Electron cryo-tomography (cryo-ET) reconstructs electron microscopy images of frozen large molecules, complexes or cells at high-resolution in 3D. Here, an image processing pipeline within the open source software RELION 5 used for such reconstructions readily enables data to go from unprocessed to high-res model building, aiding standardisation and newcomers to the field

We refilled Kiddo's insulin today.

This is what we paid. That secondary "cash" number is what we're GOING to be paying in a couple of months when her insurance ends. That's $767 dollars. There are manufacturer discounts but they often have stipulations that are tricky to meet. The Lantus is a fixed daily dose and will last ~5 months. The Relion is the fast-acting, and that's variable depending on how much she eats, how many high blood sugars she has and how bad they are, etc. And we burn through two of those at a time, since she has to have one at school. Additionally, they do have a shelf life of about one month after they've been brought to room temperature, so even if she doesn't use it up we have to toss it because it's simply no good anymore.
So when I ask for help in paying for their medication, this is what I mean. These are the numbers we're looking at. The only reason we were even able to pay for this is because my parents gave me birthday money, because my partner was just in the hospital for a week so we're facing down a short check AND hospital bills, which is its own whole separate battle.
So yeah. This is why capping the cost of insulin at $35 would be a godsend (even though it should be free). This is why you hear about diabetics rationing their insulin. If you didn't have a practical example of that before, now you do.

So I am going to do this for Edenye (big surprise!) thanks for tagging me @chubbyooo! (also I am very sorry for the delay)

1. She has little to no memory of her mother since she was taken when she was little. Most little things she knows of her were told to her by another slave, Uom, who suspected her sensitivity. ‘Slaves do not get the luxury of knowing their parents.’ - Master Relion when she asked once.
2. She has a hearing disorder affecting her auditory sensory processing. High-pitched noises are extremely painful for her, she cannot filter out background noise, cannot locate noise sources in a room and if there is too much painful noise she can have the beginnings of a meltdown (I am borrowing from my own experiences on this). Wearing hearing aids can help in terms of noise location, but noise-cancelling headphones are her saviours.
3. Her entire back is littered with whip scars. Got her bones broken multiple times during her time in the Ziost academy before her trials.
4. During the trials on Korriban, she barely stayed at the academy to sleep/eat. Being a former slave is not just a danger with the wrong overseer, other acolytes will pick up on these sentiments. Mostly stayed with the various excavation teams around the academy. 
5. Learned a lot about archaeology and Sith history from those stays. Most Sith let her stay as long as she made herself useful. Acquired some basic skills in tool repairing and helping preserve artefacts for transport.
6. She is fairly indiscriminate in the food she likes. Sometimes she forgets to eat and then eats huge amounts in one sitting. Never learned how to cook either. Though, her absolute favourite snack is anything lemon flavoured. She will eat an entire lemon if she could.
7. She enjoys swimming or being generally around bodies of water. It is a source of calming down (it also helps that Marr likes it too so they spend a lot of their rare down times near a lake, ocean etc.)
8. Her most crippling obstacle is her fear related to her visions. What they show, what they mean and when they come. It can leave her incapacitated without a lot of warning. Makes her vulnerable to others and conscious who she is around and where. 
9. Her and Temare (her best friend who will be the Sith Warrior) love to star gaze. Their favourite way to catch up in between missions (and be out of earshot).
10. She is incredibly ticklish. A fact Marr sometimes exploits to have her laugh. Though, proceed with caution. She will kick and punch without discretion.

this past week, i have  1) been waking up around 3:30am because i have trouble staying asleep? trying to embrace this and eating a leisurely breakfast while also having time to go to the gym before lab 2) sent my protein directly into the waste during ion exchange chromatography 3) reached over job440 on relion, with no major improvement in particle quality 4) attempted to read for leisure 5) realized my PI is starting to get worried about my lack of progress (not worried that i’m not being productive, just worried that my efforts haven’t yielded any significant results so far)

On relie d’abord la masse à la borne 8 du composant.
La borne 6 à une résistance R_t = 2200 Ω et un condensateur C_t a la borne 6 de 10 nF. On a déterminé ces valeur par F = 1 / (R_t*C_t) et F = 40 000 Hz.
Nous relions les bornes 4 et 5 a la masse vu qu’elles ne nous sont pas utiles. La sortie 3 nous permet de visualiser le signal sur l’oscilloscope.
La borne 1 (-) et 2(+) sont reliées a un AOP dont leur sortie est reliée a la borne - d’un autre AOP ajouté a la borne + qui représente la borne 7.
La borne 16 est placée sur V_ref dont la tension de sortie est de 5v que nous relions a un potentiomètre lié a la borne 2 afin de faire varier la rapport cyclique du signal voulu. Cette borne 16 est également reliée à un condensateur qui va à la masse.
La borne 15, Vin est alimentée en 15 volts (tension d’entrée).
Nous plaçons les bornes 14 (émetteur B) et 11 (émetteur A) des transistors reliées à la masse.
Les bornes 13 et 12 sont reliées a une résistance R2 = 99 kΩ qui va a l’alimentation 15 volts.

