Biomolecules, Vol. 13, Pages 1537: Comprehensive Transcriptomic Profiling of m6A Modification in Age-Related Hearing Loss

Age-related hearing loss (ARHL), also known as presbycusis, is one of the most common neurodegenerative disorders in elderly individuals and has a prevalence of approximately 70–80% among individuals aged 65 and older. As ARHL is an intricate and multifactorial disease, the exact pathogenesis of ARHL is not fully understood. There is evidence that transcriptional dysregulation mediated by epigenetic modifications is widespread in ARHL. However, the potential role of N6-methyladenosine (m6A) modification, as a crucial component of epigenetics, in ARHL progression remains unclear. In this study, we confirmed that the downregulation of m6A modification in cochlear tissues is related to ARHL and found that the expression of the m6A methylation regulators Wilms tumour suppressor-1-associated protein (WTAP), methyltransferase-like 3 (METTL3), ALKB homologous protein 5 (ALKBH5) and fat mass and obesity-associated protein (FTO) is decreased significantly at the #mRNA and protein levels in ARHL mice. Then, we used methylated #RNA immunoprecipitation sequencing (MeRIP-Seq) and #RNA sequencing (#RNA-Seq) to identify the differentially m6A-methylated genes in the cochlear tissues of ARHL mice. A total of 3438 genes with differential m6A methylation were identified, of which 1332 genes were m6A-hypermethylated and 2106 genes were m6A-hypomethylated in the ARHL group compared to the control group according to MeRIP-seq. Further joint analysis of #RNA-Seq and MeRIP-Seq data showed that 262 genes had significant differences in both #mRNA expression and m6A methylation. GO and KEGG analyses indicated that 262 unique genes were enriched mainly in the PI3K-AKT signalling pathway. Hearing Loss: Patient-caregiver Dyad Effects On Quality Of Life

Background and aims Patients with age -related hearing loss (ARHL) and their natural caregivers have to confront a disability that produces progressive lifestyle changes. There is an interest in studying the ability of patients and their caregivers to cope with the difficulties that affect quality of life (QoL). In a sample of patient-caregiver dyads in the specific context of ARHL, we examine whether the QoL of patients and caregivers is influenced by the coping processes they use from a specific actor–partner interdependence model (APIM). Methods This cross-sectional study involved dyads with patients having a diagnosis of ARHL. The self-reported data included QoL (WHOQoL-BREF) and coping strategies (BriefCope). The APIM was used to test the dyadic effects of coping strategies on QoL. Results A total of 448 dyads were included; the patients and caregivers were love partners for 59% of the dyads. Coping strategies, such as social support, avoidance, problem solving, and positive thinking, exhibited evidence of actor effects (degree to which the individual’s coping strategies are associated with their own QoL). Effects on the partner (degree to which the individual’s coping strategies are associated with the QoL of the other member of the dyad) were found, i.e., when the patients mobilized their coping strategy based on social support and problem-solving, their caregivers reported higher environmental QoL. Conclusion This study emphasizes that the QoL for patients and their caregivers was directly related to the coping strategies they used. This finding suggests that targeted interventions should be offered to help patients and their relatives to implement more effective coping strategies. Read the full article

Classification, Pathophysiology, Genetics, And Role of Lifestyle Medicine in Presbycusis

Abstract

Presbycusis, which is age-related hearing loss is hearing loss usually seen in the elderly due to advancing in age, marked by a higher hearing threshold usually worse at higher frequencies, which impairs speech discrimination in noise. Genetic predisposition is an important factor in the aetiology of presbycusis. Presbycusis was previously classified into 4 different types, and recently into 6 types with the addition of mixed, and indeterminate types of presbycusis, with each type reflecting a peculiar audiometric configuration and speech discrimination percentage. Artificial intelligence has utilized this peculiar audiometric feature in diagnosis of genetic hearing loss. and recently, lifestyle medicine is being employed to highlight the benefit of preventive medicine in the management of hearing loss. Human studies have remained the trend of genetic hearing loss research in recent years. There is an overwhelming interest, with reasonable insight gained into the pathophysiology and genetics of ARHL, but the interaction of genetics, lifestyle, and environment certainly complicates our ability to separate their individual contributions to this pathology. There is need to improve research in the role of lifestyle medicine in curbing presbycusis.

