Abstract Long noncoding RNAs (lncRNAs) have recently been identified as crucial biomarkers of papillary thyroid cancer (PTC). In this study, we aimed to investigate the biological function and potential clinical role of lncRNA ENST00000494165 in PTC. From January 2019 to December 2019, 226 PTC patients who underwent preoperative thyroid US-fine needle aspiration biopsy(US-FNAB) and confirmed by postoperative pathology were enrolled at our hospital. Thyroid tissues were collected from FNAB samples and stored in the refrigerator at –80°C. qRT-PCR (quantitative reverse transcriptase-polymerase chain reaction) analysis was performed to detect the relative expression level of ENST00000494165. CCK-8 (Cell Counting Kit-8) and colony formation assay were performed to detect the cell proliferation ability. Cell migration and invasion abilities were evaluated by transwell and scratch assay. We also evaluated the relation between the expression level of ENST00000494165 and the clinicopathological features of PTC. Functional assays demonstrated that the cell proliferation, migration and invasion abilities were all promoted in PTC cell lines when ENST00000494165 expression was overexpressed. The expression of ENST00000494165 in the lymph node metastasis group was significantly higher than that in non-lymph node metastasis group. High level expression of ENST00000494165 was significantly associated with lymph node metastasis of PTC (P < 0.001). The overexpression of ENST00000494165 promoted the progression, migration and invasion abilities of PTC and was significantly correlated with lymph node metastasis in PTC. ENST00000494165 could act as a possible promoter gene and a potential biomarker for PTC.

My Cancer Story #1
When I was 12 (2017) late one night I noticed a lump on my neck that wasn’t there before. My first sleepy thought was that it was an adam’s apple... so I googled to see if girls had one, and if it was meant to be on the side of the neck. Eventually I deemed my neck lump completely fine and healthy then forgot about it until this year (2020). One afternoon I was laughing hard at something my mum said and she noticed the huge lump on my neck for the first time since you can’t really see it unless I tilt my head back like I did just then. We went to the doctors to get it checked out. Eventually that lead to getting an ultrasound, then an ultrasound guided fine needle biopsy.
That biopsy happened on Wednesday last week, and we weren’t expecting the results until Monday (Tomorrow) but we got them sooner than expected.
On Friday after school, I get home and my mum told me to sit down on the couch. “I have something to tell you. The doctors called about your results.” She told me. She then proceeded to tell me that I have Papillary Thyroid Carcinoma or more commonly known as Papillary Thyroid Cancer.
The doctors are already organising to get my whole thyroid surgically removed and we should find out a surgery date by the end of the week.
I have unknowingly had cancer for three years... which means it is likely that it has spread to my lymph nodes now.
The most surprising part about this to me is the fact that this is a cancer that is most common in 30-50 year olds, not 12-15 year olds.
Honestly I wasn’t even surprised when my mum told me I have cancer, I guessed it about a month ago. I just don’t care anymore, but at least this cancer has a very high survival rate from my research.
Also I have named the lump on my neck! Her name is Gertrude.
Tumblr media
^^ Gertrude, I gave her a face. She is as big as the circle for her head, so about the size of a golfball. She is a toxic bitch and is no longer welcome in my body.
I know you can’t really see the lump, but it is there and it hurts.

Happy #Friday everyone! Time for #fridayintroductions ❤️ This week I thought I'd share some fun facts about me! 1. I've worked at @starbucks for 4+ years (off and on again). I did the Coffee Masters program and I am a certified #coffeemaster ...I also have my own black apron for work 😍 2. 99.9999% of my wardrobe consists of black, grey, or white clothing. I rarely wear colors. 3. I recently had a heart procedure done to correct issues I've had with my heart for the past 10 years. Last year I was diagnosed with #papillarythyroidcancer and am now cancer free! 4. I've never seen The Matrix movies, Back to the Future, or the Terminator movies. (But I am currently watching The Matrix since I'm in bed rest!) 5. One of my favorite stories of all time is Alice in Wonderland. I also did a photo series based on the book! Tell me about you! Leave a comment below of some of your favorite things! 😊❤️ (at Houston, Texas)

