I'm kinda confused. What exactly is a yokai? Is there any instance of yokai and human falling in love in Japanese folklore or is that an anime thing?

There are three major supernatural groups in Japan, the Kami/Gods, the Yurei/Ghosts and the youkai. 

There is no clear definition of what a youkai is and depending on who’s book you read there are many attempts to clarify it. Basically youkai is a blanket term that covers many kinds of supernatural entities. Some are dead, some are monsters in fairy tales and some are born from humans evil deeds. 

The best western comparison I can give for youkai is Faeries and elves. Not all are evil but very very few of them are benevolent. The folklore of each in turn carry the same message: You don’t want to get involved with them!

Of course that being said there is folklore and tales of humans and youkai falling in love. 99.9% of them do not end well. Many of these stories involve Kitsune and Obakineko who can shape shift to look like humans and in turn marry unsuspecting lovers. 

Then there are more cautionary tales such as the Futa-kuchi-onna, a youkai who looks like an ordinary woman with a second, voracious mouth hidden behind her head. A very stingy but rich man did not want a wife because he didn’t want to pay for another mouth to feed. So when he learned of a poor village woman who did not eat anything yet could work very hard, he demanded to marry her! Little knowing that when he left the house, she would let her hair loose and the vicious mouth would devour all the food in the pantry! 

Not realizing what was going on, the scrooge suspected his wife was giving the food away to the poor and one day he spied on his wife to see for himself. When he discovered what she truly was, the Futa-Kuchi-onna ensnared him with her hair and that gruesome mouth devoured him! 

So no, much of the romantic stuff is really just Anime plot tropes at best, but that doesn’t mean the actual folklore isn’t entertaining as well. 

If you want to learn about some folktales involving youkai, there is a book titled: Tales of Old Japan by A.B. Mitford.
