Kanpur Train accident: Four coaches of the Train derailed at Kanpur Central RLY Station
Kanpur / New Delhi : four LC coaches coming from Lucknow derailed at the changing point of Kanpur Central Railway Station. Due to this, trains coming from Lucknow and Prayagraj have been stopped wherever. The train accident is reported to be around 7 am Wednesday. No casualties were reported from any passenger. DRM Amitabh Kumar of Allahabad division has left for Kanpur.

The passenger train coming from Lucknow was given a signal on platform number 3. Two coaches of the train derailed as they turned towards platform number 3 near the scissors of platform number 2 and 3. Due to the loud voice, there was chaos in the passengers. Due to low speed, the car stopped at some distance.

As soon as the train stopped, the passengers, who believed in the accident, jumped and started running. Some people were making a cry that if not get down early, the whole coach would be overturned. Being an incident near the station, the soldiers of GRP and RPF arrived shortly. The police assured the passengers that there were no accidents. After this, the passengers have breathed a sigh of relief.
Prayagraj and Lucknow rail routes closed, 18 trains stopped
The tracks of Prayagraj and Lucknow are also currently closed due to derailment of the train at the changing point. 18 trains have been stopped, including Pushpak Express coming from Mumbai, Kanpur-Lucknow Memu, Prayagraj Intercity, Jabalpur and Chitrakoot Express. All Rajdhani and Shatabdi trains had passed in the morning.

TÓM TẮT
Mục đích của nghiên cứu này là để sản xuất steviosid và rebaudiosid A có độ
tinh khiết cao, chiết xuất từ lá cây làm chất tạo ngọt cho thực phẩm và dược
phẩm. Lá Cỏ Ngọt được sấy khô và nghiền nhỏ thành bột, bột lá Cỏ Ngọt
được chiết xuất bằng nước, dịch chiết được kiềm hóa bởi canxi hydroxit và
sau đó bằng sắt (III) clorua, tiến hành khử muối, khử ion, khử màu và làm
bay hơi dịch lọc đến khô thu được hỗn hợp glycosid thô. Hỗn hợp glycosid
thô tiến hành phân lập và tinh sạch steviosid và rebaudiosid A bằng cách sử
dụng metanol, etanol để kết tủa, thu được steviosid và rebaudiosid A có độ
tinh khiết cao. Tỷ lệ thích hợp của bột lá Cỏ Ngọt và nước trong quá trình
chiết xuất giới hạn khoảng 1:20 đến 1:25 w/v. Nhiệt độ và thời gian chiết xuất
tốt nhất là 75oC trong 240 phút và 100oC trong 30 phút. Sự phân lập và tinh
sạch steviosid từ hỗn hợp glycosid thô đã được tiến hành bằng cách sử dụng
metanol để kết tủa. Tỷ lệ hỗn hợp glycosid thô và metanol là giữa 1:2 đến 1:7
w/v, tốt nhất là 1:5, hàm lượng steviosid thu được trong khoảng 91,1-91,5%.
Sự phân lập và tinh sạch rebaudiosid A được tiến hành bằng cách sử dụng
etanol để kết tủa. Tỷ lệ xirô và etanol là 1:2 đến 1:7 w/v, tốt nhất là 1:5.
Rebaudiosid A với 90,7–90,9% độ tinh khiết thu được. Steviosid và
rebaudiosid A là hai chất tạo ngọt không sinh năng lượng, không đắng, áp
dụng trong thực phẩm và dược phẩm.

ABSTRACT
The purpose of the study is to produce highly purified stevioside and
rebaudioside A from the extract of Stevia rebaudiana Bertoni plant use in
food and drugs. Stevia rebaudiana Bertoni leaves were dried and powdered
leaves being subjected to water extraction and the resulted extract is purified
using treatment with a base such as calcium hydroxide and then iron
chloride, desalting, decolorizing, and evaporating the filtrate to
drynessobtained glycoside. The mixture of glycosides, conducting isolation
and purification of stevioside and rebaudioside A were developed using
methanol, alcoholic precipitation.The highly purified stevioside and
rebaudioside A were obtained.The preferable ratio of leaves to water is
within the limits of about 1:20 to 1:25, wt/vol.The extraction temperature and
time preferably were at 75oC for 240 min and and 100oC for 30min. The
isolation and purification of Stevioside was developed using methanol
precipitation. The proportion of glycosides and methanol was between 1:2-
1:7 w/v, preferably 1:5, the powder contents were around 91.1-91.5% of
stevioside. The isolation and purification of rebaudioside A was developed
using ethanol precipitation. The proportion of syrup and ethanol was between
1:2-1:7 w/v, preferably 1:5. Rebaudioside A with 90.7- 90.9% purity was
obtained. Steviosid and rebaudiosid A are two sweeteners non-calorie, noncariogenic, non-bitter, applied in foods and drugs.

CHƯƠNG I GIỚI THIỆU
1.1 Đặt vấn đề
Ngày nay trên thế giới, con người tiêu thụ một lượng đường rất đáng kể.
Tuy nhiên ngoài mặt lợi đường cũng có những mặt trái của nó do cung cấp
nhiều năng lượng nên có thể gây béo phì, cho dù liều lượng sử dụng không
đáng kể. Vì vậy để tránh béo phì, tiểu đường thì người ta buộc phải dùng
đường ít lại hay dùng một chất tạo ngọt khác để thay thế. Hiện nay, trên thị
trường đã có rất nhiều sản phẩm hoá học tạo vị ngọt dùng để thay thế đường
như saccharin, sodium cyclamate, sucrolose, acefulfame potassium nhưng phổ
biến nhất là chất aspartame. Mặc dù các loại đường này rẻ tiền và tiện dụng
nhưng người ta rất e ngại khi sử dụng vì chúng có thể gây ảnh hưởng xấu đến
sức khoẻ con người về lâu về dài. Qua một số nghiên cứu cho thấy saccharin
gây ung thư bàng quang ở chuột. Vì vậy saccharin đã bị cấm sử dụng ở một
số quốc gia. Trước các nguy hại của đường hoá học, tâm lý chung của người
tiêu dùng là muốn tìm lại sản phẩm đường tự nhiên ly trích từ thực vật. Cây
Cỏ Ngọt (Stevia rebaudiana Bertoni) là loại thảo dược có nguồn gốc từ Nam
Mỹ, lá cây Cỏ Ngọt có chứa các chất ngọt tự nhiên như steviosid (4-13% trên
trọng lượng khô), rebaudiosid A (2-4%), rebaudiosid C, rebaudiosid B (1–
2%), rebaudiosid D, rebaudiosid E và dulcosid A (0,4–0,7%) (Kinghorn et al.,
1991).Trong đó độ ngọt của rebaudiosid A gấp 250 – 450 lần so với đường
mía và steviosid khoảng 300 lần so với đường mía (Cramer et al.,1986). Chất
tạo ngọt chứa trong cây Cỏ Ngọt là loại chất tạo ngọt không sinh năng lượng.
Ngoài ra, chất tạo ngọt trong cây Cỏ Ngọt còn điều trị được bệnh tiểu đường,
hạ đường huyết (Lailerd et al.,2004), béo phì, sâu răng, tăng huyết áp
(Dyrskog et al., 2005), kháng vi khuẩn, kháng nấm, kháng virus, kháng viêm
(Ghosh et al., 2008). Đặc biệt trong cây Cỏ Ngọt có hai loại chất tạo ngọt
steviosid và rebaudiosid A với độ ngọt cao và chiếm hàm lượng lớn nhất.Từ những lợi ích to lớn đó đã dẫn đến nhu cầu con người muốn sử dụng sản
phẩm chất tạo ngọt tự nhiên, đặc biệt là hai loại chất tạo ngọt steviosid và
rebaudiosid A. Do đó việc ly trích và tinh sạch để có hai loại chất tạo ngọt
steviosid và rebaudiosid A với độ sạch cao, an toàn cho thực phẩm và dược
phẩm là rất cần thiết. Xuất phát từ thực tế này, “Nghiên cứu chiết xuất và
tinh sạch steviosid và rebaudiosid A từ cây Cỏ Ngọt (Stevia rebaudiana)
làm chất tạo ngọt trong thực phẩm và dược phẩm” được thực hiện nhằm
góp phần nâng cao chất lượng cuộc sống của con người.
1.2 Mục tiêu nghiên cứu
Xây dựng quy trình chiết xuất, tinh sạch 02 hợp chất tự nhiên đặc trưng từ cây
Cỏ Ngọt là steviosid và rebaudiosid A làm chất tạo ngọt trong thực phẩm và
dược phẩm.
1.3 Nội dung nghiên cứu
Để đạt được mục tiêu trên, đề tài đã tiến hành 3 nội dung chính sau đây:
– Khảo sát tỷ lệ chiết giữa nước và nguyên liệu trong quá trình ly trích hỗn
hợp glucosid thô.
– Khảo sát thời gian và nhiệt độ trong quá trình ly trích hỗn hợp glucosid
thô.
– Phân lập và tinh sạch hai loại chất tạo ngọt steviosid và rebaudiosid A
CHƯƠNG II LƯỢC KHẢO TÀI LIỆU
2.1 Lịch sử về cây Cỏ Ngọt
Vào năm 1887, MS Bertoni, một nhà thực vật học là người châu Âu đầu tiên
nghiên cứu về Cỏ Ngọt. Sau đó vào năm 1931, nhà hóa học Pháp đã ly trích
được steviosid. Steviosid là hợp chất tinh thể màu trắng có độ ngọt rất cao.Từ
đó Cỏ Ngọt được sử dụng như một chất làm ngọt ở Anh trong thế chiến II.
Nhật Bản đã sử dụng Cỏ Ngọt để thay thế cho saccharin sau khi nó bị cấm sử
dụng vào những năm của thập niên 70 và chất ngọt từ Cỏ Ngọt được tiêu thụ
tại Nhật Bản với số lượng rất lớn so với bất kỳ quốc gia nào khác. Tại Bắc
Mỹ và Châu Âu, Cỏ Ngọt bắt đầu được sử dụng như là sản phẩm thảo dược
và có sẵn trên thị trường vào những năm 1970 và 1980. Cỏ Ngọt đã được
chiết xuất và cho phép sử dụng ở Mỹ vào năm 1994.
2.2 Đặc điểm, tính vị và công dụng của cây Cỏ Ngọt
2.2.1 Thực vật học (Đỗ Huy Bích et al., 2004)
Tên khoa học: Stevia rebaudiana (Bertoni) Hemsley.

