DBCO Click Chemistry
DBCO click chemistry is a process of applying surface functionalization to a compound in order to change its structure and properties. This process involves a variety of methods, including PEI-DBCO and Azide-alkyne click chemistry. DBCO is a highly effective chemical agent that can be used to form solids, liquids, and polymers.
PEI-DBCO
DBCO (dibenzocyclooctyne) PEG acid is a heterobifunctional reactive PEG derivative. Its fast kinetics make it suitable for Click Chemistry reactions without metal catalysts. The heterobifunctionality of DBCO PEG derivatives enables them to be used for spontaneous biomolecule labeling. They are also stable in aqueous buffers. Here, we report the use of this compound to label aCD44 antibodies on mesoporous silica nanoparticles (MSNPs).
For the labeling of aCD44, aCD44 peptide was conjugated onto the aCD44 antibody. This peptide was attached onto a PEI nanoparticle through copper-free click chemistry. In this process, the azide moiety is covalently linked to the PEI-MSNPs via NHS ester linker. The azide moiety gives the peptide a positive charge. The PEI nanoparticles were then characterized by atomic force microscopy (AFM), scanning electron microscopy (SEM) and zeta potential measurements.
The aCD44-PEI-MSNPs displayed a high internalization rate, which may account for the increased cytotoxicity. ACD44-PEI-MSNPs were more cytotoxic than PEI (25 kDa) and PEI-SS-CLs. The aCD44-PEI-MSNPs also showed a higher efficiency in gene transfection, while the PEI-SS-CLs were less cytotoxic.
Azide-alkyne click chemistry
During the past decade, copper catalyzed azide-alkyne click chemistry has become an increasingly popular reaction in the science community. This reaction provides a method for linking different types of peptides, antibodies, and drugs. This method can also be used to develop biomolecules that can be targeted in a complex chemical environment.
Click chemistry has gained popularity because it is a versatile and modular reaction, which is useful for a variety of applications. Click reactions are also quick, reliable, and selective. They can be used to target selected cellular organelles, such as mitochondria. They are also an effective tool for studying the biological effects of Pt-based anticancer drugs.
Click chemistry is also used for drug discovery and development. ABP Biosciences has developed fluorescent dyes for use in click chemistry. They also have developed a biotin that is CLICKable.
Azide-alkyne click chemistry is classified into two main categories, the Cu(I)-catalyzed reaction and the strain-promoted azide-alkyne click (SPAAC) reaction. In SPAAC, the cycloalkyne precursor is oxidized by a reagent such as a gem-fluorine to release its ring strain, thereby increasing the reaction driving force. This enables the high fidelity ligation of alkynes with azides in a variety of chemical environments. The SPAAC reaction also allows for high efficiency ligation without the use of a catalyst.
Surface functionalization
DBCO click chemistry is a novel bio-conjugation strategy. This strategy provides a simple way to conjugate molecules such as enzymes and antibodies to surfaces of nanoparticles. These molecules can be used as signal probes for imaging and biosensing applications. Click chemistry also provides a flexible framework for controlling the conjugation process. It can also improve the analytical performance of nanosensors used in bio-analysis.
DBCO compounds react with azide functionalized compounds without the need of a Cu(I) catalyst. The resulting triazole linkage provides a stable connection between azide groups and azide ligands. This reaction was first identified in 1953 by Blomquist and Liu. Later, Wittig and Krebs confirmed the reaction in 1961.
Bioorthogonal click chemistry has been used to conjugate recognition elements to SMNPs. Using this strategy, researchers have developed QDs as multiple signal probes. These QDs are able to detect individual virus particles. They can also be used to detect circulating tumor cells.
Bioorthogonal click chemistries have made it possible to detect multiple biomarkers in a single sample. In addition, their combination with lateral flow devices can reduce the cost of nanosensors and shorten the analysis time.
oYo-Link DBCO
oYo-Link is a site-specific protein labeling solution that is activated by non-damaging black light. This product is designed to eliminate the shortcomings of conventional protein labeling techniques. Instead of requiring sample concentration and centrifugation, oYo-Link enables you to label just a single ug of antibody at a time, resulting in a high affinity and 100% sample recovery.
oYo-Link can be used in buffers, and is compatible with Tris, BSA, and BSA-Tris buffers. When illuminated by the non-damaging black light, the oYo-Link reagent forms a covalent bond with the antibody. This bond forms in just two hours, and is ready for storage. Once the reaction is complete, the antibody will be labeled with the desired label. It is also a copper-free click chemistry solution. It can be used with azide-tagged molecules, and it can be run in aqueous and organic solvents.
AlphaThera's oYo-Link Conjugation Technology can be used to label the Fc region of an antibody using click chemistry or enzymes. It can also be used to label an antibody with oligonucleotides or biotin.