Her core trait is her loyalty. It is her best trait alongside with her compassion. Her worst trait would be her uncertainty. Even before she was freed, Relion would always tell her that she could never be Sith. A slave could never be Sith and not one who had “alien blood” tainting her. The academy did not help much either and Malgus gets involved too late to undo most of the damage. It’s a mess when they interact... and it is going to cost her dearly. Very dearly. 
56. What’s your overall goal with this character? Will they get a happy ending or will they succumb to their faults?
My overall goal? Not one in particular, but I am trying to get a happy ending with her.

À la question de savoir pourquoi nous existons, la science n'en a pas la moindre idée car les questions morales sont en dehors des paramètres scientifiques qui soutiennent que nous sommes juste un arrangement accidentel de molécules. En d'autres termes, nous existons par hasard car nous ne sommes que le produit de notre environnement immédiat. Il n'y aurait alors aucune raison de restreindre l'être humain vu que la mort n'est que néant sans continuité. Par conséquent, nous sommes libres de vivre comme bon nous semble car notre existence est sans but, obligations ou responsabilités.À la question de savoir pourquoi nous existons, la Bible dit que nous sommes là pour des buts bien précis qui ont été établis par un Concepteur Intelligent. Nos vies ne sont ni banales ni sans importance et nous devront répondre des choix que nous avons fait durant notre court séjour sur terre.Si la science ne peut pas expliquer pourquoi nous existons, la Bible par contre nous enseigne 3 choses fondamentales:

, YAHWEH a démontré sa gloire dans Sa Création plus précisément celle de l'humain qu'Il a fait à son image. La seule fois où YAHWEH a utilisé à la fois "image" et "ressemblance" dans la Bible, c’est durant la création de l’être humain et avant qu'Il ne souffle la vie dans les narines de l'homme.
Genèse 1:26 "Puis Dieu dit: Faisons l'homme à notre image, selon notre ressemblance..."
De manière profonde cela signifie que l'humain est façonné à la forme intérieur et extérieure de Dieu lui-même. YAHWEH a donné cette forme unique à l'humain seul et Il lui a aussi accordé de dominer sur toutes autres créatures. Pour ce faire, Il lui a transmit des capacités comme les siennes.
Il a permit à l'homme de communiquer, penser, exprimer des concepts complexes, raisonner, prendre des décisions, planifier, inventer et évaluer des idées.
Exode 35:35 " Il les a remplis d'intelligence, pour exécuter tous les ouvrages de sculpture et d'art, pour broder et tisser les étoffes teintes en bleu, en pourpre, en cramoisi, et le fin lin, pour faire toute espèce de travaux et d'inventions."
Il a imprimé dans le cœur de l'homme, l'oeuvre de la loi qui repose sur ses concepts basiques et complémentaires d'amour et de justice pour que nous puissions avoir le sens de moralité.
Romains 2:15 'Ils montrent que l'oeuvre de la loi est écrite dans leurs cœurs, leur conscience en rendant témoignage, et leurs pensées s'accusant ou se défendant tour à tour."
Il a aussi donné à l'homme l'unique capacité spirituelle à questionner, imaginer et désirer la vie après la mort. Les animaux n'ont pas ces capacités.
Ecclésiaste 3:11 "Il fait toute chose bonne en son temps; même il a mis dans leur cœur la pensée de l'éternité"

, l’être humain est tripartite c'est à dire nous sommes constitués d’un corps, d’une âme et d'un esprit.