Họ: Cúc (Asteraceae).
Cây thảo nhỏ, sống nhiều năm, cao 0,5 – 0,6 m và có khi cao tới 1m. Thân
cứng mọc thẳng, có rãnh dọc và nhiều lông mịn, ít phân nhánh.Lá mọc đối,
hình mác hoặc bầu dục, gốc thuôn, đầu tù hoặc hơi nhọn dài 5 -7 cm, rộng 1 –
1 , 5 cm, có 3 gân, 4-6 đôi răng nhọn ở phần nửa về phía đầu lá, hai mặt có
lông trắng mịn, nhấm lá thấy có vị ngọt rất đậm, cuống lá rất ngắn. Hoa
lưỡng tính, tụ họp thành đầu màu trắng ở ngọn. Quả bế, không có mào lông,
hạt không có nội nhũ. Mùa hoa khoảng tháng 5-9
2.2.2 Phân bố, sinh thái (Đỗ Huy Bích et al., 2004)
Chi Stevia Cav không có một đại diện nào ở vùng châu Á. Cỏ Ngọt đang
được trồng phố biến ở Việt Nam hiện nay, được nhập nội từ một nước Nam
Mỹ (nguồn gốc ở Paraguay) năm 1988. Cỏ Ngọt là cây ưa ấm và ưa sáng, có
thể chịu bóng hoặc ưa bóng vào thời kỳ cây con. Vốn là cây ở vùng nhiệt đới,
Cỏ Ngọt trồng ở Việt Nam sinh trưởng và phát triển tốt vào vụ xuân – hè. Về
mùa đông, cây có hiện tượng rụng lá và hơi tàn lụi. Cây ra hoa, quả nhiều
hàng năm. Tuy nhiên, người ta thường áp dụng cách nhân giống bằng cắm
cành.
2.2.3 Bộ phận dùng (Đỗ Huy Bích et al., 2004)
Phần trên mặt đất của cây Cỏ Ngọt. Thu hái lúc cây xum xuê.
2.2.4 Tác dụng dược lý (Đỗ Huy Bích et al., 2004)
2.2.4.1 Tác dụng hạ đường huyết: thành phần steviosid trong lá cây Cỏ Ngọt
có tác dụng làm đường huyết giảm rõ rệt ở thỏ, chuột cống trắng và bệnh
nhân mắc bệnh đái tháo đường.
2.2.4.2 Tác dụng giãn mạch: steviosid có tác dụng làm giãn tĩnh mạch toàn
thân rõ rệt.
2.2.4.3 Tác dụng trên thận và huyết áp: thành phần steviosid trong lá cây Cỏ
Ngọt có tác dụng làm tăng lưu lượng huyết tương qua thận (RPF), tăng tốc độ
lọc cầu thận (GFR), hạ huyết áp, tăng bài niệu và tăng thải trừ natri.
2.2.4.4 Tác dụng kháng khuẩn
Cao lá Cỏ Ngọt có tác dụng đối với Pseudomonas aeruginosa và Proteus
vulgaris. Chưa thấy tài liệu công bố về tác dụng trên các vi khuẩn
Streptococcus inutans, Lactobacillus plantarum và Lactobacillus casei là
những vi khuẩn có liên quan đến quá trình sún răng ở trẻ em, vì hiện nay có
dùng steviosid thay thế đường trong chế biến bánh kẹo.
2.2.4.5 Liều dùng an toàn ở người
Nồng độ steviosid trong một số nước giải khát ở Nhật Bản là 0,005 – 0,007%.
Nếu một ngày uống 1 lít thì lượng steviosid đưa vào cơ thể là 0,05 – 0,07g
tương ứng với khoảng lg lá Cỏ Ngọt. Điều đó có thể được chấp nhận vì là liều
an toàn, đã được Nhà nước Nhật cho phép.
2.2.4.6 Độc tính cấp: Cho chuột uống steviosid với liều 2g/kg không thấy có
chuột chết và cũng không thấy có biểu hiện độc sau 2 tuần theo dõi.
2.2.4.7 Độc tính bán cấp: Cho chuột cống trắng ăn với liều dùng hàng ngày
0,5g/kg trong 56 ngày, các thông số theo dõi gồm cân nặng, các chỉ tiêu huyết
học, các chỉ tiêu hoá sinh và xét nghiệm tổ chức học gan đều bình thường.
2.2.4.8 Độc tính trên thận:
Thí nghiệm trên chuột cống trắng, liều cao steviosid có thể gây độc với thận,
làm tăng urê và creatinin huyết thanh.Tiêm tĩnh mạch cho chuột cống trắng,
liều 0,4; 0,8; 1,2; l,6g/kg rồi theo dõi độ thanh thải của một số chất có so sánh
với lô đối chứng, thấy từ liều 0,8g/kg trở lên độ thanh thải inulin không thay
đổi, chứng tỏ chức năng lọc cầu thận không bị ảnh hưởng. Độ thanh thải của
glucose tăng, chứng tỏ chức năng tái hấp thu của ống lượn gần bị suy giảm,
một phần glucose không được tái hấp thu và đã bị thải trừ qua nước tiểu. Độ
thanh thải steviosid cao hơn độ thanh thải inulin, chứng tỏ steviosid còn bị
thải trừ qua niêm mạc của ống thận. Ngoài ra, steviosid cũng gây bài niệu
tăng thải natri niệu và làm tăng độ thanh thải của para-aminohippuric.
Tiêm dưới da cho chuột cống trắng với liều 1,5g/kg, kết quả thấy những thay
đổi giống thí nghiệm trên là tăng nồng độ urê và creatinin trong huyết thanh,
thấy glucose trong nước tiểu. Ngoài ra về tổ chức bệnh học, thấy có biến đổi
hình thái ở tế bào niêm mạc ống lượn gần. Những tổn thương này là do hoạt
động của 2 enzym phosphatase kiềm và gama-glutamyl-transpeptidase tăng
lên. Những enzym này khu trú chủ yếu ở điểm bàn chải của tế bào ống lượn
gần.
2.2.4.9 Khả năng gây đột biến
Nhiều thí nghiệm nghiên cứu khả năng gây đột biến trên các chủng
Salmonella typhimurium TA 98, TA 100, TA 1535, TA 1538 và TM 677,
hoặc trên chủng Escherichia coli WP2 đều xác định cao Cỏ Ngọt không gây
đột biến. Có nghiên cứu cho biết một số chất chuyển hoá của steviosid lại có
thể gây đột biến. Mặc dù steviosid không bị chuyển hoá trong cơ thể và thải
trừ nguyên vẹn qua nước tiểu. Song người ta lo ngại một số tạp khuẩn vùng
màng tràng có thể phân giải được steviosid.
2.2.5 Tính vị, công năng (Đỗ Huy Bích et al., 2004)
Cỏ Ngọt có vị ngọt rất đậm, có ích cho người đái tháo đường và người béo
phì, thuốc ít độc.
2.2.6 Công dụng (Đỗ Huy Bích et al., 2004)
Chữa đái tháo đường: Cỏ Ngọt và steviosid có vị ngọt sẽ làm giảm nhu cầu
chất đường và chất bột của người bệnh, vì thế sẽ làm giảm đường huyết. Liều
dùng theo thử nghiệm ở Braxin là mỗi lần 0,25g steviosid (hoặc 2,5g lá Cỏ
Ngọt), ngày 4 lần, uống nhiều ngày. Ngoài ra Cỏ Ngọt và steviosid còn chữa
béo phì do có vị ngọt sẽ làm giảm nhu cầu chất đường và chất bột của cơ thể,
nên cũng có tác dụng chữa béo phì. Liều dùng 0,5 – lg steviosid chia ra 3 – 4
lần trong ngày, uống nhiều ngày. Lá Cỏ Ngọt hoặc steviosid thường dùng làm
chất điều vị cho các loại trà thuốc, trà túi lọc tỷ lệ lá Cỏ Ngọt hoặc steviosid
trong đó thường thấp. Tại Việt Nam, cũng đã có một số chế phẩm trà thuốc có
Cỏ Ngọt như trà actisô – stevia. Trà sâm quy – stevia có sâm khu 5, tam thất,
đương quy, thục địa, táo, long nhãn, ngũ gia bì và Cỏ Ngọt.Trà nhân trần,
thảo quyết minh, Cỏ Ngọt.Trà túi lọc Sotevin có dừa cạn, hoa cúc, hoa hoè và
Cỏ Ngọt. Chữa cao huyết áp hàng ngày uống trà sotevin. Dùng thay thế
đường sacharose trong công nghiệp thực phẩm để làm chất điều vị cho bánh
mứt kẹo, nước giải khát.
2.3 Thành phần hóa học của cây cỏ ngọt
Trong các loại đường thay thế, chất ngọt tự nhiên đã được xem xét rất nhiều
bởi các nhà nghiên cứu và đường trong cây Cỏ Ngọt được giả định là nguồn
chất tạo ngọt tự nhiên chính. Một loạt các loài cây Cỏ Ngọt đã được nghiên
cứu bởi các nhà hóa sinh và công nghệ sinh học về các thành phần hóa học
của cây Cỏ Ngọt và chỉ ra có khoảng 110 loài, trong đó có 18 loài có tính
năng tốt (Soejarto et al.,1982). Steviosid là một thành phần chính trong cây
Cỏ Ngọt và steviosid chứa nhiều nhất trong lá của cây Cỏ Ngọt (Geuns,
2003). Cùng với steviosid, một số hợp chất ngọt khác như steviobiosid,
rebaudiosid A, B, C, D, E và dulcosid A cũng đã được tìm thấy trong lá của
Cỏ Ngọt (Savita et al.,2004). Việc xây dựng cấu trúc của các dẫn xuất
steviosid và vị ngọt của nó so với sucrose được nghiên cứu đầy đủ (Cramer et
al.,1986 ), (Geuns, 2003). Kết quả được ghi nhận ở Bảng 2.3 cùng với vị
ngọt, cây Cỏ Ngọt cũng có một số dư vị đắng do sự hiện diện của một số loại
tinh dầu, tannin và flavonoids và đã nhận thấy rằng steviosid và rebaudiosid A
có trách nhiệm ở một mức độ cho dư vị đắng này, mặc dù vai trò của
rebaudiosid A là ít hơn đáng kể so với các steviosid (Phillips, 1987)

