Swine fever is a highly contagious disease among pigs and poses a severe threat with a very high mortality rate. It, however, does not affect humans.

Swine Fever Vaccine: A Potential Lifesaver for Pigs Around the World
Origins and Spread of Swine Fever African swine fever virus (ASFV) is a large DNA virus that was first identified in the early 20th century in Kenya. It is highly contagious among pigs and wild boar. The disease has since spread to many sub-Saharan African countries as well as parts of Europe and Asia. In recent years, ASFV has caused severe outbreaks and significant economic losses in Eastern Europe and China. Its ability to survive for long periods in uncooked pork products has aided its transmission over long distances. With globalization and increased movement of people and goods, the threat of ASFV spread to other pig-raising regions remains high. Challenges of Developing an Effective Vaccine Developing a vaccine against Swine Fever Vaccine has proven exceptionally challenging for several reasons. First, ASFV is a very large and complex virus unlike other common livestock pathogens. This complex structure makes it difficult for the immune system to generate protective antibodies against it. Second, there are no cell culture systems or small animal models available for consistent vaccine testing. Researchers must rely on infected pig trials for efficacy evaluations. These limitations have hindered traditional vaccine approaches using killed or attenuated virus versions. Novel vaccine strategies targeting specific ASFV genes are now being explored but require extensive safety testing before use in massive pig populations. Considerable resources and time are needed for vaccine development, testing, and regulatory approval. Promising New Vaccine Candidates Emerging In recent years, advances in viral genomics and gene-editing technologies have provided new opportunities for ASFV vaccine research. Chinese and European scientists have identified particular ASFV genes responsible for virulence. By deleting these genes, they have generated attenuated live vaccine candidates that induce protective immunity without disease in pig trials. Separately, researchers at the University of Florida developed a subunit vaccine containing a key ASFV protein formulated with an oil-based adjuvant. In preliminary tests, this synthetic vaccine demonstrated the ability to protect 100% of immunized pigs against lethal challenge. While more studies are still required, these modified live and subunit vaccine approaches show the most promising results to date. Large-Scale Production Challenges Ahead Even if an effective swine fever vaccine is proven through extensive field trials, major challenges remain for its widespread adoption and use in practice. Mass production under stringent quality control will need to meet the enormous demand from huge swine industries, especially in China and Europe. Ensuring proper cold chain management during transport and storage will be critically important for temperature-sensitive live vaccines. Achieving widespread coverage through multiple immunizations will pose logistical difficulties. Farmers may be reluctant to adopt an unproven new vaccine in places currently unaffected by the disease. Establishing an affordable pricing structure acceptable to different countries and farm sizes will also take rigorous economic modeling and negotiation with manufacturers. Overcoming these challenges will likely require unprecedented international cooperation between researchers, industry partners, and regulatory bodies. Widespread Immunization - The Ultimate Goal Ideally, an effective and affordable Swine Fever Vaccine distributed on a global scale could significantly curb the devastating impacts of this swine fever. With sufficient population immunity established through immunization programs, outbreaks may become more localized and amenable to control through quarantine and culling practices.

