Meet the Amatruda Lab!
James Amatruda, MD, PhD

Dr. James Amatruda is the Head of Basic and Translational Research for the Cancer and Blood Disease Institute at Children’s Hospital Los Angeles. He’s the inaugural holder of the Dr. Kenneth O. Williams Chair in Cancer Research. Dr. Amatruda is a Professor of Pediatrics and Medicine for the Keck School of Medicine of USC. He attends on the Solid Tumor oncology service at CHLA.
Dr. Amatruda received his MD and PhD from Washington University School of Medicine. He completed his internship and residency in Internal Medicine from Brigham and Women’s Hospital. He was a Visiting Fellow at the Institute of Cell Biology in Consiglio Nazionale delle Ricerche in Rome and completed his Medical Oncology fellowship at Dana-Farber/Partners Cancer Care in Boston, Massachusetts.
When not in the lab, Dr. Amatruda enjoys running, reading, music-making and exploring around Los Angeles.
Ashley Jean, MD

Dr. Ashley Jean is a Clinical Fellow in the Amatruda Lab. Dr. Jean graduated from Tufts Medical School in Boston and completed her Pediatric Residency at Maine Medical Center. Dr. Jean started her Pediatric Fellowship at Children’s Hospital Los Angeles in 2019.
Her research focuses on pediatric Ewing Sarcoma. She is currently studying the TAK1 pathway in the tumor genesis of this condition.
Dr. Jean likes to spend her free time outdoors. She enjoys activities such as hiking, paddle boarding and snowboarding.
Christopher Kuo, MD

Dr. Christopher Kuo is a Clinical Fellow in the Amatruda Lab. Dr. Kuo received his Medical Degree from Rush University and completed his Pediatric Residency from Children’s Hospital Los Angeles. Dr. Kuo started his Pediatric Fellowship at Children’s Hospital Los Angeles in 2020.
His research interest is in osteosarcoma. He is currently working on a project that involves the investigation of the tumor microenvironment of Ewing sarcoma.
Dr. Kuo’s hobbies include breakdancing, swimming and going to coffee shops.
Adam Marentes, MSc., PhD Candidate

Adam Marentes is a Graduate Student Researcher in the Amatruda Lab. Adam received his Bachelor of Science in Neuroscience from the University of California, Riverside. He then completed his Master of Science from California Polytechnic University Pomona. Adam is currently attending University of Southern California Keck School of Medicine to earn his PhD in Cancer Biology and Genomics.
Adam’s research focus is in mitochondrial DNA variants in Ewing Sarcoma. He is currently working on a collaboration that involves editing mitochondrial DNA in cancer cell lines in zebrafish.
Adam enjoys baking, playing video games with his fiancé and catching a show at the local comedy club.
Tanya Mosesian, MHA

Tanya Mosesian received her Bachelor of Science in Public Health from California State University of Northridge. She then completed her Master of Health Administration at the University of Southern California.
Tanya is Project Associate for the Amatruda Lab. She provides on-site support for all administrative matters and project facilitation.
Tanya enjoys spending time with her family and friends. She likes to play tennis and hike during the weekends.
Elena Vasileva, PhD, MSc.

Dr. Elena Vasileva is a post-doctoral fellow in the Amatruda Lab. Dr. Vasileva received her Bachelor of Science and Master of Science from Peter the Great St. Petersburg Polytechnic University in Applied Mathematics and Physics. She received her PhD in Molecular Biology from the Institute of Cytology, Russian Academy of Sciences.
Dr. Vasileva is interested in studying the molecular mechanisms of cancer development and progression. She has developed an inducible zebrafish model of EWS-FLI driven Ewing Sarcoma as a platform for biologic discovery and preclinical testing of novel therapies.
Dr. Vasileva enjoys running and hiking in Los Angeles.
Mona Wu, PhD

Dr. Mona Wu is a post-doctoral fellow in the Amatruda Lab. Dr. Wu received her Bachelor of Science from the University of British Columbia, a Master of Science from Université de Montréal, and a PhD from McGill University.
Dr. Wu is interested in understanding the cell of origin for pediatric neoplasms because she believes that this knowledge could lead to better early biomarkers and more effective treatment. She is particularly interested in understanding how aberrant ncRNA (especially miRNAs) may play a role in pediatric disease.
Dr. Wu likes reading and visiting different libraries. She enjoys “foodie-related” activities including trying restaurants, cooking, baking and watching (far too many) cooking shows.