2.4 Hoạt tính sinh học
Quá trình sử dụng hai thành phần chính của cây Cỏ Ngọt là steviosid và
rebaudiosid A uống bằng đường miệng để kiểm tra độc tính thì được chấp
nhận không độc trong quá trình sử dụng (Medon et al., 1982), (Toyoda et al.,
1997). Sử dụng steviol glycosid với liều cao cho vào miệng của chuột và nuốt
vào bụng cho thấy trọng lượng cơ thể của chuột giảm (Curry et al., 2008),
nhưng không có bằng chứng mạnh mẽ về độc tính (Carakostas et al., 2008),
(Hagiwara et al.,1984), nghiên cứu tác dụng gây ung thư của steviosid trên
bàng quang thấy rằng không có tiền ung thư hoặc ung thư.
Dịch chiết cũng như các chất tách được từ Cỏ Ngọt có tác dụng mạnh
trong việc điều khiển quá trình chuyển hoá glucose và insulin trong cơ thế.
Theo Chen et al., 2005, khi sử dụng steviosid với liều 0,5 mg/kg sẽ làm giảm
lượng đường glucose trong máu đồng thời hạn chế sự kháng insulin ở chuột
bị tiểu đường. Tác giả Ferreira cũng chỉ ra rằng dịch chiết nước Cỏ Ngọt (20
mg/kg/ngày) cũng có tác dụng làm ức chế quá trình chuyến hoá glucose trong
gan trên chuột thí nghiệm (Arayjo et al.,2006). Trước đó vào năm 1986, Curi
đã thử nghiệm tác dụng của dịch chiết nước Cỏ Ngọt trên một nhóm người
tình nguyện. Kết quả cho thấy với dịch chiết 5 g lá dùng liên tục trong 3 ngày,
hàm lượng đường glucose trong huyết tương giảm rõ rệt (Curi et al.,1986).
Các nghiên cứu này mở ra khả năng ứng dụng Cỏ Ngọt trong việc chữa trị
bệnh tiểu đường và các bệnh liên quan đến sự chuyển hoá glucose. Ngoài ra,
cùng với tác dụng giảm đường huyết, steviosid còn có tác dụng hạ huyết áp
trên lô chuột bị tiểu đường (Xiao et al., 2003).
Tác dụng chống ung thư và chống viêm của Cỏ Ngọt cũng được nghiên
cứu chi tiết trên mô hình chuột thí nghiệm gây u bằng 12-0-
tetradecanoylphorbol-13-acetate (TPA). Các hợp chất tách được từ cây này
gồm steviosid, rebaudiosid A và C, và dulcosid A ức chế mạnh quá trình gây
viêm đồng thời hỗn hợp các hợp chất này ngăn ngừa tốt sự hình thành ung
thư da trên chuột với liều 1,0 và 0,1 mg/con (Yasukawa et al.,2002).
Hợp chất steviosid còn được sử dụng trong trị liệu chữa tiêu chảy hiệu
quả. Khởi nguồn từ kết quả nghiên cứu tác dụng kháng khuẩn và kháng virút
của dịch chiết nóng cây Cỏ Ngọt cho thấy tác dụng diệt mạnh chủng E. coli là
nguyên nhân gây ra bệnh ỉa chảy (Tomita et al.,2002). Các nghiên cứu tiếp
sau đó khẳng định tác dụng và khả năng ứng dụng rất cao của Cỏ Ngọt cũng
như nhóm chất steviosid trong việc chữa trị ỉa chảy (Pariwat et
al.,2008),(Shiozaki et al.,2006),(Takahashi et al.,2001).
Ngoài các hợp chất đitecpen, trong cây Cỏ Ngọt còn chứa nhóm chất
phenol và flavonoit. Tác giả Ghanta đã định lượng được hàm lượng nhóm
chất này trong dịch chiết etyl axetat là tương đương 0,86 mg axit galic và
0,83 mg quercetin trên 1 mg. Bằng phân tích LC-MS và phổ NMR, nhóm tác
giả đó nhận dạng được sự có mặt của quercetin-3-O-arabinoside, quercitrin,
apigenin, apigenin-4-O-glucoside, luteolin, and kaempferol-3-O-rhamnoside.
Dịch chiết metanol và etyl axetat cho thấy có khả năng diệt các gốc tự do
bằng phương pháp thử DPPH và lipid peroxidation. Nghiên cứu này chứng tỏ
cây Cỏ Ngọt còn là một dược liệu thiên nhiên chống oxy hoá hiệu quả
(Ghanta et al., 2007).
2.5 Các phương pháp chiết mẫu thực vật
Sau khi tiến hành thu hái và làm khô mẫu, tuỳ thuộc vào đối tượng chất
có trong mẫu khác nhau (chất phân cực, chất không phân cực, chất có độ
phân cực trung bình…) mà ta chọn dung môi và hệ dung môi khác nhau.
2.5.1 Chọn dung môi chiết
Thông thường các chất chuyển hoá thứ cấp trong cây có độ phân cực khác
nhau. Tuy nhiên những thành phần tan trong nước ít khi được quan tâm.
Dung môi dùng trong quá trình chiết cần phải được lựa chọn rất cẩn thận.
Điều kiện của dung môi là phải hoà tan được những chất chuyển hoá thứ cấp
đang nghiên cứu, dễ dàng được loại bỏ, có tính trơ (không phản ứng với chất
nghiên cứu), không độc, không dễ bốc cháy.
Những dung môi này nên được chưng cất để thu được dạng sạch trước
khi sử dụng. Nếu chúng có lẫn các chất khác thì có thể ảnh hưởng đến hiệu
quả và chất lượng của quá trình chiết. Thường có một số chất dẻo lẫn trong
dung môi như các diankyl phtalat, tri-n-butyl-axetylcitrar và tributylphosphat.
Những chất này có thể lẫn với dung môi trong quá trình sản xuất hoặc trong
khâu bảo quản như trong các thùng chứa hoặc các nút đậy bằng nhựa.
Metanol và chloroform thường chứa dioctylphtalat [di-(2- etylhexyl)
phtalat hoặc bis-2-etylhexyl-phtalat]. Chất này sẽ làm sai lệch kết quả phân
lập trong các quá trình nghiên cứu hoá thực vật, thể hiện hoạt tính trong thử
nghiệm sinh học và có thể làm bẩn dịch chiết của cây. Chloroform, metylen
clorit và metanol là những dung môi thường được lựa chọn trong quá trình
chiết sơ bộ một phần của cây như: lá, thân, rễ, củ, quả, hoa…
Những tạp chất của chloroform như CH2C12, CH2ClBr có thể phản ứng
với một vài hợp chất như các ancaloit tạo muối bậc 4 và những sản phẩm
khác. Tương tự như vậy sự có mặt của lượng nhỏ axit clohiđric (HCl) cũng có
thể gây ra sự phân huỷ, sự khử nước hay sự đồng phân hoá với các hợp chất
khác. Vì chloroform có thể gây tổn thương cho gan và thận nên nó cần được
thao tác khéo léo, cẩn thận ở nơi thoáng và phải đeo mặt nạ phòng độc.
Metylen clorit ít độc hơn và dễ bay hơi hơn chloroform.
Metanol và etanol 80% là những dung môi phân cực hơn các
hiđrocacbon thế clo. Người ta cho rằng các dung môi thuộc nhóm rượu sẽ
thấm tốt hơn lên màng tế bào nên quá trình chiết với các dung môi này sẽ thu
được lượng lớn các thành phần trong tế bào.Trái lại, khả năng phân cực của
chlorofrom thấp hơn, nó có thể rửa giải các chất nằm ngoài tế bào. Các etanol
hoà tan phần lớn các chất chuyển hoá phân cực cùng với các hợp chất phân
cực trung bình và thấp. Vì vậy khi chiết bằng etanol thì các chất này cũng bị
hoà tan đồng thời. Thông thường dung môi etanol trong nước có những đặc
tính tốt nhất cho quá trình chiết sơ bộ.
Tuy nhiên cũng có một vài sản phẩm mới được tạo thành khi dùng
metanol trong suốt quá trình chiết (Goad et al.,1997). Thí dụ trechlonolid A
dung môi như các diankyl phtalat, tri-n-butyl-axetylcitrar và tributylphosphat.
Những chất này có thể lẫn với dung môi trong quá trình sản xuất hoặc trong
khâu bảo quản như trong các thùng chứa hoặc các nút đậy bằng nhựa.

