CALDO LMX

Uso para identificación de coliformes fecales y Escherichia coli

Este caldo permite la determinación bioquímica de tres enzimas: la triptofanasa, β-galactosidasa y β-Dglucuronidasa. En este caso, tiene un sustrato cromógeno que es 5-bromo-4-cloro-3-indolil-β-D-galactopiranósido (Xgal) que es escindido por la enzima galactosidasa produciendo una coloración verde azulado. Por otra parte, tiene un sustrato fluorógeno, que es 4-metilumbeliferil-β-Dglucurónido (MUG) el cual es escindido por la enzima glucuronidasa produciendo metilumbeliferona (compuesto susceptible a transiciones electrónicas por parte de la radiación UV) produciendo fluorescencia. Como inhibidor tiene lauril sulfato de sodio que es un detergente aniónico que denatura proteínas, tiene cloruro de sodio para el balance osmótico y fosfato monopotásico y dipotasico como buffer. Finalmente, también permite confirmar si la bacteria produce indol a partir del aminoácido triptófano.

ABAJO: CONTROL POSITIVO PARA FLUORÓGENO

La presencia de la coloración verde azulada confirma la presencia de Coliformes totales, pues esta enzima es específica de este grupo. Por otra parte, la presencia de la enzima glucuronidasa (producción de fluorescencia a partir de la luz UV) confirma a Escherichia coli, puesto que, esta enzima es altamente específica para esta bacteria.