Abusing all of the modules of MySewNet Platinum to make patches in a way far from how the makers intended:
(This is an embroidery software tutorial. MSNP is on the more expensive side if you look at buying the software outright, but if you're doing the subscription thing it's actually a lot less, so I don't actually feel as bad about putting a tutorial about a really pricey software on my sewing blog as I think I maybe should)
Most people shopping in my store buy Platinum for the digitizer, but Platinum is actually more like Adobe Creative Suite in that it's like 25 programs that work together. So here's me using other modules to make my life easier.

part 1 is to go into digitizing and do whatever you want there. Everyone at my store really pushes the digitizer, so I'm assuming that most people who get this software already kind of have an idea on digitizing. This is a cat-themed nightmare of a circle, but it's what we're using.
You can also just use a premade design or a SuperDesign if you want.
You can make whatever you want, as long as it'll fit on your patch. Oh, also, knowing how big your patch will be is kind of important. Make your design a little smaller than that.

Get that into the embroidery module and fix your colors. This is also where I get it into the hoop that's the size I want it to be. I'm making a lot of patches, so I want to use my big hoop.

Grab the Project in the Hoop generator, select coaster, and then pick a blank one in the shape you want your patch to be in.

Center your design in the coaster (the generator will make the project in the hoop directly in the center of your canvas, so you can just click your design and hit "center in hoop") and drag it so that it's on top of the coaster and visible. Ungroup the coaster group it generated.

The coaster is made of two shapes. One is the main shape, and one is the final stitching line. You need to stick your design between these two layers. Then, group these three things together into one group.
NOTE: If you want to get fancy with your stop commands, do that before you duplicate the coasters. If you convert each applique stop to a color change, you can do the applique a bit faster. I forgot to do that, so we're just going to do applique the boring way.

Duplicate this group however many times you need to fill your hoop. Stick one in each corner of the hoop.

I'm making 10 here, so I select the two in the top corners, and then three more. It can be any three, as long as I have those top two corners.
Then, go into alignment, and align them all to the top. After that, hit "distribute horizontally", and they'll all line up nice and neat. Do that with the bottom row, and you've saved yourself a ton of attempting to re-arrange them.
Mine were touching when I did this, so I had to go in and delete two of them. Again, I can delete one from the top row (not a corner), and one from the bottom row (again, not a corner), and then distribute horizontally. I've found that a ton of people don't take advantage of that distribute tool.

Now, if we select the entire thing, we will see that it's 40 color blocks. This will have the machine make one coaster at a time, and we'll need to stop what we're doing to go add applique fabric for each piece individually. I don't have time for that, so let's color sort. Combine the entire thing into one, go for color sort, and then it'll drop down into a perfectly manageable five color blocks.

Now, go into the stitch editor and grab your design separator. Select one of your patches and divide it out from the rest of the design. Once you're back in embroidery editing, you can make sure that your separated patch is going to stitch out first.
This is important, because if there is any major flaw with your design, you want to catch it BEFORE it sews the ground work for every patch. It'll save you a lot of applique fabric and thread.
(If there is a major flaw and the first patch is a botch, fix the other 7, and then delete the bad one, without moving any other parts of the design. You can then stick the same hoop in the machine, and it'll ignore your botch one and sew the remaining 7. You might want to grab the design separator and single one off, like you did with this, to make sure that version 2 is okay).
Hoop some shit up and let's go. If you're doing a sew-on patch, and you're confident enough, you can do this on tear-away and it'll tear away all nicely for you. If you're making an iron-on or stick-on patch, you'll have to cut the bottom anyway, so you won't benefit from tear-away. Cut-away will work just fine and be a little stronger for the actual stitching process.
Then, if you want to be just like me, you turn on your expensive problem child of a machine, find out that (despite being less than two months old) the USB port isn't working, go into the craft store on your day off, and once again be grateful that you got the service contract as you resign yourself to not getting this done any time soon.
[yes, it'd be covered under warranty, but 1) our tech is my friend and he gets paid more for service maintenance agreements than he does for warranty work and 2) when processing it for warranty, the tech has to look for evidence that this was caused by a client misuse. It wasn't, but he's got to look. Service contract covers me fucking shit up just because I don't know what I'm doing, which results in faster service]

