COVID vaccine efficacy against severe illness just under 50%, per early estimates from 2023 - Published Aug 20, 2024
Two European observational studies estimate the vaccine effectiveness (VE) of the COVID-19 XBB.1.5 vaccine approved in fall 2023 against hospitalization, the first one finding 49% overall VE in adults, and one showing good protection—but uneven uptake—among pregnant women.
Steep declines in VE after 1 month For the first study, published in Influenza and Other Respiratory Viruses, researchers with the European Hospital Vaccine Effectiveness Group conducted a test-negative case-control study on the VE against hospitalization of the XBB.1.5 COVID-19 vaccine in the first months of its rollout from October 2023 to January 2024.
The XBB.1.5-like+F456L variant was dominant in Europe from fall to mid-December 2023, when BA.2.86 supplanted it.
The participants were 622 adults hospitalized for COVID-19 and 3,457 control patients admitted for a different severe respiratory infection to 41 hospitals at 7 sites participating in the Vaccine Effectiveness, Burden and Impact Studies (VEBIS) study. In total, 27% of COVID-19 patients and 44% of controls had been vaccinated after fall 2023, most with the Pfizer/BioNTech vaccine.
The median time between vaccination and symptom onset was 58 days for COVID-19 patients and 52 days for controls.
Overall VE was 49% against hospitalization (range, 69% at 14 to 29 days to 40% at 60 to 105 days postvaccination), although confidence intervals overlapped for all estimates, indicating uncertainty. VE was over 70% in adults ages 65 years and older up to 1 month after vaccination. VE in patients ages 80 and older declined from 76% in the first 29 days to 39% after 60 days.
The study authors noted that their VE estimates are lower than those of three previous European studies but in line with a fourth, except for a greater decline in VE with increasing time since vaccination (TSV), proposing that the differences could be attributed to different TSVs bands.
"The findings of our study suggest that the adapted COVID-19 XBB.1.5 vaccines provided protection against hospitalisation in the first 3.5 months post vaccination, by reducing the risk of COVID-19 hospitalisation by approximately half among the vaccinated individuals," they concluded.
Uptake highest in those with chronic conditions The second case-control study, published in Vaccine, included all 47,046 pregnancies ending from June 2021 to August 2022 among 39,213 women seen at Northwest London general practices. In 57% of pregnancies, the mother had received at least one dose of COVID-19 vaccine during their pregnancy, with 91% of them receiving a second dose.
Pregnant women, the study authors noted, are at higher risk of COVID-19 complications such as hospitalization, intensive care unit admission, invasive mechanical ventilation, and death.
"They are also at increased risk of pregnancy-related complications such as preeclampsia and emergency cesarean delivery, and their infants are at higher risk of being preterm or stillborn," they wrote, adding that uptake has been suboptimal.
Most pregnancies (80%) were among women aged 25 to 39 years, those who were White (39%) or Asian (34%), and those living in areas in the second (35%) or third (29%) most deprived quintiles of socioeconomic deprivation. The most common of the five risk factors studied was asthma (9.9%), and chronic heart disease was the least common (0.68%).
A total of 180 women were hospitalized for COVID-19. Admission for COVID-19 was much less likely among vaccinated women than among the unvaccinated. A conditional logistic regression model suggested a five-fold decrease in the chances of COVID-19 hospitalization in vaccinated women, compared with their unvaccinated peers (odds ratio [OR], 0.22).
Vaccine uptake was lowest in women ages 18 to 24 years (33%), Black women compared with White (37%; OR, 0.55), and those in socioeconomically deprived areas (50%). Coverage was higher among women with chronic conditions, with the greatest uptake among those with chronic heart disease (75% for first dose, 94% for second).
Among women with chronic conditions, women with asthma (OR, 1.20), chronic heart disease (OR, 2.34), diabetes (OR, 1.54), and morbid obesity (OR, 1.15) had significantly higher odds of receiving at least one vaccine dose during pregnancy, compared with those without the condition.
The probability of receiving a first dose of the vaccine was nearly six times higher in the 45- to 49-year) age group than in those ages 25 to 29, compared with 18- to 24-year-olds.
Second-dose patterns were similar. Hospitalized women were much less likely to have been vaccinated than those not hospitalized (22% vs 57%; OR, 0.22). "This association reinforces evidence on the real-world effectiveness of COVID-19 vaccines in preventing severe disease and consequent hospital admission among pregnant women," the researchers wrote.
"COVID-19 vaccine uptake among pregnant women is suboptimal, particularly in younger women, Black women, and women in more deprived areas," they added. "Interventions should focus on increasing uptake in these groups to improve health outcomes and reduce health inequalities. Future vaccination programmes should engage pregnant women earlier and communicate with them clearly and carefully."

