miR-206 Inhibits Stemness and Metastasis of Breast Cancer by Targeting MKL1/IL11 Pathway

Purpose: Effective targeting of cancer stem cells is necessary and important for eradicating cancer and reducing metastasis-related mortality. Understanding of cancer stemness-related signaling pathways at the molecular level will help control cancer and stop metastasis in the clinic.

Experimental Design: By analyzing miRNA profiles and functions in cancer development, we aimed to identify regulators of breast tumor stemness and metastasis in human xenograft models in vivo and examined their effects on self-renewal and invasion of breast cancer cells in vitro. To discover the direct targets and essential signaling pathways responsible for miRNA functions in breast cancer progression, we performed microarray analysis and target gene prediction in combination with functional studies on candidate genes (overexpression rescues and pheno-copying knockdowns).

Results: In this study, we report that hsa-miR-206 suppresses breast tumor stemness and metastasis by inhibiting both self-renewal and invasion. We identified that among the candidate targets, twinfilin (TWF1) rescues the miR-206 phenotype in invasion by enhancing the actin cytoskeleton dynamics and the activity of the mesenchymal lineage transcription factors, megakaryoblastic leukemia (translocation) 1 (MKL1), and serum response factor (SRF). MKL1 and SRF were further demonstrated to promote the expression of IL11, which is essential for miR-206's function in inhibiting both invasion and stemness of breast cancer.

Conclusions: The identification of the miR-206/TWF1/MKL1-SRF/IL11 signaling pathway sheds lights on the understanding of breast cancer initiation and progression, unveils new therapeutic targets, and facilitates innovative drug development to control cancer and block metastasis. hi! do you know if there have been any updates on the findjackwalten website recently?

Hi! I've been keeping an eye on findjackwalten for months but as far as i know there hasn't been an update since August of 2022. Here's my post about that update from when it first dropped, for anyone interested. Though it's significantly less content-rich than the update that had come just before it.

There's been a new Findjackwalten update! From what I can tell, this one alters one page (or two depending on how you count it) and adds one new one.

The main page has been updated: https://www.findjackwalten.com/

it strongly resembles the '/copia-de-nueva-pagina' page we discovered back in July, which now, in fact, redirects to it. The old homepage, /cyberfuntech82, is now a completely blank webpage.

Some images have been added to the new landing page that did not exist on the old /copia-de-nueva-pagina page, like the Cyberfun Tech poster we saw Martin post in his community tab is what I believe was May of last year? And three as yet unseen images of Little Bon, Little Bon and Little Sha, and then of the animatronic Bon. There's a fourth image not visible in this screenshot, of the exterior of Bon's Burgers (the same shot of it we seen in TWF1).

Clicking on the "JOB OFFER" image leads to a new webpage, findjackwalten.com/caretakerlibrary.

This page contains a greyed-out version of the banner from the homepage, a large greyed-out version of the mounted buck's head known for being on findjackwalten.com/jackwalten, and a picture of someone we can assume, thanks to later context, is named Richie. It looks like the background illustration of an anthropomorphized family of rabbits is still on this page, it's just mostly hidden by the other page assets. Something interesting to note about this page is that all of its images are actually in regular full color, the page just puts a black & white filter over them. It's thanks to that we can have this regular, full-color image of Richie.

This page claims to be a "Caretaker Audio Archive", which seems to be the same archive Brian mentioned in TWF1, when he mentioned being asked to record himself during his job "for the archives". It contains two pieces of audio attributed to an "R.", who worked for BSI between 1974 and 1975. It refers to him also as the "Night Shift CARETAKER A".

The first piece of audio, titled "richie 1" is dated 05-23-74, or the 23rd of May, 1974. I've transcribed it as follows, but this is pretty rough: Hey there! Its your, uh, it's your coworker Richie from uh, B-Uh, BSI. Uh, Felix probably mentioned me to you before, I-uh-I don't believe we've actually, yknow, met face-to-face before, but I-uh, look forward to working with you! Uh, so, uhm. Yeah, uh! I work the, uh, I work the- I'm working the night shift at the moment. Uh, and, uh, [UNINTELLIGIBLE] uhm, sorry you're not in any, uh, trouble or anything, heh. Uh-m just- saying hi. It's uh, company policy, all employees gotta, like, know each other so we can work more efficiently, I guess. I dunno. Uh, from what I've heard I take it you've been working here quite a while, right? Uh, summer job? Same here! Uh, so uh, where was I? Hmm.. Ah- uh, some notes from last week's shift, uhm, the arcade machines should be on their way by Tuesday, I think? We uh, we made a call with Starleys, uh, now they're going to be doing the installations themselves, so you don't gotta worry about that anymore. Just try and keep [UNINTELLIGIBLE] and plus, you now owe me a favor! Nah, just kidding. Uhh, so! Tech supervisor asked us to present the documentation of the animatronics' state by tomorrow! So, uh, yeah, better get that out of the way soon. And, uh, huh! I guess that's it. Ahh- sorry, and uh, one more thing, uh, I think we'll, uh, yknow, properly meet on, uhm, Thursday! I think. [UNINTELLIGIBLE] a tour of the installations. And, not-not the ones that smell like tobacco. The other one. So, uh, yeah, uh, see you there!

