THE CORRUPTION.      insects.     a    close    imitation    of    the    natural    course    of    life.     an    illness    in    a    community.     a    rag    that    dirties    more    than    it    cleans.     an    untreated    wound.     containment.     breaching    containment.     unbreathable    air.     fungi.     one    with    what    you    love.     one    with    what    loves    you.     a    corpse    unfit    for    a    glass    case.     hearing    a    song    in    the    sound    of    tiny    wings    and    legs.     honeycomb    patterns.     an    ecosystem    within    a    person.     a    curse    passed    on.     the    hubris    of    a    scientist.     an    ugly    death    where    a    glorious    one    is    owed.     blood    on    a    handkerchief.     parasites.     something    pushing    up    the    sewer.     a    mask    to    keep    something    out.     trypophobia.     knowing    you    belong.     death    weeks    after    impact.     fever.     food    that’s    gone    off.     pandora’s    box.     death    behind    a    glass.  

THE DARK.      shadows.     lights    that    turn    off    by    themselves.     the    feel    of    cold    marble.     a    beaked    creature    in    the    night.     the    difference    between    seeing    darkness    and    seeing    nothing.     touch    of    something    you    can’t    see.     hiding    under    a    blanket.     white,    clouded    eyes.     months    without    going    outside    during    sunlight.     pouring    dark.     unscrewing    lightbulbs.     black    matter.     light    sensitivity.     a    starless    night.     time    before    light    was    created.     a    shadow    on    the    wall    without    a    body    to    attach    to.     withering    plants.     a    world    without    a    sun.     footfalls    in    an    empty    house    in    the    night.     a    light    that    doesn’t    reach    as    far    as    it    should.     desperate    reach    for    a    flashlight.     clothes    that    hide    your    shape.     staying    imperceivable.     winter    months    in    the    north.     an    empty    church.  

THE DESOLATION.      senseless    pain.     warmth    of    faith.     wax    where    skin    should    be.     a    blazing    fire.     heat    without    a    source.     the    third    or    fourth    tragedy    in    the    family.     losing    everything    you’ve    ever    held    dear.     so    much    to    live    for,    gone    so    soon.     the    smell    of    gasoline.     touch    that    scars.     coffee    cup    that    never    goes    cold.     scorch    marks    on    wood.     inescapably    warm    air.     a    child    born    in    fire.     death    of    a    loved    one.     a    candle    without    a    flame.     an    altar    in    the    middle    of    the    woods.     animals    with    burnt    fur.     plastic    explosives.     burning    hot    metal.     sweating    in    an    interrogation    room.     never    touching    a    loved    one.     disfigurement.     a    kiss    that    ruins    you.     the    scent    of    burning    fat.     a    tattoo    that    terrifies    its    viewer.     the    agony    of    hellfire    displayed    as    art.     auburn    hair.     little    clothing    in    cold    weather.     a    ripple    in    the    air.     trying    to    cool    down    in    vain.  

THE FLESH.      body    horror.     factories.     a    hunger    for    something    more    filling.     never    quite    happy    with    how    you    look.     the    terror    of    an    animal    waiting    for    slaughter.     a    very    good    meal.     the    liquid    of    a    perfect    steak.     fighting    your    worst    survival    instincts.     a    twisted    bone.     long    nights    working    out.     more    than    one    heart.     appearance    that    shapes    like    clay.     a    bag    of    bones.     bone    broth    in    a    pot.     knowing    to    fear    pigs.     the    butcher’s    shop.     plastic    surgery.     something    alien    inside    your    body.     a    hunger    in    the    gaze    laid    upon    you.     unwitting    cannibalism.     forgetting    what    you    used    to    look    like.     being    admired    for    your    appearance    and    appearance    only.     teeth    marks    on    skin.     scars    from    wounds    that    should’ve    killed    you.     cooking    in    scarcity.     fenced    in    with    one    way    to    go.  