Generally its a good mod, I really do enjoy pups+ I have like, minor issues? but thats really it. I'll put a read more and a more in depth look.
Pups+ is a mod that adds new versions of pups to your game based on some of the playable campaigns, this includes abilities, personalities, and food needs. Yes I like it, it has its issues but I do like it. It's a great mod. /gen
Tundrapup -Saint variant. -Gives the player saint tongue. -Unable to throw spears. -Can't store food. Aquaticpup -Rivulet variant. -Increased speed and agility. -Fast in water. -Gives a boost underwater. Hunterpup -Hunter variant. -Able to throw spears at Survivor strength. -Carnivorous, eats large corpses. -Taller then an average pup.
Rotundpup -Gourmand variant. -Highest food requirement, eats basically anything. -Throws spears at Hunter strength, exhausts very easily. -Large, slams on creatures, carrying one will let you do the same. -Spits up items when given a disliked food. The basic spawn chance for these pups is 18% for a Aquaticpup 11% for a Tundrapup 25% for a Hunterpup 16% for a Rotundpup 30% for a regular pup
You can adjust these in the remix menu settings. PROS: -This mod allows for pups to be waaaaay more fun and useful. -The variety breaks up gameplay, its nice, each pup has its own look and food requirements.

-The mod is compatible with multiple different mods, Slugpup Safari, Rainbow pups (eeeeeh), Dress My Slugcat, and Dev Console. -Also allows for slugpups to store items! -Also works with Genetic Slugpups! (for everyone but survivor and monk, lol) CONS: -Because of the way the color generation works, and how its linked to personalities, a lot of variants will be the same color scheme, so yes it works with Rainbow Pups, but, variants tend to look the same.

As you can see, a lot of the colors match up. even if you do adjust the percentage typically they'll still group up into colors- why is there a routund pup in the tundra pup cage wait. -Hunter pup is almost indistinguishable from a regular pup, technically they are meant to be "taller" but, typically the only way to tell is 1. the color (see con above) or two, throwing a corpse at them to see if they eat it. /lh

The left pup is the regular pup, the right is the hunter pup. -This would be fine if you could see the food pip bar, before hibernating, but what ends up happening es a lot of time my hunter pups starve for a cycle, because they will still eat plants. -Hunter pups I find typically very useless to me, and more frustrating, if you're good at combat, they tend to just get in the way. -Rotund pups can be INCREDIBLY frustrating, but that's just because of the exhaustion mechanic when they're on your back. I think if it was a little less often it'd be better.
All in all I DO enjoy this mod, and do use them in my own playthrus, I do recommend it, but highly suggest adjusting the ratios on spawns.