The second audio, titled "richie 2", is dated 06-18-74, or the 18th of June, 1974. This is, very notably, a week after Jack Walten's disappearance. I've transcribed it as follows: Hey, uh, just wanted to update you on everything that's been going on the past few days. So, uhh, for starters I got paid the extra hours for last week, so that's nice. Uhmm, right, uh, right. So, uh, did you get the memo, or whatever? Apparently the lockdown was because of some infestation or something? Yknow, like, bugs? Heh. So just a heads up if you feel kinda- smell chemicals and whatnot. Uhh, what else, what else, what else? Right, uh, right- so here, from next week onward, all animatronics will be cleaned and checked by authorized personnel. Nobody else is allowed- sorry- Nobody else is permitted to go near them until further notice. So, that's one less thing to worry about with our jobs, right? Uhhh, right- ah, oh right, uhm. Don't come to work on Monday. They're doing one more big cleanup to make sure there's no bugs crawling around or whatever. We, uh, we wouldn't really want our customers eating a beetle sandwich, yknow? Haha, sorry- hmm. Uhh, yeah! That's all for tonight. If you see any bugs in the next few days, just like, wack 'em with a newspaper or somethin'. You'll be fine. Anyways, uh, see ya around! Take care! Bye!

A very notable peculiarity with this is that the second audio sounds very... dank? Like it's being recorded in a cave? I have absolutely no idea what this means.

New Things in TWF1:

Most of us probably know by now that Company Introductory Tape was reuploaded last night, but a bit more has been done to it than just replacing the offending footage that got it removed in the first place. Short walkthrough of everything new and notable under the cut:

The first thing we see is black & white footage of a rabbit in a forest grazing on some grass, with text in the top left corner reading ">STOP" and then ">REC", which I think might imply this was a pre-existing tape that was/is being recorded over with BSI's Company Introductory Tape.

The video is otherwise identical until just after the six minute mark, where the previous footage of Brian driving on a grass-and-car-tracks makeshift road to Saint Juana's is replaced with with different footage of him driving on what could pass as either a dirt road or a really shitty asphalt one. It's the same again until we cut to the inside of K-9, where we get shakeycam footage of Brian looking at pictures on the wall in ornate yellow-gold frames. These are immediately evocative of some similar pictures we've seen hung up in Bon's Burgers in previous episodes and in teasers for TWF4, but none of them are the same as any we've seen so far. (These photographs likely were hung up in Bon's at some point, though, hence their relocation to the K-9 facility.)

The first we see is this photo of Felix, with what my brain wants to assume is an mostly-uncovered animatronic mech next to him. (The arm is easy enough to make out, but the vague blurry mass of colors near where the head should be makes me want to call this Sha, with just her head on?)

Next, Brian pans his flashlight over to this photo of Edd & Molly. Not too much to note about this, except i see what might be a picture on the wall behind them? Wherever this photo was taken, I want to assume it's either the inside of the Walten residence or the inside of Bon's Burgers.

The camera cuts to static for a few seconds before we're finally shown this photo. In my opinion, from right to left, it seems to contain Jack, Charles, Rosemary, Susan, Felix, and someone I have to assume is either Chris or the mysterious N. Pooltrick. There's a figure in front of them, either an adult sitting down or a very short person, who I want to say is Linda. Other possibilities could be a young Sophie with her hair up in pigtails (this seems the least likely to me, we've never seen her dressed like this at this age.) or the Lily mentioned by Boozoo in TWF3.

The footage as Brian runs away from Bon is also changed, where he appears to be running blindly into the forest, rather than running down any sort of path. We continue as usual until the last image of the episode before the credits roll. This picture of Jack Walten: the clearest one we've ever seen of him.

