Canine peptides
matrix metalloproteinases (MMP) induced by the alteration of the proteolysis of proteins of the extracellular matrix and the basement membrane is the key to tumor progression and metastasis. metalloproteinase-7 (matrilysin matrix), the smallest member of the MMP family also performs quite similar; therefore serves as a potential candidate for anti-tumor immunotherapy.
Conversely, being an antigen associated with an endogenous tumor (TAA), targeting MMP-7 for immunization is difficult. But MMP-7 xenovaccine base can overcome the obstacle of low immunogenicity and immune tolerance, often encountered in conventional vaccines based TAA for anti-tumor immunotherapy. This opens the way for the study of the potential of MMP-7 derived from major histocompatibility complex (MHC) binding peptides to elicit responses of specific T cells specific epitope for possible inclusion in formulations anti-tumor vaccines.
Perhaps also paves the way towards achieving broad-based multi-epitope and universal cellular immunity. In the current experiment, a immunoinformatics approach was used to identify the putative canine matrix matelloproteinases-7 (CMMP-7) derived peptides with class I MHC motifs binding that can elicit antigen-specific immune responses powerful in BALB / c.
Immunization with CMMP-7 DNA vaccine induces a strong CD8 + cytotoxic T lymphocytes (CTL) and Th1 type response with a high level of production of interferon gamma (IFN-γ) in mice BALB / c . Both peptides nonameric putative MHC-I-binding identified (Peptide32-40 and Peptide175-183) of CMMP-7 induces proliferation of lymphocytes with the significant production of IFN-γ from CD8 + T lymphocytes in mice immunized with a DNA vaccine CMMP-7.
The current observation represented the immunogenic potential of the two nonapeptides CMMP-7-derivatives for their possible use in anti-tumor immunotherapy mediated xenovaccine in the mouse model.
