Nononono it’s totally fine! I’ve found that it’s actually really hard to learn about this stuff unless you’ve actually been to a live concert or viewing. Which… as some of you may know from experience, is excruciatingly difficult if you’re an overseas fan (꒪⌓꒪)

The short answer: UO stands for “Ultra Orange”, and is a chemical glowstick that glows bright orange, often only for a couple of minutes before it burns out. Because of how short-lived they are, people usually save them for their favorite songs at concerts.

Glowsticks are a staple of anisong and idol concerts, where audience participation is super important. The audience section is dark during concerts, and waving around glowsticks helps the performers know that you’re enjoying the show!

There are two types of glowsticks: battery-powered penlights and cyalumes.

Penlights

These are battery-powered LED glowsticks. Many of them can change color with the click of a button. It’s pretty much a plastic tube mounted onto a handle with buttons. You can almost always unscrew the tube off, and then the handle is basically a multi-colored flashlight. Most of them also come with wrist straps, so that the penlight doesn’t come flying out of your sweaty hands at a concert. The brightness and colors will die as the batteries drain, so I always recommend packing some fresh batteries for concerts.

If you’ve ever watched a live (whether in-person or not), you’ve probably seen plenty of these. I’ll cover some of the more known brands from the perspective of a Love Liver.

King Blade

One of the most popular brands of penlight. The newest version (x10iii) comes with an app that allows you to tune the penlight to have whatever color palette you want (e.g. you can make specialized blades with the colors for μ’s, Aqours, or whatever you favorite group is!)

Most of the official Love Live! penlights are King Blades. They’re often colloquially referred to as “kbs” or “blades”. The King Blade is so common (at least in the Love Live! community) that sometimes you’ll hear people use the word “kb” as an umbrella term for all penlights.

Another good penlight brand, and a somewhat cheaper alternative at $25 USD vs. the King Blade’s $30-35. The default color palette on the latest version, Lumiace 2 Omega (abbreviated L2O) is 24 colors rather than the King Blade’s 15 colors. However, the L2O cannot be programmed to have a custom palette like the King Blade x10iii.

The official Aqours in Los Angeles penlight from Anime Expo 2017 was a Lumiace 2, and comes with the nine official Aqours image colors rather than the Lumiace default palette. As far as I know, this is the only official Love Live! penlight that isn’t a King Blade.

The two different styles are the same as the King Blade ones — the former has a glittery tube, while the latter has a matte tube.

PENLa

A slightly different type of penlight. PENLas don’t actually change color, but they have three different brightness settings. The first brightness setting is about as bright as your King Blade or Lumiace at full battery. The second and third are much, much brighter. The primary advantage of PENLas is that they are just as bright as chemical glowsticks (covered later), but they are reusable.

They are suuuuper bright. My eyes.

A fan compares a PENLa UO to a KB x10ii. Jeez they are bright.

PENLas come in three tube sizes (S, M, L), but I recommend the S because same brightness + smaller tube = brighter penlight :)

Unlike KBs and Lumiaces, they also come with a small bag. I’ve heard complaints that the wrist strap that comes with PENLas isn’t as secure.

Cyalumes

These are chemical glowsticks. You crack them to cause a chemical reaction that makes the glowstick… glow. And boy do they glow. They’re much brighter than penlights, but they only glow for a couple of minutes before dying. And they’re one-time use only, so they’re useless after that. By the way, cyalumes are also kind of fragile. Don’t drop them unless you want to crack them by accident and have them dead before you actually need them (꒪⌓꒪)

Cyalumes are almost always colloquially referred to as the abbreviated form of “Ultra [color]”, regardless of brand. Ultra Oranges are UOs, Ultra Blues are UBs, Ultra Greens are UGs, etc.

The UO in particular has chemicals that make it much brighter than any other color of cyalume, making it the most popular color by far (regardless of who your best girl might be!). They’re an irreplacable part of idol concerts. Because of how bright and short-lived they are, people will save UOs for special moments such as solos or the choruses of their favorite songs. They’re also smaller and thinner than penlights, so it’s easier to go crazy and hold like 8 of them at once.

For even more hype, you can take two UOs and do a rapid cycling motion above your head. This is called “cycloning”. At concerts you will often see people cycloning during the chorus of fast-paced crowd favorites such as Strawberry Trapper or Thrilling・One Way. You can also cyclone using your kbs, but it’s more exciting if you use brighter glowsticks like UOs or PENLas. It’s also way easier to cyclone with UOs — if you try to cyclone with bulkier glowsticks like kbs without practice you’ll probably end up hitting them on each other.

Perhaps the most famous use for the UO is during Honoka’s solo in Snow Halation. At Final Live, everybody popped a UO. No kidding:

Other colors are less common because they’re not as bright. Even so, in recent years, more and more Love Livers are starting to use colors other than Orange. I’ve seen people cracking UBs for both Koi ni Naritai Aquarium and MIRAI TICKET, and I know at least a couple of others who are considering UGs for HAPPY PARTY TRAIN.

