I like that scene because it's so funny actually. They were highly confident that their plan would work. Bradford and Xever, when they set aside their rivalry make actually a great formidable team together. Once they fight alongside each other, they actually make a terrifying duo.

So they see these two constantly together, fighting together and assume that they must be friends. But they are just partners who were forced to work together and only tolerate each other.

Xever laughs and says they are not friends and the turtles are instantly caught off guard:

They all even keep staring at Bradford in confusion. They absolutely did not expect that. It backfired on them.

They are absolutely uncertain about the situation now. Leonardo has no longer the confidence in his voice and becomes squeaky while also stuttering.

Xever really has no reason to help out Bradford and just keeps cutting the ropes. Regardless, he definitely picked up on their shift in behavior.

Xever is attentive of things like that because he is good at reading people and the situation at hand. They were entirely caught off guard and that was his first cue they would not actually go through with it. The sudden loss of confidence, squeaky voice and stuttering were just other hints. They were standing there trying to make empty threats and Xever was aware of it.

Which makes this funnier:

I find it interesting that he confronts Xever about it - because he definitely was mad about the situation. You can tell on his smirk that he is lying - the turtles made no secret about it. It feels like he is saying that just to mess with him further.

You can tell he was mad.

He went right for Donatello after he said that. Regardless, they were the ones who put him into the humilating situation in the first place. He was tied up inside a trash can, rolled across the city, dropped and the turtles teasing him about it. He was at the mercy of his enemies, terrified for his life and everything depended on Xever.

He fully intends to settle the score here, you can see it on the way he fights and he does take Mikey, Donnie and Leo (the last one with Xever's help) out. A lot of his current frustrations was taken out on them.
