Effect of Growth Regulators on Rooting of Andrographis neesiana: A Valuable Endemic Medicinal Plant of India

Abstract
The present study deals with the vegetative propagation prospects of an endemic medicinal plant Andrographis neesiana Wight, which is familiarly used in herbal medicine for the treatment of several diseases. Due to over exploitation this plant is disappearing from original habitat hence its cultivation on commercial scale is suggested. Stem cuttings of Andrographis neesiana are advantageous to root. Treatment with Indole-3-Butyric Acid (IBA) and a-Napthalene Acetic Acid (NAA) raised rooting and increased shoot development in greenhouse under intermittent misting. IBA and NAA treated cuttings performed improve in all development parameters compared to control. The highest percentage of rooting was observed in IBA 1500 ppm (68.10%). The maximum root length was noted in IBA 2000 ppm (11.20 cm). The roots were profuse and branched in characteristic. Besides this, the survival rate of IBA treated plantlets was 86.45% and NAA treated plantlets was 72.61% in comparison to control where it was 7.32%. The percentage of rooting and root length upgraded by using plant growth hormones, either separate or together. The present investigation concludes that clonal multiplication of these an endemic medicinal plant is feasible through application of plant growth regulators. The significance of these findings on the propagation and conservation of A. neesiana is discussed.

Introduction
Medicinal plants contain importances that can be used for biotherapeutic applications or which are used as precursors for the synthesis of beneficial (Sofowora 1993). The medicinal characteristics of plants lies in their phytochemicals namely alkaloids, flavonoids in addition to other phenolic compounds that co-opt as to producing particular physiological properties to the body of man and animals (Che Man, 2010). However, analysis of extract of Andrographis neesiana was found to be rich in flavones to the expensive phytoconstituent potentialities of the plant. Andrograhis neesiana is therefore grown as a medicinal species for the curing of many ailments particularly in India. (Alagesaboopathi and Balu 1999; Alagesaboopathi and Sivkumar 2011; Ponvinobala et al., 2012a). Andrographis neesiana Wight belongs to the family Acanthaceae and has been widely used in healthcare traditions. The rooted plants by cottage have various advantages such as faster growth rate (Ooyamma and Toyoshima, 1965), greater stock stand uniformity, improve site matching and true-to-type planting material production (Fielding, 1969). Cuttings can be categorized into 3 groups as comfortable to root, difficult to root and inflexible to root (Nanda, 1970).

Species of Andrographis Wallich ex Nees (Acanthaceae) are used in the Indian systems of medicine namely, Siddha, Ayurveda, Unani, Naturopathy, Homeopathy, Amachi and Modern (Alagesaboopathi and Balu, 1999). The genus Andrographis as a whole is of potentialities significance to India. The genus exhibits antipyretic characteristics (Kirtikar and Basu, 1975). This genus consists 40 species distributed in Tropical Asia (Anonymous, 1948). About 21 species are distributed in India (Gamble, 1982) and all of them available in Tamilnadu. (Henry et al., 1987). Among the 21 species 18 species are reported to be endemic to India (Ahmedullah and Nayar, 1986). Andrographis neesiana Wight (Fig. 1) is an endemic medicinal species (Ahmedullah and Nayar, 1986) found in wild in Shevaroy Hills of Salem district of Tamilnadu (11o45 and 11o55 N and 78o11 to 78o20E) upto 1500 m.
The pharmaceutical industries is widely dependent upon the wild populations for providing these plant species for extraction of their intrinsic phytochemicals. Moreover, herbal medicine practitioners, local medicine men, village dwellers, vaidyas, tribals (Malayalis), forest dwellers, and other traditional healers often use these collection of the plant materials from forests and lacking experiments either to allow the replenishment or propagation, these critical flora are rapidly vanishing. As a importance, A. neesiana declared as an endemic plant in India (Ahmedullah and Nayar, 1986). Therefore, there is an immediate need to enlarge serviceable cultivation techniques for propagation of these noteworthy medicinal floras which will excessively commercial application. Andrographis neesiana has been used in the treatment of aphrodisiac and antifungal property (Alagesaboopathi and Balu, 1999; Alagesaboopathi and Balu, 2000). Several medicinal properties such as cough, edema, laxative, bitter and overcomes trouble in breathing, worms, liver complaints, acidity, burning sensation, syphilitic ulcers, skin disorders and also veterinary medicines have been attributed to this plant in the traditional systems of Indian medicine (Sivarajan and Balachandran, 2001). It is used in the treatment of antimicrobial, antioxidant and anticancer activity (Alagesaboopathi and Sivakumar, 2011; Ponvinobala et al., 2012 a, b).

