PART 4 - Drum Magazine, May 2013 ; Sean Kinney: From The Ashes After the VH1 concert, they did a few club dates around the U.S., Europe, and Japan, with DuVall’s place in the lineup becoming more concrete all the time. The stars seemed to be aligning in their favor, for the first time in a long time. But for Kinney, one unfinished piece of business remained. And until he could resolve it, it posed a fatal threat to whatever future this new incarnation might have. BREAKING THE CHAINS Staley’s death, says Kinney, was a “pinnacle moment,” forcing him to pull back on the reins of his own destructive impulses. But with each passing year the list continued to grow of friends whose lives had been claimed under similar circumstances (including, chillingly, one friend he’d just learned had hung himself the night before our interview). “I live everyday missing a lot of people and, you know, ten times a day with knots in my stomach missing somebody that was like a brother to me,” Kinney says. Still, none of it had been enough to scare him totally straight. Not yet. That moment finally came in July 2007, a couple of weeks before they were set to go out on a high-profile tour with Velvet Revolver. “I just couldn’t feel right about what was going on because of what I was still doing and I didn’t have a way out and I damn-near OD’ed again and just about died, which wasn’t the first time. And it just kind of hit me, it’s like, ’You can’t do this. How important is this?’ It’s so important to me to pass on our legacy to have Layne’s music and this music remembered, and to honor the fact of what we had done. And like anything, when s–tty things happen, you’re going to get knocked down. But you’ve got to get up at some point. "UniCAR"-modified off-the-shelf NK-92 cells for targeting of GD2-expressing tumour cells.
Related Articles "UniCAR"-modified off-the-shelf NK-92 cells for targeting of GD2-expressing tumour cells. Sci Rep. 2020 Feb 07;10(1):2141 Authors: Mitwasi N, Feldmann A, Arndt C, Koristka S, Berndt N, Jureczek J, Loureiro LR, Bergmann R, Máthé D, Hegedüs N, Kovács T, Zhang C, Oberoi P, Jäger E, Seliger B, Rössig C, Temme A, Eitler J, Tonn T, Schmitz M, Hassel JC, Jäger D, Wels WS, Bachmann M Abstract Antigen-specific redirection of immune effector cells with chimeric antigen receptors (CARs) demonstrated high therapeutic potential for targeting cancers of different origins. Beside CAR-T cells, natural killer (NK) cells represent promising alternative effectors that can be combined with CAR technology. Unlike T cells, primary NK cells and the NK cell line NK-92 can be applied as allogeneic off-the-shelf products with a reduced risk of toxicities. We previously established a modular universal CAR (UniCAR) platform which consists of UniCAR-expressing immune cells that cannot recognize target antigens directly but are redirected by a tumour-specific target module (TM). The TM contains an antigen-binding moiety fused to a peptide epitope which is recognized by the UniCAR molecule, thereby allowing an on/off switch of CAR activity, and facilitating flexible targeting of various tumour antigens depending on the presence and specificity of the TM. Here, we provide proof of concept that it is feasible to generate a universal off-the-shelf cellular therapeutic based on UniCAR NK-92 cells targeted to tumours expressing the disialoganglioside GD2 by GD2-specific TMs that are either based on an antibody-derived single-chain fragment variable (scFv) or an IgG4 backbone. Redirected UniCAR NK-92 cells induced specific killing of GD2-expressing cells in vitro and in vivo, associated with enhanced production of interferon-γ. Analysis of radiolabelled proteins demonstrated that the IgG4-based format increased the in vivo half-life of the TM markedly in comparison to the scFv-based molecule. In summary, UniCAR NK-92 cells represent a universal off-the-shelf platform that is highly effective and flexible, allowing the use of different TM formats for specific tumour targeting. [ Authors ] Edward Farhi, Shelby Kimmel, Kristan Temme [ Abstract ] We study a quantum algorithm that consists of a simple quantum Markov process, and we analyze its behavior on restricted versions of Quantum 2-SAT. We prove that the algorithm solves this decision problem with high probability for n qubits, L clauses, and promise gap c in time O(n^2 L^2 c^{-2}). If the Hamiltonian is additionally polynomially gapped, our algorithm efficiently produces a state that has high overlap with the satisfying subspace. The Markov process we study is a quantum analogue of Sch\"oning's probabilistic algorithm for k-SAT.