African swine fever (ASF) is an acute and severe disease transmitted among domestic pigs and wild boars. This disease is notorious for its high mortality rate and has caused great losses to the world’s pig industry in the past few years. After infection, pigs can develop symptoms such as high fever, inflammation, and acute hemorrhage, finally leading to death. African swine fever virus (ASFV) is the causal agent of ASF; it is a large DNA virus with 150–200 genes. Elucidating the functions of each gene could provide insightful information for developing prevention and control methods. Herein, to investigate the function of I267L, porcine alveolar macrophages (PAMs) infected with an I267L-deleted ASFV strain (named ∆I267L) and wild-type ASFV for 18 h and 36 h were taken for transcriptome sequencing (#RNA-seq). The most distinct different gene that appeared at both 18 hpi (hours post-infection) and 36 hpi was F3; it is the key link between inflammation and coagulation cascades. KEGG analysis (Kyoto encyclopedia of genes and genomes analysis) revealed the complement and coagulation cascades were also significantly affected at 18 hpi. Genes associated with the immune response were also highly enriched with the deletion of I267L. #RNA-seq results were validated through RT-qPCR. Further experiments confirmed that ASFV infection could suppress the induction of F3 through TNF-α, while I267L deletion partially impaired this suppression. These results suggest that I267L is a pathogenicity-associated gene that modulates the hemorrhages of ASF by suppressing F3 expression. The factors that influence the pathogenicity of African swine fever (ASF) are still poorly understood, and the host’s immune response has been indicated as crucial. Although an increasing number of studies have shown that gut microbiota can control the progression of diseases caused by viral infections, it has not been characterized how the ASF virus (ASFV) changes a pig’s gut microbiome. This study analyzed the dynamic changes in the intestinal microbiome of pigs experimentally infected with the high-virulence ASFV genotype II strain (N = 4) or mock strain (N = 3). Daily fecal samples were collected from the pigs and distributed into the four phases (before infection, primary phase, clinical phase, and terminal phase) of ASF based on the individual clinical features of the pigs. The total DNA was extracted and the V4 region of the 16 s #rRNA gene was amplified and sequenced on the Illumina platform. Richness indices (ACE and Chao1) were significantly decreased in the terminal phase of ASF infection. The relative abundances of short-chain-fatty-acids-producing bacteria, such as Ruminococcaceae, Roseburia, and Blautia, were decreased during ASFV infection. On the other hand, the abundance of Proteobacteria and Spirochaetes increased. Furthermore, predicted functional analysis using PICRUSt resulted in a significantly reduced abundance of 15 immune-related pathways in the ASFV-infected pigs. MANILA – Department of Agriculture (DA) Secretary William Dar on Tuesday announced that starting next month, the rapid test kits developed by the Central Luzon State University (CLSU) for African swine fever (ASF) would be mass-produced.
“Mayroon nang resulta ang pinondohan nating development of rapid test kits in Central Luzon State University (We finally got results for the development of rapid test kits in CLSU, which we funded). This February, we will be mass-producing it for our monitoring and surveillance,” Dar said in a virtual presser.
He said hogs would be tested even without showing symptoms of ASF.
“Mas makikinabang ang hog raisers (Hog raisers will benefit from the rapid test kits),” Dar said, adding that some of the industry players have opted not to raise their average number of hogs due to the threat of ASF, resulting in the shortage of pork supply in the market.
He said the rapid test kits developed by the state university would hasten the surveillance and monitoring operations of the DA’s Bureau of Animal and Industry (BAI) and ASF-affected local government units (LGUs).
The rapid test kits will render results within an hour or less and will be much cheaper, the DA said.
Dar said his department would spend some PHP80 million for the development and mass production of Filipino-made rapid test kits.
“We are allocating an initial PHP80 million through the DA - National Livestock Program and BAI to mass-produce and distribute to LGUs the test kit, called ‘ASFV Nanogold Biosensor’,” he said.
The budget for the Bantay ASF sa Barangay (BABay ASF) program will be provided by the DA -Bureau of Agricultural Research to augment the mass production of test kits in partnership with other interested private firms and state universities and colleges.
“We can even have these test kits on standby at the port of entries for a quick sampling of the meat products entering the country,” Dar added.
CLSU-developed test kits
Dr. Clarissa Yvonne J. Domingo, professor of CLSU’s College of Veterinary Science, developed the rapid test kits in collaboration with the DA-BAI, noting that these are easy to use and can differentiate the ASF virus from hog cholera and other swine-related viruses.
The DA said the rapid test kits use nucleic acid-based tests.
They have a built-in DNA extraction and molecular amplification process that uses primers or markers, whose gene sequence was designed from the P72 gene of the ASF virus isolated from the province of Rizal.
“It can detect the presence of ASF even through surface swabbing of pig barns and delivery trucks, saliva, and nasal swabs, feces, water, semen, feeds, aspirated whole blood, or blood-soaked swabs, and even domestic flies,” the department said.
The ASF test kit, which comes with all the consumables, costs PHP3,500, good for 10 samples at PHP350 per sample.
To ensure efficacy, the test kits were tested on surface swabs taken from nipple drinkers, walls, railings, floors, and pig ways of barns; water samples of farms; ASF contaminated floor sweepings and feeds; saliva, nasal swabs, and feces of pigs; processed, fresh, and canned meat that were confiscated at the airport, and domestic flies that touched dead carcasses.
