Almadusk Woods - A dark forested area that is believed to be haunted, thanks to all the plantlife having a penchant for growing to resemble the screaming souls of the damned. Home to Nightfall Manor, an old gothic mansion which once belonged to infamous serial killer The Countess Antumbra. ↳ Known residents: Glasya Elliss, The Countess Antumbra (former)

Arctic North - A polar region up in the northernmost point of Alternia. It snows sweep-round, with large swathes of area being covered in ice sheets and permafrost. Further inland is more forested, and pockets of villages are spread out amongst the region. Larger villages are responsible for delivering goods to the smaller, more remote villages, although frequent blizzards can affect the supply chain. ↳ Known residents: Davitt Cesare (former), Leutia Izotze, Sigrun Apteno

Arctic North, Northern Lights Village - A small community within Alternia's Arctic North region. Only a few log cabins have been built here due to its isolated location, however the residents who live here are all very close. Ashwood Bushland - A large eucalyptus forest. The climate is quite arid and sees little to no rainfall in the warmer seasons, making the possibility of bushfires a serious risk. Few trolls live within this forest for that reason, however it is home to many unusual species of flora and fauna native to its highly unique ecosystem. ↳ Known residents: Kalpan Sewick

Bayside Keyes - An exclusively-seadwelling neighbourhood along the coastline. Mega mansions line the streets of this small gated community, each with their own section of private beach. While quieter and less territorial than other seadweller communities, its residents are hostile towards their own neighbours and keep to themselves. Block 136 - One of Alternia's major cities. A cultural melting pot of which trolls of all castes call their home, however social segregation is still rampant thanks to unofficial caste-based residential districts. Evergreen Woods - A forested area of reasonable size. The platonic ideal of a forest, with large evergreen trees and a temperate climate, and absolutely no notable features. Grünesmeer Rainforest - A swamp-like rainforest with a slightly humid climate. Oils secreted from the trees give the bodies of water within the forest a distinct bright green hue, which is how the region got its name. Few trolls live in this area, and there's been multiple incidents of unexplained disappearances and drownings. ↳ Known residents: Vivyin Kafkah

Northwestern Highlands - A mountainous region in Alternia's northwest, characterised by its long green rolling plains and cliffs that end abruptly at the coastline. The number of hives in the area are sparse, but those who make the highlands their home witness spectacular views of both the ocean and the countryside. Based on both the Scottish and Irish Highlands. ↳ Known residents: Charon Acerbi, Somerl Alfero

Northwestern Midlands - A region in Alternia's northwest, less mountainous than the highlands and characterised by its many lakes and moorlands. The midlands is home to many castle ruins, and tends to be a popular location for hikers, history enthusiasts, and fans of romantic literature alike. Pescaruzzia - A region out in Alternia's countryside, known for its large green fields and warm Mediterranean climate. It is home to many vineyards and orchards, as the weather and soil quality is perfect for agricultural endeavours. It is not uncommon for residents of this region to sell their wares at farmers markers, or export them in bulk to nearby towns. Phosphor Forest - A phosphorous mine that was abandoned thousands of sweeps ago and has been long since reclaimed by nature, growing into a lush forest. The traces of phosphorous in the soil have caused the plants and wildlife to develop bioluminescence, at the cost of the air being mildly toxic to those who have not grown accustomed to it. ↳ Known residents: Ananta Cahaya

Revoire City - Officially designated as Block 50. A glitzy coastal city that is a highly popular tourist location for rich socialites and celebrities alike. It is known for the famous Revoire Casino, the largest casino in Alternia, as well as the historic Revoire City Grand Prix. Those with permanent residence in Revoire are almost exclusively highbloods, and live within gated hivestem communities. Sommerset Beach - A stretch of coastline and surrounding islands that are famous for its pink-tinted sand and beautiful coral reef. The seadwellers who live within this region are highly territorial and squabble over ownership of their private beaches and portions of the ocean. Vernrot Harbour - Officially designated as Block 870. A small fishing village plagued by eternal rainfall due to the existence of Those Who Slumber In The Deep. The majority of its residents are unaware that it is a hotspot for both eldritch and supernatural activity. However, the town experiences very little interference from the Empire due to the horrorterrors protecting it. Circ_0001529 Expression Abnormalities Mediate the miR-578 CDC25A Axis Involved in the Growth of Ovarian Cancer