Two new flavonoids, 2, 4, 6, 2, 3, 4- hexamethoxychalcone and 5-hydroxy -7, 2, 5 - trimethoxyflavone together with a known flavone glycoside, echioidinin 5-O-beta-D-glucopyranoside were isolated from the whole plant extract of Andrographis neesiana (Muntha Kesava Reddy et al., 2003). There is no previous report on use of growth hormones in vegetative propagation of this useful plant. The work was undertaken to result rooting response of A. neesiana under greenhouse using growth regulators and outcomes reported. Auxin is one of the factors for stimulating root productions in cuttings (Hartman et al., 1990). Thakur and Gupta (1998) reported that cutting of Alnus nitida with various concentration of IBA and obtained the maximum root percentage at IBA 800 ppm.
Rooting of stem cutting through action of growth hormones has been undertaken as a protocol for broad scale propagation of this plant following the approach of Senthilkumar et al., 2009; Akwatulira et al., 2011; Saradha and Paulsamy, 2012). Auxins, a category of plant evolution substances are often called as plant growth hormones and performance an important role in coordination of many growth and behavioral technique in the plant life cycle (Tiwari and Kuntal Das, 2010). Indole-3-Acetic Acid (IAA), -Naphthalene Acetic Acid (NAA) and Indole -3-Butyric Acid (IBA) are conventionally the leading auxins which are usable commercially and can be applied with liquid (liquid formulation) or in talc (powder formulation) for rooting and sprouting of stem cuttings (Hopkins, 1999). The present study therefore aimed at ascertaining the most suitable propagation A. neeisana using stem cuttings. In the present study, the stem cuttings of A. neesiana were treated by plant growth regulators (PGRs), IBA and NAA. The aim of this investigation was to test the potential outcome of plant growth regulators on the stem cuttings and to prefer an optimal concentration of plant growth regulators for stem cuttings of A. neesiana

AYUSH MEDICINES: Magic or Logic (2024)
Magic is the use of supernatural forces to create effects that defy natural laws and evoke a sense of wonder.
Magic is an effective use of laws of physics to create illusion and virtual reality to make people believe things that they can't imagine under normal circumstances with common sense.

Many a times due to scientific shortcuts and lack of wisdom or vested intentions, traditional practices are termed Magics.
AYUSH medicines, which include Ayurveda, Yoga, Naturopathy, Unani, Siddha, and Homeopathy, are sometimes colloquially referred to as "magic remedies" because of their holistic approach to health, centuries-old practices, and the perceived "miraculous" benefits they offer in managing various ailments by the abusers and consortium of evidence based practitioners. This label is often rooted due to popular perception rather than clinical evidence, reflecting how traditional healing systems are seen to work gently and naturally, sometimes achieving results where conventional treatments may fall short. And surprise at most times evoke doubts.
Hence, the term "magic remedies" can be misleading, as AYUSH therapies are grounded in specific philosophical and medicinal principles, and not in supernatural effects. The label sometimes diminishes the scientific foundations and the extensive yet unfortunately undocumented traditional knowledge that backs these systems. The Indian government, through the Ministry of AYUSH, has been promoting scientific research and clinical validation to establish the efficacy of AYUSH therapies more formally.
The term "magic remedies" has not been historically attributed to Ayurvedic medicines by any specific person. However, the phrase gained legal significance in India through the Drugs and Magic Remedies (Objectionable Advertisements) Act, 1954, which regulates advertisements claiming miraculous cures in Allopathic, Ayurvedic, and other traditional medicines to prevent misleading claims. The act used "magic remedies" broadly to address exaggerated claims rather than any official or scientific designation of Ayurvedic or traditional medicines as magical.
Few examples of companies that faced action for violating the Drugs and Magic Remedies (Objectionable Advertisements) Act in India:
(2016): Company XX was scrutinized for promoting certain products with exaggerated health claims, including remedies for diabetes, blood pressure, and joint pain. The Advertising Standards Council of India (ASCI) flagged multiple ads for making unsubstantiated claims, leading to corrective actions.
YY Ayurveda (2021): YY faced legal issues when its advertisements claimed their products could effectively "cure" serious conditions like diabetes and hypertension. The claims were deemed misleading as they lacked scientific validation.
ZZZZ: Known for producing traditional Ayurvedic products, ZZZZ received warnings for advertisements that overstated the effects of some formulations, including claims to cure serious illnesses, violating the Magic Remedies Act.
These cases emphasize the importance of responsible advertising and adherence to regulations in the AYUSH sector.
Let's understand what specific issues that require tightening of administration.
(2020): AAAA promoted Proxozeen as a COVID-19 "cure" without sufficient scientific evidence. The claim received widespread criticism, leading to government intervention. Authorities required AAAA to revise its labeling and advertise Proxozeen as an immunity booster instead of a cure for COVID-19.
YY Ayurveda Diabetes Cure Claim: YY Ayurveda advertised certain products as a permanent cure for diabetes, without clinical support for the claim. This led the ASCI to issue a warning, highlighting that such ads could mislead patients into abandoning conventional treatment for diabetes management.
ZZZZ's Anti-Asthma and Cancer Treatment Claims: ZZZZ faced scrutiny for ads claiming that certain products could treat asthma and even cancer effectively. These advertisements lacked clinical evidence and were flagged for making unverified promises about serious health conditions.
These examples underscore the importance of evidence-based products and communication with adherence to introductory regulatory standards in licensing AYUSH products responsibly.
To avoid being tagged as "magic remedies," companies manufacturing AYUSH products should:
Follow Scientific Validation: Invest in clinical trials and scientific research to validate claims with evidence-based outcomes at product introduction.

