we started "running" m1 and by running i mean we make it to bonzo and then die bc it's giving more class and cata exp than we get for winning against livid. but we've tried to kill bonzo anyways bc What If We Get Lucky. we can survive for a while but eventually something kills us both at the same time
but the long survival has led to some great stats

no but it’s just That Time Of Year Again where because AO3, the website that is ranked globally in the top 2000 websites out of an active ~1.85B sites, and maintains server stability and growing site operability 100% through user funding and collaborative generous volunteer time, while defending the legal right for fanworks to exist, is doing it bi-annual donations drive.
there's the usual 'ao3 exceeded their goal why can't they donate the rest to charity' discourse floating around which others have written about much more clearly and concisely than i can, but for me, this particular round of reaction has been pretty next-level vitriolic in that i've had to unfollow several people in the past day for reblogging posts that have explicitly said that people donating to ao3 during a global pandemic instead of to any number of donation requests crossing their dash should kill themselves. the less vitriolic version of this being that people who do so are 'going to hell', but the call for suicide in giant, bold, headline letters was the one that was the most frequent and obviously visible on my dash.
generally i can let most criticism of ao3 slide by as a difference of opinion - some people do have genuine good-faith criticism of wanting more features such as blocking and muting added to the site to protect users from harassment. and yeah. absolutely. 100% get that. also 100% understand how things like that, which seem simple, have incredible back-end implications that take a huge amount of time to code for and ensure stability for, especially with the collaborative nature of the website. i also think that ao3 should be more communicative about the process of developing these features to be as transparent with the process as they are with their finances.
even the more bad-faith criticisms i can usually just shrug at, maybe get a flicker of passing disbelief and annoyance, and scroll on.
but i'm sorry this co-option of genuine oppression and suffering that's been systematically concentrated under the capitalist handling of the COVID-19 pandemic, while calling for individual guilt over that system and goading people to self-harm for donating to ao3 instead of every donation drive?
that kind of takes this out of the bad-faith category and into the explicitly malicious territory for me.
as i've said in other reblogs and tags on the subject, the fact that these people aren't telling people to kill themselves for using their spare money to keep up their netflix, spotify, amazon, hulu, disney+, etc. subscriptions, instead of donating to every personal post on their dash – companies which literally cause active global harm and oppression, abuse their workers and spearhead anti-worker legislation and campaigns, bend to incorporate imperialist and anti-worker messages into their media products, and contribute to capitalist actual neo-colonialization.......kind of puts this whole social justice framework of these calls for people to self-harm and feel immense personal guilt for the crusade-determined purity of their individual choices, into relief as the complete empty garbage that it is.
this isn't about social justice. it's about co-opting and predating on feelings of pain, anxiety, and empathy that people have due to rising systematic inequality, into the guilt-mongering, reactionary, conservative purity culture crusade against a site that was founded on being explicitly against commercialization, monetization, and corporate determinism over a creative resource, and the policies that keep it that way.
to see the propaganda technique of making systematic injustice seem dependent on the individual, viciously weaponized by ppl on this site for a conservative purity campaign, with explicit calls for self-harm and suicide and claims of deep moral deficiency, is just....yeah it's absolutely intentionally malicious and noxious, and an escalation that i have absolutely zero tolerance for.