THE END.      the    last    page    of    a    book.     nightmares    that    don’t    feel    like    nightmares.     a    skeletal    hand.     the    grip    of    the    grim    reaper    around    your    throat.     existential    pain.     ivory    dice.     flatlining    in    a    hospital.     gambling    with    death.     as    old    as    the    universe.     soul    and    spirit    tied    to    an    object.     a    dream    where    you    die.     closing    your    eyes    for    the    last    time.     the    plead    of    a    dying loved    one.     knowing    the    fate    of    someone    you    know    and    being    unable    to    prevent    it.     a    thousand    cords    tugging    you    towards    your    end.     skin    that’s    freezing    to    the    touch.     an    act    of    desperation.     someone’s    life    for    yours.     an    eternity    spent    alive.     the    cost    of    your    selfishness.     watching    your    own    burial.     causing    your    own    burial.     the    smell    of    death.     numbness    to    fear.     words    from    someone    gone.     meaninglessness    of    the    actions    or    lives    of    single    people    in    the    universe.     multiple    near-death    experiences    you    refuse    to    die    from.  

THE EYE.      googling    something    you    shouldn’t    have.     eureka    moments.     the    unforgiving    lens    of    a    camera.     witness    reports.     hidden    libraries.     eyes    of    different    colours.     feeling    of    being    watched.     a    death    recorded    in    tape.     a    tragedy    you    can’t    walk    away    from.     endangering    yourself    for    knowledge.     truth.     analog    records.     a    symbol    of    an    eye.     a    watch    tower.     compulsion    to    document.     turning    on    recording    devices    without    thinking    about    it.     saving    the    evidence    before    the    person.     extracting    information.     truth    or    dare,    without    the    dare.     a    thirst    for    knowledge.     books    that    speak    to    you.     coordinated    shelves.     cataloguing    systems.    voyeurism.     police    report    you    can’t    put    down.     reasoning    your    way    out.     smell    of    old    papers.     books    that    read    you    back.

You don't hold hands with klu Klux klan and sing koombaya my Lord. Nope. Every evil that rises against you a greater evil contends against the enemy 10 fold. Stand in your power. Leave what does not resonate with your brilliant soul anymore. Let people be. You're above shadows now. Always were. Walk in your authority. Do not accept pearls from people. No matter how beautiful they are. Do not accept curses wrapped as a gift.

Beware of witchcraft embossed items. Beware. You are on levels that you cannot be harmed. However, you still should take precautions to not allow the enemy into your life so personally. Handshakes can hide snakes. Pats on the back can hold you back. Shake off the dust and keep going. Keep rising. You will thrive in this season and anyone who dares to challenge and mock you, There's a severe punishment coming in immediately. You're not evil, you've forgiven but God does not. His wrath pours down upon the head of your enemy. You are no longer doubting your abilities.

This time, you are powerful and giving yourself the credit for it. You are teaching others how to respect you by loving on yourself. It’s not bragadocious. It’s self empowerment. The Forest Raven is a message that people are going to copy you, because you are the blueprint. This is something they’ll do, to things that make no sense to them. They can never duplicate your energy and they won’t see the forthcoming challenges after they try to consume what doens’t belong to them. Your energy is poison and self destructive to those whom it does not belong to.. Do not worry, the more they try to emulate, the more powerful you become. And unfortunately, coming up against you in this manner is spiritual and natural suicide. The Forest Raven is a sign and a prophecy that you are one with the divine and all of creation. It does not matter what path you choose, (religion, Santeria, voodoo, Ifa, etc) that power means nothing if you do not have God in it. Know that all power comes from source. Even the devil came from God/almighty. Right now you are on the right path, as you continue this is the prophecy and promise to you:

You are being gifted wisdom beyond the natural, you’re swimming in the power of the supernatural. And this has given you authority over the upper, middle and lower realms. Command and authorize your days. Your prophecies, will never fail. The witches, demons, warlocks and everything else, will hang themselves. A flock of Ravens will be the opposite of unkindness for you and just that for everything and anyone that tries to challenge you.

Immune checkpoint blockade (ICB) therapies can be very effective against some cancers by helping the immune system recognize cancer cells that are masquerading as healthy cells. 

T cells are built to recognize specific pathogens or cancer cells, which they identify from the short fragments of proteins presented on their surface. These fragments are often referred to as antigens. Healthy cells will will not have the same short fragments or antigens on their surface, and thus will be spared from attack. 

