As part of the COVID-19 International Research Team, researchers at the Johns Hopkins Kimmel Cancer Center, Children's Hospital of Philadelphia, the University of Pittsburgh and Weill Cornell Medicine discovered a novel cause of cytokine storm -; the extreme inflammatory response associated with increased risk of death in COVID-19 infection.

Their findings were reported Nov. 27 in the online issue of Proceedings of the National Academy of Sciences.

In an intensive genomic search for causes of cytokine storm, the research team used autopsy samples obtained from 40 patients who died from COVID-19. They performed genome analysis on samples taken from multiple sites, including the lung, heart, liver, kidney, lymph nodes in the chest that initially filter the virus, and the nasal cavity where the virus enters the body.

They zeroed in on some 50 upregulated immune genes in the samples obtained from nasal swabs and followed through in the genomics for the autopsy tissues.

Stephen Baylin, M.D., Virginia and D.K. Ludwig Professor for Cancer Research and co-senior author, and first author Michael Topper, Ph.D., Evelyn Grollman Glick Scholar and instructor in oncology, were

familiar with many of the genes as part of the inflammasome, a protein signaling network they helped define that is activated to rid the body of virus or bacteria-infected cells.

"Some of the same genes involved in overactivation of the inflammasome appear to be key immune gene regulators of the hyperinflammatory process that leads to a new view of how these subsequently activate the "cytokine storm syndrome" and severely damage multiple tissues, says Topper.

The genes should turn on and off, Baylin explains, but when they stay on, it results in cytokine storm, the very severe inflammation that can be lethal to patients with COVID.

Essentially, immune genes in the nasal cavity, where the virus enters, send signals downstream through a system called renin-angiotensin-aldosterone system (RAAS) to initiate cytokine storm.

RAAS, a hormone system that normally turns on and off to help regulate blood pressure, body fluids and electrolytes, is the spark that pushes the immune response into overdrive, the researchers found, compromising the infection-fighting function of lymph nodes and causing severe damage to the lungs, kidneys, heart, liver and other organs.

The researchers also believe their findings may have implications for long COVID, a chronic condition following COVID-19 infection that is characterized by a wide range of symptoms, including fever, fatigue, coughing, chest pain, heart palpitations, headaches, joint and muscle pain, gastrointestinal issues and more. This is a focus of ongoing research, Topper and Baylin say.

About the Author

Chris Hedges, a graduate of Harvard Divinity School, was a foreign correspondent for nearly 20 years. He was the bureau chief in the Middle East and the Balkans, and worked in other foreign posts, for The New York Times from 1990 to 2005. He worked previously for The Dallas Morning News, National Public Radio and The Christian Science Monitor in Latin America and the Middle East. He has reported from more than 50 countries. Hedges was a member of the New York Times team that won the 2002 Pulitzer Prize for Explanatory Reporting for the paper’s coverage of global terrorism, and he received the 2002 Amnesty International Global Award for Human Rights Journalism. He holds a B.A. in English Literature from Colgate University and a Master of Divinity from Harvard Divinity School. Hedges has taught at Columbia University, New York University and Princeton University, where he is currently a Visiting Lecturer in the Council of the Humanities and the Program in American Studies as well as the Anschutz Distinguished Fellow. He has written for Foreign Affairs, Granta, Harper’s, Mother Jones and The New York Review of Books. Hedges is the author of War Is a Force That Gives Us Meaning—a finalist for the National Book Critics Circle Award for Nonfiction. His other books are What Every Person Should Know About War and Losing Moses on the Freeway: The 10 Commandments in America. He lives in New Jersey.

It's Fine Press Friday!

Today’s fine press book is an English translation of ancient Roman playwright Terence's romantic comedy of errors Andria, hand-printed in Verona in an edition of 170 copies by Giovanni Mardersteig (1892-1977) at his Officina Bodoni in 1971. The book uses Richard Bernard’s 1598 translation of the play from the Italian, with revisions and edits by Betty Radice.

In 1492, legendary German artist Albrecht Dürer (1471-1528) drew illustrations on wood blocks for a planned edition of Andria, but they were never cut. The blocks have been preserved, however, in the Kupferstichkabinett of the Kunstmuseum Basel. For this edition, those drawings were copied onto 25 new blocks and cut for the first time by noted German American artist and graphic designer Fritz Kredel (1900-1973).

The text was handset in Dante type, designed by Mardersteig, and printed on handmade Magnani paper, with the edition signed by Fritz Kredel. The edition maintains all the qualities Mardersteig established for his press when he founded it in 1922: "The book in its noblest kind should always be a work of art. And the instrument with which this end can be achieved most fully is the press.”

Then he went to Germany where, for a few years, there was more interest in him than in his “pick up band” i.e. The Beatles—who called Sheridan “The Teacher” despite (or because of) his crazy ways and occasional fisticuffs. After Beatlemania, Sheridan kept on as a successful German-based pop star, segueing into blues and jazz and involving himself in bizarre side hustles, anti-Communist platforms, and for a time he ran with Bagwhan Sri Rajneesh. Sheridan was secretive about his personal life, but he was wide open on the public stage.

The Tony Sheridan & The Beatles cover of “My Bonnie” prompted me to impulsively record it in a grunge-y San Francisco warehouse loft: Fast, dirty, and just for fun. Riceve in dono da lui un’edizione completa dei Saggi di Montaigne e il primo esemplare di un adattamento per pianoforte del primo atto del Sigfrido, da lei Le passeggiate di Roma di Stendhal.

Richard(1473 - 1540). Duke of York. Husband of Anne Mowbray and father of 10 children: John, Eleanor, Jane, Humphrey, Ursula, Geoffrey, James, Elizabeth, Joan and Anne. Regularly quarreled with his elder brother over his politics, for which he was banished from court. After his nephew's accession to the throne he was forgiven.

Anne(1475 - 1540). Queen of Navarre. Wife of Jean III of Navarre. Mother of 6 children: Louise, Henry II, Madalena, Arnaud, Gerard and Francoise. The marriage of Anne and Jean was not a happy one as the king had love affairs on the side. Because of her soft character, Anne had to put up with her husband's infidelities. She also did not settle at court because she was a foreigner.

George(1477 - 1546). Duke of Bedford. Husband of Elizabeth Howard and father of 13 children: Elizabeth, Edward, Catherine, Marcella, Dorothy, Charles, Edmund, Agnes, Douglas, Ralph, Jasper, Emma, Lionel. He had a sociable nature and loved jokes, and in his spare time wrote poetry. George and Elizabeth's marriage was a happy one.

Catherine(1479 - 1549). Queen of Spain. Wife of John III. Enjoyed great popularity at the Spanish court. She was a patroness of writers, musicians, poets and artists. Had great influence on her husband. The marriage of John and Catherine was very happy. They had 7 children: Luis I, Isabella, Ramona, Ferdinand VI, Blanca, Garcia and Jaime.

Briget(1480 - 1550). Empress of the Holy Roman Empire. Wife of Philip I of Austria and mother of 9 children: Maximilian II, Frederick, Mary, Rudolf, Elizabeth, Eleanor, Philip, Margaret, Albrecht. She exercised great influence not only on her husband but also on her son Maximilian II. Under her, the imperial court became the center of European culture.