He was last to get off and he saluted in greeting to the field commander, “Commander Harrison.” he briefly jerked his head in a nod and allowed him and his men to be escorted into their base. “General Moore requested we assisted your team on this operation, I need your plans.” 

Aden had seen far more at his point in his career then he knew to never trust a man outside of his team. His gut feeling was correct was the days went by, he heard the jabs thrown at his team, at Ryan, at Thema and at the other civilians they were protecting. “Arab scums.” rang out during one night, the mess hall was crowded. Local militia attending a security conference and joining them for a meal. Aiden looked up to catch the following words that were clearly meant to be heard. “Can’t wait to go out and there and headshot those fucking jihads..fuck the Russians man, these fuckers are the real enemy. They want to preach their sermons then they’ll do it under my boo-” 

He didn’t finish as Aiden slammed his fist down on the table, his eyes filled with rage as he glanced that the field commander who did nothing but smirk alongside his subordinate's words.”The fuck you want jarhead? this isn’t your business.” Aiden stood and calmly but radiating a deadly aura, the soldier in question stood, towering over Aiden and smirked. “You want to go princess? did-” once again he was cut off by Aiden moving fast, his hand grabbing out and twisting it in an odd angle, his boot wrapping around the other’s leg and forcing him on his knees. 

“You are a United States soldier, you made an oath to protect the innocent, to fight for what's right. Did you forget that Solider? did you forget why you’re wearing this uniform?! You see those men right there? They welcome your pathetic ass to their country for help and you treat them like dogs? You dare disrespect them, my team, my men and women?” He gripped his jaw with his free hand and yanked him back. “You are not worth a single speck of respect from me, you’re not worth single respect from those men and sure as hell are not worth respect to our flag. Be grateful I’m not your commander for I will teach you respect.” 

He shoved the soldier to the ground and turned to the field commander, “You requested assistance, you allowed a soldier to be insubordinate, you are complicit to these attacks to these innocent men and woman. Ryan, send what you have seen to General Moore, Commander Harrison as of this instant I will be handling this operation.” 

He turned and left to sit back at his table, anger seething from him as Ryan accomplished his task before looking at him, a spark of something flashing in his systems. “Alright?” he gruffed out and Aiden physically relaxed his shoulders, “Let’s just get this over with.”

They flew into battle, the other soldiers already on the ground as they flanked the compounds right flank. He noticed scared women and children huddled in a corner and he waved for them to leave, “arhl alana!” (leave now!) the sounds of gunfire outside growing closer as they made it up each floor, shooting down targets and setting women and children free. He saw too much blood in this war and he didn’t hesitate to pull a trigger when an enemy jumped from around a corner and fired. He couldn’t hesitate, not in this line of job. He couldn’t afford to let his mind wander as he blood quickly coated the ground red.

“Everyone split, meet on the roof, do not die today Devil Dogs.” Everything was going smoothly, almost too smoothly as he and Ryan split and reached the rooftop where their main priority was trying to escape. Before he could tell him to drop down a bullet lodged into his head, the Captain from the other night had disregarded his direct command and shot him. “Soldier! What the fuck did you do!! he was supposed to be taken alive!!” 

He wasn’t prepared for the disgraced soldier to kick him down, his weapon trained on him, “You dare come here and tell me what to do?? you piece of shit sympathizer!” Ryan was quick to dislodge the soldier off of Aiden before he snapped his head up and shoved Aiden behind a half wall as an RPG round barely missed them, hitting the building behind. The blast caused all three to be thrown aside. Aiden being so close to the edge was hanging from a rapidly falling beam, his eyes looking down to the sharp debris waiting for him below. 

He could hear shouting, his men should be coming soon but they were quickly losing their advantage before the beam completely broke off and making him fall seven floors to his death, Ryan’s arm should out and grabbed him. Thirium leaking out of a headwound. “Ryan! What-” 

“No one left behind” He grunted, “I-I won’t let you die...not on my watch.” It was strange to not hear the once stoic android look fearful and worried, “Ryan...” he yelp as a rouge bullet skimmed his leg as Ryan began to pull him up. The disgraced soldier creeping behind him with the intent to kill a soldier with their back to them. Before the trigger was pulled, Gabriel had made it to the roof and fired into the defected soldier’s shoulder before rushing to help Ryan. 

