Anticancer effect of (S)-crizotinib on osteosarcoma cells by targeting MTH1 and activating reactive oxygen species
MTH1 has become a new rising star in the field of ‘cancer phenotypic lethality’ and can be targeted in many kinds of tumors. This study aimed to explore the anticancer effect of MTH1-targeted drug (S)-crizotinib on osteosarcoma (OS) cells. We detected MTH1 expression in OS tissues and cells using immunohistochemistry and western blot. The effects of MTH1 on OS cell viability were explored using the siRNA technique and CCK8. The anticancer effects of the MTH1-targeted drug (S)-crizotinib on OS cells were explored by in-vitro assays. The intracellular 8-oxo-dGTP level and oxygen reactive species (ROS) of OS cells were detected by Cy3-conjugated avidin staining and dichlorofluorescein diacetate staining, respectively. The expression of MTH1 was significantly higher in OS tissues and cell lines than that in the corresponding adjacent tissues and osteoblastic cell line. The proliferation of OS cells was significantly inhibited through knockdown of MTH1 by siRNA technology. (S)-Crizotinib could inhibit the proliferation of OS cells with an increase in the apoptosis levels and causing G0/G1 arrest by targeting MTH1 and activating ROS. In addition, (S)-crizotinib could inhibit the migration of OS cells. (S)-Crizotinib could suppress the proliferation and migration, cause G0/G1 arrest, and increase the apoptosis level of OS cells by targeting MTH1 and activating ROS. This study will provide a promising therapeutic target and the theoretical basis for the clinical application of (S)-crizotinib in OS. Relationship Between GDF5 +104T/C Polymorphism and the Susceptibility of Lumbar Disc Degeneration: A Meta-Analysis by Zengwu Shao* in Open Access Journal of Biomedical Science (OAJBS)
Lumbar disc degeneration (LDD) is a multifactorial and chronic disease with a complex genetic background. Previous studies have reported the strong association between growth differentiation factor 5 (GDF5) +104T/C polymorphism and the risk of LDD in different ethnic populations. The aim of this study was to analyze the association between the GDF5 +104T/C polymorphism and the susceptibility of LDD by a meta-analysis. Potentially relevant studies were selected by electronic databases (PubMed, EMBASE, Web of science, the Cochrane Library Chinese National Knowledge Infrastructure) up to 10th April 2017. Summary odds ratio (OR) and 95% confidence intervals (CIs) were used to assess the strength of association between the GDF5 +104T/C polymorphism and LDD risk. Six case-control studies, including 1107 LDD cases and 4353 controls, were included. The combined results showed that GDF5 +104T/C polymorphism was significantly associated with increased susceptibility to LDD at the allele level (T vs. C: OR = 1.52, 95%CI = 1.21-1.90) and genotype level (TT vs. CC: OR = 2.43, 95%CI = 1.81-3.25; TC vs. CC: OR = 1.84, 95%CI = 1.37-2.47; TT+TC vs. CC: OR = 2.14, 95%CI = 1.61-2.84; TT vs. TC+CC: OR = 1.42, 95%CI = 1.01-1.99) in the overall population. After stratified by ethnicity, we found a strong significant association (OR = 1.80 to 3.70, all P <0.05) in Asian population and weaker associations (OR =1.20 to 1.64, all P <0.05) in Caucasian population. This meta-analysis suggests an increased risk of GDF5 +104T/C polymorphism against intervertebral disc degeneration among populations especially in Asians.

A pilot study on clinicopathological features and intestinal microflora changes in colorectal cancer patients born over a nine-year period encompassing three years before and after the Great Chinese famine

Abstract
Background
Exposure to energy restriction during childhood is associated with a lower risk of developing colorectal cancer (CRC). To date, the association between this critical period of growth and prognosis of CRC has rarely been investigated. Changes in microbiota and epigenetic dysregulation may be key underlying mechanisms.
Methodology
Tissues collected from patients born between 1956 and 1964 were grouped based on time-period. The differences in overall survival among patients from the three time-periods were examined via univariate analysis. The 16S rRNA gene sequencing approach was to determine differences in microbiota among the groups. Samples were randomly selected to detect BRAF mutations, microsatellite instability (MSI) and promoter CpG island methylator phenotype (CIMP) status. The chi-square test was to assess the relationship between alterations in these molecules and microbiota differences.
Results
Patients from the three groups differed in terms of location of CRC (P = 0.034) and carcinoembryonic antigen (CEA) level (P = 0.036). A survival advantage was observed in the famine group compared with the other two groups. Fusobacterium nucleatum, Bacteroides fragilis and Escherichia coli were more abundant in the two comparing groups. Abundance of B. fragilis was associated with BRAF mutations, microsatellite instability (MSI) and abundance of E. coli. Moreover, the incidence of CIMP and MSI was higher in patients with greater abundance of F. nucleatum.
Conclusions
Limitation of energy intake during childhood may affect the composition of gut microbiota, resulting in persistent epigenetic changes that subsequently influence the prognosis of patients with CRC.
