“My dear Onor,
A thousand thanks for your sweet long letter, which really cheered me. The photos, too, gave me such joy as they give me a good idea of your charming little chapel. How delightful it must be to attend Mass in the middle of the wood. Besides, I think it is a clever way to meet various members of the clergy from Hesse. - Wolfsgarten gets lovelier by the year. It is a pity, though, that the temperature there was still 30 degrees at 10 o’clock in the evening. It has turned much cooler again here - Nicky is at the army camp almost every day, comes home at 1 o’clock at night and leaves again at 8 o’clock in the morning. 
It is very tiring for him but he does not want to leave me on my own for too long. I also had to send off my boy without me. - You can imagine how hard it was for me not to be able to take him there myself. I placed him in the care of big Olga three times and Tatiana twice. The boys in the family start wearing officer’s uniforms with star and ribbon at the tender age of 7 - it was his regiment’s anniversary - and then another inspection (in this case, though, as a sailor). -
Arthur has come to the manoeuvres, and it is such a joy to see the dear boy here; - he dined with me and the girls yesterday (N.[icholas] was at Krasnoe). It was really quite a strain as my heart is enlarged again - got overtired, ladies reception. - Since our return I have been out once for a drive for half an hour plus 1/2 an hour in my wheelchair. - I am doing a lot of work for our bazaar (all of it lying down) - and I have already painted 80 items - suddenly I find myself able to paint flowers. This surprises me very much. I am also finishing my fourth cardigan. Am ordering and thinking up all sorts of things. - Would you be able to send me Wiebelchen and small salted pretzels to sell? - Could you be so kind as to ask Georgina to send me 10 pillow cases (chintz ones, with buttons). Römheld can pay the bill. -
I have been seeing people and answering queries regarding the weddings and discussing Kiev for days on end. These are actually two ‘new style weddings’ in the family, so there is a lot to think about. Unfortunately I shall not be able to attend these weddings but the children are going and Anastasia will wear a Russian dress for the first time at Ioanchick’s wedding.
They all have grown a lot. Tatiana is already taller than Olga, whose dresses almost reach the floor now. - Skirt hemlines drop and hair goes up when they reach the age of 16 - how time flies. Tatiana K.’s wedding will be a very quiet and simple affair at Pavlovsk. - I will have to collect the Serbian bride from the railway station - and hope to be able to turn up for the family dinner on the eve of the wedding. She will not be wearing a crown or cloak; I will help her dress and be there to congratulate them. -
On the 28th we will leave for Kiev for a week - for the unveiling of a statue, 2 parades ect. - I will barely put in an appearance; will have to see how strong I am, and that won’t be much. - We will then board the Standart at Sevastopol on 7th September, Old Style. We cannot stay at the house till the 20th as not everything is dry and ready yet. The photos we receive every month give us a good impression of the house, and I am grateful to Ernie for giving me so much good advice. There is still much to be done before we can settle in. -
Forgive me for writing in pencil but I am still in bed - but should get up now. Tenderest kisses to you and darling Ernie. - God bless you, Onor dear. Your devoted Alix.

Comprehensive genomic profiling of head and neck squamous cell carcinoma reveals FGFR1 amplifications and tumour genomic alterations burden as prognostic biomarkers of survival