PMID: 31499940 Abstract Title:  Thymoquinone loaded mesoporous silica nanoparticles retard cell invasion and enhance in vitro cytotoxicity due to ROS mediated apoptosis in HeLa and MCF-7 cell lines. Abstract:  Thymoquinone (TQ) loaded monodispersed mesoporous silica nanoparticles (TQ-MSNPs) with size of 188 ± 3 nm were prepared and characterized using DLS, TEM and FTIR. These TQ-MSNPs overcome the limitations of free TQ like hydrophobicity, low aqueous and photo stability and thus enhance its anticancer activity. In vitro release kinetics showed biphasic drug release where up to 50% was released in first 8 h and subsequently 98% released after 48 h. Enhanced cytotoxicity of TQ-MSNPs was observed against MCF-7 and HeLa cell lines as compared to free TQ. DAPI and Annexin V-FITC/PI staining confirmed the induction of apoptosis in cancer cells following treatment with TQ-MSNPs. Also, TQ-MSNPs exhibited enhanced anti-invasion properties against both cell lines as very low concentration of loaded TQ imparts similar benefits as free TQ. Both TQ and TQ-MSNPs exerted their cytotoxicity via reactive oxygen species (ROS) generation, as addition of an antioxidant NAC attenuated their killing activity.

Why Colorado is a Must for All Adventure-Seeking Travelers

Hey there, fellow travelers!
Ah, Colorado… where the air is so pure that my polluted Jersey lungs went into shock. It’s a breathtaking state with countless adventures to be had. When I went to Colorado it was a part of my first big road trip in 2016. We went to Colorado from Wyoming, where we visited Yellowstone and Grand Teton National Parks (coming soon in another blog). It was a pretty long trek from Grand Teton at about 8 hours from there to our first stop, Estes Park. I totally loved this town. It’s got a great main street area with tons of gift shops, restaurants, and cool stuff to check out. If you are visiting, make sure to check out the town area before leaving!
Rocky Mountain National Park
Estes Park, Colorado is the gateway to Rocky Mountain National Park. During these road trips we are usually on a pretty tight schedule and very rarely ever stay anywhere for more than a night. Due to the time constrictions we usually try to find shorter hikes to do that are still considered great trails. A lot of trails in the Rockies are on the lengthier side, but if you are looking for a short trail with some awesome views then you’re in luck because there’s two great ones. The first is Forest Canyon Trail, and you really can’t even call it a trail, it’s more of a 0.2 mile sidewalk leading to a stunning view of the Rockies. We did this trail as soon as we got into Colorado. Just driving through the park also boasts phenomenal views and there are plenty of view points to pull over and take some pictures.

The highest point of elevation in RMNP is 12,183 feet above sea level, which is at Alpine Ridge Trail. Keep in mind that at this altitude there is 35% less oxygen in the air than there is at sea level. There are stairs leading up to the viewpoint so it is very easy to become winded. Although I was lucky enough not to experience altitude sickness, it can happen to some people. Symptoms include nausea, headache, fatigue, shortness of breath, and decreased coordination. The best thing to do if you do experience altitude sickness is to descend as quickly as possible. In order to try and prevent this from affecting you during your hike you should stay hydrated, eat a high calorie diet the day of, avoid tobacco and alcohol, and take it slow. If you can make it to the top without losing your breath, the view from the top is sure to take it away anyway!

On the descent back down to the exit of the park we encountered an area still completely covered in snow... on the 4th OF JULY! This was absolutely mind-blowing to us Jerseyans, although it’s probably normalcy to those who reside in Colorado. Naturally, we had to get out of the car and play in the snow! How many other times could we say we played in the snow on the 4th of July?

The Stanley Hotel
If you are a horror fan like me, The Stanley Hotel is a must visit! The Stanley Hotel is located in Estes Park, as well, and it is the hotel that inspired Stephen King to write “The Shining.” The hotel is very expensive to stay in so we opted for a cheaper option about 5 minutes away, but they do offer guided tours of the hotel which we knew we couldn’t miss. The Stanley fully embraces its horror inspiration with ghost tours, an entire area dedicated to Shining memorabilia, and some pretty awesome sounding Halloween events. We opted for the ghost tour ($28 per person) and it was really cool to be able to see the hotel and learn some of its secrets.