My Cancer Story #2
So it’s been a week...
I don’t really believe I have cancer yet, but I’ve read the papers from my doctor’s office and it says it right there. We also still don’t have a surgery date, they said to wait another week. Also I’m not going to be seeing a paediatrician since they have no experience in thyroid surgery, so I get to go to the big boy hospital with all the big scary adults.
Honestly I’m starting to get a little scared and stressed, I mean before I found out about the cancer my dog got cancer, my dad got arrested, and my grandma past away, so it’s been a crazy past few months. Also the doctors are worried about my cancer because my tumour is way bigger than 1 cm and when it’s bigger than that the risk of it spreading is 75%. So that information added with the fact that I’ve had it for three years they don’t know how far it’s spread or how bad it is and they won’t be able to find out until the surgery.
Another thing, my family doesn’t exactly have a lot of money, especially with my dad in jail (who earnt the most out of everyone) and my mum still having to look after my younger brother.
On a lighter note; my older sister and mum have both accidentally let it slip that my grandad is coming up from Tasmania to come support me. He and I used to be really close when I was younger, and before his wife (my nana) passed from pancreatic cancer. as you can see cancer runs in the family. But I am very excited to see him again, it’s been so long.
I’ve also been having the urge to do something kinda crazy but not too crazy ya know? like cut my hair short or something (I have pretty long hair). I don’t know about that though since I really like my long hair. I’ll take any suggestions though if anyone sees this and/or reads all the way through this.
So yeah, see you in the next update on my tumour, Gertrude, I guess.

The larynx is an inferior continuation of the oropharynx:
It extends from the epiglottis (namely the glossoepiglottic and pharyngoepiglottic folds) to the inferior aspect / border of the cricoid cartilage.
Inferiorly:
It continues as the cervical trachea.
Gross anatomy:
The larynx consists of a cartilage “skeleton”, as well as internal structures that are divided into three subsites, mainly for the purposes of laryngeal cancer staging:
Supraglottis:
Which extends from the tip of the epiglottis to the laryngeal ventricle
Subsites:
Epiglottis:
Divided into suprahyoid and infrahyoid portions
Pre-epiglottic space
Arytenoid cartilages
Aryepiglottic folds
Ventricles
False vocal cords
Paraepiglottic space
Glottis
True vocal cords
Anterior commissure
Posterior commissure
Subglottis:
Extends from the inferior surface of the true vocal cords / 5mm below the free margin of the true vocal cord to the inferior aspect / lower border of the cricoid cartilage
Relations:
Anterior:
Strap muscles
Anterior jugular vein
Posterior:
Esophagus
Laryngopharynx
Superior:
Hyoid bone
Laryngopharynx
inferior:
Trachea
Arterial supply:
Above the vocal cords:
Superior laryngeal artery:
Branch of the superior thyroid artery
Below the vocal cords:
Inferior laryngeal artery:
Branch of the inferior thyroid artery
Venous drainage
Above the vocal cords:
Superior laryngeal vein accompanies its artery and drains into the superior thyroid vein:
A tributary to the internal jugular vein
Below the vocal cords:
Inferior laryngeal vein accompanies its artery and drains into the inferior thyroid vein:
A direct tributary to the brachiocephalic vein
Lymphatic drainage:
Supraglottic:
The supraglottic lymphatic network follows the superior laryngeal artery:
Piercing the thyrohyoid membrane and then draining into the superior deep cervical nodes and the pre-epiglottic nodes.
Subglottic:
The infraglotttic lymphatic network drains to the inferior deep cervical nodes, following the inferior laryngeal artery, and/or the prelaryngeal (Delphian) nodes via the conus elasticus (aka the lateral cricothyroid ligaments).
Both the superior and inferior deep cervical nodes then drain into the right and left jugular trunks:
Which subsequently empty into the right lymphatic duct and the thoracic duct on the left.
Innervation:
Motor:
The recurrent laryngeal nerve supplies all the intrinsic muscles of the larynx apart from the cricothyroid muscle:
Which is supplied by the external branch of the superior laryngeal nerve.
Sensory:
Above the vocal cords (supraglottic):
Internal branch of the  superior laryngeal nerve
Below the vocal cords (infraglottic):
Recurrent laryngeal nerve
Development:
The larynx is formed from the laryngotracheal groove:
Which is related to the caudal aspect of the floor of the primordial pharynx
The cartilages of the larynx develop from:
The 4th and 6th pharyngeal arches