How long do cyalumes last? A good rule of thumb is that the brighter they are, the more short-lived they are. The Lumica Daisenkou Kiwami Orange is one of the brightest brand of UOs available, and one stick lasts 2-3 minutes. Nice duration if you want to crack for a solo or chorus and have the UO last through the rest of the song before dying.

You can also buy standard Lumica Daisenkou Orange, which are not as bright as the Kiwamis (side note: kiwami means “extreme” lol), but last for 5 minutes instead. On the other hand, UBs are not much brighter than a KB at full battery, but they last for a good 15 minutes. In my experience, Kiwami UOs are always better than the normal UOs. The extra brightness is 500% worth it (•̀ ∀ •́)ﻭ ✧

The best brand of cyalume for buying in bulk (and trust me, you don’t want to buy just a single chemical glowstick. Especially if you’re importing from Japan.) is the Lumica Daisenkou. Why buy cyalumes at all when PENLas exist? They’re short-lived, and don’t seem very cost-effective.

The way I see it, the point of having chemical glowsticks is to show both the performers and the people around you how excited you are for your absolute favorite songs. As a novice concert-goer I’ve found that I really enjoy cracking UOs (and UBs) in the heat of the moment and seeing other people get fired up cracking UOs for the songs they like best. They may not be very cost-effective, but at the end of the day, they truly enhance your concert experience.

The progression in cure, early detection and degeneration of oral squamous cell carcinoma (OSCC) remain a key factor to improve the survival rate of patients, in which an elevated proportion of patients are diagnosed at an advanced stage. Current developments in molecular biology research enhanced the understanding of molecular process in OSCC progression and led to identification and characterization of numerous biomarkers. These biomarkers are expected to facilitate the early detection of primary and relapsed tumors. In the present research, we evaluate potential biomarkers for the early detection of OSCC.

Introduction

Oral cancer is a subgroup of head and neck cancer which affects various regions within oral cavity i.e., lips, tongue, salivary glands, and gums. It is the sixth most common cancer, approximately 3% of the total cancer burden and results in 128,000 annual deaths globally [1,2]. The most common type of oral cancer is oral squamous cell carcinoma (OSCC), which accounts 90% of all oral cancer cases. Patients with OSCC are often diagnosed at a late stage, thus high recurrence rate occurs after treatment, especially in those with neck lymph node metastasis [3]. Despite clinical and treatment advances, the overall 5-year survival rates for oral cancer remains low and stagnant during past few decades [4,5]. Although OSCC is commonly diagnosed through oral examination followed by histopathology and computed tomography/positron emission tomography scanning, there has been continuous interest in developing serum protein biomarkers to aid the diagnosis. Tumor antigens are promising diagnostic biomarkers for human cancers showing the clinical utility of tumor antigens, such as carcinoembryonic antigen (CEA), CA-50, CA19-9 and squamous cell carcinoma antigen (SCCA) for OSCC detection [6-9].

Serum SCCA appeared to be more sensitive than the other tumor antigens and positive in 38.1% and 41.4% of OSCC patients under testing [8,10]. Studies also revealed other potential serum protein biomarkers i.e., CYFRA 21-1 (cytokeratin 19-fragments), tumor polypeptide antigen (TPA) and insulin-like growth factor binding protein 3 [9,11]. CYFRA 21-1 has been demonstrated as biomarker for other solid tumors, whereas TPA is a serine protease found in rapidly growing tissue due to its role in forming intermediate filaments of the cellular cytoskeleton, making it a promising candidate for cancer detection. Both CYFRA 21-1 and TPA levels were found to be significantly higher in OSCC patients compared to healthy controls and benign tumor patients, and both reduced in 2–3 weeks after surgical resection of their OSCC lesions [11]. Although the testing serum protein biomarkers as a simple diagnostic tool for oral/head and neck cancer has been well demonstrated but still needs to be further validated in large clinical trials. In this study, proteomics analysis of serum for early detection, evaluation, aggressiveness and occurrence of OSCC were summarized. The emphasis was placed on early detection by serum with histological defined oral carcinoma patients. Protein in tissues, saliva and serum may more accurately reflect the progression of OSCC, so novel approach for the depth research strategy and the sample choice for proteomics are of importance in OSCC biomarker discovery.

Material and Methods

Sample collection

Information about chewing habits and other characteristics of the study participants was acquired. Those with the habit were questioned for the frequency of the habit in number per day and duration of the habit in years. The type of lesion in the cases was decided on the basis of careful observation of the oral cavity and then blood samples were collected from People’s College of Dental Science and Research Centre, Bhanpur Bhopal, over a period of one month. 2-3ml of blood were taken as samples from the patients having oral premalignant lesions (OPL) (leukoplakia and/or erythroplakia) and stored in sterile vials containing 1.5mg/ml EDTA and stored at -20 °C. Individuals who were not addicted to smoking, tobacco or not habitual areca nut consumer and were not having any prior history of cancer and who had never smoked before in his or her lifetime are defined as a never smoker and were considered as control.