tính tốt nhất cho quá trình chiết sơ bộ.
Tuy nhiên cũng có một vài sản phẩm mới được tạo thành khi dùng
metanol trong suốt quá trình chiết (Goad et al.,1997). Thí dụ trechlonolid A
2.5.2 Quá trình chiết
Hầu hết quá trình chiết đơn giản được phân loại như sau:
– Chiết ngâm.
– Chiết sử dụng một loại thiết bị là bình chiết soxhlet.
– Chiết sắc với dung môi nước.
– Chiết lôi cuốn theo hơi nước.
Chiết ngâm là một trong những phương pháp được sử dụng rộng rãi nhất
trong quá trình chiết thực vật bởi nó không đòi hỏi nhiều công sức và thời
gian. Thiết bị sử dụng là một bình thuỷ tinh với một cái khoá ở dưới đáy để
điều chỉnh tốc độ chảy thích hợp cho quá trình tách rửa dung môi, dung môi
có thể nóng hoặc lạnh nhưng nóng sẽ đạt hiệu quả chiết cao hơn. Trước đây,
máy chiết ngâm đòi hỏi phải làm bằng kim loại nhưng hiện nay có thể dùng
bình thuỷ tinh.
Thông thường quá trình chiết ngâm không được sử dụng như phương
pháp chiết liên tục bởi mẫu được ngâm với dung môi trong máy chiết khoảng
24 giờ rồi chất chiết được lấy ra. Thông thường quá trình chiết một mẫu chỉ
thực hiện qua 3 lần dung môi vì khi đó cặn chiết sẽ không còn chứa những
chất giá trị nữa. Sự kết thúc quá trình chiết được xác định bằng một vài cách
khác nhau.
Như vậy tuỳ thuộc vào mục đích cần chiết lấy chất gì để lựa chọn dung
môi cho thích hợp và thực hiện quy trình chiết hợp lý nhằm đạt hiệu quả cao.
Ngoài ra, có thể dựa vào mối quan hệ của dung môi và chất tan của các lớp
chất mà ta có thể tách thô một số lớp chất ngay trong quá trình chiết.
CHƯƠNG III NGUYÊN LIỆU VÀ PHƯƠNG PHÁP NGHIÊN CỨU
3.1 Nguyên liệu nghiên cứu
3.1.1 Nguyên liệu thí nghiệm
Nguyên liệu dùng để chiết xuất các đối tượng nghiên cứu của đề tài: lá cây
Cỏ Ngọt dùng để chiết xuất steviosid và rebaudiosid A. Cỏ Ngọt thuộc họ cúc
được mua tại nhà thuốc Minh Phát, 73B Hải Thượng Lãng Ông, phường 10,
quận 5, Thành phố Hồ Chí Minh.
3.1.2 Hóa chất, chất chuẩn
– Metanol (99,99%), (Merck KGaA, 64271 Darmstadt, Đức)
– Acetonitrile (99,9 %), (Merck KGaA, 64271 Darmstadt, Đức)
– Etanol (99,9%), (Merck KGaA, 64271 Darmstadt, Đức)

3.2 Phương pháp nghiên cứu
3.2.1 Sơ đồ nghiên cứu
Sơ đồ nghiên cứu.
3.2.2 Thiết kế thí nghiệm
– Cỡ mẫu: tùy theo hàm lượng hợp chất đặc trưng trong dược liệu, khối lượng
dược liệu (cỡ mẫu) sử dụng đủ để chiết xuất nhiều mẻ nhằm thu được 20 g
hợp chất.
– Chiết xuất, phân lập, tinh chế: thí nghiệm được tiến hành tại phòng thí
nghiệm Khoa Sinh học Ứng dụng và phòng thí nghiệm Công nghệ Thực
phẩm Trường Đại học Tây Đô.
– Phân tích và kiểm nghiệm: phòng thí nghiệm trung tâm Khoa Dược và Điều
Dưỡng Trường Đại học Tây Đô.
3.2.3 Thời gian thực hiện đề tài
Đề tài được thực hiện từ 10/2014 đến 10/2015.
3.2.4 Bố trí thí nghiệm
3.2.4.1 Thí nghiệm 1: khảo sát tỷ lệ chiết giữa nước và nguyên liệu trong quá
trình ly trích hỗn hợp glycosid thô.
* Mục đích: tìm ra tỷ lệ chiết xuất thích hợp giữa nguyên liệu và nước mà ở
đó hàm lượng glycosid thô cao nhất.
Thí nghiệm được bố trí hoàn toàn ngẫu nhiên 1 nhân tố, 5 mức độ với 5
nghiệm thức và 3 lần lặp lại cho mỗi nghiệm thức. Kết quả phân tích hàm
lượng glycosid thô được xử lý thống kê.
Nhân tố A: tỷ lệ nước
A1: 1:6
A3: 1:15 A2: 1:9 A5: 1:25
A4: 1:20
Tiến hành:
Mẫu được mua về tiến hành tách lá ra khỏi thân cây, lá mang phơi khô
dưới ánh sáng mặt trời trực tiếp ở nhiệt độ dao động từ 25°C đến 30°C trong
24 giờ đến 48 giờ. Các lá khô được nghiền thành bột. Bột lá Cỏ Ngọt được
cho vào túi polyethylene, buộc kín và bảo quản ở nhiệt độ 4 ± 1°C trong tủ
lạnh cho đến khi sử dụng (Abou-Arab et al., 2010). Bột lá Cỏ Ngọt cân 1g
cho vào cốc 50mL, cho thêm 6mL nước vào ta có tỷ lệ 1:6. Tương tự các cốc
còn lại bố trí với tỷ lệ 1:9, 1:15, 1:20, 1:25. Sau đó cho tất cả các cốc vào bếp
đun cách thủy với nhiệt độ 75oC trong vòng 120 phút, mỗi tỷ lệ được tiến
hành 3 lần lặp lại. Khi kết thúc quá trình đun cách thủy, các cốc chứa dịch
chiết sẽ được lọc qua giấy lọc. Tách lấy phần dịch lọc cho vào tủ sấy ở nhiệt
độ 105oC. Sau khi sấy xong mang cốc ra cho vào bình hút ẩm, chờ ổn định và
tiến hành cân cốc. Tính được lượng glycosid thô.
3.2.4.2 Thí nghiệm 2: khảo sát thời gian và nhiệt độ trong quá trình ly trích
glycosid thô.
* Mục đích: chọn thời gian và nhiệt độ chiết xuất thích hợp để thu được hàm
lượng glycosid thô tốt nhất.
Thí nghiệm được bố trí hoàn toàn ngẫu nhiên 2 nhân tố, với 16 nghiệm thức
và 3 lần lặp lại cho mỗi nghiệm thức. Kết quả phân tích glycosid thô.
Nhân tố B: nhiệt độ chiết Nhân tố C: thời gian chiết B1: 45oC
B3: 10oC B2: 75oC C1: 30 phút
B4:1000C C3:120 phút C2: 60 phút
C4:240 phút
Tiến hành
Từ khảo sát tỷ lệ chiết giữa nước và nguyên liệu trong quá trình ly trích
hỗn hợp glycosid thô ở trên ta chọn được tỷ lệ tốt nhất để tiến hành cho thí
nghiệm khảo sát thời gian và nhiệt độ trong quá trình ly trích hỗn hợp
glycosid thô. Cân 1g bột lá Cỏ Ngọt cho vào cốc 50mL thêm nước vào với tỷ
lệ được chọn như trên. Tiến hành gia nhiệt các mẫu ở nhiệt độ 45oC, 75oC,
100oC và làm lạnh ở 10oC với thời gian lần lượt là 30, 60,120, 240 phút. Các
mẫu sau đó được lọc qua giấy lọc. Phần rắn bỏ đi, phần dịch lọc cho vào cốc
50mL đã biết trước trọng lượng. Tiếp tục cho cốc chứa dịch lọc vào tủ sấy ở
nhiệt độ 105oC và sấy đến khối lượng không đổi. Sau đó cân khối lượng, tính
được hàm lượng hỗn hợp glycosid thô có trong mẫu.
3.2.4.3 Thí nghiệm 3: phân lập và tinh sạch hai loại chất tạo ngọt steviosid và
rebaudiosid A
* Mục đích: dùng metanol, cồn tinh sạch hai loại đường steviosid và
rebaudiosid A
+ Phân lập, tinh sạch steviosid:
Thí nghiệm được bố trí hoàn toàn ngẫu nhiên 1 nhân tố là dung môi metanol
với 6 nghiệm thức và 3 lần lặp lại cho mỗi nghiệm thức. Việc phân tích hàm
lượng steviosid bằng sắc ký lỏng hiệu năng cao (HPLC).
Nhân tố D: dung môi metanol
D1: 1:1 D2: 1:2 D3: 1:3

+ Phân lập, tinh sạch rebaudiosid A:
Thí nghiệm được bố trí hoàn toàn ngẫu nhiên 1 nhân tố là dung môi etanol với
6 nghiệm thức và 3 lần lặp lại cho mỗi nghiệm thức. Việc phân tích hàm
lượng rebaudiosid A bằng sắc ký lỏng hiệu năng cao (HPLC).
Nhân tố E: dung môi etanol

* Tiến hành:

quá trình ly trích hỗn hợp glycosid thô. Sau đó cho mẫu vào đun cách thủy ở
nhiệt độ và thời gian đun tốt nhất đã được chọn ở thí nghiệm khảo sát thời
gian và nhiệt độ trong quá trình ly trích hỗn hợp glycosid thô. Kết thúc thời
gian đun, tiến hành cho mẫu qua giấy lọc, phần rắn bỏ đi, phần dịch lọc thu
được cho Ca(OH)2 vào đến khi pH của dịch lọc khoảng 10, tiếp tục cho FeCl3
vào dịch lọc để tạo kết tủa. Tiến hành lọc lấy phần dịch, bỏ kết tủa nhằm loại
bớt màu của dung dịch. Sau đó, tiếp tục cho phần dịch lọc thu được qua các
cột chứa hạt Amberlite lần lượt Amberlite FPC23 H, Amberlite FPA51 và
Amberlite FPA98Cl nhằm khử muối, khử ion, khử màu. Phần dịch sau khi
cho qua các cột Amberlite mang sấy hoặc sấy phun thu được hỗn hợp
glycosid thô. Hỗn hợp glycosid thô thu được đem hòa tan trong metanol và
duy trì ở nhiệt độ 20-25°C, trong thời gian khoảng 30 – 60 phút. Tại thời gian
này sự kết tủa được hình thành, lọc và làm khô kết tủa ta thu được steviosid,
còn phần dịch lọc cho vào tủ sấy để loại bỏ metanol thu được được hỗn hợp ở
dạng xirô. Hỗn hợp xirô này được pha loãng với nước và cho qua màng siêu
lọc polysulfone. Tiếp tục thu dịch lọc đem sấy khô. Dịch lọc sấy khô thu được
thêm etanol và duy trì ở nhiệt độ 45-50°C trong khoảng 30 phút. Trong thời
gian này sự kết tủa được hình thành, lọc và làm khô kết tủa ta thu
được rebaudiosid A tinh khiết
* Tiến hành:

Thí nghiệm được tiến hành như đã nêu ở phần nguyên liệu và phương pháp
nghiên cứu có 5 nghiệm thức thay đổi giữa tỷ lệ dung môi khác nhau được
tiến hành với 3 lần lặp lại. Bột lá khô ngâm với nước nóng ở nhiệt độ 75oC
theo tỷ lệ khác nhau giữa bột và lá lần lượt là 1:6, 1:9, 1:15, 1:20, 1:25. Ở kết
quả nghiên cứu này chỉ ra rằng khi bổ sung lượng nước từ tỷ lệ 1:6 – 1:25 thì
nhận thấy lượng chất tạo ngọt tăng dần từ tỷ lệ 1:6 – 1:20 và từ tỷ lệ 1:20 –
1:25 thì lượng chất tạo ngọt không tăng nữa. Kết quả thống kê giữa hai tỷ lệ
1:20 và 1:25 không có sự khác biệt có ý nghĩa thống kê ở mức độ tin cậy
95%. Ở tỷ lệ 1:20 – 1:25 lượng chất tạo ngọt không tăng là do lượng chất tạo
ngọt và lượng nước đã đạt đến trạng thái cân bằng cho nên khi lượng nước
tăng thêm thì lượng chất tạo ngọt thu được cũng không tăng. Từ các kết quả
thu được có thể kết luận rằng hàm lượng chất tạo ngọt cao nhất là ở tỷ lệ từ
1:20 – 1:25, tốt nhất là ở tỷ lệ 1:20 được minh họa ở Hình 4.1
4.2 Kết quả khảo sát thời gian và nhiệt độ trong quá trình ly trích hỗn
hợp glycosid thô
Trong quá trình chiết xuất dược liệu, nhiệt độ và thời gian chiết xuất là
hai nhân tố ảnh hưởng rất lớn đến việc chiết xuất. Nhiệt độ chiết xuất có ảnh
hưởng đến khả năng chiết tách hàm lượng chất tạo ngọt trong cây Cỏ Ngọt.
Khi nhiệt độ tăng sẽ làm giảm độ nhớt đồng thời làm tăng vận tốc khuếch tán,
do đó tạo điều kiện thuận lợi cho việc chuyển các chất hòa tan từ trong
nguyên liệu vào dung môi. Mặt khác nhiệt giúp cho quá trình trích ly dễ dàng
hơn bằng cách phá hủy màng tế bào làm tăng khả năng hòa tan của bột lá Cỏ
Ngọt và chất cần trích. Vì vậy nhiệt độ phù hợp sẽ cho hàm lượng chất tạo
ngọt rất cao. Quá trình chiết xuất cũng phụ thuộc vào thời gian chiết xuất.
Nếu thời gian trích ly quá ngắn thì không đủ để dung môi hòa tan chất tạo
ngọt. Khi thời gian càng tăng thì hàm lượng chất tạo ngọt trong và ngoài tế
bào sẽ càng đi đến trạng thái cân bằng. Khi đạt đến trạng thái cân bằng thì quá
trình trích ly không xảy ra, do đó hiệu suất trích ly sẽ giảm.

Trong thí nghiệm khảo sát nhiệt độ và thời gian chiết xuất tỷ lệ bột lá Cỏ Ngọt
và nước chọn được tỷ lệ tốt nhất là 1:20 là nhân tố cố định, các nhân tố thay
đổi là nhiệt độ và thời gian. Ở thí nghiệm này nhiệt độ khảo sát là 45oC, 75oC,
10oC, 100oC, thời gian khảo sát cho mỗi nhiệt độ là 30, 60, 120, 240 phút. Kết
quả trình bày ở Hình 4.2 cho thấy lượng chất tạo ngọt của các nghiệm thức
dao động từ 0,065-0,189g và giữa các nghiệm thức có sự khác biệt có ý nghĩa
thống kê với mức độ tin cậy 95%. Từ đó cho thấy quá trình chiết xuất ở nhiệt
độ và thời gian khác nhau thì kết quả hàm lượng chất tạo ngọt khác nhau, nên
trong quá trình chiết xuất nhiệt độ và thời gian là hai nhân tố ảnh hưởng rất
lớn. Nhìn chung, quá trình khảo sát sự ảnh hưởng của thời gian và nhiệt độ có
hai nghiệm thức cao nhất là 75oC, 240 phút có hàm lượng đường 0,189g và
nghiệm thức 100oC, 30 phút có kết quả 0,185g, được minh họa ở Hình 4.2
Hai nghiệm thức này không khác biệt có ý nghĩa thống kê với mức độ tin cậy
95%.
4.3 Kết quả phân lập và tinh sạch hai loại chất tạo ngọt steviosid và
rebaudiosid A
Dịch chiết được kiềm hóa bởi canxi hydroxit và sau đó bằng sắt (III)
clorua, tiến hành khử muối, khử ion, khử màu bằng Amberlite và làm bay hơi
dịch lọc đến khô thu được hỗn hợp glycosid thô. Hỗn hợp glycosid thô tiến
hành phân lập và tinh sạch steviosid và rebaudiosid A bằng cách sử dụng
metanol, etanol để kết tủa thu được steviosid và rebaudiosid A có độ tinh khiết
cao. Kết quả hàm lượng steviosid và rebaudiosidA sau phân lập, tinh sạch
được đo bằng sắc ký lỏng hiệu năng cao (HPLC) và trình bày ở Hình 4.3.1,
4.3.2, 4.3.3 và 4.3.4.
– Phân lập và tinh sạch steviosid
Phân lập và tinh sạch hàm lượng steviosid từ glycosid thô bằng cách sử
dụng metanol, tỷ lệ glycosid thô và metanol được khảo sát trong khoảng 1:2
đến 1:7. Xác định hàm lượng steviosid bằng HPLC ta thu được kết quả ở
Bảng 4.3.1. Qua kết quả thu được ta thấy hàm lượng steviosid dao động từ
74,3% đến 91,4% và giữa các nghiệm thức khác biệt có ý nghĩa thống kê ở
mức độ tin cậy 95%. Theo đó, ở tỷ lệ 1:2 đến 1:4 khi hàm lượng metanol tăng
dần thì hàm lượng steviosid có độ tinh khiết cũng tăng và giữa các nghiệm
thức từ 1:2 đến 1:4 có sự khác biệt có ý nghĩa thống kê ở mức độ tin cậy 95%.
Ở tỷ lệ 1:5 đến 1:7 mặc dù hàm lượng metanol tăng dần nhưng hàm lượng
steviosid không tăng và giữa các nghiệm thức này không khác biệt có ý nghĩa
thống kê. Nhìn chung, từ kết quả ở trên cho thấy rằng tỷ lệ tốt nhất cho quá
trình phân lập và tinh sạch hàm lượng steviosid là tỷ lệ 1:5 và độ tinh khiết
của steviosid là 91,1% – 91,5%
Các giá trị có mẫu tự giống nhau không khác biệt về mặt thống kê ở độ tin
cậy 95%
– Phân lập và tinh sạch rebaudiosid A
Phần dịch lọc từ thí nghiệm tinh sạch steviosid cho vào tủ sấy để loại bỏ
metanol và tiến hành tinh sạch rebaudiosid A. Dịch lọc sau khi loại bỏ
methanol được pha loãng với nước và cho qua màng siêu lọc polysulfone.
Dịch lọc thu được đưa đi sấy khô thu được xirô. Xirô thu được thêm etanol và
duy trì nhiệt độ ở mức 45-50°C, khoảng 30 phút. Trong thời gian này sự kết
tủa được hình thành, lọc lấy kết tủa và làm khô. Hàm lượng chất tạo ngọt thu
được đem đi xác định rebaudiosid A bằng sắc ký lỏng hiệu năng cao.
Rebaudiosid A có độ tinh khiết thu được trình bày ở Bảng 4.3.2.
Trong quá trình phân lập và tinh sạch hàm lượng rebaudiosid A, tỷ lệ hỗn
hợp xirô và etanol dùng giới hạn ở khoảng 1:2, 1:3, 1:4, 1:5, 1:6, 1:7. Qua kết
quả trình bày ở Bảng 4.3.2 cho thấy hàm lượng rebaudiosid A dao động từ
88,5% đến 90,8%, giữa các nghiệm thức khác biệt có ý nghĩa thống kê ở mức
độ tin cậy 95%. Bên cạnh đó, cũng thấy rằng ở các tỷ lệ 1:2, 1:3, 1:4 hàm
lượng metanol tăng dần thì hàm lượng rebaudiosid A cũng tăng theo. Ở các tỷ
lệ 1:5, 1:6, 1:7 hàm lượng etanol tăng nhưng tỷ lệ rebaudiosid A không tăng.
Vì thế, trong quá trình phân lập và tinh sạch rebaudiosid A tỷ lệ bột và etanol
tốt nhất là 1:5 và độ tinh khiết của rebaudiosid A là 90,7% – 90,9%
CHƯƠNG V. KẾT LUẬN VÀ ĐỀ NGHỊ
5.1 Kết luận
Qua nghiên cứu này, quá trình khảo sát tỷ lệ thích hợp của bột lá Cỏ Ngọt
và nước trong quá trình chiết xuất giới hạn khoảng 1:20 đến 1:25 w/v. Nhiệt độ và
thời gian chiết xuất tốt nhất là 75oC trong 240 phút và 100oC trong 30 phút. Sự
phân lập và tinh sạch steviosid từ hỗn hợp glycosid thô đã được tiến hành bằng
cách sử dụng metanol để kết tủa, tỷ lệ glycosid thô và metanol là giữa 1:2 đến 1:7
w/v, tốt nhất là 1:5 w/v, hàm lượng steviosid thu được trong khoảng 90,1% –
91,5%. Sự phân lập và tinh sạch rebaudiosid A được tiến hành bằng cách sử dụng
etanol để kết tủa rebaudiosid A, tỷ lệ xirô và etanol là 1:2 đến 1:7 w/v, tốt nhất là
1:5 w/v, rebaudiosid A với 90,7% – 90,9% độ tinh khiết thu được. Hai chất ngọt
thu được không sinh năng lượng, không đắng, áp dụng trong thực phẩm và dược
phẩm.
5.2 Đề nghị
– Tiếp tục tinh sạch hợp chất steviosid và rebaudiosid A trên đạt tiêu chuẩn
chất chuẩn đối chiếu dùng trong lĩnh vực hóa phân tích và kiểm nghiệm.
– Tiếp tục phân lập các chất còn lại rebaudiosid B, C, D, E và Dulcosid

PHỤ LỤC 1.PHƯƠNG PHÁP PHÂN TÍCH HOÁ LÝ
Sắc ký lỏng hiệu năng cao (HPLC)
HPLC phân tích được thực hiện bằng cách sử dụng cột HIQSIL 100 C18 (25
cm x 4,6 mm ID, 5 micron). Nhiệt độ cột được duy trì ở mức 27-28°C và đầu
dò UV điều chỉnh ở 210 nm. Pha động là acetonitril HPLC và nước khử ion
(80:20) và độ pH được điều chỉnh đến 3,0 bằng acid phosphoric 85%. Các
mẫu được lọc qua bộ lọc 0,22 µm hoặc tương đương. Thể tích tiêm đã được
thiết lập để 10μL với tốc độ dòng của 1ml / phút. Các chuẩn steviosid hydrate
(98%) và rebaudiosid A (96%), (Sigma – Aldrich) đã được sử dụng.
Phân tích thống kê
Các thí nghiệm được tiến hành ba lần lặp lại. Phân tích phương sai (ANOVA)
được thực hiện và sự khác biệt có ý nghĩa (p <0,05) giữa các giá trị trung bình
được đánh giá bởi Duncan, sử dụng SPSS phiên bản 19.0 (SPSS, Chicago, IL,
USA).

