Ever tried talking while doing a handstand? It's tricky. Your body works best the right way up. Just like cells in your body do. Cell shape, structure and how components inside cells are organised help them figure out their orientation – this is called cell polarity. Disrupted cell polarity can cause cancer and other diseases, including gut disorders. Cells lining your intestine are 'polarised' – with one side facing the gut tube interior (apical), the other the blood stream (basal) – along an apicobasal axis. In mice, RNA and proteins in these cells are asymmetrically spread along this axis. Researchers investigate if this happens in humans using laser-capture microdissection to isolate top and bottom segments from intestinal epithelial cells (pictured using H&E staining). Analysing the location of genes actively making proteins in these segments revealed that RNA and proteins are asymmetrically spread along this axis. This research provides a way to better understand the biology of the human intestine.

Do I know anyone here with a saltwater reef tank? Ever had to deal with a flatworm infestation?
I recently got some biopsies from Caribbean Acropora that were losing tissue and dying. These guys are full of flatworms, and while there are several species of flatworm described on corals in the Pacific and in aquaria on Pacific species, I can't find any references that mention a Caribbean variety, or even an Atlantic one, at all. It's pretty difficult to get an ID beyond "flatworm" from routine histology but I might get tissue to sequence using laser capture microdissection that I can compare to other known species.
I think these might be escapees from someone's hobby aquarium, just possibly. It's also possible that they are a native species no one noticed before but the fact that they are such a big aquarium pest has me worried.
This is why we should never dump an aquarium into wild waters or release any kind of pet that isn't native, or even that is but has had contact with non-native species, into the wild. You never know what else you're unleashing. You might be killing coral reefs.

Refining Spatial Proteomics by Mass Spectrometry: An Efficient Workflow Tailored for Archival Tissue
Formalin-fixed paraffin-embedded (FFPE) tissue, while excellent for preserving tissue for extended periods of time, poses a challenge when extracting molecular information. We therefore developed an easily adaptable and highly efficient workflow for extracting high levels of proteins from low-input material. We compared sensitivity between two stains, EpCAM and H&E, across material inputs of ~2 and 1,600 cells. In the context of EpCAM-stained tissue, our investigations unveiled a range from ~1,200 unique protein groups at the lowest input to ~5,900 at the highest. For H&E, the spectrum covers ~900 to ~5,200 protein groups. We found an optimal balance between maximizing detected proteins and minimizing input material to be within the range of ~100 to ~200 cells. With this knowledge, we tested the spatial capabilities by isolating specific cell populations, through Laser Capture Microdissection (LCM), from three different tissue types, where we were able to identify tissue-specific signatures and prominent clustering of all cell populations. http://dlvr.it/T23m3p

Cells, Vol. 12, Pages 2478: Eye Lens Organoids Made Simple: Characterization of a New Three-Dimensional Organoid Model for Lens Development and Pathology
Cataract, the opacification of the lens, is the leading cause of blindness worldwide. Although effective, cataract surgery is costly and can lead to complications. Toward identifying alternate treatments, it is imperative to develop organoid models relevant for lens studies and drug screening. Here, we demonstrate that by culturing mouse lens epithelial cells under defined three-dimensional (3D) culture conditions, it is possible to generate organoids that display optical properties and recapitulate many aspects of lens organization and biology. These organoids can be rapidly produced in large amounts. High-throughput #RNA sequencing (#RNA-seq) on specific organoid regions isolated via laser capture microdissection (LCM) and immunofluorescence assays demonstrate that these lens organoids display a spatiotemporal expression of key lens genes, e.g., Jag1, Pax6, Prox1, Hsf4 and Cryab. Further, these lens organoids are amenable to the induction of opacities. Finally, the knockdown of a cataract-linked #RNA-binding protein encoding gene, Celf1, induces opacities in these organoids, indicating their use in rapidly screening for genes that are functionally relevant to lens biology and cataract.