Determination of Protein Content

Lowry method

The protein content of serum was determined 1mg/mL stock solution of Bovine serum albumin (BSA) was prepared. BSA was dissolved in 0.1 N NaOH and used as standard [12]. For preparation of standard curve of BSA different concentration that is 100μg/mL to 1000μg/mL of BSA was prepared in clean glass tubes and volume was made up to 1mL using Millipore water. To each tube, 5mL of alkaline solution was added and the tubes were incubated at room temperature for 10min. After incubation, 0.5mL of Folin’s reagent was added and was incubated at room temperature in dark for 30 minutes. Violet to blue colour was developed. The intenity of colour was propotional to protein concentration. After incubation the absorbance was measured at 660nm. Standard curve was prepared by plotting protein (BSA) concentration on X-axis and O.D on Y-axis. A blank was prepared by taking 1mL of Millipore water in tube in which 5mL of alkaline solution and 0.5mL of Folin’s reagent were added. All the procedures were carried out in triplets. Estimation of protein contents was done in a same manner as BSA standard curve.

Sodium dodecyl sulphate polyacrylamide gel electrophoresis (sds-page)

Poly acrylamide gel electrophoresis in the presence of sodium dodecyl sulphate was performed according to LaemmLi [13]. The SDS-PAGE was performed using a 12% separating gel and 4% stacking gel. The samples were heated for 5min at 100 °C in capped vials with 1% (w/v) SDS in the presence of β-mercaptoethanol. Electrophoresis was performed at a 125 V for 4h in Tris- HCl buffer of pH 8.3. After electrophoresis, proteins in the separating gel were made visible by staining with Coomassie Brilliant Blue R-250. The standards were used to make a plot of log molecular weight versus mobility of the protein band were myosin (200kDa), β-galactosidase (120kDa), bovine serum albumin (91kDa), glutamate (62kDa), ovalbumin (43kDa), glyceraldehyde-3-phosphate dehydrogenase (36kDa), carbonic anhydrase (29kDa), myoglobin (26kDa), and lysozyme (14kDa) as markers [6] (Table 1).

Results and Discussion

The OPLs (leukoplakia/erythroplakia) case sample was collected from OPLs patients of PCDS&RC, People’s University, Bhopal the sample size was 20 in which 3 were females and 17 were males under the age group of 18 to 60. Taking into consideration the addictions, 11 of them were habitual of smoking either bidi or cigarette, 6 were betel nut consumers whereas most of them were pouch/gutkha consumers. Among the sample population, female: male ratio observed was 3:17, where females account for 30% and males 70%. Total population (OPLs cases) age range varies from 18 to 68 years, of which 18- 68 and 25-54 was age range of males and females respectively. Among the observed population 38% were smokers, 21% were betel nut consumers and majority i.e., 41% were pouch and gutkha consumers. Biomarkers have been wide accepted in other disciplines but there is no consensus for their use in oral malignancies. Despite recent advances in surgical, radiotherapy, and chemotherapy treatment protocols, the survival of patients with OSCC still lacks significant improvement. This unsatisfactory treatment may be explained by the fact that OSCCs frequently present with extensive local invasion and advanced stages [14,15]. That makes necessary the development of new tools for the diagnosis and prognosis.

Protein Profiling

Total protein

From the known concentration of protein, the standard curve was plotted, and protein content of the clinical isolates were observed. The total proteins were estimated which revealed the difference in Oral premalignant lesions. The protein concentration ranged from 3.2-11.8mg/mL. Control group the protein concentration ranged from 2.87-8.35 (Figure 1). Oxidation of protein plays an important role in pathogenesis of cancer and studies have demonstrated decreased protein levels in cases of OPMD’s and oral malignancy [16,17]. In oral cancer, tobacco and areca nut related habit leading to tissue damage and resultant free radicals play a major role as an aetiologic factor. These habits are seen commonly in all the ages and both the sex. The serum protein levels were decreased in OSMF, OL and NS but increased in OM. This difference was statistically significant. These findings are fill agreement of with the findings of Patidar et al. [18] and Rajendran et al. [19] in OSMF participants and Dawood et al. [20] in OM participants. In contrast our results did not simulate with the finding of Chandran et al. [16] in OM group where the plasma protein levels were found to be decreased. The increase in serum protein levels may be explained in terms of inflammatory reaction associated with oral malignancy.

Sodium dodecyl sulfate polyacrylamide gel electrophoresis

SDS-PAGE analysis of various samples revealed at least 06 types of banding patterns, with the number of bands ranging from 25 to 40. The maximum number of sample 26.6%) had a banding pattern with 04 bands. Proteins with molecular weights 66kDa, 75.0kDa, 95,110 and 140kDa were consistently present in the in the pattern. The pattern 2 showed number of 16.6 % sample had a banding pattern with 03 proteins bands with molecular weights 11.5,23, and 90.5kDa. The pattern 3 (10% sample) showed 04 proteins bands with molecular weights 118.5, 23, 66 and 97kDa. Pattern 4 showed number of 23.3 % sample had a banding pattern with 04 proteins bands with molecular weights 14, 18, 42 and130kDa. Pattern 05 also showed number of 10 % sample with 04 proteins bands with molecular weights 67.5,75,95 and110kDa. Pattern 6 revealed 03banding pattern with molecular weights14.5, 38.5, 55.5kDa. Feng et al. [21] measured the level of some biomarkers (SCCA, Cyfra 21-1, epidermal growth factor receptor (EGFR) and Cyclin D1) in an attempt to determine the usefulness of their combined determination in the diagnosis of OSCC. They concluded that Cyclin D1, the product of the CCND1 gene located on chromosome 11q13, had the highest diagnostic specificity. Moreover, the combined detection of EGFR and Cyclin D1 (36kDa) had the highest sensitivity, specificity and accuracy. A previous study Capaccio et al. [22] demonstrates that Cyclin D1expression was significantly associated with the presence of occult lymph node metastases. These data suggest that the immunohistochemical analysis of Cyclin D1 expression in diagnostic biopsy samples may be an additional tool for selecting patients to be treated with elective neck dissection.