La façon la plus simple de comprendre la constitution de l'être humain par rapport au dessein de YAHWEH est la suivante:
Avec nos corps nous avons une affinité avec la nature et nous nous rattachons à l'univers matériel.
Avec nos âmes nous avons une identité de soi et pouvons interagir avec d'autres humains.
Avec nos esprits nous avons la vie et nous relions à YAHWEH et le monde spirituel. ​

la vie n'est pas à propos de nous; c'est à propos de YAHWEH. Il ne nous a crée ni pour combler une solitude quelconque ni pour nourrir son ego ou un désir ardent d'être adoré. I’ve been thinking about what some of my trolls would be like in a fighting game, and hyperfocus won’t let me forget about it, so here’s what I was able to come up with.
Corali Primer
Difficulty: 9 out of 10
She’s a real tough one because he primary strategy is ATTACK ATTACK ATTACK. She gives very little opportunities to counter, and she will combo you like a mother fucker. And she hits HARD. If she gets you in a corner, you’re fucking dead kid. You have to time it damn near perfectly to get openings, and she can take a shit load of damage and still come right at you. And that’s not even taking into account her armblade.
In addition, if her health gets below a certain threshold or you land a particularly good hit, she’ll go into a rage and her attacks become more damaging and she’ll be able to take a lot more hits, though she’ll also become more erratic so you’ll be able to land more hits.
The best strategy is to keep your distance so she can’t combo, but she also has a spear she’ll hurl at you attached to a cable so she can reel you back in if you stay out of range for too long, and she will spam the fuck out of that.
Thiomi Shiaka
Difficulty: 3 out of 10
She’s the easiest fight. Hits from her do minimal damage, and you only need to hit her maybe five times before she’s down for the count. The only obstacle is actually hitting her. Speed is the name of her game, and she’s extremely good at dodging, blocking, and running. She’ll do her damnedest to keep you at a distance and defend. Her fight is less difficult and more annoying.
Best strategy is to corner her and keep her from running away. She’ll still dodge and duck away, but if she can’t run she’ll leave more openings until you can get her.
Shadah Relion
Difficulty: 7 out of 10
Shadah has a lot at her disposal that makes her a difficult and time-consuming fight, and with her time is something you’ll very soon be running low on. Her main weapon is a whip laced with highly toxic poison that causes you to lose more health over time the more it hits you until (If you do really badly) you’re losing health at a constant rate. She also has an electrified whip that does extended damage and temporarily paralyzes you for about three seconds. If you do manage to get close enough that she can’t use her whips, she has a fucking gun that she will constantly fire (And as per video game rules, it has a bottomless magazine).
Despite the fact she has a gun, your best bet for beating her is getting in too close for her to use her whips. The gun does less damage than her whips do, and if you time it right you can get in several hits.
Karell Rohsco
Difficulty: 6 out of 10
Like Thiomi, fighting Karell is more annoying than actually difficult. Not only is he just as fast as her and equally good at dodging, he’s constantly regenerating health. Unlike Thiomi, however, he actually has a decent weapon and is reasonably strong. Getting hit with the nail bat causes a lot of damage, and he will not let up once he manages to land a hit. He regenerates health at twenty health per second; in ten seconds, any damage you’ve done is practically completely healed away.
To beat him, you have to adopt Corali’s strategy and just keep on laying on the hits without giving him a break, which is difficult since he blocks and dodges a lot. Just keep whittling down his health by whatever means you can until he runs out.
Allmah Liftir
Difficulty: 10 out of 10
I won’t lie, Allmah is OP as hell. She has incredibly potent magic, and she’s partially demon and corrupted, which boosts her stats. Using her blood magic, she can take control of you and hurl you around for massive damage, and she can use her magic to create barriers and defend and attack with her prehensile hair (Which she uses to impale or strangle, as well as block attacks). She also steals a lot of health from you to add to her own.
Just like Corali, her fight also has a second phase if you get her to a certain point: If you wear her down enough, she transforms into a demonic version of herself that’s much stronger, has more endurance, and increased defenses. Her attacks get more fierce and do a lot more damage, and her combos get more intense. She can also summon her familiar, another demon, to do added damage.
Best strategy to beat her? I wouldn’t be able to hazard a guess. She’s over powered as all hell.
Varoll Kaydur
Difficulty: 5 out of 10
First of all, shame on you for fighting a child. Varoll’s fight is more frustrating than difficult. Because of her small size, she’s very agile and fast, which makes her difficult to hit. If she bites you -- and she will bite you -- she will latch on and do continuous damage until you manage to dislodge her. She can also use a bow and arrow, though hits from that do minimal damage compared to her bites and scratches. She’s also able to call for her lusus for help during a fight for massive damage.
Although she does a lot of damage, she can’t take many hits. You’re definitely going to want to keep her at a distance because of what her bites can do, and she can only do minimal damage from far away.
