Would the non-aging effect of cainite vampirism affect one's brain chemistry and psychological development? Because if your neurons can't shift and change properly then shifts in mental state (positive or negative) would be really slow and require more effort long term. That would partially explain why it's so hard to rebel against the Beast and stay on the path of Humanity long term.
It'd also go a long way in explaining why kindred are just Like That in general. 'Cause let's be honest there's no real way for the embrace to not be mentally traumatic and trauma very much affects the human brain. So instead of that Ventrue just being a petulant ass because Ventrue it could be that despite the embrace they're still basically in their early 20's and their brain isn't really developing even after a century. Instead it's stacking trauma on top of unhealthy coping mechanisms like a lasagna made of mental illness until they snap.

Would anyone have ideas for how hunters would go after gangrel specifically? So far all I've been able to think of is pursuit tactics and to have them open fire on any wolves that show up because even if it ain't a vampire it's still invasive where my game is set.

Here's an idea. What if one were to use a table to keep a general track of where characters would fall in Triat alignment. Where you could have one as primary and one as secondary but balancing the third is rare.
So a Gangrel and Tremere might both be of the Wyrm but the Gangrel might have a lot of the Wyld about them; because Gangrel. The Tremere though, will probably have a larger slant towards the Weaver if they deal with technological magic enough. This could create room for interesting mechanics where a character with enough leaning in one direction could shift in characteristics similarly to the Black Spirals or a powerful Mage. Not sure how it'd affect mechanics but it sounds fun in concept.

As an idea I can appreciate this and I do know of instances where great changes in social standing like you're describing has happened. However the actual framework that a society puts in place has way more impact than you seem willing to acknowledge. We should probably take a moment to look at the actual probability of those shifts happening as well.
We are all self made. Only those that are happy with where they are tend to admit it.

A fun little tip for vampire characters born in or around colonial America is that "yankee" was actually an insult back in the day for colonists who were seen as unrefined or uncivilized. Even in the modern U.S. it's still really not okay to call someone that depending where you are. So having an english NPC casually call someone a "yank" or some other version of the term and running into trouble because that person was born in 1754 and still has a bone to pick may lead to some interesting in game moments.

I like to think the idea of civil, borderline passive Ventrue is only really a thing among corporate neonates. Because the whole idea of the "Clan of Kings" kind of comes with a martial tradition of war and dueling that is much older than boardrooms and tailored suits.
And the further back you go the less true that stereotype is. Even in the States, "talk shit get hit" had legal precedent in the form of fighting words until dueling was outlawed in most of the union. So if your Brujah walks into Elysium and starts to get a bit too fresh with the Prince you just might get wrecked by one of the primogen just to prove a point.