This is the 3rd and last part of the series The Rwandan’s occupation and land grabbing strategy since 1950 In the series we explore how the Rwandans who came seeking for refuges in the 1950’s turned Uganda’s indigenous communities into refuges in their country by taking over their political, economic, social and now land heritage.
This is a long article detailing the history and facts linking Museveni Tutsi/Rwanda merciless strategies to grab land and literally wipe out all indigenous societies in the above region.

The Banyarwanda’s are registered at each sub-county and mandated to hold highly classified monthly get together meeting. They do this so secretly that local community members never know, yet even if one of them is a house girl in your home knows of this meeting and attends it.
By 1991 it was estimated that Banyarwanda in Uganda were 1.3M out of a total population of 18M in the country. Of these, 450,000 were Uganda indigenous Banyarwanda who became Ugandans after Kisoro became part of Uganda in 1918 and 650,000 were the Banyarwanda economic immigrants who came to look for work and a few who came to look for pasture for their cattle between 1926 and 1959 of whom 84,000 were the Banyarwanda Tutsi refugees registered under UNHCR.
Many had left the refugee settlement camps and integrated into local society. In 1993 the then Minister of Local government, Dr. Steven Chebrot estimated that about 300,000 Banyarwanda refugees were spontaneously resettled amongst the locals outside the camps.
The able bodied had left the camps to the elderly and settled in urban centres while others had acquired land outside camps and settled there using money provided by government. The UNHCR had handed the management of the camps to the government though food aid continued coming in. Even within the camps, the refugees had become self sustaining by producing enough to feed themselves and surplus for sale.
In July 1994 when the RPF took over power in Rwanda, a reconnaissance party of Banyarwanda civilians left for Rwanda so as to get hold on political fortunes. By end of 1995, an estimated 226,000 Banyarwanda refugees had returned to Rwanda. They deserted the refugee camps and the countryside as far as Ngoma, Kyankwanzi, Masindi, Nakasongola and Luwero and moved en masse and headed for Rwanda.
As the Tutsi were leaving Uganda for Rwanda, the Hutu were leaving Rwanda in millions for Tanzania and Congo. A small contingent of 11,000 Hutu entered Uganda and were camped in Kisoro and Ntungamo before being transferred to Oruchinga camp. A departing Tutsi at Oruchinga camp attempted to spear a child of an arriving Hutu but only to injure his fellow Tutsi.
The 1993 Arusha Accord between the Tutsi RPF rebels and the Hutu government of Rwanda had stipulated that returning refugees after ten years were not to seek to reclaim previous properties but were to be resettled on unoccupied land. Once the RPF took over power, the returning Tutsi occupied property left behind by fleeing Hutu. However, as time went on, reconciliation efforts dictated that returning Hutu ought to reoccupy their property. The issue was so controversial to an extent that it accounted for the false accusations of genocide against some Hutu just as a way of keeping them off the property.
To strike a compromise, a villagisation (Imidugudu) scheme where services would be centralized and modern agricultural technology made accessible was initiated. The international development partners supported the construction of Imidugudu. Meeting land and housing needs of returning refugees proved quite a challenge. The situation was worse with the pastoralists Tutsi who needed huge chunks of land for their cattle.
Many returned and settled in Uganda where grazing land was plenty. Those who had anticipated a land of milk and honey, they too were disappointed and retreated to Uganda where land, availability of social services, and economic opportunities including employment were easily forthcoming more than in their new-homeland. For the majority of others who had not left for Rwanda, they opted to stay in Uganda. Even the elites in influential positions in Rwanda have been acquiring land in Uganda through proxies.
With the sound availability of cash and government favors, the so called Balaalo have evolved from the traditional pastoralist and casual laborers to owners of thousands of heads of exotic cattle, huge chunks of land, are armed and determined to spread to as far as West Nile and Acholi at the border with Sudan.
Museveni managed to push through the 1995 Constitution a provision that recognised Banyarwanda as one of the 56 indigenous ethnic communities. It was not made clear if the constitutional Banyarwanda ethnic group referred to the Banyarwanda who were added into Uganda by colonial boundaries in Kisoro, the economic immigrants of the early 20th century who came to look for work, or those who came as refugees during the 1959 – 1974 exodus escaping political turmoil. By 2017 Rwanda had a polling station in Uganda, whose eligibility none knows whether Ugandan Banyarwanda voted or not.
In October 2001, the Uganda government announced that it was probing the composition of 1,252 army Cadet Officers after discovering that several Rwandan undercover spies posing as Ugandans had been recruited for training. The then Army Spokesman, Col. Keitirima confirmed that they had also turned away several Rwandans who had sought to be recruited.
In July 2002, a delegation of 30 Banyarwanda elders met Museveni for a petition over the alleges of harassment of Banyarwanda by security agencies. They claimed that over twenty Banyarwanda were being held in safe houses by intelligence services. This was during the time when relations between Uganda and Rwanda were sour following the bloody clash of the two armies in Congo. To the security agencies, the detained Banyarwanda were Rwandese spies. Appearing on Andrew Mwenda Live radio talk show, Kagame had complained that; “I was told how Rwandese are being arrested in Kampala and in Kikuko wherever they are found.”
On 17th June 2007, the government owned New Vision ran a letter by one Mutesi of Kabale in which she was complaining about Banyarwanda being denied passports. That she had had to consult her friends in security services who advised her not to state that she was a Munyarwanda but should instead call herself a Mufumbira.
She went ahead to disclose that since she had worked in Kisoro as a teacher, she travelled there and using the voters card she filled forms, had them approved by the LCs and the area M.P and eventually got her passport. She disclosed that her grandfather had migrated from Rwanda to Tanzania 100 years ago, and that her father relocated to Uganda, died and was buried in Ntungamo, Uganda.
In 2007, the Local Government of Kyankwanzi district offered to take in and accommodate the 100 Banyarwanda Balaalo families that had been evicted from Buliisa following clashes with Bagungu. The move was spearheaded by the LC3 Chairman, Fred Mpora and escorted by Police. Fred Mpora has been a key figure in the incursions by Balaalo into Acholi.
In April 2008 Banyarwanda living in Uganda under their umbrella organisation, UMUBANO held the annual assembly at Lugogo Indoor Stadium under the chairmanship of Erick Kyamuhangire who is Museveni’s Senior Presidential Advisor on Culture. The assembly expressed grave concern over denial of passports, loss of land, denial of recruitment into public service, denial of scholarships and general discrimination by government.
Their Chairman lamented thus; “…. if one happens to have a contact in Rwanda, then it’s enough reason for disqualification.” They gave the example of Balalo who were being evicted in Masindi describing it as paid harassment before accusing the government of being biased against the Balaalo.
Note: one of the key state witnesses in the Kyadondo terror attack, Muhammad Mugisha claimed to be a Ugandan who was born in Rwanda but relocated to Uganda in 1998.
In 2009, the Uganda Citizenship and Immigration Control Act was amended to put it in line with the constitutional provision on Banyarwanda Tutsi. Section.12 provides for citizenship by birth for any person born in Uganda whose parents or grandparents is or was a member of any of the indigenous communities existing in and residing within the borders of Uganda as at the first day of February 1926, as set out in the Third Schedule to the 1995 constitution.
It went ahead to stipulate that every person born in or outside Uganda one of whose parents or grandparents was at the time of birth of that person a citizen of Uganda by birth, is eligible for citizenship. S.14 (1) (a) (ii) eliminates Hutu refugees from accessing citizenship thus; “every person born in Uganda who at the time of birth neither of his or her parents and none of his grandparents was a refugee in Uganda”.
Since it’s now the Hutu who are refugees, the provision bars them. S.14 (b) grants citizenship to anyone who has continuously lived in Uganda since 9th October 1962.
Interestingly, the Act under S.14 (2) (b) gives a blank cheque to Tutsi Banyarwanda who are always on the move migrating to and settling in Uganda thus ” any person who has legally and voluntarily migrated to Uganda and has been living in Uganda for at least twenty years is eligible for citizenship”.
The other requirements include a good command of the English language or ‘prescribed local language’.
In July 2010, the Museveni regime in connivance with the Kagame regime in Kigali forcefully returned to Rwanda 1,700 Hutu Asylum seeker and refugees. The victims have escaped Gacaca community courts, land wrangles and general repression.
Note: The UPC government was accused of forcefully returning Tutsi refugees to Rwanda in the early 1980s but in the instant case it looked okay because the victims were Hutu. In 2010, a group of Banyarwanda refugees in Uganda petitioned the Constitutional Court over acquisition of citizenship. They claimed that the Immigration Department was refusing to give them citizenship application forms on grounds that they were not eligible on account of their being refugees.
They based their petition on a constitutional provision that made refugees eligible for citizenship by naturalisation and registration. Article 12 (2) (c) and 14 (2) (c) provides that “a person who on commencement of this constitution has lived in Uganda for the last 20 years is eligible for citizenship by registration”. The petitioners had lived in Uganda since 1985. In October 2015 court ruled that refugees were eligible for citizenship not by registration but naturalisation but two of the petitioners had already been relocated to the USA by UNHCR while the 3rd could not be traces.
The 2013 Land Policy condemns those who classify “cross border population movement as refugees or internally displaced people because of shared common heritage and culture”. With Museveni’s treacherous populist refugee policy, what can stop Rwandans faced with land shortage back in Rwanda from coming to Uganda claiming to be refugees from Burundi or Banyamulenge of Congo.
In 2012 the government set up a committee to discuss naturalisation for refugees especially Rwandese and Congolese of 1990s and 1960s lot respectively. The Director of Refugees stated that at least 5,000 refugees had applied for naturalisation. In January 2015, the Ministry of Internal Affairs ran a countrywide awareness campaign to enable any non-citizens that meet the conditions stipulated under S.14 of the Uganda Citizenship and immigration Control Act to become citizens. The Minister made it clear that refugees were not eligible to apply. In both instances, nothing concrete was arrived at and the scheme was simply abandoned.
In 2012, Museveni and Kagame met members of the Banyarwanda community in Uganda (UMUBANO) at State House in Kampala. They were feuding over leadership with one faction recognising Donant Kananura while another one recognized Dr. Eric Kyamuhangire who also doubles as a Senior Presidential Advisor on Culture.
The Kananura group was accused of extorting money from Kagame under the guise of assisting Banyarwanda in Uganda. Museveni promised to mediate in resolving the leadership wrangles. On his part, Kagame advised the Banyarwanda not to focus on those small issues of leadership but work for their dominance of the two countries.
In 2013 Tanzania expelled Rwandese who were illegally staying in Kagera region back to Rwanda. Thousands of them chose to come to Uganda where the Museveni regime accorded them VIP reception, free land and citizenship. The Chairman of the Banyarwanda in Uganda who is also a Presidential Advisor on Culture, Dr. Eric Kyamuhangire wrote a long missive in The New Vision condemning Tanzania’s action. He argued that the people expelled were Banyarwanda who had migrated there in the 1970s and 80s in search of pasture and were therefore both Ugandans and Tanzanians because that region is suitable for cattle grazing.
The expulsion also fermented serious friction between Museveni and Kagame on one hand and Tanzania’s Jakaya Kikwete. Kyamuhangire’s argument confirms the belief by some people that the so-called cattle corridor is for their exclusive occupation. How come they couldn’t trace their previous homes if they were Ugandans? In July 2015 Museveni paid a visit to a one Johnson Nyinondi in Tanzania’s Kagera region whom he described as his relative who had helped a lot in intelligence gathering and recruitment of fighters against the Iddi Amin regime.
In April 2016, the then Minister of Internal Affairs, Rose Akol visited Mirama Hill border post at the Uganda-Rwanda border. She ordered Rwandan nationals in Uganda who were holding both Ugandan and Rwandese national IDs to surrender one. During the same occasion the DPC Ntungamo and Immigration Officers disclosed that the irregularity was making it difficult to fight cross border crime. Immigration Officers further revealed that they were confiscating an average of ten Ugandan IDs per day from Rwandan travellers.
Given the porous borderline, it is obvious that many more Uganda ID card holders move freely to and from the two countries. During the 2016 general elections in Uganda, the opposition decried that the Museveni regime had been issuing national IDs to Rwandans in order to enable them to vote for Museveni.
The Minister’s stand did not go well with the regime agenda and she was not only relieved of her ministerial position but made to lose in the parliamentary elections. Death of high Aronda Nyakairima is allegedly linked to this project
It’s no wonder that even the Uganda national ID is institutionally baptized ENDAGAMUNTU vs INDANGAMUNTU – a Kinyarwanda word for Rwandan national ID.
Last week the Rwandan High Commissioner to Uganda called upon Rwandans residing in Uganda to get ready to cast their vote at the embassy during the forthcoming August general elections. He disclosed that there are 55,000 Rwandans in the diaspora of which about 6,000 live in Uganda.
From the above long narration, it can be authoritatively argued that the so called Banyarwanda include all the Rwandans who find themselves in Uganda including the Rwandese embassy staff. They are bonafide citizens of Rwanda and Museveni’s Uganda (they vote Kagame and Museven)
The spirit of the constitution was manipulated to cover up for a wider scheme for Tutsi domination of Uganda. The recognition of Banyarwanda as one of the indigenous communities of Uganda was not meant for the migrant Banyarwanda who came to Uganda at the beginning of the last century.
Moreover, this category had already integrated into the local Ugandan society and those from Kisoro district who were made part of Uganda by colonialists have their own identity as Bafumbira. Therefore, the Banyarwanda of Uganda are the former Tutsi refugees of 1960s and 70s and the Tutsi immigrants both of whom have found two homes in both Uganda and Rwanda. They are bent on systematically achieving dominance and have the political and financial muscle to achieve this.
Currently all Banyarwanda wherever they are in Uganda are secretly registered by highly classified officials. They are given money for upkeep, pay fees for their children in big schools and even given start up funds to do different projects in their areas especially buying massive land/properties. By design, all Banyarwanda’s in Uganda whenever they are, are paid state agents/spies.
The Banyarwanda’s are registered in each sub-county and mandated to hold highly classified monthly get together meeting once in that sub-county. They do this so secretly that local community members never know, yet even if one of them is a house girl in your home knows of this meeting and attends it.
Under the guise of modernization, land reforms and commercial farming, Museveni’s scheme is to deprive indigenous communities of their land, hoard them into concentration camps in urban centres so that they lose their respective community identities to Banyarwanda.
That is the price of the false so called national unity and region integration

