just stumbled across your art for the first time & im very curious as to what exactly your starting point for creating soul variants was? by that i mean, what made you begin creating them in the first place, what served as the inspiration for all of this? id also like to add i scrolled your entire archive and i love how expressive your art is! apologies if youve already answered something like this as well. thank you, have a nice day!

Hey, thank you very much! I'm always so glad when people enjoy my varianting, and honestly that's really heartwarming that you went through my archive (especially since I still remember how long it took me to get all of my art up at the time onto here haha)

I've been asked similar questions to this before, but I don't mind it at all, it's always fun to answer even if the history on variants is actually a bit messy haha (to the point I had to kinda just pick a day for keeping track of the Variantverse's anniversary, which I actually use this drawing for reference since it's the first real time there were multiple AU Souls in one art haha).

I'm not joking about the history being messy by the way, you better strap in because this is not a question I'm usually able to answer in just a few neat sentences (for your time there will be art at the end though haha)

So basically, aside from the really old (I'm talking middle school old) AUs I had of Soul (which... don't really have updated art aside from being retroactively really important to Variantverse lore hah), variants initially started as me just finally joining the Soul Eater fandom and just doodling canon Soul in different ways. After that it basically went like I want to draw him in a different outfit, to I want to draw him as Pokemon (context like this involved Discord but that's another story hah), to okay, I want to draw him weird and no one can stop me, and then came one morning where I did a doodle session of drawing him in a lot of non-human designs for the fun of it.

This is the shortened version of it, because there were doodles in between including an entire comic saga and an entire AU that didn't end up on the Variant List (*until much later anyway) because the list hadn't been established yet. Have I mentioned the "List of Soul Variants," by the way? Some know way back when I used to keep a list of all the Soul variants I was making and even gave them all numbers (it was kind've a big deal to the point it was actual in-Variantverse lore), however shortly after it reached 1,000 variants the list went defunct (also a big deal) and I no longer keep track of how many there are. The numbers don't matter anymore, but it contextualizes some things haha.

Now to actually answer the question, where does "variants" start: #019. After the previously mentioned doodle session, I got a Thought™ after waking up to start a [secret project], however I needed a handful more variants for it, and ended up doodling some more. I honestly have no idea if I should still be calling it a secret considering it's been four years now and I never ended up drawing it up despite- Anyway, this is also exactly the point in time I started to call them all variants (theorized by me to be in reference to "regional variant" Pokemon, ones that take a new appearance when adapting to a new region).

So what inspired me? Simply because I just kept going. I started thinking of a reason the variants I'd made so far were all together during the events of [secret project??], I started thinking of a couple Souls watching over the event (that ended up becoming the main characters of the overarching story) and new unrelated variants that were goofing off in the background, I started thinking of an antagonist, not just tied to the events of this, but to the multiverse at large, and I started thinking up what happened afterwards. I ended up giving the first 25 variants lore when I wanted to flesh out their designs, I added new variants to interact with the overarching story, and I just kept doodling new variants for the hell of it, all because it was fun and even if I never really revealed what [secret project] was (at least to only more than a handful of people hah) people in the circles I was in really enjoyed seeing them. Need I remind you that I stopped keeping track of how many there were after about 1090 variants. So yeah, it just kept going.

I guess in a way this is more answering where does the Variantverse start, but it's such an integral core concept that I think is heavily responsible for this madness snowballing to begin with haha, and things like the "Soul Variant Emporium" (an entirely different inside joke) and different Souls interacting out in the multiverse still resurfaces from time-to-time in the little blurbs I give art. I'm just particularly fond of capturing variants from an angle as if writing Pokedex entries, which I think kinda loops back to that idea of where the word variant comes from.

Honestly I just really love character design, and Soul genuinely is my favourite character of all time, so it's very fun to combine the two (especially when integrating concepts I find I like, such as the mass amount of werewolves hah) and I couldn't be happier that people also get a kick out of it even if I feel just a little guilty I don't really draw non-Soul designs much anymore haha (and I will say the madness brought upon my ask box as been an absolute hoot). So yeah, to sum it up: It started because I just had doodle ideas, and I got inspired by never stopping having ideas and everyone unfortunate enough to be caught in my ramble radius happened to enjoy said ideas.