21 Ideas for Wild Statement Ceilings
Usually, the ceiling in any space goes fairly unnoticed. Seen as just a way to hide insulation, roofs and attics, ceilings tend to get painted a light color and then left alone. But if you’re looking to create a space that really pops, you might want to consider adding some artistic flair to the ceiling. Wild statement ceilings come in all types, from funky geometric designs to creative colors to three-dimensional creations. Rather than letting the ceiling fade into the background, take a look at the statement ceilings below to find an idea that might just be part of your next room remodel.
Three-dimensional statement ceilings that literally pop
One of the most popular ways to create statement ceilings is to choose three-dimensional designs. Three-dimensional ceilings could be as simple as raised beams around lighting fixtures, with traditional white or cream paint to keep things subtle. However, you also have the option of going wild by incorporating bold colors your design. Either way, these ideas are wonderful ways to add visual interest to a room and customize your living space.

Geometric statement ceilings
Another way to get statement ceilings is to seek out geometric designs that draw the eye. Many of these also have a three-dimensional element; however, the dimensions tend to be flatter, and the focus is more on the geometry of the design.
Many geometric designs make use of beams to create an eye-catching pattern. Other designs simply use cleverly placed paneling. Still others use paint, making them a more accessible option if you’re looking to update a space without a significant overhaul in structure and design.

You can also paint around the ceiling for a geometric design. Image: Olga Adler Interiors
Fun statement ceiling colors
The easiest way to get a statement ceiling is to paint the ceiling a bright color. The color of the ceiling tends to match accent walls or elements in the room décor. In that way, you can use your statement ceiling to create cohesiveness in the room. However, you can also use a statement ceiling as a point of contrast for a space with a bright, artsy flair. Take a look at some of the photos below for bright statement ceiling ideas. (And you can see additional painted ceiling ideas here.)

Artistic statement ceilings
Yet another option for statement ceilings is to go for artistic vistas. These can be geometric patterns, natural flowers, foliage, a skyscape and much more. It’s a great way to bring unique art into the space. And if you get creative enough, the ceiling painting can be part of the room’s larger theme. Some designs even make use of an added light fixture or art element that goes with the painted design. And even if you don’t have an artist’s touch, you can commission someone to do these designs for you.

ICO Analysis: Grain
As human capital continues to rise worldwide and supply chains gain international footprints, companies are increasing looking for ways to have cross-border collaboration in terms of employment and freelance work. However, current payroll processes create regulatory and compliance hurdles and also come with significant currency exchange fees when companies look to hire abroad.
// -- Discuss and ask questions in our community on Workplace. One company, Grain is looking to revolutionize the current payroll process by moving it to a blockchain-enabled platform that reduces operational costs and third-party vendor costs as well as currency exchange fees.
Token The Grain token (GRAIN) will be implemented using the Ethereum ERC20. GRAIN holders generate value from the token in a single distinct way: the payment of labor contracts facilitated on Grain’s protocol. The premise of the GRAIN token is based on its utility to fuel the smart contracts implemented on Grain’s blockchain – GRAIN “is able to facilitate transactions for work in a safe and transparent manner.
According to the white paper, 35% of the funds raised will go towards growing the development team (tech HR), 35% towards marketing and business development, 20% towards reserves for unforeseen costs, 7% towards operational costs, 2% towards third-party legal and security providers, with the remainder of the pot (2%) going towards token sale costs.
// -- Become a yearly Platinum Member and save 69 USD and get access to our secret group on Workplace. Click here to change your current membership -- // The GRAIN tokens are valued at 0.02778 EURO per 1 GRAIN. The total amount of tokens to be sold is capped at 3,600,000,000 which equals 30,000,000 euros.
The company intends to list the GRAIN tokens on major crypto exchanges by Q2 2018.
Team Grain’s core team consists of three senior professionals, two of which have backgrounds at Microsoft and the other with a background in blockchain consultancy. As compared with the majority of ICOs, the team has a light staff and is lacking a strong development team.
The company’s CEO & President, Onno Hektor is a former Senior Director for Microsoft. While Onno’s Microsoft background is great, there’s a red flag concerning his current roles. According to Onno’s LinkedIn, he’s currently the co-founder of NG Group, tao.company, Atlantic Crossings, and FleXentral. Onno is also currently the Executive Vice President of EMEA at Sitrion. The company’s Board Member, Andre Bonvanie also has a background at Microsoft. Yet, the red flags concerning Andre’s current roles are consistent with Onno’s. The company’s Blockchain Strategist, Erik Koster is currently the CEO of Global Screen.