The Rwandan’s land grabbing strategy since 1950 [part 3]
With military, financial, political and administrative powers achieved, the Rwandese are at the last part of occupation using either force, money or both against indigenous communities
This is the 3rd and last part of the series The Rwandan’s occupation and land grabbing strategy since 1950 In the series we explore how the Rwandans who came seeking for refuges in the 1950’s turned Uganda’s indigenous communities into refuges in their country by taking over their political, economic, social and now land heritage.
This is a long article detailing the history and facts linking Museveni Tutsi/Rwanda merciless strategies to grab land and literally wipe out all indigenous societies in the above region.
This part looks at their current level of settlement and forcefully wiping out indigenous communities [Forceful and Legalizing land grabbing]

The Banyarwanda’s are registered at each sub-county and mandated to hold highly classified monthly get together meeting. They do this so secretly that local community members never know, yet even if one of them is a house girl in your home knows of this meeting and attends it.
By 1991 it was estimated that Banyarwanda in Uganda were 1.3M out of a total population of 18M in the country. Of these, 450,000 were Uganda indigenous Banyarwanda who became Ugandans after Kisoro became part of Uganda in 1918 and 650,000 were the Banyarwanda economic immigrants who came to look for work and a few who came to look for pasture for their cattle between 1926 and 1959 of whom 84,000 were the Banyarwanda Tutsi refugees registered under UNHCR.
Many had left the refugee settlement camps and integrated into local society. In 1993 the then Minister of Local government, Dr. Steven Chebrot estimated that about 300,000 Banyarwanda refugees were spontaneously resettled amongst the locals outside the camps.
The able bodied had left the camps to the elderly and settled in urban centres while others had acquired land outside camps and settled there using money provided by government. The UNHCR had handed the management of the camps to the government though food aid continued coming in. Even within the camps, the refugees had become self sustaining by producing enough to feed themselves and surplus for sale.
In July 1994 when the RPF took over power in Rwanda, a reconnaissance party of Banyarwanda civilians left for Rwanda so as to get hold on political fortunes. By end of 1995, an estimated 226,000 Banyarwanda refugees had returned to Rwanda. They deserted the refugee camps and the countryside as far as Ngoma, Kyankwanzi, Masindi, Nakasongola and Luwero and moved en masse and headed for Rwanda.
As the Tutsi were leaving Uganda for Rwanda, the Hutu were leaving Rwanda in millions for Tanzania and Congo. A small contingent of 11,000 Hutu entered Uganda and were camped in Kisoro and Ntungamo before being transferred to Oruchinga camp. A departing Tutsi at Oruchinga camp attempted to spear a child of an arriving Hutu but only to injure his fellow Tutsi.
The 1993 Arusha Accord between the Tutsi RPF rebels and the Hutu government of Rwanda had stipulated that returning refugees after ten years were not to seek to reclaim previous properties but were to be resettled on unoccupied land. Once the RPF took over power, the returning Tutsi occupied property left behind by fleeing Hutu. However, as time went on, reconciliation efforts dictated that returning Hutu ought to reoccupy their property. The issue was so controversial to an extent that it accounted for the false accusations of genocide against some Hutu just as a way of keeping them off the property.
To strike a compromise, a villagisation (Imidugudu) scheme where services would be centralized and modern agricultural technology made accessible was initiated. The international development partners supported the construction of Imidugudu. Meeting land and housing needs of returning refugees proved quite a challenge. The situation was worse with the pastoralists Tutsi who needed huge chunks of land for their cattle.
Many returned and settled in Uganda where grazing land was plenty. Those who had anticipated a land of milk and honey, they too were disappointed and retreated to Uganda where land, availability of social services, and economic opportunities including employment were easily forthcoming more than in their new-homeland. For the majority of others who had not left for Rwanda, they opted to stay in Uganda. Even the elites in influential positions in Rwanda have been acquiring land in Uganda through proxies.
With the sound availability of cash and government favors, the so called Balaalo have evolved from the traditional pastoralist and casual laborers to owners of thousands of heads of exotic cattle, huge chunks of land, are armed and determined to spread to as far as West Nile and Acholi at the border with Sudan.
Museveni managed to push through the 1995 Constitution a provision that recognised Banyarwanda as one of the 56 indigenous ethnic communities. It was not made clear if the constitutional Banyarwanda ethnic group referred to the Banyarwanda who were added into Uganda by colonial boundaries in Kisoro, the economic immigrants of the early 20th century who came to look for work, or those who came as refugees during the 1959 – 1974 exodus escaping political turmoil. By 2017 Rwanda had a polling station in Uganda, whose eligibility none knows whether Ugandan Banyarwanda voted or not.
In October 2001, the Uganda government announced that it was probing the composition of 1,252 army Cadet Officers after discovering that several Rwandan undercover spies posing as Ugandans had been recruited for training. The then Army Spokesman, Col. Keitirima confirmed that they had also turned away several Rwandans who had sought to be recruited.
In July 2002, a delegation of 30 Banyarwanda elders met Museveni for a petition over the alleges of harassment of Banyarwanda by security agencies. They claimed that over twenty Banyarwanda were being held in safe houses by intelligence services. This was during the time when relations between Uganda and Rwanda were sour following the bloody clash of the two armies in Congo. To the security agencies, the detained Banyarwanda were Rwandese spies. Appearing on Andrew Mwenda Live radio talk show, Kagame had complained that; “I was told how Rwandese are being arrested in Kampala and in Kikuko wherever they are found.”
On 17th June 2007, the government owned New Vision ran a letter by one Mutesi of Kabale in which she was complaining about Banyarwanda being denied passports. That she had had to consult her friends in security services who advised her not to state that she was a Munyarwanda but should instead call herself a Mufumbira.
She went ahead to disclose that since she had worked in Kisoro as a teacher, she travelled there and using the voters card she filled forms, had them approved by the LCs and the area M.P and eventually got her passport. She disclosed that her grandfather had migrated from Rwanda to Tanzania 100 years ago, and that her father relocated to Uganda, died and was buried in Ntungamo, Uganda.
In 2007, the Local Government of Kyankwanzi district offered to take in and accommodate the 100 Banyarwanda Balaalo families that had been evicted from Buliisa following clashes with Bagungu. The move was spearheaded by the LC3 Chairman, Fred Mpora and escorted by Police. Fred Mpora has been a key figure in the incursions by Balaalo into Acholi.
In April 2008 Banyarwanda living in Uganda under their umbrella organisation, UMUBANO held the annual assembly at Lugogo Indoor Stadium under the chairmanship of Erick Kyamuhangire who is Museveni’s Senior Presidential Advisor on Culture. The assembly expressed grave concern over denial of passports, loss of land, denial of recruitment into public service, denial of scholarships and general discrimination by government.
Their Chairman lamented thus; “…. if one happens to have a contact in Rwanda, then it’s enough reason for disqualification.” They gave the example of Balalo who were being evicted in Masindi describing it as paid harassment before accusing the government of being biased against the Balaalo.
Note: one of the key state witnesses in the Kyadondo terror attack, Muhammad Mugisha claimed to be a Ugandan who was born in Rwanda but relocated to Uganda in 1998.
In 2009, the Uganda Citizenship and Immigration Control Act was amended to put it in line with the constitutional provision on Banyarwanda Tutsi. Section.12 provides for citizenship by birth for any person born in Uganda whose parents or grandparents is or was a member of any of the indigenous communities existing in and residing within the borders of Uganda as at the first day of February 1926, as set out in the Third Schedule to the 1995 constitution.