As promised (thank you for enduring, I hope this was at least interesting haha), here's the most notable early variants that got me rolling the snow around:

Awakened Soul is the one I mentioned starting off the later doodles with the idea of "variants" first in mind, but Trickster (giving Soul a new outfit), the Pokemon variants, and other Souls of other doodlings are also notable to the entire messy history as to where this all started. I'm still planning to continue trying to draw all of the 1000 variants of the old list eventually and I look forward to sharing all these silly Souls as always.

I swear I'll give the first AU Soul I'd ever drawn real art one day.

Destiny #017

1)  Hi! I know, long time no see. I’m incredibly sorry for not reviewing sooner, but I guess better late than never, huh? In any case I’m here to stay! Reading this chapter reminded me of how interesting this story really is and I can’t wait to read more. But seeing as it’s been some time I’m going to reread it in order to freshen my memory (I decided to review anyways since I can still review the quality of your fic, which is as I remember, very good). I admit when Peter first mentioned that Ruby was easy to control because of his lack of control over his emotions (and whatnot) unlike Sapphire, I was a bit confused. Wouldn’t it be the other way? I thought to myself, but then I remembered that Sapphire had done some growing up herself. She might’ve not had as much control previously but she certainly does now. As this was kinda proven later on when she was contemplating on what emotion she should be feeling at the moment. And this can be seen as indecision, but I rather thought that this was proving your previous statement correct and that this was her way of finding a little control of the situation. Speaking of what happened a bit later, I was pleasantly surprised of Blue sparing Sapphire’s sanity. I always had hope in Blue despite her obvious turning... but this really proves that Blue can (and most likely will) realize that despite Peter’s kindness she’ll have to betray him because what he’s doing simply isn’t right. I realize now that she probably also followed him because of him brainwashing her, but I can’t remember this particular detail... damn maybe I should’ve reread this before reviewing. Ah well, I’m this far into it anyways. In any case, although this chapter was short I fully enjoyed it. Especially Y’s inner monologue at the end. I do have a soft spot for light angst :’) (I’m sure you can call it that, right...?) so, thank you!

For Ruby and Sapphire’s “control” issue, I was actually thinking about their little Hidden Power interaction. Sapphire has strong intuition and instincts, and is in full control of her senses. Or at least that was a part of it, anyways. This chapter was written years ago, so I don’t remember all the details. Hehe.

As for Blue... There’s a little character arc for her across all of my stories. If you read my stories in order of creation, you can kind of see it. First is when she’s neglected and is just rolling with it while feigning cheeriness (SE/SA/SR), but then it just piles up and she goes into depression (SA/SL/Destiny), then she kind of snaps and goes rampant (SL/Destiny), then she finds the one piece of true happiness in her life and starts to turn it around (Destiny), and eventually successfully attains happiness and stays that way (Destiny/Legacy). So you’re at the moment in Destiny where Blue has gone rampant but is trying to turn it around.

... I think I need to reread this story too. Hehe. I don’t really remember what happens in each chapter... except for a certain few, that is.

Destiny #018

1)  That’s... awful... Poor Y, who was already suffering so much, both physically and mentally. She didn’t even get to hear X say what he wanted to say to her. And if they don’t meet again she’d live the rest of her life not truly knowing where she stood with X. Always filled with guilt. Such is the sadness of the situation, but I can still hope! Hope that Y will meet X again and he’ll be able to tell her his thoughts. Though come to think of it, poor X too. He’ll have to speak with his seniors and he’s obviously really uncomfortable with that and doesn’t even have Y around to help him. Welp, to go to another depressing topic there’s also Sapphire’s situation to discuss. It is at the point not that disobeying Peter *would* be interfering with his plans, since it’s crucial that Sapphire be broken. Will Blue be able to disobey? Impossible to know, you can only hope that she does. On another topic, after rereading the fic (oh god I’d forgotten how long this was, it’s truly amazing the dedication you’ve had for this fic) I kept thinking back to Peter’s “blank eyes” you mentioned in the earlier chapters. This is a vague and rather shoddy theory, but the only thing I could think that would be the cause of the blankness is him being brainwashed as well. This is rather obvious, but this leads to who might be brainwashing him. Clearly this would be someone (or something..?) that would benefit from the restoration of the legendary’s and this would bring us to suspect #1: Zygarde. Now, I haven’t actually read the XY arc nor played the game (ik ik but I can still have a fellow feeling for X and Y) so I don’t know what this dude is capable of, but I can assume that this is within the realm of possibilities. And it would make sense with the whole blinking lights thing that Blue noticed that was going on a couple chapters ago. ‘Cause I mean, why else would this random guy help the legendaries, it’s all just a bit too sketchy. I am no detective, so I think these are fairly obvious, but they’re all I have going for me right now. This was a lovely chapter (writing of course, I can hardly apply that to the atmosphere), and I can’t wait to read the next one.