Bieche BackgroundWe aimed at identifying deleterious genomic alterations from untreated head and neck squamous cell carcinoma (HNSCC) patients, and assessing their prognostic value.Patients and methodsWe retrieved 122 HNSCC patients who underwent primary surgery. Targeted NGS was used to analyse a panel of 100 genes selected among the most frequently altered genes in HNSCC and potential therapeutic targets. We selected only deleterious (activating or inactivating) single nucleotide variations, and copy number variations for analysis. Univariate and multivariate analyses were performed to assess the prognostic value of altered genes.ResultsA median of 2 (range: 0–10) genomic alterations per sample was observed. Most frequently altered genes involved the cell cycle pathway (TP53 [60%], CCND1 [30%], CDKN2A [25%]), the PI3K/AKT/MTOR pathway (PIK3CA [12%]), tyrosine kinase receptors (EGFR [9%], FGFR1 [5%])��and cell differentiation (FAT1 [7%], NOTCH1 [4%]). TP53 mutations (p = 0.003), CCND1 amplifications (p = 0.04), CDKN2A alterations (p = 0.02) and FGFR1 amplifications (p = 0.003), correlated with shorter overall survival (OS). The number of genomic alterations was significantly higher in the HPV-negative population (p = 0.029) and correlated with a shorter OS (p < 0.0001). Only TP53 mutation and FGFR1 amplification status remained statistically significant in the multivariate analysis.ConclusionThese results suggest that genomic alterations involving the cell cycle (TP53, CCND1, CDKN2A), as well as FGFR1 amplifications and tumour genomic alterations burden are prognostic biomarkers and might be therapeutic targets for patients with HNSCC. http://ift.tt/2Fk1pPj

AUNTIES (Nadia Sablin) 
Nadia Sablin’s approach to her personal story wraps the subjects, her “Aunties,” in an idyllic setting. Sablin’s subject itself –the life of her aunties—is enough to set her work apart: the auntie’s constant stoic expressions denying us a view into their humanity, their timeless rustic world. But more than the uniqueness of her subject, Sablin skillfully renders every scene’s mood using the available light to, at times, create tension and division in the images, or else present a welcoming atmosphere.

In the above image, a normal interaction between her aunties is lit by dramatic dappling of warm window light. The harshness of this side light produces deep shadows in the background and highlights the texture of the women’s faces. This deep contrast creates drama, suggesting tension, which is heightened by the symbolism of their pastime: working over a puzzle. Two women sipping tea under such dramatic lighting would not communicate the same message, and under less dramatic lighting circumstances, the mood of this exact same subject would be milder and still. In fact, Sablin produces a very similar scene In direct contrast to this image, this time, under soft, window light pouring in from above the subject.

This scene, auntie-less, bears less tension than the previous. The window light produces soft, graduated shadows; there are no hard lines; colors are muted. No action is taking place within the image. What makes this still life so successful is the alignment of its elements: stillness (no action/tension) with softness (graduated shadows and muted color).

Sablin also successfully uses harsh light as a dividing tool in her images, separating subjects and creating shapes. One such example is the above image, which makes use of full daylight pouring in through an open car-port or shed. The shadow line places one auntie in the shade, the other in the bright sun.
ATMOS (Naoya Hatakeyama)
Naoya Hatakeyama explores atmosphere in her printed book, Atmos, through a two-sided volume that features a collection of images featuring smoke and steam that fills a scene’s atmosphere on the left side, and another collection of images featuring landscapes on the right. With the left side, several of the images make use of direct light to highlight a billowing cloud’s texture, while several make use of strong backlighting to illuminate cloud.

The cloud in the above image is lit by strong side light. The shadows cast by this light create our perception of texture.

This image is an example of a backlit cloud, which produces a soft glow rather than the harsh shadows of the first example.
Both methods produce different moods. While we get a sense of the grandness of a large, billowing, textured cloud as in the first image, the second image with soft diffused light is more serene. 
EYEWITNESSES (Kennan Ward)
In Kennan Ward’s image of polar bears in front of a setting sun, the warm backlighting serves to add a dynamic range of colors as it filters through the atmosphere. Ward made use of the sun’s position to create a rimlight on the bears and to highlight the texture of the ice.

The inclusion of the background not only provided context for the viewer and established location for the bears, it allowed a grander view of the strong light source. The mood of this scene is ethereal –not only by the saturation, but by the luminance of the colors. Had the light shifted slightly by either peaking out from behind the clouds more, or further being covered by them, the feel of the image would be lessened. As it is, the balance between high-key sky and shadow is pleasant and light, but still full of detail.
