i missed a neuroscience lecture and apparently it was the hardest one of the module bc they packed like so much content into 1 lecture to the point where they set up an extra support session/tutorial thing just for this lecture and i haven't watched it and it's in a few mins and the professor was like i will engage all of you in this meeting :) please dont !!!

i have difficulty speaking on the effectiveness of the spanish scifi melodrama-thriller the skin i live in (2011) in the time and culture of its release, because in the thirteen years since its time in theaters the conversation around its core themes has changed significantly. the film clearly attempts to explore questions of violation, identity, and femininity—its gratuitous presentation of sensuality and sexual violence makes this clear enough. however, the key lenses through which it presents these themes are transgenesis and sex reassignment surgery, both familiar practices today but much more exotic and dangerous topics at the time of the film’s release. whether the mere invocation of these concepts truly was enough to send a shiver up a 2011 audience’s spine, i do not know; in any case, the film certainly believes it to be sufficient. although the film roughly shares a premise with the french arthouse horror film eyes without a face (1960)—a plastic surgeon, obsessed with reshaping his severely scarred daughter to the image of beauty he recalls, kidnaps and experiments upon people—eyes without a face (1960) presents an almost claustrophobically tightly woven plot and a well-realized, atmospheric setting to develop and add poignancy to its themes of obsession and dehumanization. by contrast, this film relies on the mere invocation of vaginoplasty and genetic research to do that work, allowing its plot to meander soap opera-like and its sense of time and place to scatter. apart from a laser-intense performance by antonio banderas, much of what the film’s tone and critic’s reactions imply to be gut-churning horror has melted away with the years, leaving only its framework of transphobia, misogyny, and camp exposed and reminiscent almost of the assignment (2016). although i can see and reconstruct what the skin i live in (2011) is attempting to do, its reliance on period-specific shock factor undercuts it, leaving a film that feels unrealized, numbing where it seeks to thrill

We reached the edge of the abyss, as far as I’m concerned the options are we fall into it or we managed to leap over it and continue creating chimeras as many races of extraterrestrial have already done it and reached a wall called by us as Transgenesis, who knows what aliens called this meddling with DNA. As humans we have already broken the cardinal rule of evolution and ethical behavior although we deny it and chastise those scientists that have gone underground to continue with these practices and experimentation, our government and big corporations are indeed engaged in such practices that they continue to to conduct secretly despite those purist that fervently fight against the lawlessness of immoral practices by scientists that do not believe in god and religious dogma. Well ,folks we do know where this mentality is driving us back to the abyss. Understanding CRISPR-Cas9’s capacity to produce native overexpression (OX) alleles would accelerate agronomic gains achievable by gene editing. To generate OX alleles with increased RNA and protein abundance, we leveraged multiplexed CRISPR-Cas9 mutagenesis of noncoding sequences upstream of the rice PSBS1 gene. We isolated 120 gene-edited alleles with varying non-photochemical quenching (NPQ) capacity in vivo—from knockout to overexpression—using a high-throughput screening pipeline. Overexpression increased OsPsbS1 protein abundance two- to threefold, matching fold changes obtained by transgenesis. Increased PsbS protein abundance enhanced NPQ capacity and water-use efficiency. Across our resolved genetic variation, we identify the role of 5′UTR indels and inversions in driving knockout/knockdown and overexpression phenotypes, respectively. Complex structural variants, such as the 252-kb duplication/inversion generated here, evidence the potential of CRISPR-Cas9 to facilitate significant genomic changes with negligible off-target transcriptomic perturbations. Our results may inform future gene-editing strategies for hypermorphic alleles and have advanced the pursuit of gene-edited, non-transgenic rice plants with accelerated relaxation of photoprotection.

Stem cells ability to repair, replace and regenerate cells. In most animals and humans, stem cells are limited to generating only specific types of cells.
Pluripotent (aPSCs) meaning they can regenerate virtually any missing cell type, a process known as whole-body regeneration.
Many distantly-related invertebrates like sponges, hydras, planarian flatworms, acoel worms, and some sea squirts have stem cell populations that are pluripotent in adult animals. The study focuses on H. miamia, also known as the three-banded panther worm, is a species that can fully regenerate using aPSCs called “neoblasts.”
The mechanism of how (aPSCs) are produced remains unknown in any species. The focus of past scientific research was the adult organism.
The researchers knew that worm hatchlings contain aPSCs, so reasoned they must be made during embryogenesis. Ricci used transgenesis to create a line that caused embryo cells to glow in fluorescent green due to the introduction of the protein Kaede into the cell.
Is used Hofstenia miamia, a regenerative worm that possesses collectively pluripotent aPSCs called neoblasts and produces manipulable embryos. Lineage tracing and functional experiments revealed that one pair of blastomeres gives rise to cells that resemble neoblasts in distribution, behavior, and gene expression.