Poor Y indeed. Really. Poor Y... hehehe...

Destiny’s not THAT long, is it? ... 220 000 words... well, it’s not longer than SA which has 225 000, but... that difference is practically negligible. ... I wonder how long Legacy would end up... Anyways.

I don’t know if it’s obvious or not. I have a hard time with that. Sometimes I put in obvious hints in my stories and no one catches on. Sometimes I put vague hints in my stories and no one catches on. So I can’t tell if it’s subtle or blatant. I think for the Mega Hunter, there were a lot of subtle and blatant ones.

Now for Zygarde... well, the Neural Para... er... mind control is not really in its arsenal. Destiny’s plans were written shortly after XY games were released. Zygarde was kind of worthless there, but given what happened with Kyurem, I suspected that it would get a cooler new form. But then in SM, it turned out that its cooler new form was just a massive health buff. And it still gets annihilated by Xerneas, so... pfft. But I think I used its signature moves pretty well. Especially, say... Core Enforcer. Hahaha.

Destiny #019

1)  Ok, first of all, WHAT KINDA QUESTION IS THAT? How can I choose? Both options of your review survey thing are so sad... but after a moment’s thought, I think the second option is better. If Y were to continuously push herself then she’d just die. End of story. But with the second option, although incredibly risky and kinda pointless for X to go save her, the chances of them both staying alive are possibly higher, and Y would know Ax’s true feelings. This I think, would be what you referred to as the “happy ending”. Unless you actually said it outright that it’s the opposite and I just completely missed it. But anyways I’ve spent enough on Y’s situation lmao. I think I might switch over to Blue now, because something has been nagging me for a while. I feel like slapping this woman. I know she’s partially under the control of Peter (or assuming that my previous theory is correct, Zygarde’s), but, murder? Thankfully she admitted this chapter that she was, in fact, NOT wanting to murder anyone. But when she let anger blind her she was quite willing to do away with Green. And she has reason! Was Green did to her was beyond shitty, but killing him is just a little overtop. And she tried to justify it by saying, “Peter is the only one that has been kind to me” so it’s ok that I betray even my closest friends whom I’ve known for years even if I haven’t kept much in contact (besides Silver smh). She clearly knows what Peter is doing is wrong and while I don’t think that anyone is a saint and everyone makes mistakes, I’m still thinking “come on”. Plus, it’s not difficult to see how Peter’s kindness is really just a way to benefit him. Though I can let that one slide since it’s easy to fool yourself. Despite all that, later on in the chapter after feeling annoyance at Blur for seriously trying to blame Y for wanting to give X back *his* Mega Ring (plus why do they call it ring) just because it’d been a gift to her (and reminding myself that she also had reason to feel that way but whatever) it was very nice to see her being selfless. Which might be an awful thing to ask of her now, but it was the right thing to do. And that’s gratifying on its own, right? I feel as if these reviews have started to just be me ranting at this point. Can you still enjoy these..? But honestly there isn’t much to review at this point. I’ve touched a lot on your actual writing in past reviews, and since it’s the same fic the style hasn’t exactly change. Though I can still admire how seamlessly you seem to write, even while changing point of views. Quickly changing scenes from something a little peaceful, to a battle, which you are able to describe in detail and yet still be engaging. Ah, I got kind of sidetracked, didn’t I? In any case all I have yet to do is speculate what’s going to happen next, but alas, I have no idea. I assume there’s a somewhat happy ending, with a final battle with Peter and the legendaries, not to mention the plot twist. Guess I’ll just have to wait and see.

Huh? ... Oh right. The review survey. Hahaha. Second option, huh? I think I remember most people choosing that option. But... if you look at the choices, it seems pretty clear, right? And I just love to break expectations. Hehe...

Now, as for what’s happening with Blue, it’s explored a lot more in the upcoming chapters, with everything being explained in... 25? 26? And I do believe that all the issues you have will be resolved.

Hehe. Asking a girl, who after being neglected for years is finally trying to find some happiness for herself, to be selfless? How awful! Hahaha.

headcanon #019: hair through the eras

Mama (2012)

Ash debuted with what I would like to call shaggy emo hair.  It was a little longer and wavier than he’d had normally in the most recent times pre-debut, but Ash quite liked this style for his hair at the time and had no complaints.