It holds on him for an entire 20 seconds as Clair de Lune by Claude Debussy plays in while the background behind the image slowly blooms to white with an intensifying static sound. There's what appears to be a bit of red coloration in the center of his face, though it could only be discoloration from the VHS filter. Then there's only static with text reading ">REWIND"

The last small change we see is Martin censoring the credits of his own name to read only as "Martin", which I think pretty definitely confirms that his previous pseudonym is defunct and no longer accurate.

iirc you said martin apologized for the edited alcala picture in the past. was it via the twf discord or something? i believe he'll likely retcon it but it still troubles me a lot. i know one could argue if he just replaced it in the reupload it would or wouldn't have "fixed" things but idk i really wish he did. keeping it in mind is far from a problem but it's frustrating. not something you can really help me with, i'd just like to hear another fan's opinion on the situation

I actually don't recall him ever directly apologizing for the Rodney Alcala thing, but when I was checking the discord server just to see if I could find anything for you, I did see a secondhand account mentioning Bravvy saying something about how Martin did not know who Rodney Alcala was when he created Jack's missing poster in TWF1. Which I obviously can't verify in any meaningful way, so make of that what you will. (But I don't think it's an unreasonable claim to make. Martin isn't even American, I don't expect him to have an encyclopedic knowledge of American serial killers, even though I don't think ignorance necessarily excuses him of any judgement here.)

That said, Martin has talked before about wanting to broach topics related to real-world tragedy sensitively in The Walten Files, so I don't think this is something he'd like, dismiss as not being a big deal, even if he's never gone out of his way to bring him up. But I absolutely agree with you about wishing he had replaced it in the Company Introductory Tape reupload. I was honestly really expecting him too? And it left a pretty bad taste in my mouth that he didn't

ok now im really curious on what was the not new thing you noticed

I've held off on sharing this because I'd wanted to somehow edit the audio to make the part that I'm hearing a little more clear without headphones and a really good ear, but I know less than nothing about audio editing so oh well. This specific clip I did put through an incredibly basic pass of noise reduction in Audacity, but that was all I could figure out before I started getting frustrated.

But enough of my excuses. Do you hear the music? It's very subtle, and it sounds sorta like something from a music box, though maybe lacking the tinny quality you'd get from that. It's from here: [timestamped link to Company Introductory Tape, usually I would embed the video but embedded youtube links in tumblr posts cant be timestamped. imminent jumpscare warning.] just before the part where Bon jumpscares Brian and kills him. And like I said in my tags and you alluded to in this ask, this music is actually not new to the reuploaded version of the episode. It's been there, unchanged, since the original version from 2020.

This is extremely interesting to me! Though I can't make a claim on what it does or doesn't mean. I'm vaguely reminded of the 'river music' stuff we first heard about on Anthony's findjackwalten page and then from William with that rivermusic mp3, but the river music there was always depicted as more of a 'singing' or 'whistling' noise, when this sounds distinctly like an instrument. Much to think about

read your most recent post on the Findjackwalten update and there's a part that confused me. Bon has an ai consciousness? I think I may have missed something.

I mean, pretty much he does, yeah. This is info we got from /Cyberfuntech82 (this was the main website landing page for a long while) like forever ago.

The page is all CyberFun talking about their current and future projects, and it has stuff like this on it:

I do think the "recognition system" refers to the facial recognition that at least Bon is equipped with, that we see him using on Brian in TWF1. But I don't know what "-develop a mind of their own [with] a cutting new method of Artificial Technology" or "Machines able to resemble the actions of a real life performer... interacting with it's audience and surroundings like a real person would." would mean other than Bon (and the other animatronics too, honestly) having some sort of artificial intelligence system?

man actually i cant believe i never made a post about how actually unironically hilarious the molten catalogue is. i watched it forever ago now. like ok one of the actually shitty things in the walten files is the fact that one of the images that was edited to make Jack Walten’s face in TWF1 is an image of serial killer Rodney Alcala. 

And like. That’s Rodney Alcala as in The Dating Game Killer, yknow that’s like a genuinely really gross and tasteless thing to do to make Scary Assets for your horror series, and I’m glad Martin Walls has moved past doing that for The Walten Files

And like that’s something i’d known for a while by the time i sat down to watch The Molten Catalogue (which, for those who aren’t familiar, is a TWF parody series which exists on youtube). and it was so insanely fucking funny to me to be sitting there watching this series introduce it’s own mysterious founder John Molten, and then he comes up on screen and it’s just a 100% unedited image of Rodney Alcala. like thats just straight up a google images picture of serial killer Rodney Alcala