Comply with Regulations: Adhere to guidelines set by regulatory authorities like the Drugs and Magic Remedies (Objectionable Advertisements) Act, 1954 and the Ministry of AYUSH, avoiding exaggerated or misleading health claims.

There's no shortcuts to success. Ayurvedic companies should not be excused for proper documentation and compliance at product introduction in local markets under Ministry of AYUSH. The companies continue to struggle with the followings even in 2024:
Traditional Knowledge vs. Modern Documentation: Many Ayurvedic formulations are based on traditional recipes without modern validation processes, which may not align with the rigorous documentation standards required for modern healthcare. This results in gaps when substantiating claims with scientific evidence, as traditional practices focus more on holistic benefits than specific, measurable outcomes.
Cost and Complexity of Research: Conducting clinical trials, maintaining strict quality control, and complying with documentation standards require significant investment. Many smaller companies lack the resources to conduct such trials, leading them to bypass extensive documentation to reduce costs. Govt. should come up with a policy to support if the innovation is unique as per committee validation.

Limited Regulatory Oversight: Although the Ministry of AYUSH has set regulatory guidelines (scope for improvement remains and that's an ongoing process), the enforcement of these standards is inconsistent across states. Inspections and audits are often irregular, and smaller companies can sometimes operate without comprehensive scrutiny. This lack of strict oversight allows some companies to launch products with minimal compliance (this needs a strict stoppage).
Lack of Penalties and Follow-Through: The penalties for non-compliance are often not substantial enough to deter companies from cutting corners. The Ministry of AYUSH and local regulators sometimes lack the resources or coordination to impose effective penalties, especially in cases of misleading advertisements or low-quality production practices.
To strengthen the system, the Ministry of AYUSH will come up with stricter compliance checks and enforcements, increase penalties for violations, and improve collaboration with state-level authorities for more consistent oversight. Additionally, providing grand subsidies or incentives for scientific research on AYUSH formulations could encourage companies to invest in proper documentation and clinical validation.
Global Red Flags is a regular phenomenon by multiple countries across developed world makes manufacturers decategorize products to expand global presence and that's mostly because the complex regulation of traditional medicines across different countries. That is largely due to multiple factors, with no single party solely to blame.
Historical and Cultural Variability: Each country has its own historical and cultural ties to traditional medicine, such as Ayurveda in India, Traditional Chinese Medicine in China, and Native American herbal practices in the U.S. These unique roots have led countries to develop independent regulatory standards based on local needs and beliefs rather than a unified global standard.
Lack of Global Regulatory Authority: The World Health Organization (WHO) provides guidance but lacks the power to enforce regulations globally. As a result, countries are free to adapt WHO recommendations to suit their own regulatory frameworks and health policies, leading to varied levels of regulation.