Wolf (2013)

It took over a year for Knight to comeback and during that time Ash had mostly kept his hairstyle from Mama era. It was only when it came time to shoot the music video and promotional photos they he was given a new hairdo for the era, which was a reddish-brown coif. The aim was to make him look a little older and a little sportier without making him look too mature since he was still one of the younger members of the group. Though he was only seventeen, he began to earn more attention for his visuals this era than he had in debut. Despite his negative opinions on nearly everything else about this era, he once again didn’t have any major complaints about his hair style, though he was attached to his debut era hair.

Growl (2013)

Growl was the first time Ash had an obviously unnatural hair color, as he was styled with a red bowl cut with angular bangs. As was the styling of the era, the style was chosen to look youthful and easy to style with a backwards baseball cap if they chose to put him in one. He went back to a natural color very quickly after this era finished, as he found the hair dye made his hair more brittle due to being particularly sensitive to it.

Overdose (2014)

For Overdose, Ash went back to an unnatural color with dark purple hair, usually styled in a comma hairstyle. It was a popular look for fans at the time, but Ash wouldn’t personally consider it one of his favorite hairstyles he’s ever had and he went back to a normal dark brown halfway through promotions due to the hair dye irritating him again. It was one of the more popular eras for him visually again, though. Fans seemed to like his hair styled to show his forehead.

Call Me Baby (2015) 

For Call Me Baby, Ash’s hair was somewhat similar to the previous era with dark brown hair, though his bangs were shorter. He didn’t have much of an opinion on his styling this era, to be honest.

Love Me Right (2015)

For Love Me Right, Ash’s hair was lightened to a light brown with long bangs and some blonde highlights. Ash quite liked the longer bangs looks

Sing For You (2015)

While this is one of Ash’s favorite era’s musically, he probably couldn’t tell you what his hairstyle had been for it, but he had brown hair with asymmetrically parted bangs. His hair was pretty healthy at this point and it would continue to be saved from bleach for several more eras.

Monster / Lucky One (2016) & Lotto (2016)

His first two eras after his scandal brought one of his fans’ favorite hairstyles of all time, and Ash was happy that they had something to talk about other than his recent bad press. For Monster/Lucky One promotions, as well as the repackage promotions with Lotto, Ash had black comma hair.

Fanfare (2016) & Gorilla (2016) & Hey Mama! (2016)

The next three eras came incredibly close together with almost no break for Ash, so his hairstyle remained the same throughout all three — a brown wavy style that was easy to maintain.

Can You Feel It? (2016)

This time around, Ash got a medium brown bowl cut. Ash wasn’t a huge fan, but it was a popular style at the time that many other idols had and he didn’t say anything to stylists since he was just grateful he hadn’t been punished for his scandal with outrageous hairstyles.

Roar (2017)

Roar brought the first major hair color change for Ash since Overdose three years prior  —  a light brown/dirty blonde color generally styled with a wavy texture. The bleaching required to lighten his hair had taken a toll on it again, so the color didn’t last long past the end of music show promotions.

Easy Love (2017)

His hair was back to a light brown for Easy Love, one of his favorite eras musically, but again pretty forgettable as far as hairstyle. Stylists did sometimes give him pink highlights this era, namely for the music video and a select few music show performances and fan signs early in the promotion period.

Ko Ko Bop (2017)

For Kokobop, Ash’s hair was pretty much the same as it had been for Easy Love, light brown, though they had long ago given up on the pink highlights by this point. He was also on We Got Married’s special summer season during this time, so they wanted him to have an approachable and friendly look.

365 Fresh (2017) & Power (2017)

For these two eras, which included the first round of promotions Ash had ever done outside of his group, he went slightly darker with the brown and had some highlights to add texture. Unlike the past few eras, his hair was finally styled to expose his forehead again and was often styled for a wavy coif of bangs to frame his face. It was, again, a more popular era for him visually, as most eras where his bangs are styled out of his face tend to be.

I’m Young (2017) & O Sole Mio (2018) & Blooming Days (2018)

For his solo debut as well as the Knight era and the White Knight era following it, Ash ditched the shades of brown to go for a solid black that was generally styled with his bangs parted in the middle to show his forehead again. It could be argued this was the beginning of a more mature styling for him.