The dendrogram showed that the samples [12] were grouped in two closely related clusters. The clusters of Oral Premalignant lesions were significantly different from an unrelated to that of the control. It was also seen that sample from control tended to fall close together on cluster analysis. In our study, the SDSPAGE pattern revealed several characteristic bands common to all samples.

The sample under study was divided in to 2 clusters with cluster 1 having 12 sample and cluster 2 with 18 samples having a similarity 82.5% and dissimilarity of 17.5% in the jaccard’s coefficient scale ion the dendogram (Figure 2). The cluster 1 was further divided into 1a and 1b with 3(OPLs-18, OPLs-08 OPLs-14,) and 9 (Control-10, Control-07, Control-6, Control-3, OPLs-16, OPLs-13, OPLs-07, OPLs-03, OPLs-05) sample respectively. The cluster 2 was also divided in to two sub-cluster 2a (1) (Control-1) and 2b (17) (OPLs-17, OPLs-04, OPLs-12, OPLs-20, OPLs-02, OPLs-10, Control-09, Control-08, Control-04, Control-02, OPLs-06, OPLs-15, Control-5, OPLs- 19, OPLs-11, OPLs-01, OPLs-09). Cluster 2a having 1 sample showed similarity 88.7% and dissimilarity of 10.5%. 2b showed similarity 87.3% and dissimilarity of 12.3% (Figure 2).

The sub-cluster 2(b) was also sub divided into sub sub cluster as 2b1 (OPLs-17, OPLs-04, OPLs-12) showed similarity 87.9% and dissimilarity of 12.1%. Cluster as 2bII (OPLs-20, OPLs-02, OPLs-10, Control-09, Control-08, Control-04, Control-02, OPLs- 06, OPLs-15, Control-5, OPLs-19, OPLs-11, OPLs-01) showed similarity 86.1% and dissimilarity of 13.9%. These differences between the samples are due to the difference in their protein profile which can be mediated due to the difference in the oral premalignant lesions therapy. The study clearly indicates that the profile of total protein from Oral Premalignant lesions can be used for developing classification pattern. The cluster 2bII was also sub divided into sub cluster as 2b1Ia and 2b1Ib (Control-09, Control-08, Control-04, Control-02, OPLs-06, OPLs- 15, Control-5, OPLs-19, OPLs-11, OPLs-01) showed similarity 87.9% and dissimilarity of 12.1% (Figure 2). Cluster analysis has been used, allowing one to make a more objective interpretation of immunoprofiles, based on staining with multiple antibodies, and holding great promise for the immunohistochemical classification of tumors [23].

Ideally, a good clinical test requires high sensitivity and specificity. The oral cavity is commonly subject to inflammation from a variety of causes, including trauma, dental plaque, infection and certain mucocutaneous inflammatory diseases. Whether such oral inflammation (non-neoplastic conditions) affects the levels of the potential OSCC serum biomarkers is essentially unknown, because most studies investigated the potential biomarker levels only in OSCC and non-OSCC, without regard to other inflammatory conditions present [24]. If any OSCC biomarkers levels increases in the presence of oral inflammation to the level of OSCC patients, it would result in a high false positive rate and greatly reduce the value of that biomarker in clinical use for detection. Many of reported potential OSCC biomarkers, such as IL-6 [25], IL-8, IL-1 β [26], basic fibroblast growth factor [27] and molecules related to oxidative stress [27] are known to be important factors involved in inflammation and/or wound healing [28]. Indeed, the levels of some of these constituents have been reported to be significantly higher or lower in periodontitis or OLP patients who did not have OSCC [29]. Therefore, research that validates any potential OSCC biomarker with individuals having common non-neoplastic oral inflammatory diseases is necessary in order to establish the reliability of that salivary OSCC biomarker

Conclusion

Serum biomarkers obtained represent a promising approach for oral cancer detection, and an area of strong research interest. However, some issues/challenges needs to be determined in order to establish this approach as a reliable, highly sensitive and specific for clinical use, including lack of consistency of serum sample collection, processing and storage; wide variability in the levels of potential oral carcinoma serum biomarkers in both non-cancerous individuals and oral carcinoma patients; and further validation of oral carcinoma serum biomarkers with individuals either a chronic oral inflammatory disease or other types of cancers, but do not have oral carcinoma. Research for eventual standardization especially biological and physiological variance affecting the potential biomarkers gained importance in serum diagnostics. This approach can be useful in monitoring non-cancerous disease activity applying serum biomarkers for other forms of cancer.

