Study on the Anti-Hepatocellular Carcinoma Effect of Selenized Algal Blue Protein Biomaterials
Abstract: Selenocyanin was extracted and purified from selenium-enriched cultured Spirulina obtusususus, and immobilized onto tissue culture polystyrene (PSt) substrates by photoimmobilization method to prepare biomaterials. The biomaterial was prepared by immobilizing selenized alginate onto tissue culture polystyrene (PSt) substrate by photoimmobilization method. The inhibitory effect of this biomaterial on hepatocellular carcinoma cell 7402 was investigated and compared with that of alginate immobilized material and sodium selenite.

Selenium is a trace element that plays an important role in life activities. Selenium is most abundant in liver tissue, and selenium deficiency leads to liver disease as well as endemic, cardiovascular, cancer, and viral diseases (e.g., AIDS) . Low blood selenium levels were first observed in patients with metastatic liver cancer, hepatitis, and liver sclerosis. Subsequent studies have confirmed that lower blood selenium levels are associated with the severity of liver disease. Patients with chronic active hepatitis, cirrhosis and hepatocellular carcinoma showed a decreasing trend in blood selenium levels. Salonan, a Finnish scholar, pointed out that individuals with blood selenium < 45 μg/mL had a risk factor of 3 : 1 for cancer [1], and in recent years, Clak [2] and kawamori [3] found that selenium supplementation could inhibit tumorigenesis.
Phycocyanin is an important bioactive component of Spirulina. Many literatures have reported that phycocyanin has the functions of improving immunity, anti-tumor, anti-radiation, anti-aging, preventing anemia, and nutrition, etc. In terms of anti-tumor, Iijima et al. In terms of anti-tumor, in 1982, Iijima et al. [4] firstly found that the lymphocyte activity of experimental mice injected with liver tumor cells was significantly higher than that of the control group after oral administration of alginate to the experimental mice, so Iijima et al. believed that alginate could promote the immune system to resist various diseases.
Schwartz of Harvard Medical School [5] also reported that phycocyanin has inhibitory effect on some cancer cells.  Due to the special properties of phycocyanin in absorbing and transmitting light energy, it has been used as a photosensitizer in tumor laser therapy. He Liming et al [6] explored the killing effect of phycocyanin on colorectal cancer cell line HR8348 cells and S180 transplantation tumors, and compared with the photosensitizer Porphyromonas serratiae CPD4, and found that phycocyanin's effect of killing tumor cells was better than that of CPD4. Recently, we also found that the inhibitory effect of phycocyanin on hepatocellular carcinoma cells in vitro was significantly enhanced after light fixation [7].
The aim of this paper is to investigate the co-immobilization of selenium and bioactive algal cyanoproteins (i.e., immobilization of selenated algal cyanoproteins), which are effective in liver cancer, and their inhibitory effects on the growth of hepatocellular carcinoma cell 7402 in vitro .
1 Materials and methods
1 . 1 Sources of samples, materials and reagents
Selenium-enriched Spirulina platensis was cultured in our institute, N-(4 azidobenzoic acid-based) succinimide was purchased from SIGMA, 24-well plates were purchased from GREINER, liver cancer cells 7402 were provided by the Animal Center of Sun Yat-sen University of Medical Sciences, and the RPMI Medium 1640 and trypsin were products of GIBCOBRI. RPMI Medium 1640 and trypsin were from GIBCOBRI. Neonatal bovine serum was purchased from Hangzhou Four Seasons Spring Bioengineering Material Research Institute.
1 .2 Preparation and purity test of selenized phycocyanin (Se a SPC)
After repeated freezing and thawing, the supernatant was graded and precipitated by ammonium sulfate, and then analyzed by DEAE I 52 cellulose and Sephadex G I 100 to obtain the pure product of selenocyanin, which is a blue powder after lyophilization. The purity of selenocyanin was detected by Shimadzu UV-1 240 ultraviolet spectrometer.
1 .3 Detection of selenium
Quantification of selenium in selenated algal cyanoproteins was carried out by 2,3 a diaminonai (2,3 a DAN) fluorescence spectrophotometry [8] . 1 . 4 Preparation of photoactive selenated phycocyanin (Se a SPC) [9, 10].
Se I SPC (200 mg, 1 μmol) was added to a solution of N I (4 azidobenzoic acid) succinimide (26 mg, 0.5 mmol) in dimethylformamide DMF/PBS (pH 7.4) (4:1, 20 mL) at 4°C for 48 h with stirring in an ice bath. 1 mmol) in a solution of dimethylformamide DMF/PBS (pH 7.4) (4:1, 20 mL) at 4 °C for 48 h. The reaction was carried out at 4 °C for 2 h. After the synthesis, the reaction was carried out on an ultrafiltration membrane.  At the end of the synthesis, the azidophenyl-derived Se a SPC was purified by overfiltration membrane (Milipore Molecut " , 5KDa) .
1 .5 Light fixation
Before photofixation, the photoactive Se 1 SPC was first covered with tissue culture polystyrene (PSt) substrate, dried at low temperature, and then irradiated with UV light to fix the Se 1 SPC on the PSt material (as shown in Figure 1). After light fixation, the substrate was washed repeatedly with PBS(I) solution until no Se- SPC was leaked out.
1.6 Growth inhibition of isolated hepatocellular carcinoma cell 7402 by Se an SPC photoimmobilized materials
A newborn bovine serum containing 10% neonatal bovine serum (V/V), penicillin 0 . 03 mg/mL, streptomycin 0 . 05 mg/mL of RPMI Medium 1 640 containing neonatal bovine serum 10% (V/V), streptomycin 0 .
