What do you think about all these "big roles" like Captain America and 50 Shades of Grey that Jensen supposedly turned down to do Supernatural?

Eh, Jensen didn’t turn down those two roles because he was never offered.   Like every actor in Hollywood, Jensen auditioned for Capt America and may have even gotten on the short list, but he was never offered.  The confusion  was because Chris Evans played a character name “Jensen” in the movie The Losers.  

Do you think they would just offer Jensen a lead to a potential Grey franchise without auditioning?  One of 50s producer is a friend of Jensen’s and asked if he wanted to audition and Jensen said nope.

No TV actor in their right mind would miss out on a chance to be in a tentpole movie so don’t believe the AA’s fanlore that Jensen turned down lead roles or high profiled supporting roles in huge tentpole movies because he’s loyal to a middling-rated show on an almost netork.  It took Jensen 11 years to say he actually likes his character, Dean Winchester.  The funniest theory I read from the AAs is WB is planning a secret Marval project for Jensen but it’s secret because they don’t want to unleash the power of his fans just yet.  Yeah, okay how is that even supposed to make sense?  Has the AAs even seen the comic-book fandom?  Absolutely brutal, comic fanboys will eat AAs alive.