The company’s advisory board consists of Temme Sikkema (a professor at the University of Groningen), Roberto de Freitas (an actuarial risk management specialist), and René Hendriks (a serial entrepreneur of several early-stage start-ups).
Verdict Grain presents a highly speculative buying opportunity for investors interested in long-term capital appreciation.
While two of the members of Grain’s core team have a Microsoft background, their time is currently invested elsewhere (multiple positions at various companies). Another red flag is that the company doesn’t have a dedicated development team – how can you launch a blockchain company without a team of developers and engineers? The company’s advisory board is essentially a joke – they don’t bring anything of value to the table.
The concept behind having a blockchain-based platform to pay employees and freelancers is great. However, the cautionary tape surrounding Grain as a company and specifically its ICO is worrisome. Tread cautiously if you’re considering an investment.
Risks Grain suffers from a high amount of technology risk. From a bare bones perspective, Grain doesn’t have a dedicated development and engineering team. The company has stated that it won’t have a team of 10 developers until Q4 2018 – meaning there’s a significant drag on product development. Additionally, 30 million euros is too much money for a company that has yet to fully develop a product and go through beta testing. -2.5 While the concept of blockchain technology underpinning payroll for employees and freelancers is sound, it’s likely that this product is too early for the market. Blockchain technology is still in its early-stages in terms of enterprise adoption. Many of the large companies in the EEA (including Microsoft) are developing their own private Ethereum-based blockchains for internal use. It’s likely that if they wanted a product like Grain, it would be cheaper in the long-run for them to develop this concept internally. -2.5 With Grain’s core team juggling a variety of positions at various companies, execution risk runs extremely high. Launching a start-up (especially in the blockchain space) is time consuming and requires complete devotion. Without devotion and focused effort it’s likely this company will only reach limited scale. -2 Growth Opportunity The movement towards global, distributed teams is a positive trend that Grain may benefit from. As companies continue to embrace resource arbitrage and reap the economic benefits that come with it, a centralized third-party provider for both payroll and freelancer payments will be needed. +4 Since two members of Grain’s core team have worked at Microsoft in the past, the company has the potential to engage in a Microsoft partnership. Microsoft has been a strong backer of ethereum-based solutions for enterprises (as seen by its current internal efforts and cloud offerings). If Grain is able to secure a Microsoft partnership, it would open the doors for acceptance by a variety of smaller SMBs and other large enterprises. +3 Provided the company is able to develop a fully-fledged product, Grain would likely become a potential acquisition opportunity by indirect competitors, think: Upwork, freelancer.com, Toptal, etc. Upwork acquired Elance in the past and will likely want to move to the blockchain over time – an acquisition would be the easiest way to implement this. +3 Disposition Moving payroll for employees and payments for freelancers to the blockchain is an exciting idea that will likely take hold over the next five years. However, Grain isn’t likely to be the company leading this revolutionary change. There are too many risks (including: technology, adoptiopn, and execution) surrounding this company.
If Grain had a dedicated team and was raising a 3-5 million euro Series A through the traditional VC route, then maybe this company would have a chance. However, the company is raising a 30 million euro ICO for which a token isn’t even warranted yet (there’s no functioning product).
Tread lightly if you’re considering investing.
Against this backdrop, we believe that a score of 3.0 out of 10 is warranted.

Businesses cautious in installing patches to fix chip flaw
New Post has been published on https://new800numbers.com/business/businesses-cautious-in-installing-patches-to-fix-chip-flaw/
Businesses cautious in installing patches to fix chip flaw
Local vanity Numbers:
(Reuters) – Chances that a fix to a major microchip security flaw may slow down or crash some computer systems are leading some businesses to hold off installing software patches, fearing the cure may be worse than the original problem.
Researchers this week revealed security problems with chips from Intel Corp (INTC.O) and many of its rivals, sending businesses, governments and consumers scrambling to understand the extent of the threat and the cost of fixes.
Rather than rushing to put on patches, a costly and time-intensive endeavor for major systems, some businesses are testing the fix, leaving their machines vulnerable.
“If you start applying patches across your whole fleet without doing proper testing, you could cause systems to crash, essentially putting all of your employees out of work,” said Ben Johnson, co-founder of cyber-security startup Obsidian.
Banks and other financial institutions spent much of the week studying the vulnerabilities, said Greg Temm, chief information risk officer with the Financial Services Financial Services Information Sharing and Analysis Center, an industry group that shares data on emerging cyber threats.
The flaws affect virtually all computers and mobile devices, but are not considered “critical” because there is no evidence that hackers have figured out how to exploit them, said Temm, whose group works with many of the world’s largest banks.