Critical Beauty (2018)

For Critical Beauty, Ash kept the black, though it was styled with his bangs to one side and out of his face and he had a bit of an undercut. Compared to his usual middle part, he had a side part this era. Ash liked this style, but it only lasted as long as music show promotions before he changed it again.

Tempo (2018) & Untitled, 2014 (2018)

Ash’s hair was initially dyed pink/red for the filming of his BC Portal “New Heroes”, but due to an overwhelmingly positive response, he kept it through the end of the year. By the time he was promoting Untitled, 2014, it was more of a faded pink or even simply blonde to spare his hair from all of the dye on top of the bleach.

Love Shot (2019)

After the damage his hair had suffered in the back half of 2018, it was obvious Ash would need to go back to a dark brown for Love Shot era. It matched the dark and sexy but sophisticated vibes of the song, anyway.

Troublemaker (2019)

For Troublemaker, Ash’s hair was still dark brown or black, but he had begun letting it grow out.

Now or Never (2019) & Now (2019)

For Knight’s next comeback as well as his second release with Yoona, he had continued letting his hair grow out and it was now dark and curly. Ash was really loving growing his hair out longer than he’d ever been allowed to have it be and if he hadn’t been so exhausted at time time, he would have been really feeling himself. As it hair got longer, it got more divisive, though, and there was a vocal minority of fans (and, even more so, anti-fans jumping on the opportunity to hate) who absolutely hated the new look and were more than happy to make it known in online comments and to his face.

Romanticism / D (Half Moon) (2019) & Enough (2020)

For his third solo album through now, Ash’s hair has been black and wavy, and at its longest. He’s not a very vain person, but he really does like how he looks these days and thinks he’s the closest to how he’d choose to look if he wasn’t an idol and had more of a say in how he presents himself.

Woo Ah (2020)

Ash cut his hair shortly before the release of his song. It was far more BC’s idea than his, but he sported a shorter hair style with blue-green ends for the short promotions of this song, often in a forehead-exposing, comma hair style. For several months after, as his hair began to slowly grow out again, he would still make appearances with green ends.

How Can I Love The Heartbreak, You’re The One I Love (2020)

The sound of this song was more of a classic sound, so Ash lost the dyed ends and instead stuck with dark brown/black hair that had grown out to a similar length as it had been around the same time the year before and generally styled wavy.

Bittersweet (2020)

For this comeback, Ash kept the longer dark hair that he’d had in his viral fancam a month prior, which was pretty similar to his hair from the previous solo album. His hair was permed during this time and throughout this period, he often styled his hair up in buns or ponytails.

My last #gradblrchallenge post was on Thursday. I’ve been really bad about posting the past few days because I’ve just felt like I didn’t have much to say... I also just haven’t felt very productive school-wise, since I don’t even start until October and there’s only so much pre-planning I can write about before I start to feel like a broken record. I sort of feel silly posting about grad school when I have zero experience with grad school so far (other than interviews), and I haven’t even started my program. I know that will change very soon, so it does matter. It’s also interesting to go through the pre-grad school process, where you’re worried about all the wrong things, learning about everything for the first time, etc. But I feel a little silly posting about it because I feel like I’m just clogging up the real grad student’s posts. Lol, I know I’m being ridiculous. I’ll try to just get back to posting, and hopefully I have something interesting to offer regardless of where I’m at right now. If anyone in grad school for psych or social work or something is reading this, I would love to chat with you about it! I’ve talked to a few people so far about their programs, and it’s been helpful to hear how things work, what they like and don’t like, what they’re going through. I want to have a better understanding of what I’m getting myself into.

Some things I’ve done recently:

spent a lot of time online window shopping (when you “shop” all day but don’t end up buying any of the stuff you added to your cart),

researching new phones (both online and at the phone store),

waiting for my maintenance guy to come and then dealing with all that (clogged sink and bath, clean up),

taught Trev how to play Skyrim,

went out to eat once, and to a coffee shop once,

cooked some yummy meals,

bought some notebooks I didn’t need on Amazon,

took Trev to the theosophical society library,

and generally just being kinda lazy...

Yesterday, we ordered some new phones... Ours were getting old and worn out (battery life, general use, camera failing, etc), so we decided to just bite the bullet and get some new ones. Since phones are RIDICULOUSLY PRICED, it ended up being a better deal to get a brand new Note 9 for me, and an S9 for Trev, because they had a BOGO deal going where you get one phone free and a free pair of headphones and stuff like that. I had to change my phone number though to be eligible, so that’s going to be a pain in the ass to go through and update on every form and account I own..........