The sample was analyzed with a mixture containing trypsin 0 . 025% (w) and EDTA0.02% (w). 0.025% (w) trypsin and 0.02% (w) EDTA (i) PBS was used to digest hepatocellular carcinoma cells, and the cells were washed with serum medium and serum-free medium successively, and finally a cell suspension of 15 × 104 cells/mL was made with serum-free RPMI Medium1640 medium.
A 70% (V/V) ethanol solution was used for the immobilization of the various concentrations (0 . 01, 0 . 02, 0 . 05, 0 . 10, 0 . 25, 0 . 50, 1 . The cells were sterilized in 24-well polystyrene Petri dishes with Se I SPC (00 mg/ Well), and then, 1 mL of serum-free cell solution (15× 104 cells/mL) was added to each well. Cells were incubated at 37 ℃ in a 5% CO2 incubator for 12 h. At the same time, cells were also incubated in 24-well polystyrene dishes immobilized with the same concentrations of SPC and SPC + Se (sodium selenite), and in 24-well polystyrene dishes containing SPC (0 . 50 mg/Well), Se i SPC (0.50 mg/Well), selenite (0.50 mg/Well), and Se (0.50 mg/Well). 50 mg/Well), sodium selenite (0 . 04 μg/Well) and pure serum-free medium in parallel .
At the end of the culture, the number of cells was determined by measuring cellular DNA [11] .
2 Results and Discussion
2 . 1 Results of purity and selenium content of selenized algal cyanoproteins
The absorption spectrum of selenized algal blue protein sample was measured by UV spectrometer and the maximum peak was located at 620 nm, A620/A280 > 4 . 0, indicating that the purity of selenocyanin has reached the requirements of pure product [12]. The selenium content of selenized algal blue protein was measured to be about 80 μg/g SPC .
2 .2 Inhibition of hepatocellular carcinoma cells by immobilized concentrations of selenated algal blue protein

Figure 3 shows the inhibition of cancer cell growth by fixed low concentrations of Se - SPC, SPC . The inhibitory effects of immobilized Se - SPC, SPC and SPC + Se were nearly the same at concentrations lower than 0.05 mg/well. At a concentration lower than 0.05 mg/well, the inhibitory effects of immobilized Se - SPC, SPC and SPC + Se were nearly the same, and the optimum concentration was 0.02 mg/well. The optimum concentration was 0.02 mg/well. Therefore, it can be analyzed that the main inhibitory factor for hepatocellular carcinoma cells at low concentrations was immobilized phycocyanin (SPC), and Se had no effect on hepatocellular carcinoma cells. The selenium content of the selenized algal cyanoblasts measured by us was 80 μg/g SPC, and the level of Se was much lower than the blood selenium level of liver cancer patients at a lower protein concentration [1].
The inhibitory effects of Se-SPC and SPC were different in the concentration range above 0.05mg/well. In the concentration range above 0.05 mg/well, the inhibitory effects of Se-SPC and SPC were different. As shown in Figure 4, immobilized Se - SPC had the strongest and fastest inhibitory effect . At an immobilized concentration of 0.25mg/well, the inhibition of Se-SPC was the strongest and fastest. At an immobilized concentration of 0.25mg/well, the inhibition rate of Se-SPC on cancer cells reached 56%, while that of SPC was only 26%, indicating that the effect of Se was more prominent. With the gradual increase of the concentration, the reduction of cells in the culture wells fixed with Se-SPC and SPC was consistent, indicating that selenium and phycocyanin together inhibited liver cancer cells in the high concentration range.  At present, the inhibitory mechanism of selenium on hepatocellular carcinoma cells has been relatively clear, researchers believe that selenium has a selective effect on the cAMP metabolism of liver tumors, resulting in the control of the internal environment of cell division and reproduction. It has also been suggested that selenium may play a role in the early growth of hepatocellular carcinoma cells by regulating mitochondrial function.
Organic selenium behaves differently than inorganic selenium, immobilized selenium behaves differently than selenium in solution . Immobilized selenium, even at low concentrations, has a certain inhibitory effect on cancer cells, such as immobilized 0 . For example, if 0.25 mg/well of selenized algal blue protein is immobilized, the maximum amount of selenium immobilized on the surface of polystyrene material is 0.02 μg. 02 μg of selenium immobilized on the surface of polystyrene material is at most 0.02 μg, but due to the synergistic effect of selenium immobilized on the surface of polystyrene material with phycocyanin, the inhibition rate of the cancer cells reaches 56%, which is 30% higher than that of phycocyanin alone. The inhibitory effect of immobilized SPC + Se was very close to that of immobilized SPC, indicating that unfixed inorganic selenium did not have a significant effect.
As a control, sodium selenite (0 . 04 μg/well) solution did not inhibit cancer cells significantly .
2 .3 Inhibition rates of immobilized Se $ SPC and natural Se $ SPC on hepatocellular carcinoma cells
Hepatocellular carcinoma cell growth was not inhibited when cultured in parallel in serum-free medium without Se-SPC . The growth of hepatocellular carcinoma cells was not inhibited in parallel culture in serum-free medium containing natural Se-SPC (0.50 mg/well). When cultured in serum-free medium containing natural Se-SPC (0.50 mg/well), vacuolation and disintegration were observed in a small number of cells, and the inhibition rate was about 6% (Figure 5), which was similar to that of the same concentration of natural SPC on cancer cells [7]. In addition, compared with the same dose of natural selenocyanin and immobilized selenocyanin, the inhibition rate of the latter was obviously higher (3, 4 vs. 2).
In our previous study, we found that the growth-promoting effect of cured EGF on CHO-k1 cells was stronger than that of natural EGF [13].  The reason may be that the immobilized EGF on the substrate interacts with the EGF receptor on the cell surface for a long period of time. In the present study, the enhanced inhibitory effect of immobilized selenocyanin on hepatocellular carcinoma cells suggests a similar possibility.