It went ahead to stipulate that every person born in or outside Uganda one of whose parents or grandparents was at the time of birth of that person a citizen of Uganda by birth, is eligible for citizenship. S.14 (1) (a) (ii) eliminates Hutu refugees from accessing citizenship thus; “every person born in Uganda who at the time of birth neither of his or her parents and none of his grandparents was a refugee in Uganda”.
Since it’s now the Hutu who are refugees, the provision bars them. S.14 (b) grants citizenship to anyone who has continuously lived in Uganda since 9th October 1962.
Interestingly, the Act under S.14 (2) (b) gives a blank cheque to Tutsi Banyarwanda who are always on the move migrating to and settling in Uganda thus ” any person who has legally and voluntarily migrated to Uganda and has been living in Uganda for at least twenty years is eligible for citizenship”.
The other requirements include a good command of the English language or ‘prescribed local language’.
In July 2010, the Museveni regime in connivance with the Kagame regime in Kigali forcefully returned to Rwanda 1,700 Hutu Asylum seeker and refugees. The victims have escaped Gacaca community courts, land wrangles and general repression.
Note: The UPC government was accused of forcefully returning Tutsi refugees to Rwanda in the early 1980s but in the instant case it looked okay because the victims were Hutu. In 2010, a group of Banyarwanda refugees in Uganda petitioned the Constitutional Court over acquisition of citizenship. They claimed that the Immigration Department was refusing to give them citizenship application forms on grounds that they were not eligible on account of their being refugees.
They based their petition on a constitutional provision that made refugees eligible for citizenship by naturalisation and registration. Article 12 (2) (c) and 14 (2) (c) provides that “a person who on commencement of this constitution has lived in Uganda for the last 20 years is eligible for citizenship by registration”. The petitioners had lived in Uganda since 1985. In October 2015 court ruled that refugees were eligible for citizenship not by registration but naturalisation but two of the petitioners had already been relocated to the USA by UNHCR while the 3rd could not be traces.
The 2013 Land Policy condemns those who classify “cross border population movement as refugees or internally displaced people because of shared common heritage and culture”. With Museveni’s treacherous populist refugee policy, what can stop Rwandans faced with land shortage back in Rwanda from coming to Uganda claiming to be refugees from Burundi or Banyamulenge of Congo.
In 2012 the government set up a committee to discuss naturalisation for refugees especially Rwandese and Congolese of 1990s and 1960s lot respectively. The Director of Refugees stated that at least 5,000 refugees had applied for naturalisation. In January 2015, the Ministry of Internal Affairs ran a countrywide awareness campaign to enable any non-citizens that meet the conditions stipulated under S.14 of the Uganda Citizenship and immigration Control Act to become citizens. The Minister made it clear that refugees were not eligible to apply. In both instances, nothing concrete was arrived at and the scheme was simply abandoned.
In 2012, Museveni and Kagame met members of the Banyarwanda community in Uganda (UMUBANO) at State House in Kampala. They were feuding over leadership with one faction recognising Donant Kananura while another one recognized Dr. Eric Kyamuhangire who also doubles as a Senior Presidential Advisor on Culture.
The Kananura group was accused of extorting money from Kagame under the guise of assisting Banyarwanda in Uganda. Museveni promised to mediate in resolving the leadership wrangles. On his part, Kagame advised the Banyarwanda not to focus on those small issues of leadership but work for their dominance of the two countries.
In 2013 Tanzania expelled Rwandese who were illegally staying in Kagera region back to Rwanda. Thousands of them chose to come to Uganda where the Museveni regime accorded them VIP reception, free land and citizenship. The Chairman of the Banyarwanda in Uganda who is also a Presidential Advisor on Culture, Dr. Eric Kyamuhangire wrote a long missive in The New Vision condemning Tanzania’s action. He argued that the people expelled were Banyarwanda who had migrated there in the 1970s and 80s in search of pasture and were therefore both Ugandans and Tanzanians because that region is suitable for cattle grazing.
The expulsion also fermented serious friction between Museveni and Kagame on one hand and Tanzania’s Jakaya Kikwete. Kyamuhangire’s argument confirms the belief by some people that the so-called cattle corridor is for their exclusive occupation. How come they couldn’t trace their previous homes if they were Ugandans? In July 2015 Museveni paid a visit to a one Johnson Nyinondi in Tanzania’s Kagera region whom he described as his relative who had helped a lot in intelligence gathering and recruitment of fighters against the Iddi Amin regime.
In April 2016, the then Minister of Internal Affairs, Rose Akol visited Mirama Hill border post at the Uganda-Rwanda border. She ordered Rwandan nationals in Uganda who were holding both Ugandan and Rwandese national IDs to surrender one. During the same occasion the DPC Ntungamo and Immigration Officers disclosed that the irregularity was making it difficult to fight cross border crime. Immigration Officers further revealed that they were confiscating an average of ten Ugandan IDs per day from Rwandan travellers.
Given the porous borderline, it is obvious that many more Uganda ID card holders move freely to and from the two countries. During the 2016 general elections in Uganda, the opposition decried that the Museveni regime had been issuing national IDs to Rwandans in order to enable them to vote for Museveni.
The Minister’s stand did not go well with the regime agenda and she was not only relieved of her ministerial position but made to lose in the parliamentary elections. Death of high Aronda Nyakairima is allegedly linked to this project
It’s no wonder that even the Uganda national ID is institutionally baptized ENDAGAMUNTU vs INDANGAMUNTU – a Kinyarwanda word for Rwandan national ID.
Last week the Rwandan High Commissioner to Uganda called upon Rwandans residing in Uganda to get ready to cast their vote at the embassy during the forthcoming August general elections. He disclosed that there are 55,000 Rwandans in the diaspora of which about 6,000 live in Uganda.
From the above long narration, it can be authoritatively argued that the so called Banyarwanda include all the Rwandans who find themselves in Uganda including the Rwandese embassy staff. They are bonafide citizens of Rwanda and Museveni’s Uganda (they vote Kagame and Museven)
The spirit of the constitution was manipulated to cover up for a wider scheme for Tutsi domination of Uganda. The recognition of Banyarwanda as one of the indigenous communities of Uganda was not meant for the migrant Banyarwanda who came to Uganda at the beginning of the last century.
Moreover, this category had already integrated into the local Ugandan society and those from Kisoro district who were made part of Uganda by colonialists have their own identity as Bafumbira. Therefore, the Banyarwanda of Uganda are the former Tutsi refugees of 1960s and 70s and the Tutsi immigrants both of whom have found two homes in both Uganda and Rwanda. They are bent on systematically achieving dominance and have the political and financial muscle to achieve this.
Currently all Banyarwanda wherever they are in Uganda are secretly registered by highly classified officials. They are given money for upkeep, pay fees for their children in big schools and even given start up funds to do different projects in their areas especially buying massive land/properties. By design, all Banyarwanda’s in Uganda whenever they are, are paid state agents/spies.
The Banyarwanda’s are registered in each sub-county and mandated to hold highly classified monthly get together meeting once in that sub-county. They do this so secretly that local community members never know, yet even if one of them is a house girl in your home knows of this meeting and attends it.
Under the guise of modernization, land reforms and commercial farming, Museveni’s scheme is to deprive indigenous communities of their land, hoard them into concentration camps in urban centres so that they lose their respective community identities to Banyarwanda.
That is the price of the false so called national unity and region integration