Riding Nektar’s wave: cytokine immunotherapies in oncology
Throughout the course of recent months, there has been tremendous attention on cytokines in oncology after Bristol-Myers Squibb (BMS) paid a dazzling $1.85B for rights to Nektar's lead investigational immuno-stimulatory asset, NKTR-214.
NKTR-214 is an engineered (pegylated) cytokine targeting the interleukin-2 (IL-2) receptor beta subunit, which can possibly turn "cold" tumors — tumors inadequately infiltrated with immune cells, and thus less responsive to immunotherapies — immunogenic. On account of this feature, IL-2-based therapies are touted as one of the most promising combinatorial methodologies with immune checkpoint barricade.
The sector is bustling with activity and various high-profile assets are in development. For instance, Immunservice's inhaled biomimetic IL-2 Pulmoleukin is in Phase II for lung metastases in renal cell carcinoma.
Likewise, Roche's RG7461, an IL-2 variant (IL-2v) linked to an anti-fibroblast activation protein (FAP) antibody, is in Phase II in combination with in-house modified death-ligand 1 (PD-L1) inhibitor Tecentriq (atezolizumab) for various solid tumors.
The biggest study is an ongoing pivotal trial testing Nektar's asset with BMS's Opdivo (nivolumab), a customized cell death protein-1 (PD-1) inhibitor, in 250 patients with various solid malignancies, as part of a collaboration wrote in 2016 between the two companies.
Although IL-2-based therapeutics are ahead with respect to clinical development, various other interleukin 1 receptor like 2 drugs development market projects in development are gaining traction, energized by the interest displayed in the sector following Nektar's prosperity.
Treading closely following IL-2 are IL-10 and IL-12, which have seen rekindled interest in oncology despite being previously tarnished by systemic toxicity, pharmacokinetic limitations, and an extremely thin therapeutic index.
A few different delivery mechanisms or plasmid vector technologies currently aim to address the limitations of these methodologies and have previously advanced into various oncology projects.
Armo Biosciences is in front of the load with pegilodecakin (AM0010), is a long-acting type of recombinant human IL-10. Attaching IL-10 to polyethylene glycol (PEG) drags out the presence of the particle in the circulation and within a tumor.
IL-10 stimulates the expansion and cytotoxic activity of CD8+ T-cells, which directly connect with and kill tumor cells. The readout of pegilodecakin's pivotal trial in pancreatic adenocarcinoma will be one of the main biotech events during this year's subsequent quarter.

Bain Capital Ventures has a fresh $1B to invest in startups
Bain Capital Ventures (BCV), the venture capital arm of the private equity giant Bain Capital, has brought in $1 billion to invest in startups across industries and stages.
The capital has been spread across three funds: $650 million for its eighth flagship vehicle, $250 million for a co-investment fund focused on growth-stage investments and an additional $100 million directly from the partners at Bain Capital, which will be deployed on every investment out of the latest fund.
Founded in 1984, BCV is known for its investments in LinkedIn, Rent the Runway, SurveyMonkey, SendGrid and DocuSign. Initially, the firm was more of a growth-stage investor, though it’s warmed to early-stage companies, making a total of 106 early-stage investments, 52 of which were at the seed stage, since 2013. The firm says that’s triple its total volume of seed and Series A investments from previous years, thanks to the members of its West Coast investment team, who are responsible for striking a majority of BCV’s early-stage bets.
The firm opened its first office in San Francisco in 2016. BCV also has offices in New York, Boston and Palo Alto.
BCV’s seventh flagship fund closed on $600 million in 2016; its latest core vehicle is even larger. Ajay Agarwal, one of BCV’s eight managing directors since 2003, said that’s because of the bull market and lack of liquidity in venture capital, which has caused several VC funds to raise larger and larger pools of capital. Just look at Thrive Capital or GGV Capital, for example, VC funds that have surpassed $1 billion with recent fund closes.
Bain Capital Ventures is headquartered in San Francisco.
“We, like many of our peers, have far more demand for our funds than we have the capacity for,” Agarwal told TechCrunch. “Companies are taking longer to go public and therefore require more capital, but we don’t want the fund size to get too large.”
“It’s certainly an environment where lots and lots of capital is coming into the market … It puts even more of an emphasis on firms that have a real competitive differentiation — something unique to offer entrepreneurs.”
BCV invests in 12 to 14 companies per year, not including its seed-stage portfolio companies. Recent investments include Ribbon, a real estate tech startup that brought in $225 million in October; Basis, a cryptocurrency startup that raised $133 million in April; and messaging startup Attentive, which closed a $13 million round in February.
With the three additional funds, BCV has $4.9 billion in assets under management.