Abstract Purpose: To investigate the effect of Circ_0001529-mediated miR-578-CDC25A axis on the growth of ovarian cancer cells in vitro and in vivo. Method: The expression differences of Circ_0001529 in ovarian cancer tissues and paraneoplastic tissues were first analyzed; Circ_0001529 overexpression plasmids and interference plasmids were constructed and transfected into SKOV3 cells and SiHa cells, respectively, followed by MTT assay, flow cytometry, and immunofluorescence to detect cell growth, proliferation, and apoptosis. The binding relationship between Circ_0001529 and miR-578, or miR-578 and CDC25A was verified by online prediction site and dual luciferase assay, and CDC25A overexpression plasmid and interference plasmid were constructed and transfected into SKOV3 cells and SiHa cells, respectively. qRT-PCR and Western Blot were used to to detect the expression of ACP5, followed by cell growth as well as apoptosis. A mouse xenograft tumor model was constructed, and the growth of ovarian cancer cells in vivo was assessed by calculating tumor volume growth with KI67 and Caspase-3 immunohistochemistry. Result: Circ_0001529 was highly expressed in ovarian cancer tissues and cells; silencing Circ_0001529 inhibited ovarian cancer cell growth and promoted apoptosis in vitro and in vivo. circ_0001529 could bind to miR-578 target, and miR-578 bound to CDC25A mRNA target. Overexpression of Circ_0001529 promotes the expression of CDC25A and promotes the growth and inhibits the apop- tosis of ovarian cancer cells. CDC25A can upregulate the ERK/MAPK signaling pathway, thus promoting ovarian carcinogenesis. Conclusion: circRNA_0001529 promotes CDC25A expression through targeted binding of miR-578, thereby promoting the growth and metastasis of ovarian cancer cells. Introduction Ovarian cancer (OC) is one of the deadliest gynecological cancers, with a significant fatality rate. It has a poor prognosis due to late diagnosis and medication resistance [1]. In the treatment of ovarian cancer, it is critical to distinguish between patients who will benefit from primary full cytoreductive surgery and those who will benefit more from neoadjuvant chemotherapy [2]. TK1 could be used as a biomarker to diagnose ovarian cancer. When it comes to differentiating malignant ovarian tumors from benign ones, ROMI outperforms ROMA [3]. The use of cytoreductive surgery (CRS) with hyperthermic intraperitoneal chemotherapy (HIPEC) has been demonstrated to be effective in a variety of cancers, and it may also provide a prognosis benefit for patients with advanced ovarian cancer [4]. The intracellular signal transducer and regulator of gene expression and protein turnover, Focal adhesion Kinase (FAK), increases tumor progression and is a promising therapeutic target for a variety of cancers, including ovarian cancer. [5] There have been some results suggesting that ERRα may promote ovarian cancer progression and may serve as a promising predictive biomarker [6]. Ovarian Cancer (OC) is a type of gynecological cancer with high mortality rate, and Circ_000152. (PM9 plays an important role in it. However, the clinical significance of circular RNA and its molecular mechanism in OC are mostly unknown. Circ_0001529 (hsa_circAFF4_016), is a circular RNA formed by reverse shearing of AFF4, is a circular RNA newly discovered by us [7] Circular RNAs (circRNAs) are increasingly recognized as important regulators in cancer including ovarian cancer (OC). This work focuses on the effects of circ_0000745 on the OC development of and molecules involved [8]. Imaging intracellular microRNAs (miRNAs) demonstrated an essential role in exposing their biological and pathological functions [9]. The induction of microribonucleic acid (miRNA) in cells is of great significance for early disease diagnosis and treatment monitoring. DNA is an interesting building material that can be programmed into components with rigid and branched structures and is particularly suitable for intracellular biomolecular imaging or therapeutic drug delivery [10]. Moreover, it has been shown that targeting the hsa_circ_0005785/miR-578/ APRIL regulatory pathway may be a promising diagnostic and therapeutic strategy in the clinical practice of HCC [11]. Similarly, for the ovarian cancer we studied, we combined Circ_0001529 with miR-578 targeting to see if it could promote cancer cell changes and metastasis functionally. Given the importance of silencing Circ_0001529 in ovarian cancer treatment, we aimed to identify an excellent inhibitor and investigate its potential therapeutic effects, thereby knocking down the role of Circ_0001529 expression to promote ovarian cancer cell growth and inhibit its apoptosis. Circular RNA (circRNA) is recently found to participate in the regulation of tumor progression, including ovarian cancer. Methods Tissue samples Eighty patients with ovarian cancer diagnosed and treated in our hospital from September 2012 to June 2013 were selected from surgically resected cancerous tissue and normal paracancerous tissue (at least 2 cm away from the ovarian cancerous tissue), with a median age of 49-79 years and a mean age of (64.9 ± 7.3) years. And all patients were followed up every three months for five years. Inclusion criteria: 1, all patients were diagnosed with ovarian cancer by postoperative pathological examination; 2, all patients did not receive radiotherapy or chemotherapy before surgery; 3, clinical data were complete. Exclusion criteria: 1, combined with chronic systemic diseases; 2, combined with other malignant tumors. The study was approved by our ethics committee and supervised by our ethics committee, and the subjects all signed the informed consent form while enjoying the right to information. Cell Culture Human ovarian cancer cell lines Hela, SKOV3, SiHa, SPC-A-1, L9981 and human normal ovarian epithelial cells