The android rapidly scanning Aiden’s wound, a tight curl of something, of fear, of ...concern flared his system to act. He saw a red wall and tore it down to save Aiden ( not Staff Seargent Barnes) instead of apprehending the soldier. He found that he made the right call, that he was free to feel and he frowned at the intrusive thought of wanting to take Aiden into a private room and personally attend to his injury. 

A curse snapped him out of his newfound thoughts as he turned to look at the soldier, his hand curling around his handgun. He held a hand out, aiming his gun at him only for Aiden to place a hand against his forearm, “He’s not worth your decommission...let The Brass deal with him. Ryan...remember our mission.”

When I started writing Anomaly in 2014 I had an idea of where I wanted The Soul Prophecies series to go. I wanted to write about Arhl’s world and I had a vision for how the series would end. The series was always going to be a trilogy but half way through Anomaly I realised that I needed to explain Sophia’s story too. I wrote Hidden Variables as a prequel to Anomaly for that purpose.

Four years later and I have finished writing the series; I know where the story travels and its ultimate destination. Outlet Publishing offered me the release of a deluxe edition of Hidden Variables/Anomaly to include my science notes (Sophia’s journal) and suggested editorial changes to tie in with the rest of the series.

This was the perfect opportunity for me to make some changes to the original books and tidy up any loose ends. Thus Another Path was born.

The basic stories behind the original Anomaly and Hidden Variables remain unchanged, though some sections and ideas have been modified. For those of you who have already read Anomaly and Hidden Variables I hope you will also enjoy reading Another Path. We plan to release the next books in the series all within a year of this one.  The original Anomaly and Hidden Variables books will be discontinued after the current press run ends, so I guess that will make them collectable in the future.

Chen et al reported that the expression of Cav1.3 in cochlea of C57BL/6J was gradually decreased when the mice got older accompanied by increased hearing threshold, which indicated a possible association between decreased expression of Cav1.3 and ARHL, but the underling mechanism is still unknown. Moreover, it has been found that Cav1.3 calcium channel in the stria vascularis contribute to the generation and maintenance of the end cochlear potential. It is mainly expressed in CNS, mediated persistent Ca 2+ influx, sustain plateau potential, support peacemaking, and has a role in hair cell development. However, the underlying mechanism for ARHL are still obscure.Ĭav1.3 is a voltage-gated calcium channel which plays an important role in diverse cell functions.

It has been found that oxygen free radical, glutamate toxicity, and Ca 2+ overload are closely related to ARHL. The high prevalence of presbycusis causes severe social and health problems. Overall, 10% of population experience hearing loss which is sufficient to influence communication, and the rate increases to 40% in the population older than 65 years. Presbycusis or age-related hearing losses (ARHL) is the most common cause of hearing loss. Mechanistically, Cav1.3 knock out resulted in decreased intracellular calcium which subsequently reduced the inactivation of ROS from complex I, and finally resulted in increased intracellular ROS and enhanced senescence.Ĭollectively, these findings confirmed that Cav1.3 could protect cells in auditory pathway from oxidative stresses, and decreased expression of Cav1.3 in auditory pathway could contribute to hearing losses by enhancement of calcium-mediated oxidative stress. The effect was further confirmed in 3D organ culture, increased loss of hair cells after Cav1.3 was knocked down under ROS stresses. The effect was further confirmed in vivo, after Cav1.3 knocked down by injection of AAV, hearing impairment was observed in C57BL/6J male mice subjected to senescence and this was accompanied by increased loss of hair cells in cochlea. Further, Cav1.3 knock down also interfered with the electrophysiology of hair cells. The correlation between decreased expression of Cav1.3 and age-related hearing losses was studied in vitro, after Cav1.3 was knocked out, the rate of apoptosis of hair cells increased after being subjected to ROS stresses, accompanied with enhanced senescence. It was found that the expression of Cav1.3 in cochlea decreased with aging whereas this phenomenon was not observed in neuron of auditory cortex. In this study, age related Cav1.3 expression in cochlea and auditory cortex of C57BL/6J male mice was evaluated.