Making better measurements of the composition of galaxies
“We’re trying to measure the composition of gases inside galaxies,” said Yuguang Chen, a postdoctoral researcher working with Professor Tucker Jones in the Department of Physics and Astronomy at the University of California, Davis. Most elements other than hydrogen, helium and lithium are produced inside stars, so the composition and distribution of heavier elements — especially the ratio of oxygen to hydrogen — can help astronomers understand how many and what kinds of stars are being formed in a distant object. Astronomers use two methods to measure oxygen in a galaxy, but unfortunately, they give different results. One common method, collisionally excited lines, gives a strong signal, but the results are thought to be sensitive to temperature changes, Chen said. A second method uses a different set of lines, called recombination lines, which are fainter but not thought to be affected by temperature. The recombination line method consistently produces measurements about double those from collisionally excited lines. Scientists attribute the discrepancy to temperature fluctuations in gas clouds, but this has not been directly proven, Chen said. Chen, Jones and colleagues used optical and infrared astronomy to measure oxygen abundance in dwarf galaxy Markarian 71, about 11 million light years from Earth. They used archived data from the recently retired SOFIA flying telescope and the retired Herschel Space Observatory, as well as making observations with telescopes at the W.M. Keck Observatory in Mauna Kea, Hawaii. SOFIA (Stratospheric Observatory For Infrared Astronomy) was a telescope mounted in a Boeing 747 aircraft. By flying at 38,000 to 45,000 feet, the aircraft could get above 99% of the water vapor in Earth’s atmosphere, which effectively blocks infrared light from deep space from reaching ground level. A joint project of NASA and the German space agency, SOFIA made its last operational flight in September 2022 and is now headed for a museum display in Tucson. The Herschel Space Observatory, named after astronomers William and Caroline Herschel, was an infrared space telescope operated by the European Space Agency. It was active from 2009 to 2013. A surprising result With data from these instruments, Chen and Jones examined oxygen abundance in Markarian 71 while correcting for temperature fluctuations. They found that the result from collisionally excited infrared lines was still 50% less than that from the recombination line method, even after eliminating the effect of temperature. “This result is very surprising to us,” Chen said. There is no consensus on an explanation for the discrepancy, he said. The team plans to look at additional objects to figure out what properties of galaxies correlate with this variation, Chen said. One of the goals of the James Webb Space Telescope, launched in 2022, is to make infrared observations of the composition of distant galaxies in the first billion years of the universe. The new results provide a framework for making these measurements with the JWST and the Atacama Large Millimeter Array in Chile. The work was financially supported in part by NASA. SOFIA was jointly operated by the Universities Space Research Association, Inc., and the Deutsches SOFIA Institut. The W.M. Keck Observatory is operated as a scientific partnership among the California Institute of Technology, the University of California, and NASA, with financial support from the W.M. Keck Foundation. A new oval bycatch reduction device might spell relief for diamondback terrapins
Diamondback terrapins have always found it hard to catch a break.
Up through the first third of the last century, terrapins were at the top of the list of luxury foods. Joseph Mitchell had a 1930s piece in The New Yorker reporting on a firm in the Fulton Fish Market that sold 2,000 quarts of diamondback terrapin stew a year.
A patchwork of state and federal regulations keep America's only truly estuarine turtle from being served up as seafood, variously classifying terrapin as endangered, threatened or species of concern throughout the species' East Coast range.
Randy Chambers explains that diamondback terrapins continue to be unintended victims of the seafood industry, as they end up drowning as bycatch in crab traps. Chambers, the director of William & Mary's Keck Environmental Field Laboratory, is leading a team that's trying to find a way to keep turtles out of the traps in the first place...

Malignant Metabolism
Growing among these healthy neurons (green) are malevolent medulloblastoma cells (red). While medulloblastoma is rare in the general population, it is the most common and most deadly type of brain cancer to affect children. The cancer begins in the back of the brain – a region called the cerebellum – causing symptoms such as headaches, clumsiness and vision problems. But, as the cancer creeps into other parts of the brain, or the spinal cord, so the symptoms vary and worsen. The ability of medulloblastoma to spread, scientists have discovered, depends on the cells switching their metabolic activity to allow the neurotransmitter GABA to be used as an energy source. Worryingly, this metabolic switch also makes the cells more resistant to common chemotherapies. While the findings suggest existing treatments are limited, they will ultimately aid the development of new ones, if a way to prevent or exploit the metabolic switch can be found.