HOSE-1 were purchased from the Tumor Cell Bank of China Medical College (Beijing, China), and subsequently inoculated into cell culture dishes at a density of 1×10 5 cells/cm2 using RPMI-1640 medium containing 10% fetal bovine serum, CM7-1 medium, F12K medium and LHC-9 medium (all purchased from Gibco, Grand Island, NY,USA), and cultured at 37°C , 5% CO2 for 48 h until cell growth was fused to 80%-90%, digested with 0.025% trypsin (Gibco), and passaged. qRT-PCR Total RNA of tumor tissues and cells was used using RNAiso Plus (TAKARA, Otsu, Shiga, Japan) and Trizol LS Reagent (TAKARA, Otsu, Shiga, Japan), respectively. The reliability of the obtained RNA was subsequently verified using formaldehyde denaturation electrophoresis assay to continue the subsequent experiments. Reverse transcription polymerase chain reaction (RT-PCR) experiments were then performed using the PrimeScript TMRT kit (TAKARA, Otsu, Shiga, Japan) strictly according to the instructions. The mRNA expression levels were quantified by standard real-time quantitative PCR methods using SYBR Premix Ex Taq (TAKARA, Otsu, Shiga, Japan). gAPDH was used as a reference gene. The primers for each group are shown in Table 1.

for the past few weeks ive been considering reopening some old quadrants for ships i havent done anything with in years and have fallen out of contact w. my old friend who played those characters

so. katrin's black quad will probs reopen, and her and klavir deciding to just be friends probs makes the most sense + doesnt send kat's character development spiralling

i'll probs retcon somerl's matespritship, cuz ultimately whether him and zmalar are together doesn't impact his character too much but given their incredibly specific dynamic i can't see them just being friends, lmao

there's a few ships for my side trolls i'll always keep because they're still so dear to me. aiolos' quads are forever kept as-is, and i'll never reopen kalpan and aegiel's red quads. for other side characters i'll leave their quads closed because i see no reason to reopen them up, but if i start playing those characters more frequently i might reconsider

as for viltau's black quad... it's difficult. kittau is still a ship i adore, but i find it difficult to write around the fact that it isn't an active ship and i have to keep trying to think up things they would be doing based off discussions i had with pw nearly a decade ago. but at the same time, i can't retcon it because that will affect a lot of viltau's relationships with my other trolls, and vil is not the type to end a relationship with someone unless the circumstances were incredibly serious. so, i think i'll leave that one closed for now, and have more of a think about it
