so there's these dudes jack n felix. remember them, they r important. jack n felix meet in college after bonding over the shared interest of making an animatronic burger restaurant. which is oddly specific but ooh yuh get it i guess. anyways they make their funky lil restaurant n it's good for a while. jack gets married after he leaves college to rosemary and they have 3 children. ed, molly, and sophie. molly is the youngest n sophie is the oldest.

so one day jack asks felix, his bestie, to drive edd and molly to this school event. also, felix is an alcoholic btw. ya. rlly bad alcoholic. probably was already tipsy when jack asked him to drive his kids tbh. anyways, felix seems hesitant but jack pushes him to say yes. so he does. he drives them to the school party. gets drunk af. and chrashes the car on the way home. he kills edd and molly but not himself. felix survives.

anyways, he feels so guilty about killing them that he buries them in the woods next to the road. lol moment.

anyways. jack goes missing a few weeks later. he's still alive maybe idk. we don't know. anyways, rosemary keeps looking for her husband and kids. she goes to the restaurant everyday looking for jack. spolier alert she dies. bon, one of the animatronics, kills her. bon dismembers rosemary and shoves her in sha, one of the sheep animatronics. and yes, the creator confirmed he was inspired by fnaf.

as for sophie, she is given pills to make her forget her childhood. so she doesn't remember her dad or mom or siblings or anything from her childhood. she learns it all tho through an arcade game her girlfriend shows her. oh ya she's confirmed lesbian btw.

anyway theres like 3 other subplots. they all tie together tho.

like ashley park. she is a technician who is sent to the bon's burgers storage unit to check up on the animatronics. she goes to investigate the billy the clown animatronic and get dismembered and shoved into billy by bon.

later, like 3 yrs later, brian stells is sent to the storage unit to finish the failed job of ashley and her team. he goes into the storage unit and smells ashleys rotting corpse. billy was an unused animatronic so it just stayed in the storage unit. he does a checkup on bon who goes haywire and tries to kill brain. he does kill him, and a short cutscene after brians death we see a text on screen say 'he thought i was her' meaning that bon thought that brian was sophie. why bon has it out for the waltens idk.

then theres susan. she worked as am engineer for bons burgers and tended to the animatronics. she goes to check up on the. after the show (this takes place after rosemary's death btw' and whilst checking up on them is attacked by bon and shoved into banny (another animatronic) she survives, but starves to death as felix forgot to check up on the backrooms before closing.

Today I started by trying to help Sarah with the HPLC purification of our product wire. However, due to the extreme challenge of separating compounds that only differ by the degree of alkylation, she basically kicked me off when Marco came and asked us to finish the media.

Thus today I made the rest of the media for pAF MS2. I added the 18AA solution w/o C & Y, then the 20% arabinose, a heavy metals soln, tetracycline, and ampicillin to the previously autoclaved media (pictured is all the stuff I added under flame so its sterile). I took a sample as a blank then added the overnight cultures marco prepared. I let them incubate at 36ºC in the stir thing. After they reached an OD 600 nm of 0.8 (after 5 hours at about 6 pm), I added ~10 mL of pAF solution. Tomorrow we get pAF MS2!!! Hells yeah!! and we made so much we probs won’t have to make any more until late fall. 

Meantime, Sarah was able to separate the three compounds using the magic HPLC. Papa Bless, I have no idea how or what she did, it was magic. Hopefully we get a good yield. 

Today I also took an NMR for “fun.” I went to the facility and walked in. There was nobody there except some dudes from the Hartwig Group on the AV-500. Totally Chill right? so I log on to the AVB-400 and go to remove the standard. As I’m removing it this short asian guy shows up behind me and coughs a little expectantly when I don’t hand him the floater. When I start to put my sample in it he asks “do you have a reservation??” and I’m like “WAT.” I back down obviously but I’m confused about why he was being kinda rude? I thought the procedure was to log on to the computer to claim it before putting a sample in? I am also confused about why the system let me log on to an NMR at 4:09 when it was ‘reserved’ at 4:10 pm... How am I supposed to know if this guy had a valid reservation??? What the frick???? What if I told him to fuck off I was here first???? Idk like none of this NMR shit is clear to me. Which ones can I use?? which ones are walk on?? which ones are reservation only??

More importantly, which machines are which. There are no labels. how do I know what one I’m using? Where are the cheap AF signs that say AV-300, AVB-400, AVQ-400, AV-500, etc???? I could make these signs. I volunteer. takes 5 mins. I could bring my own. I have at least 10 sheets of paper and a sharpie and tape. fuck me. 

Anyways, I ended up on the AVQ-400 and that’s when hella people started to show the fuck up. Like everyone was trying to avoid 4:20 blaze it by taking NMR at the same damn time. So my idea of trying to get some practice for ~30 was p rekt. I took the H NMR just fine and still had ~8 mins on the machine but I had made the panel with the button to start the C13 NMR go away and had to frantically look for it for 5 mins and then said fuck it I’ll wait again by the time the person behind me pointed to where I needed to click. I could have figured it out, it’s just this damn time crunch where everyone is all over the fucking place. Like calm the fuck down! We will all get NMR, I just need 20 mins straight on a machine. Ugh. I want to practice more. 

My Carbon spectra looked like crap. Not even worth it to take. And that’s what Sarah wanted from this compound. The proton NMR looked funky fresh. It didn’t match what she had already and was tripping her out. I’m not sure what was going on. chemistry is hit or miss honestly.........

This was my 2nd time taking an NMR by myself, I have taken less than 10 NMRs in my life, and I haven’t had the low field training yet. I do that on Friday. Hopefully I can level up this skill stat. 

Overall, it was a good day. I just want to get up earlier because I start school soon and need to be on that 6:30 AM wake up life.