The Ameliorating Effect of Uncooked beans Diet in Cd-1 Mice

Abstract

Beans contain serotonin and its precursor, 5-Hydroxytryptophan which have neurobehavioral actions on memory, anxiety, mood and pain. This study was therefore, designed to investigate the ameliorating effect of uncooked beans on pain sensation using three groups of Swiss white mice (control and test) weighing 18g-35g (n=10 each).The control group received normal rodent chow, while the test group received 50g of uncooked beans in 50g of rodent chow per day and serotonin precursor (5HTP) (0.2mg/50g w/w) diet. Water was given adlibitum while daily food and water intake, as well as body weight changes, were monitored during the 30-day study. The formalin tests were used to assess pain sensation. The results showed that in the formalin test, the frequency and duration of paw attention in both phases of the test was significantly lower (P<0.05) compared to the control group. The duration and frequency of paw licks (P<0.05) was also significantly lower in the uncooked beans diet and serotonin precursor group compared to the control. Therefore, consumption of uncooked beans diet may decrease pain sensation.

Introduction

Pain can simply be referred as an emotional feeling which is associated with tissue damage. It is therefore a protective mechanism in the body. Beans are considered as good source of protein content, complex carbohydrates, dietary fibers and some minerals and vitamins [1]. In addition to these nutritional components, common beans are rich in a variety of several phytochemicals with potential health benefits, such as polyphenolic compounds, flavonoids, saponins, alkaloids, glycosides, tannin, lectin, trypsin inhibitor, and phytic acids, among others [2,3]. It has also been reported that beans contain serotonin and its precursor 5-Hydroxytrytophan (5-HTP) [4]. Since beans contain neurotransmitters and chemicals that can potentially affect behavioural patterns, it may be worthwhile to find out whether long term consumption of uncooked beans diet can affect behaviour, such as pain sensation.

Materials and Methods

Experimental animals/grouping

Thirty Swiss white mice weighing between (18-35g) and bred at the animal room of the Department of Human Physiology, University of Abia State University. The animals were acclimatized under standard laboratory conditions and given free access to normal feed and clean drinking tap water. The animals were randomly assigned into two groups, control and a test group. The animals in the control group received normal feed (rodent chow) only; while the test group received mixed feed of 50g powdered uncooked beans per every 50g of rodent chow (50% of the uncooked beans diet) and (0.2mg/50g) serotonin precursor diet for 30 days.

Experimental design

The formalin test was used to test for pain sensation as developed by [5]. Mice were carried into the room in their home cages. Each mouse was picked by the base of its tail and 0.2ml of 2.5% formalin was injected into the right hind paw of the mouse using a needle and syringe. The animal was placed in the observation box and observed for 5 minutes. The animal was then returned to its cages and allowed for 30 minutes before it was taken back to the observation box to be re-observed for another 5 minutes. This procedure was repeated for each animal.

Behaviour scored during the pain test included the following;

a) Frequency of Right hind lick/scratch

b) Frequency of Right hind paw attention

c) Duration of attention.

Statistical analysis

Data Obtained from the experiments was statistically analyzed using Microsoft excel, with factorial ANOVA/T-test in the statistics programme start view version for windows or Mac. Post-hoc comparison was also done using the student ±Newmankeuls design. Values were represented as Mean ± SEM and a “P” value less than 0.05, was considered as significant.

Results

Figure 1 shows the frequency of right hind paw licks for mice fed normal, uncooked beans and serotonin precursor diets as 14.90±1.59; 7.71±0.75 and 5.71±0.42/5mins respectively in the first trial after 5 minutes of formalin administration denoting acute pain. The frequency of paw lick for the mice fed uncooked beans and serotonin precursor diet was significantly shorter (p<0.05) than that of the control fed mice. In the second trial after 30 minutes of formalin administration denoting chronic pain, the frequency of paw licks was 0.87±0.22; 0.14±0.14 and 0.14±0.14/5mins for mice fed normal, uncooked beans and serotonin precursor diet respectively. The frequency of right hind paw lick in the uncooked beans and serotonin precursor diet fed mice was significantly shorter (p<0.05) than those of the control fed mice.

The values for duration of hind paw lick following administration of normal, uncooked beans and serotonin precursor diets were 26.79±2.56; 17.75±2.32 and 8.67±2.08 seconds respectively in the first 5 minutes. The duration of hind paw lick in the uncooked beans and serotonin precursor fed mice was significantly lower (p<0.05) compared to control. In the second trial, after 30 minutes of formalin administration, the hind paw lick duration in the group of mice fed normal, uncooked beans and serotonin precursor diets were 1.30±0.52; 0.16±0.16 and 0.16±0.16 seconds respectively. The duration of hind paw lick was significantly lower in the uncooked bean and serotonin precursor fed mice compared to the control group (p<0.05) see Figure 2.