Labelling Requirements of Ayurvedic Drugs

A label of a product is one of its most notable features that leads to product identification. In the normal parlance, the process of labelling, thus, involves affixing or inscribing[1] information onto a product as a way of imparting a distinct identity to it as well as to make available certain indispensible particulars about it to the public. Therefore, uniformity in the labelling requirements of different categories of products is a necessary corollary. The labelling requirements of Ayurvedic drugs in India are governed by the Drugs and Cosmetics (Act and Rules), 1940.
The Drugs and Cosmetics Act and Rules draw out a clear distinction between a ‘drug’ and an ‘Ayurvedic, Siddha or Unani drug’. The provisions pertaining to labelling of a drug and an Ayurvedic, Siddha or Unani drug are governed by different chapter under the Drugs and Cosmetics Act and Rules. As per section 3 (2) (a) of the Drugs and Cosmetics Act, 1940 (hereinafter referred to as “Act”), an Ayurvedic, Siddha or Unani drug includes all medicines intended for internal or external use for or in the diagnosis, treatment, mitigation or prevention of disease or disorder in human beings or animals, and manufactured exclusively in accordance with the formulae described in the authoritative books of Ayurveda, Siddha and Unani Tibb systems of medicine. The list as to the authoritative books of the Ayurvedic and Siddha Systems is provided under the Schedule I of the Act.
Law relating to the labelling and packing of Ayurvedic (including Siddha) or Unani drugs under Rules
The Rule 161 of the Drugs and Cosmetics Rules, 1945 provides a detailed account of the manner in which the labelling of an Ayurvedic, Siddha or Unani drug must be undertaken. The Rule states that there shall be considerably displayed on the label of the container or package of an Ayurvedic (including Siddha) or Unani drug the following details;
However, the Rule provides that if needed, standardized abbreviations prescribed for parts and form of ingredients in the official Ayurveda, Siddha and Unani Pharmacopoeias and Formularies, may be used on the label. Further, if the list of ingredients contained in the medicine is large and cannot be accommodated on the label, the same is to be printed separately and must be enclosed with the packing with a reference to the same be made on the label.
Further, the container of a medicine for internal use made up ready for the treatment of human ailments if it is made up from a substance specified in Schedule E (1) should be labelled conspicuously with the words ‘Caution: To be taken under medical supervision’ both in English and Hindi language.
According to Rule 161 (3) the following particulars shall be either printed or written in indelible ink and shall appear in a conspicuous manner on the label of the innermost container of any Ayurvedic (including Siddha) or Unani drug and on any other covering in which the container is packed namely–
A true list of all the ingredients with the botanical names of plant based ingredients along with plant part(s) and form of ingredients, in which, these are used in the manufacture of the drug together with their quantity must be conspicuously displayed on the label of the package of an Ayurvedic (including Siddha) or Unani drug. In addition to this, a reference to the method of preparation as prescribed in the authoritative books specified in the First Schedule of the Act also needs to be mentioned on such Ayurvedic, Siddha or Unani drugs that do not fall in the category of patent or proprietary Ayurveda, Siddha or Unani drugs. In case a drug made for internal use for the treatment of human ailments contains a substance specified in Schedule E(1) of the Act, such drug has to be compulsorily labelled with the words ‘Caution: to be taken under medical supervision’ both in English and Hindi languages in a conspicuous manner.
Along with these, the following must be printed or written in indelible ink in a conspicuous manner on the label of the innermost container of any Ayurvedic (including Siddha) or Unani drug including Patent or Proprietary ones and on any other covering in which the container is packed the name of the drug:
a. statement of the net content in terms of weight, measure or number b. name and address of the manufacturer; c. the manufacturing license number, batch number, date of manufacture and date of expiry of the Ayurvedic (including Siddha) or Unani drug; d. the words “Ayurvedic medicine”or “Siddha medicine” or “Unani medicine” should also be clearly specified; e. if the medicine is for external application, the same needs to be clearly disclosed by mentioning ‘For external use only’ on the label; f. in case of drugs intended for distribution to the medical profession as a free sample, the label should be overprinted with the words ‘Physician’s sample. Not to be sold’.
Pertinently, it is to be noted that any transparent cover or any wrapper-case or other covering used solely for the purpose of packing, transport or delivery is not required to be labelled as per this rule.
Rule 161A provides certain exemptions in labelling and packing provisions for Ayurvedic (including Siddha) and Unani drugs that are to be exported, in as much as such drugs can be adapted to meet the specific requirements of the law of the country importing them.
Consequences of non-compliance in the labelling
It is also apposite to note at this stage that an Ayurvedic, Siddha or Unani drug is considered to be misbranded under Section 33E of the Act if it is not labeled in the prescribed manner.  Any Ayurvedic, Siddha or Unani drug is deemed to be a ‘Misbranded drug’:-
a. if it is so coloured, coated, powdered or polished that damage is concealed, or if it is made to appear of better or greater therapeutic value than it really is; or b. if it is not labelled in the prescribed manner; or c. if its label or container or anything accompanying the drug bears any statement, design or device which makes any false claim for the drug or which is false or misleading in any particular.
The Act under Section 33EEC prohibits the manufacture, sale or distribute a misbranded Ayurvedic, Siddha or Unani drugs. Furthermore, whoever acts in contravention of this mandate is punishable with imprisonment for a term which may extend to one year and with fine which shall not be less than Rs. 20,000/- or three times the value of the drugs confiscated, whichever is more.[2]
Conclusion
Hence, by virtue of the abovementioned provisions of the Act and Rules, it becomes amply clear that errors in the labelling of the Ayurvedic, Siddha or Unani drugs manufactured or labelled may cause the Ayurvedic, Siddha or Unani drugs to be brought in the category of ‘misbranded drug’. Therefore, it becomes pertinent to ensure that labelling on its manufactured products is in consonance with the requirements under the Act and Rules.
