) year olds. The YPD version has them as 17 (Soleil & Lune) and 16 (Étoile) year olds instead.

In DigiTimeLines, Étoile’s digimon partner is a purple Pokomon/Viximon named Renard. In the YPD version he’s acquainted with Miss Espimon.

Ah… I must say… Since some stuff came later while i watched Kamikaze Kaitou Jeanne with a friend… Noel’s hair showing his eyes when he’s doing a 180º personality flip is based on Minazuki Yamato :V

That said, Noel is based on Beyblade’s Kenny. Soleil and Lune are based on Kamen Rider Fourze’s characters (Ryuusei and Nadeshiko).

Soleil likes action movies/TV shows with cool fights (possibly Shounen anime genre); Lune likes mystery and drama (possibly because she’s very psychic and emotional); Étoile is… fascinated with everything though. But he likes Mystery shows with magic and romance involved.

Noel’s surname being Leblanc was a recent “change”, because he was like Soleil and Lune, without one in mind. Also this is a plot point. That said, I still refer to him as either Noel and Étoile only.

I feel like opening a sideblog for my Rui-Ukko muses for a while, but Various Colors (the song i re-named this acc after) is a song from Rui's POV so it would make no sense to separate him & Ukko from this blog methinks.

Still i have a deep attachment with Rui and Ukkomon so i'm not rping with doubles of them for now. Maybe Ukkomon OCs (if any), but not Rui's Ukkomon. This is the first time i get extremely wary around other peeps writing a favorite character (i usually don't mind) so please understand my wish. It might change later in the future, tho.

today I did a plate assay to get a quick (3 point) anisotropy based binding curve for Pol alpha on fluorescein labeled template/primer DNA. a couple months ago we did a whole bunch of these with different conditions and they were working great, but now that we're back to doing them, they aren't working anymore. I just did a quick 3 point one today (0nM, 6nM, 150nM Pol alpha) to test and see if we're getting the same mP values we were getting before, and we are not. I think something is up with our plate reader cause the values were like 10x less than they should be at our previously discovered binding saturation (which started around 150nM pol). big sad for today in that regard.

Another thing I did today was restreak a yeast clone I've been growing. I transformed it a little bit ago, screened a couple colonies, and now I'm restreaking a few of the clones positive for my DNA. The DNA in question is a linearized plasmid integration, which I integrated in the middle of the yeast Trp1 operon. The DNA also has N-Terminally STREP-tagged Sen1 flanked by a Gal promoter so we can overexpress it. I tried linearizing my plasmid with 2 different restriction enzymes in the Trp1 operon this time (being SnaB1 and BSU361), because the last time we did this linearization, some weird stuff happened. our lab's yeast strain acts weird with Trp1 integrations if you linearize with BSU361, and we had done this before. The resulting protein was inactive and during the prep we could see a whole bunch of strange truncated product. We think relinearizing and reintegrating might help that.

Anyway, I'm confirming stable integration of Trp1 by streaking onto YPD (regular yeast food) plates, and then back onto -W (no tryptophan) selection plates. Our yeast can't grow on -w without Trp integration, so growing them on that plate confirms they have what I'm looking for. Additionally once I'm done restreaking I'll do another heat-pop PCR screen to check for Gal1-Sen1. if the PCR is able to amplify that region, it confirms that the plasmid got integrated into the yeast.

I'm doing a whole bunch of other stuff right now but I think im gonna provide background for that when it actually comes up. ill share some pics too! here's the first restreak of that clone onto YPD. I took a colony from each of these guys and streaked them onto -w earlier today.

Also, here's that colony screen i was talking about. most of the clones had the integration, seeing as there are bands of a size that I expected in each one (except 5s its dumb)

holy SHIT i want to leave this job so fucking bad

like this is all compounded by the fact that im so, so close to leaving this lab and that im splitting time with another lab that i like a lot more, but oh my GOD everything is so much harder than it needs to be. had to spend 30 minutes running back to my lab to pick up some YPG plates for the colonies and he caught me as I was waiting for the shuttle to come back around and asked how it was going. told him i was only there to grab plates and we spent ten genuine minutes going back and forth over the point of streaking on YPG vs YPD and for some reason i could not get through to him that it was simply a point of comparison to ensure theyre actually respiring. like oh my fucking god im not trying to run a TMRE on this thing its literally just to see that theyre capable of respiration and not solely relying on fermentation after, you know, editing the proteins that go into allowing them to fucking respire. its not that deep because it shouldnt need to be at this stage. jesus fuck. literally had to come back at 5:30 and get it ironed out.

the fact that its only tuesday and i have to spend the rest of the week there and not at the other lab makes me want to throw up for real. i already know the presentation tomorrow is going to go poorly and im probably going to sob in my car about it. and even then itll only be wednesday. cant even pretend to have covid to get out of a week of work because only now am i actually responsible for things and i need to turn shit out at the speed of fucking light. i know that isnt his fault at least but god i could have used some of this energy back in like june and july when i was just sitting on my hands.

at least the tagging worked. baby's first western went well and honestly was pretty fun.