Even with cancer-associated antigens studding their surfaces, tumor cells can still escape attack by presenting a checkpoint protein, which is built to turn off the T cell. Immune checkpoint blockade therapies bind to these “off-switch” proteins and allow the T cell to attack.

Researchers have established that how cancer-associated antigens are distributed throughout a tumor determines how it will respond to checkpoint therapies. Tumors with the same antigen signal across most of its cells respond well, but heterogeneous tumors with subpopulations of cells that each have different antigens, do not. The overwhelming majority of tumors fall into the latter category and are characterized by heterogenous antigen expression. Because the mechanisms behind antigen distribution and tumor response are poorly understood, efforts to improve ICB therapy response in heterogenous tumors have been hindered.

In a new study, MIT researchers analyzed antigen expression patterns and associated T cell responses to better understand why patients with heterogenous tumors respond poorly to ICB therapies. In addition to identifying specific antigen architectures that determine how immune systems respond to tumors, the team developed an RNA-based vaccine that, when combined with ICB therapies, was effective at controlling tumors in mouse models of lung cancer.

Stefani Spranger, associate professor of biology and member of MIT’s Koch Institute for Integrative Cancer Research, is the senior author of the study, appearing recently in the Journal for Immunotherapy of Cancer. Other contributors include Koch Institute colleague Forest White, the Ned C. (1949) and Janet Bemis Rice Professor and professor of biological engineering at MIT, and Darrell Irvine, professor of immunology and microbiology at Scripps Research Institute and a former member of the Koch Institute.

While RNA vaccines are being evaluated in clinical trials, current practice of antigen selection is based on the predicted stability of antigens on the surface of tumor cells. 

“It’s not so black-and-white,” says Spranger. “Even antigens that don’t make the numerical cut-off could be really valuable targets. Instead of just focusing on the numbers, we need to look inside the complex interplays between antigen hierarchies to uncover new and important therapeutic strategies.”

Spranger and her team created mouse models of lung cancer with a number of different and well-defined expression patterns of cancer-associated antigens in order to analyze how each antigen impacts T cell response. They created both “clonal” tumors, with the same antigen expression pattern across cells, and “subclonal” tumors that represent a heterogenous mix of tumor cell subpopulations expressing different antigens. In each type of tumor, they tested different combinations of antigens with strong or weak binding affinity to MHC.

The researchers found that the keys to immune response were how widespread an antigen is expressed across a tumor, what other antigens are expressed at the same time, and the relative binding strength and other characteristics of antigens expressed by multiple cell populations in the tumor

As expected, mouse models with clonal tumors were able to mount an immune response sufficient to control tumor growth when treated with ICB therapy, no matter which combinations of weak or strong antigens were present. However, the team discovered that the relative strength of antigens present resulted in dynamics of competition and synergy between T cell populations, mediated by immune recognition specialists called cross-presenting dendritic cells in tumor-draining lymph nodes. In pairings of two weak or two strong antigens, one resulting T cell population would be reduced through competition. In pairings of weak and strong antigens, overall T cell response was enhanced. 

In subclonal tumors, with different cell populations emitting different antigen signals, competition rather than synergy was the rule, regardless of antigen combination. Tumors with a subclonal cell population expressing a strong antigen would be well-controlled under ICB treatment at first, but eventually parts of the tumor lacking the strong antigen began to grow and developed the ability evade immune attack and resist ICB therapy.

Incorporating these insights, the researchers then designed an RNA-based vaccine to be delivered in combination with ICB treatment with the goal of strengthening immune responses suppressed by antigen-driven dynamics. Strikingly, they found that no matter the binding affinity or other characteristics of the antigen targeted, the vaccine-ICB therapy combination was able to control tumors in mouse models. The widespread availability of an antigen across tumor cells determined the vaccine’s success, even if that antigen was associated with weak immune response.

Analysis of clinical data across tumor types showed that the vaccine-ICB therapy combination may be an effective strategy for treating patients with tumors with high heterogeneity. Patterns of antigen architectures in patient tumors correlated with T cell synergy or competition in mice models and determined responsiveness to ICB in cancer patients. In future work with the Irvine laboratory at the Scripps Research Institute, the Spranger laboratory will further optimize the vaccine with the aim of testing the therapy strategy in the clinic.