Dinosaur Ridge
As mentioned in a previous blog, I have a fascination with dinosaurs. Anything to do with dinosaurs I have to check it out. So when I found out there was a place in Colorado where I could see real fossilized dinosaur footprints I absolutely had to add it to the itinerary. From Estes Park it was only about an hour drive and it was a pretty quick stop. The hike to the actual spot where the prints are located is about a mile and a half and it’s uphill, so bring water with you. The day we went it happened to be about 90 degrees so it was a bit rough making it up there, but seeing the dinosaur prints were totally worth it. They also have a gift shop with tons of cool dinosaur related things to buy as well as a small area where kids can do a little “fossil digging”. They even have 2 life-sized dinosaur replicas to take pictures with. Definitely worth a stop! Dino Ridge is only about a half an hour from Denver.

Great Sand Dunes National Park
Our next stop in Colorado was my favorite from the state: Great Sand Dunes National Park. From Dinosaur Ridge it was about another 2.5 hours to arrive at the park, but before heading there we stopped to pick up SANDBOARDS! This was something I have always wanted to do and couldn’t wait to try it out. We rented our boards from Kristi Mountain Sports in Alamosa and for the entire day it was only $18 per board. You can visit their website (http://www.kristimountainsports.com/products/sandboarding#rental)  for more information on how to reserve a board for your visit. They also had sand sleds to rent, as well. We opted for both but found that the board was much more fun.
A few pieces of advice for the dunes… be prepared to get sand everywhere. Eyes, nose, mouth, pants… no area is off limits. If you are a contact wearer like me, you WILL get sand in your eyes. Between the boarding and the wind the sand flying all over the place was inevitable. We tried to combat it a bit by tying a bandana around our faces and wearing sunglasses, but I’d be lying if I said it did much. We found it easiest to board with our socks on as opposed to leaving our sneakers on. I have no experience with snowboarding or any other kind of boarding but I did pretty well on the dunes. It took me a few tries of falling on my butt and kicking sand in my own face to get the hang of it, but before I knew it I was sticking the landing at the bottom of the dune. The dunes are a true marvel so even if you don’t plan on boarding this park is definitely still worth a stop!

That night we stayed at the Alamosa KOA. For those of you who are not familiar with KOA (Kampgrounds of America) it is a really great thing to look into. For $30 a year you can become a KOA member which grants you discounts and lets you accrue points for each booking you make. We stayed at multiple KOA’s during this trip and loved each and every one. At most KOA’s you have the option of booking a tent site, an RV site, or a cabin which will include a bathroom or no bathroom option (the no bathroom option always being the cheaper choice). We booked a tent site and were very comfortable here for the night. The bathrooms were clean and the staff was very accommodating.

Mesa Verde National Park
On our last day in Colorado we drove 3.5 hours to Mesa Verde National Park. MSNP is a great park for those who are interested in not only great hiking but also some fascinating history. It is best known for its Ancestral Puebloan cliff dwellings. These structures date all the way back to somewhere around 1190. Yes, 1190!  Dwellings that people built about 800 years ago are STILL standing today. It is truly incredible to think about. We were really excited to do the Cliff Palace tour, the most notable dwelling in the park, but we were not fully prepared for this endeavor. The tour is only $4 per person but it can’t be booked in advance and you may only do so once you arrive at the park, and unfortunately, we arrived too late. In order to get tickets for the tour you MUST arrive early and head right into the visitors center to get your tickets before they sell out for the day. If you do miss the tour, you can’t do the cliff palace trail on your own but there is a trail that gets you close enough to the dwellings to see them from afar, so we decided to do that. We asked one of the rangers at the visitors center and he filled us in on that.

Four Corners Monument
On our way into Utah out of Colorado we made a quick stop at the Four Corner Monument. I remember learning about this place as a kid in elementary school and always thought the concept of standing in 4 states at once was so cool. It was only an hour drive from Mesa Verde and although it’s a very quick “touristy” stop it was worth it for me to satisfy my childhood dream of one day doing this. There was a bit of a line that we had to wait on to take the picture but it wasn’t too long. They also had tons of handmade Navajo things to buy there.