Merkel cell carcinoma (MCC) is the eponym for :
Primary cutaneous neuroendocrine carcinoma:
A dermal neoplasm with cytoplasmic, dense-core neuroendocrine granules and keratin filaments.
MCC is a rare primary cutaneous neuroendocrine malignancy
The incidence of MCC in the United States has recently been reported to have increased by an estimated annual percentage change of 8% from 1986 to 2001:
Approximately 1500 new cases were predicted by the American Cancer Society in the United States alone in 2008.
MCC is one of the few cancers found to be:
Associated with immune suppression
Polyomavirus has been shown to be integrated within the genome
Merkel cell carcinoma (MCC) is an uncommon and aggressive cutaneous neoplasm that lacks distinguishing clinical features.
More than half of Merkel cell carcinomas (MCCs) occur in the head and neck of elderly people in areas of actinically damaged skin:
The most common site of occurrence is the periorbital region.
Merkel cell carcinoma (MCC) has a propensity to recur and to cause:
Local (25% to 30% of the cases) and distant metastases (30% to 35% of the cases).
Distant metastases indicate a condition that is nearly always fatal.
Merkel cell carcinoma (MCC) is a deadly disease with a poor likelihood for survival:
Local recurrence occurs in:
44% of patients:
Multiple local recurrences occur in 15% of patients.
These tumors appear as:
Rapidly growing, painless nodules in elderly caucasian individuals or in young adults with ectodermal dysplasia syndromes.
Merkel cell carcinomas (MCCs) usually appear as:
Indurated plaques or violaceous (red or deep purple) solitary and dome-shaped nodules.
The surface is typically shiny, with telangiectasias and possibly ulceration.
Most tumors measure 0.7 cm to 1.2 cm in diameter.
Merkel cell carcinomas (MCCs) usually occur in sun-damaged skin.
They are often found near other lesions of actinically damaged skin, including skin involved with:
Bowen disease, squamous cell carcinoma, basal cell carcinoma, solar keratoses, or lentigo maligna.
The mean age at presentation is 68 years:
No gender bias is observed.
Merkel cell carcinoma (MCC) has also been linked to:
Previous radiation exposure and B-cell lymphoma.
The nonspecific characteristics of Merkel cell carcinoma (MCC) lead to a lengthy differential diagnosis that includes:
Basal cell carcinoma
Squamous cell carcinoma
Keratoacanthoma
Amelanotic melanoma
Epidermal cysts
Lymphoma
Metastatic carcinoma of the skin
As a result:
Merkel cell carcinoma (MCC) is rarely diagnosed until biopsy is performed
Approximately 53% of Merkel cell carcinomas (MCCs) occur in the head and neck:
35% occur in the extremities.
In the head and neck:
46% of tumors occur in the periorbital region
29%, on the cheek
18%, on the eyelid
17%, on the forehead.
Other sites in the head and neck include:
The lips (9%)
Ears (7%)
Nose and neck (5.4%)
Scalp (4%)
Tumors have also been reported in areas not exposed to sun, such as the:
Nasal cavity
Buccal mucosa
Gingiva
Hard palate
Postauricular skin
Approximately 3% of patients with Merkel cell carcinoma (MCC) have tumors at several sites.
MCC is an aggressive tumor:
With an overall five-year survival of 40%
Reported five year survival rates of local, nodal and metastatic disease are:
64%, 39% and 18%, respectively
Reported rates of regional lymph node involvement at the time of presentation vary from 10% to 45%:
Around 50% of patients with lymph node metastases harbor concurrent distant metastases:
Most often in the:
Liver, lung, brain, bone or skin
MCC has high rates of local recurrence:
25% to 30% of patients.
MCC has high rates of distant metastasis:
30% to 35% of patients.