The values for the frequency of hind paw attention following administration of normal, uncooked beans and serotonin precursor diet were 24.00±2.07; 8.14±1.18 and 6.00±0.82/5mins respectively in the first trial after 5 minutes of formalin administration. The frequency of hind paw attention was significantly lower in the uncooked beans and serotonin precursor fed mice compared to control (p<0.05).In the second trial, after 30 minutes of formalin administration, the values were 1.20±0.47; 0.43±0.30 and 0.43±0.30. The frequency of hind paw attention was significantly lower in the serotonin precursor and uncooked beans group compared to control (p<0.05) See Figure 3.

The values for the duration of hind paw attention following administration of normal, uncooked beans and serotonin precursor diet were 89.38±11.33; 53.59±4.14 and 39.03±5.51 seconds respectively. The duration of hind paw attention fed with uncooked beans and serotonin precursor was statistically shorter than those fed with control diet (p<0.05). In the second trial, after 30 minutes of administration of formalin, the duration of paw attention was 2.60±0.60; 0.37±0.24 and 0.55±0.39 seconds respectively. The duration of hind paw attention was significantly lower in the uncooked beans and serotonin precursor fed mice compared to control (P<0.05) (Figure 4).

Discussion

The response of formalin-induced behaviour reflects activation of C fibre primary afferent nociceptors [6]. This test was in two phases. The response within the first 30 seconds following formalin injection is the perception of acute pain, while the later period shows chronic pain perception. Frequency of hind paw attention and hind paw-licking following injection with formalin was defined as the number of times the mice lick or shake their hind paw after injection with formalin. Lower frequencies of hind paw attention and hind paw licking indicate analgesic effect while higher frequencies indicate hyperalgesia. Our finding showed that during acute and chronic phases of pain, the beans diet- fed mice and that of the serotonin precursor fed mice had significantly less pain perception compared to control, since the frequencies and durations of hind paw lick and hind paw attention following formalin injection was significantly lower in the beans and serotonin precursor diet- fed mice than the control.

Pain reduction was observed on the first and second phases of pain following chronic consumption of beans diet. It is therefore interesting to note that beans diet can be beneficial in the reduction of chronic pain if the results in mice can be extrapolated to man. The Serotonin circuitry is a well-established pathway involved in brain’s analgesia system during transmission of pain in the central nervous system. It is known that the analgesic fibers of this system release neurotransmitters that inhibit pain transmission to the brain, and the neurotransmitters released by the fibers of analgesic pathway are serotonin and encephalin [7,8]. Our findings suggest that uncooked beans and serotonin precursor diet mice showed less sensitive to pain, when compared to those fed with the control diet. Beans diet may decrease pain sensitivity which may also be due to the presence of flavonoids and phlobatannins in the beans which has been reported to reduce pain perception due to their anti-inflammatory properties [9,10]. Finally, uncooked beans diet reduces pain sensation in mice. This may be so because beans contain 5-HTP (serotonin precursor) and 5-HT (serotonin) that plays a positive role in the brain analgesia system. A second set of experiments implicated the serotonergic pathway, as the threshold for pain perception was increased in the mice that consumed the serotonin precursor diet [11,12].

Group Brainstorm Duration: 5min

During our own time throughout the week, we added our ideas on the Asana board (an online project management system.) On Sunday, we took another additional five minutes on a conference call to put our ideas on the Asana board. We took two minutes to sort the items into three categories: events, programs, and technology. Then, we took another two minutes to tag each of our top five choices. We were able to isolate the top ideas into three columns:

Primary: We discussed that many of the ideas could fall under the resource portal idea as a function.

Secondary: These items required a bit more discussion with our team. Ultimately, we decided to use the trade fair system to empower the homeless through an eCommerce platform.

Tertiary: We think a great way to connect people locally and nationally is through education and awareness. We discussed how we could explore different campaign ideas to educate LA residents to be more accepting and participate in helping the homeless.

Here are other ideas we had that did not make the initial cut, but we might revisit based on the final method we end up selecting

III. Qualification of UV light meter LS126C and related national standards

I had a good week. There are several fixes, and I have added 'favourites' management for tag filters.

favourite tag filters

The Tag Filter is the newish whitelist/blacklist panel that manages which tags will be parsed, blacklisted, migrated, or, more recently, hidden on the new tag display system. It is being used in more and more places, so I have added the ability to save and load certain filters for quicker re-access.

So, now every tag filter edit panel has save/load/delete buttons in the top-right. They all share the same list of favourites. I also put in some example defaults. Let me know if you run into any trouble with it!

Thumbnails now sort by 'number of tags' and namespace sorts using the 'single' tag display context. So, if you have your thumbs set to hide page tags from the PTR, these page tags will no longer be included in a 'creator-series-volume-chapter-page' sort, for instance.

I fixed several smaller issues--a kind of video that would not import correctly, an unusual 'options' saving error after setting a number of ratings on several thumbnails, seeing duplicate thumbnails on some downloader highlights, gallery downloads that stopped early due to malformed post URLs, download pages sometimes secretly performing a collect and sort when a default collection is set, the media viewer being unable to show certain malformed files, and remote thumbnails not refreshing correctly when their respective file is imported.