The 1957-'62 Autobianchi Bianchina Trasformabile Offered Big Style In A Small Package
In Italy, the size and cost of an item traditionally have little bearing on the quality of its design; a consumer product can be comparably inexpensive and small, and still exude panache. The Autobianchi Bianchina Trasformabile exemplified this, as a chic, once-relatively-affordable interpretation of Fiat’s tiny Nuovo 500 that’s now in high demand.
The populist 500 had been the basis for countless variants over the decades, both coachbuilt and homemade. The introduction of the all-new postwar Fiat was quickly followed by that of a related upmarket variant representing a joint venture between that automaker, Pirelli, and the venerable firm Bianchi; the result was Autobianchi, and its first product was the Bianchina Trasformabile. Advertised as La piccola vettura di grande classe (“The small car of great class”), the Bianchina was based on the 500 platform and shared its mechanicals, but cloaked everything in very different styling credited to Fabio Luigi Rapi, head of Fiat’s special coachwork department.
Trasformabile (“transformable”) was both the name and functional description of the 117.5-inch-long car’s open-top body style. It echoed that of the prewar Ford Model A-400 Convertible Sedan and postwar Nash Rambler Convertible Landau, having a soft top that slid back on tracks to fold atop the rear deck. This design was enjoying a renaissance, similarly used in the contemporary Vespa 400 microcar. Compared to the Vespa, the Bianchina was more delicate in form and detailing, and better equipped; it had roll-down windows instead of sliding panes in its rear-hinged doors, a higher quantity and quality of interior trimmings, and greater displacement and an additional gear enhancing motivation.
Autobianchi’s two-seater — its thinly padded rear bench was for packages or small, agreeable children — was a bit more powerful than its French-built competitor, too, with a four-stroke, 479-cc OHV two-cylinder engine mounted aft of the independently suspended, driven rear wheels. The first 50 examples of that air-cooled inline-twin offered 13 hp and 20 lb-ft of torque. Two more horses were on tap for the regular-production 1957 First Series A model, and 1958-’59s (First Series B) made 16.5 hp. Starting with 1959’s Special First Series, this car’s engine was upsized to 499.5-cc; 17.5 hp and 26 lb-ft were its standard output. The sporty Special variant used a Weber 26 IMB carburetor and 8.6:1 compression ratio to make 21 hp and 25 lb-ft of torque; this was said to be capable of 110 km/h or 68 mph. On all, drum brakes hid behind 12-inch-diameter wheels mounting Pirelli tires.
Italy’s Bianchina Classic Club (bianchinaclub.com) suggests Trasformabile production totaled 35,500 units. The club website has a reasonably detailed history of this model, spelling out its evolution into Special First Series (1959), Second Series (1959-’60), the 1960 Special and 140 B export models (the latter, our feature car, sold new in the U.S.), and the Third Series D and Special D Second Series of 1961-’62. This organization also celebrates other Bianchina variants: the Quattroposti sedan, fully open Cabriolet, Panoramica hatchback and panel van, and Furgoncino truck. Decades on, the attractive looks and clever convertibility of the Trasformabile would inspire a Japanese tribute in the form of the 1991 Nissan Figaro, and in the last year, two restored Trasformabiles have sold at U.S. auctions for almost $90,000.

