A hc I have is that Orochimaru always had the natural ability to poison people through bites (due to their clans affinity to snakes), but it might have had just a slightly numbing or maybe even hallucinagenic effect, nothing truly lethal.
And they then learned how to infuse their chakra into it and inject it into others through their fangs. Due to the especially destructive chakra nature it of course made the venom more dangerous and later on was further developed into the cursed seal.