“It’s like getting a diagnosis of high blood pressure, but not having a cardiac arrest,” Temm said. “We’re taking it seriously, but it’s not something that is killing us.”
Banks are testing the patches to see if they slow operations and, if so, what changes need to be made, Temm said. For instance, computers could be added to networks to make up for the lack of processor speed in individual machines, he added.
Some popular antivirus software programs are incompatible with the software updates, causing desktop and laptop computers to freeze up and show a “blue screen of death,” researcher Johnson said.
Antivirus software makers responded by rolling out fixes to make their products compatible with the updated operating systems, he said. In a blog posting on Friday, Microsoft Corp (MSFT.O) said it would only offer security patches to Windows customers whose antivirus software suppliers had confirmed with Microsoft that the patch would not crash the customer’s machine.
“If you have not been offered the security update, you may be running incompatible antivirus software, and you should consult the software vendor,” Microsoft advised in the blog post.
Government agencies also are watching. The Ohio Attorney General’s office is monitoring the situation, a spokesman said by email.
“Intel continues to believe that the performance impact of these updates is highly workload-dependent and, for the average computer user, should not be significant and will be mitigated over time,” the world’s No. 1 chipmaker said on Thursday in a release.
It cited Amazon.com Inc (AMZN.O), Apple Inc (AAPL.O), Alphabet Inc’s (GOOGL.O) and Microsoft as saying that most users had seen no significant impact on performance after installing the patches.
The cloud vendors are among a group of firms that quickly patched their technology to mitigate against the threat from one of those vulnerabilities, dubbed Meltdown, which only affects machines running Intel chips.
Major software makers have not issued patches to protect against the second vulnerability, dubbed Spectre, which affects nearly all computer chips made in the last decade, including those from Intel, Advanced Micro Devices Inc (AMD.O), and ARM-architecture manufacturers, including Qualcomm Inc (QCOM.O).
However, Google, Firefox and Microsoft have implemented measures in most web browsers to stop hackers from launching remote attacks using Spectre.
Governments and security experts say they have seen no cyber attacks seeking to exploit either vulnerability, though they expect attempts by hackers as they digest technical data about the security flaws.
One key risk is that hackers will develop code that can infect the personal computers of people visiting malicious websites, said Chris Wysopal, chief technology officer of cyber security firm Veracode.
He advised PC owners to install the patches to protect against such potential attacks. Computer servers at large enterprises are less at risk, he said, because those systems are not used to surf the web and can only be infected in a Meltdown attack if a hacker has already breached that network.
Microsoft has issued a patch for its Windows operating system, and Apple desktop users with the most recent operating system are protected. Google has said most of its Chromebook laptops are already protected and that the rest would be soon.
Apple said it planned to release a patch to its Safari web browser within coming days to protect Mac and iOS users from Spectre.
While third-party browsers from Google and others can protect Mac users from Spectre, all major web browsers for Apple’s iOS devices depend on receiving a patch from Apple. Until then, hundreds of millions of iPhone and iPad users will be exposed to potential Spectre attacks while browsing the Web.

Mono- and Polyassociation Processes of Pentavalent Biotinylated PEI Glycopolymers for the Fabrication of Biohybrid Structures with Targeting Properties.
Related Articles Mono- and Polyassociation Processes of Pentavalent Biotinylated PEI Glycopolymers for the Fabrication of Biohybrid Structures with Targeting Properties. Biomacromolecules. 2019 Aug 07;: Authors: Fingernagel J, Boye S, Kietz A, Höbel S, Wozniak K, Moreno S, Janke A, Lederer A, Aigner A, Temme A, Voit B, Appelhans D Abstract This study describes new mechanistic insights in the sequential polyassociation of streptavidin with biotinylated poly(ethyleneimine) glycopolymers and biotinylated PEGylated folic acid components for the preparation of biohybrid structures (BHS) for controlled targeting experiments. Characterization of the BHS revealed that during the formation and postfunctionalization of BHS, reversible dissociation and reassociation processes occur. The BHS are stable over weeks after finalizing the equilibrium-driven polyassociation process. Cellular uptake studies showed that this sequential polyassociation involving biotinylated PEGylated folic acid components does not lead to enhanced cellular uptake of the resulting BHS. In contrast, polyplexes, containing small interfering RNA and bioconjugates (1:1 molar ratio between biotinylated glycopolymer and monomeric streptavidin-lectin fusion protein), enabled us to control the targeting of tumor cells as revealed by knockdown of the tumor-associated protein survivin. Overall, this study demonstrates the high potential of (networklike) streptavidin-biotin interactions with a dynamic character in the formation of complex BHS and extracellular matrix materials. Please make me read these 10 books before (I die) 2018
Hello people! I come to you today, trying to make some sense of my ridiculously numbered (over 1100) TBR on Goodreads. God help me.