Reexaming the Performance of Passive Options Strategies
Reexaming the Performance of Passive Options Strategies More than 40 years ago, Merton et al. published two papers [1,2] examining the performance of passive options strategies. They concluded that these strategies outperformed the traditional buy-and-hold approach. At the time of their studies, options data was not widely available, so they used historical volatility to calculate options prices. Merton et al. conducted their research by simulating the impact of options on two portfolios: a broad market proxy of 136 equities and the Dow Jones 30 index. Using a twelve-year period, the backtest incorporated historical volatility and applied the Black–Scholes-Merton model to price the options. Since then, the options market has become highly liquid, with significant structural changes. A recent article [3] reexamines the strategies studied by Merton et al., along with additional strategies, using actual options data from the period 2012 to 2023. The strategies studied include Call-Write strategies (with seven variants), Put-Write strategies (with two variants), and the Protective Put (PPUT) strategy. The authors pointed out, The recent review shows that the original option strategies recommended by Merton et al. no longer provide a favorable return-to-risk ratio. It is likely that these returns were illusory, driven by their assumptions. After all, they were based on a simulation. Recent data demonstrate that simple options strategies no longer add value to a portfolio or an index. However, our research shows that three well-known and somewhat dynamic option strategies have outperformed the S&P 500 Index on a return-to-risk basis. Furthermore, we find that the favorable performance observed in previous studies can be revitalized by incorporating simple signals of the market regime in their construction. An interesting finding of this study is that PPUT consistently outperforms the S&P 500 Index on a return-to-risk basis. Even more astonishing is that by adding the simple logic of avoiding puts after a one-standard-deviation draw down, it outperforms the index on a return basis with significantly lower risk. A key reason for the PPUT index and the logic-based PPUT strategy outperformance may be that as more firms implement covered call strategies, they inadvertently reduce implied volatility levels, underpricing the risk in the tails of the distribution. This hypothesis needs to be tested with a larger set of indices. In short, none of the simple options strategies have outperformed the S&P 500. Interestingly, the PPUT strategy outperforms the buy-and-hold approach on a risk-adjusted basis, and the VIX is shown to be an effective regime filter. IJMS, Vol. 25, Pages 6629: An Efficient Method for Vault Nanoparticle Conjugation with Finely Adjustable Amounts of Antibodies and Small Molecules
Vaults are eukaryotic ribonucleoproteins consisting of 78 copies of the major vault protein (MVP), which assemble into a nanoparticle with an about 60 nm volume-based size, enclosing other proteins and #RNAs. Regardless of their physiological role(s), vaults represent ideal, natural hollow nanoparticles, which are produced by the assembly of the sole MVP. Here, we have expressed in Komagataella phaffi and purified an MVP variant carrying a C-terminal Z peptide (vault-Z), which can tightly bind an antibody&rsquo;s Fc portion, in view of targeted delivery. Via surface plasmon resonance analysis, we could determine a 2.5 nM affinity to the monoclonal antibody Trastuzumab (Tz)/vault-Z 1:1 interaction. Then, we characterized the in-solution interaction via co-incubation, ultracentrifugation, and analysis of the pelleted proteins. This showed virtually irreversible binding up to an at least 10:1 Tz/vault-Z ratio. As a proof of concept, we labeled the Fc portion of Tz with a fluorophore and conjugated it with the nanoparticle, along with either Tz or Cetuximab, another monoclonal antibody. Thus, we could demonstrate antibody-dependent, selective uptake by the SKBR3 and MDA-MB 231 breast #cancer cell lines. These investigations provide a novel, flexible technological platform that significantly extends vault-Z&rsquo;s applications, in that it can be stably conjugated with finely adjusted amounts of antibodies as well as of other molecules, such as fluorophores, cell-targeting peptides, or drugs, using the Fc portion as a scaffold. https://www.mdpi.com/1422-0067/25/12/6629?utm_source=dlvr.it&utm_medium=tumblr

Considerations for this information: This is per one infection, and it's very difficult to get long covid diagnosed (Some of the studies analyzed were only looking at medical records), meaning paring confirmed diagnosis to reduction is inherently a bit flawed. A new meta-analysis of studies involving more than 14 million people published in the Journal of Infection shows that COVID-19 vaccination is associated with a lower risk of developing long COVID, with two doses reducing the odds by 24% and one dose reducing the odds by 15%.
In the 25 studies published up to February 2024 that were included for analysis, long COVID was defined as persistent symptoms at 3 months or beyond, and all studies compared long-COVID symptoms between vaccinated and unvaccinated groups, with the number of doses received by participants specified. All studies included were observational trials and included in total 14,128,260 participants.
Best protection from 2 vaccine doses In 15 studies, two doses of COVID-19 vaccine before infection resulted in reduced odds of long COVID by 24% (odds ratio [OR], 0.76; 95% confidence interval [CI], 0.65 to 0.89). In 5 studies, one-dose post-COVID vaccination was associated with a 15% reduced odds of long COVID (OR, 0.85; 95% CI, 0.73 to 0.98).