Advanced users only: URL Classes now let you set whether requests for URLs matching the class will send a 'referer' http header. Normally, hydrus will send any 'previous' URL as the 'referer' (typically the gallery page when visiting a post page, or the post page when downloading a file), just as if you were visiting it in your web browser, but you can set it to never send any referrer, always use an auto-generated referrer, or use the auto-generated one when no natural referrer is available (which can happen on a user drag-and-drop URL import). The auto-generated referrer is a standard StringConverter operation on the URL being visited itself--if you can figure out a way to infer it from the original URL, or just replace it with a static value, I think some more perculiar downloaders should be possible.

For server administrators: The server no longer tries to slip in a quick vacuum when it is doing a backup. The vacuum command is now a forced, full vacuum command off the same admin menu. Also added are forced 'lock on', 'lock off', and 'test lock' commands that turn the 'server busy' state on and off and disconnect the db, letting you run a third-party backup if you wish.

For server janitors: You can now right-click petition rows to copy their tags. Multiple selections work, and siblings/parents offer left/right copy actions.

when a gallery page parser now adds new urls to a file import list, urls that are invalid will now be skipped (previously, they threw an error and failed the parse

fixed a bug where if a default collect is set, pages without a collect (e.g. download pages) would nonetheless initialise with collected+sorted initial media on session load

file imports now publish the same 'refresh existing media metadata' call as the file maintenance system, meaning if the import already exists in the gui session as an 'unknown thumb', it should now refresh itself correctly

if the media canvas is called to display an invalid media (due to mime mixup or a faulty parse that slips through), it should now better recognise that and skip/dump out

fixed import of videos that have 'Duration:' in their title metadata

improved the error reporting when the old options object fails to save

removed some old ratings dialog position options storage that was causing errors on certain ratings dialog ok events

url classes now support options regarding the 'referer' http header they send (their referral url). you can set an optional converter to generate a referral url based on the url class's url and choose to always use the given referrer if available, never use a referrer, use the converter if no referrer is available, or always use the converter

the network report mode now reports on referral urls used in requests

the 'quoted' referral url (a unicode workaround) is now only applied if the referral url cannot be encoded to latin-1

the janitorial petitions processing page now lets you copy tags and left/right tags of pairs with a right-click on selected checkbox rows

cleaned a little server code

improved how the server sets and releases its 'currently busy' mode

the server no longer does <5min vacuums in a backup command

added a specific 'vacuum' server POST command that forces a full vacuum

added 'lock_on' and 'lock_off' server POST commands to lock the server and shut down the db, and restart

the new vacuum, lock_on, lock_off, and a 'is server busy?' check commands are added to the services->admin menu

added 'pause and disconnect' ability to the database mainloop

added some unit tests for url classes and the new referral url conversions and server commands

cleaned some of the thumbnail banner/icon drawing code

some misc label fixes

I have one more week before the large wx->Qt migration occurs, which I expect to take about four weeks. Since there will be long break, I will try to mostly work on small things so that last wx release is a 'clean' one without any earth-shattering bugs. I'd like to have some 'session weight' counting so the client will notice and notify you if your session gets too large.

If you have missed the news, 8chan is planned to relaunch in the near future under 8kun.net. Assuming the launch goes well and they are able to stay up, I am interested in moving the main /hydrus/ board back there. They are planning to migrate board content over to the new site for BOs who ask, and I have--thankfully, I still had my BO 'shared secret' lying around in my password manager--so with luck, the old 8chan /hydrus/ should be migrated and we'll be somewhat back to normal.

What do you think Thistle’s past is, especially in regards to the pain that Iris keeps mentioning he sees? I wasn't under the impression they're close at all. Just because you pay to do the deed don't mean jack and neither have been very forthcoming without having serious strings attached/one hell of a mental game of chess going on. Similarly, will Iris still want Thistle’s help after they’ve been together for some time? Besides information, how do you foresee Thistle might help Iris? Given Thistle’s fear of his boss, do you think Thistle and Iris will suffer consequences for Thistle trying to leave? Training Block - Feb - Strength

The days are numbered and we are just 8 training weeks away from an event you won't forget.  The question is; how do you want to remember it - as the one you should have trained more for, or the glorious celebration of achievement, effort and willpower?

Moving on.

THIS IS IT, this block is really what will cement your base and multi-day training into a structured and honed race engine.  Three weeks (followed by recovery) of high-intensity training to promote strength and power will leave you tired, possibly cranky, but stronger and ready for the final block.

Take the recommended sessions, drop them onto a calendar and in 3 weeks you'll be exhausted but stronger as a result.

You will need:

6 hours of training time - don't worry if this is less than last block, the change in intensity should account for that

4-5 training days with rest days in between - try not to do more than 2 days consecutively (it's likely to do more harm than good)

Very likely a turbo trainer or gym spin session(s) 

Good hydration throughout

Good recovery protocol (i.e. carbs & protein in the first 30 min after riding)

Kind of goes without saying, but a balanced diet - i.e. reliable source of energy to support high intensity workouts

A good combination would be 3x short sessions with 1 or 2 longer sessions during the week interspersed with recovery days.