*sounds of perpetual screaming*
I know a lot of us have the problem of excessively adding to our TBR but believe me mine is worse. 1100(+) + the books I keep buying for some reason. And I hardly ever read physical books! (I read on eBook, maybe will do a post on that?) Yet, books are the only thing my mind justifies buying (with my money, other peoples money is for other trash). I’m not sure if my brain knows logic.
This means I have a SURPLUS of books I want to read. And the thing is I really want to? But alas life. So for this post, I’ve accumulated few books that I would really love to get to by the end of this year. But I probably won’t so screw me.
Sourdough by Robin Sloan
I came to know of this book, not more than a few days ago and doesn’t that tell you what kind of a person I am. Anywho this book is about fooood. And a secret society and a whirlwind of stuff. Colour me intrigued. I really like the idea of delving into a completely new world within the world we occupy which we think we know. The intricacies of little obsessive groups and how the meaning of a thing takes a completely new form for them is fascinating to me. Also, Sloan is the author of Mr Penumbra’s 24-Hour Bookstore which according to a review I read, is kinda like this but about book nerds. So yeah.
What I talk about when I talk about running by Haruki Murakami
Raven and I have forever been talking about hitting Murakami together. And this year, I hit it. But I read his short story collection, The elephant vanishes, and while I did like his writing and vibe, they very much left me unfulfilled because all his stories are about hyper vague things and end on hyper vague notes. I have a few stories still left to complete because by the end I was so sick of it. So I really want something meaningful from him. And this book sounds so up my alley. I think Ariel Bisset mentioned in one of her videos that this book might be about running in the title, but in fact alludes to much more, which is something I very much like. Also, this sounds like a very readable non-fiction book and yes, please.
A taste for monsters by Matthew J. Kirby
You need to read the synopsis of this, and you’ll get it.
Persepolis by Marjane Satrapi
I actually have the actual physical graphic novel of this and that does not happen very often. It’s so iconic and if I have it, WHY HAVEN’T I READ IT?? Also, want to watch the movie!
Any other works of Nina Lacour
Not long ago I read her We are okay and I fell very hard in love with it. And yes I’m a little terrified that her other books won’t do the same for me, but I still want that essence of another of her books so.
Radio Silence by Alice Oseman
Alice Oseman might be the cutest person on earth. And also I am completely certain her book is going to be great, so fight me. I also say also a lot.
Welcome to the monkey house by Kurt Vonnegut
It. is. high. time. i. read. some. Vonnegut!! When will I realize that owning a book is not the final step, R E A D I N G IT IS. (I was so happy that I brought my first Vonnegut, how does leaving it unread make sense to me? And yes, I do want to read Vonnegut because of the perception of it but how is it bad to want to know what’s behind that perception. Yes, I’m a blind sheep and I admit to it.)
All the Lives I Want: Essays about My Best Friends Who Happen to Be Famous Strangers by Alana Massey
I probably really need to read this book just because of my toxic brain but current celebrity culture is very interesting?
Eleanor Oliphant is completely fine by Gail Honeyman
This book sounds so cute! And fun! And some other things. My brain has turned to jelly.
The thing around my neck by Chimamanda Ngozi Adichie
The last time I walked into a bookstore (3 days ago) I picked up this and read a few pages. And I’ve been meaning to read some CNA for sooo long. And I liked it! So guess who’s going to go back and pick the book back up?
I’m going to end this by saying I just remembered another book which is The lovers dictionary which I want to read because Lorde highly praised it and also adding that I’m probably never actually getting to these books. So, yay me!
Real talk: I’m just really bad at reading. I read A LOT of news and articles because someone is in school to become a journalist or something and by the end, I’m just really drained and can’t handle anymore input. And also my brain keeps pulling the cord on me and parachuting off, which renders me a little incapable of doing anything. i.e. I’m left terrible at everything, not just reading. And I really wish I could say I should get points for trying, but most of the time I admit defeat long before that.
But sometimes, I appear!! And hoping is nice, right?
[Raven and I are currently buddy reading Turtles all the way down by the Green man. Aren’t you excited?? Temme below.]
Lots of love and perpetual angst,
Beez