Recommended Short Sessions:

Hills

Warm up (10 min steady pace with 3 sprints at 110 rpm) 

Find a local hill that you love

Aim for about 4min of climbing at strong pace (i.e. the effort should have you breathing through your mouth)

Once you've hit 4 min return to your start point (spinning quickly to flush Mr Lactic from your muscles), try to take 2-3 min over it - maybe rest at the bottom

Rinse and repeat at least another 4 times

Cool down (10 min)

Sprint Intervals (good for commutes)

Warm up (10 min steady pace with 3 full sprints at 110 rpm) 

It can be useful to start a timer and sprint on a regular interval (like on the minute)

20s Full Gas sprint (don't worry about anything except fluid circular motion and trying to rip the pedals off) [P7, Z5, Perceived 10/10]

Recover for 40s [P1, Z2, Perceived 2/10]

Repeat for 4 more reps

Recover for 2-3 mins

Repeat the sprints - aim to do the set 4 times

Warm down

Safest to do this on the wind trainer

Optionally - you can switch out 20:40s for 30:30s (i.e. 30s sprint, 30s recovery) if you're feeling strong - these are probably twice as difficult so do this wisely.

Time Trial

There's a couple of places that this safe to do on the road (WMCC Liverton road circuit, Bits of North Miramar Peninsular), but as above it is safest to do on the wind trainer

Warm up for at least 15min (include 3-4 full gas sprints - 10s or so)

20min at Gas Mark Painful (i.e. heavy breathing, but still able to see straight) [P5, Z4/5, Perceived 7/10]

Cool down for at least 10 min

Longer Hill Reps

Good warm up (15 min or so)

You should aim for a 10min hill like Horokiwi or Blue Mountains at roughly time trial pace - i.e. you don't really want it to go on for another minute [P4, Z4, Perceived 7/10]

Recover on the way back down, wait / spin at the bottom for about 5min

Repeat again

Cool down for 10min

High-Intensity Trainer Sessions

Anything from the Sufferfest https://thesufferfest.com/ (except To Get To The Other Side and The Way Out)

Spin classes - Les Mills and other gyms offer these at very reasonable prices

Recommended Sessions to fit intensity into your Longer Steadier ride (LSD)

FTP Intervals

During your longer ride, you may have time to fit in some long-duration intervals which are great for supporting and training strength and long-duration power (FTP).  If it's a group ride, maybe discuss the idea of 20min at med/high intensity before dropping your riding chums :)  Though they can always catch you up.

Assuming you are already warmed up

20min at strong pace (i.e. the pace you could keep up for just 1 hour)  [P5, Z4 eventually, Perceived 7/10]

Recover / let your group catch you up for at least 5 min before repeating

Aim for at least 2 of these long intervals in your ride

Fast climbing

If you can't fit a steady longer interval, when you hit a 5min climb aim to hit HR Z5 [Perceived 8/10] by the end of it - aim to do this at least 3 times during the ride - maybe add more hills to your long rides :)

Recovery Day options

Core

These exercises won’t exactly get you a six pack, but have a noticeable effect on your cycling performance (click on workout name for link):

Around the World – 10min 

Strava / Fitstar Core Challenge (Strava Premium only) - 20 min 

Iron Strength – more advanced (read difficult) - 50 min  

Flex / Stretch

Yoga / Pilates - good options for improving general flexibility and stretching out muscles you didn’t know you had - providing you don’t mind the perception.

The Foam Roller (your muscles will thank you later) - I could go on about the foam roller, how it releases your fascia and works out knots and sore spots in your muscles, but I won’t - check out the following cycling specific exercises (click on title) and thank me later:

IT-Band 

Quads

Adductors (aka the VMO)

Hamstrings

Note of caution (partly experienced-based)

High Intensity workouts can lead you into the dark world of over-training - how do you know?  The following notes are geared around two possible outcomes of your training (too much or not enough)

Signs you're over doing it (Overtraining) If you get start seeing any of these, it's worth flagging this early.  

Chronic fatigue

Confusion

Decreased strength

Depression

Frequent minor infections

Increased apathy and irritability

Headaches and tremors

Increased perceived exertion during a constant exercise load

Garmin Recovery Advisor is pretty good at identifying overtraining through imperceptible changes to your heart beat.  If you have a Garmin look up this feature.   Take extra rest days if you see any of these signs as chronic overtraining can severely damage your build up to The Tour.

Signs you're under-cooking it (Undertraining)

Feeling really fresh all the time - this is usually a sign you are recovered - fine if this happens during a recovery week, but if you're feeling like this in the middle of a training block then you might want to consider upping the intensity of what you are doing (perhaps meditate on Rule #5)

Short easy miles (i.e. less than 100km) - whilst there's nothing wrong with recovery riding per se, riding at low intensities regularly is unlikely to produce the improvements you are seeking.  I'm not saying you need to smash every ride, but if you are training you will almost certainly need to break a sweat - it's ok to sweat it!  Sadly, if commuting forms part of your weekly training, then you may need to adjust how you approach it - can you add a hill or steel yourself to some sprint intervals?