Oh well though. I’m super psyched about the S pen!! It’s a Wacom pen and it’s active, meaning it’s pressure sensitive, meaning I will be able to draw on my phone like it’s an actual Wacom tablet. How cool is that???? I currently own a Jot Pro, which I love, but it’s passive. There’s no pressure sensitivity, it’s not as detailed. It’s great for doodling, but it isn’t very practical for trying to seriously draw something. I know at the end of the day, it’s still a little tiny phone screen and I’m not going to spend my days training to join Clamp, but maybe I can get in enough practice to finally feel comfortable moving forward with my web manga idea! (｡･ω･｡) It’ll be nice to have a properly working camera again, too. I’d like to start filming videos for YouTube again, and currently my camera crashes even on the lowest quality option... The phone isn’t out yet but once it’s out they’ll ship it and hopefully it’ll be in my hands and working soon! It’ll be kind of sad to say goodbye to my 512 Austin area code number... I felt like it gave me some cred...... But I’ll have a new Seattle number, so I guess I’ll have Seattle cred... It makes me a little sad though. I’ve had that number for YEARS. Plus now I won’t match Trev’s number...

Today my school emailed the orientation schedule, so I’ll be posting about that soon. I’m hoping to go out and buy a few things I need for school (mainly, a decent binder for this loose leaf textbook I just dropped $100 on ಥ‿ಥ ). Hopefully my next gradblrchallenge post will be a little more relevant to gradschool...

Chemotherapy works off of a basic premise: kill all rapidly-growing cells in an effort to wipe out tumor cells. The tactic, while generally effective, has quite a few off-target casualties, including cells that produce hair and cells that line the stomach.

  Scientists have tried to skirt the problem by creating missile-like drugs that zero in on cancer cells specifically, sparing healthy cells.

These missile-like drugs, known as antibody-drug conjugates (ADCs), have been in the works for decades, but only in recent years have they made it to clinical trials, Kimberly Tsui, a genetics graduate student, told me.

The advances are at least partially due to a better understanding of how ADCs work: On the surface of every cell are proteins that poke out—some of these proteins are only found on cancer cells, making them a perfect target for tailored drug delivery. ADCs include a shuttle molecule that identifies and clamps down on these cancer-specific proteins, transporting a drug capable of incapacitating the cell.

“ADCs are showing lots of potential in the clinic, but there’s a lot we don’t understand about how they actually work,” said Tsui. “We don’t know enough about how ADCs are taken up into the cells, or how the drug is trafficked into different compartments to ultimately kill the cell.”

Now, Tsui, Michael Bassik, Ph.D., assistant professor of genetics, and a team of researchers are tapping gene editing to better understand how ADCs deliver their deathly blow to cancer cells.

A study detailing their findings appears in Nature Chemical Biology. Bassik is the senior author. Tsui is the first author. The work was done in collaboration with Carolyn Bertozzi, Ph.D., director of ChEM-H at Stanford.

Bassik and Tsui used the gene-editing technology CRISPR to figure out which genes help ADCs weasel their way into cancer cells.

“With our CRISPR screening system, we can shut off one gene at a time to figure out which ones are important for ADC toxicity,” said Tsui. Using this set up, the team is trying to see which genes either help intensify the toxic effect, or conversely, inhibit it.

The idea is to better understand how the ADC interacts with the tumor cell and use the information to make the ADC even more toxic.

“Overall, it starts with basic biology and understanding the genes that could affect ADC toxicity,” said Tsui. For example, if a specific gene is “knocked out” or rendered unable to function, and the ADC stops being toxic against the cancer cells, that’s a sign that the gene is required for the ADC to become active in the cell. “It could even have important implications for potential ADC-resistance mechanisms,” she said.

For an ADC to kill a cell, it has to check a few boxes, most of which are regulated by specific genes. But not all ADCs get into the cell and cause damage through the same mechanism, making it even more critical to understand how ADCs vary when it comes to tumor cell destruction.

It’s been thought that all ADCs have to get chopped up, or “digested,” before they inflict damage. The understanding was that any ADC binds to the surface of the tumor cell and is taken in through a vessel known as an endosome. The endosome then shepherds the drug to a new compartment of the cell called the lysosome.

There, the ADC gets broken down into its parts, and the drug separates from the antibody shuttle. Then, the drug is moved to yet another new location into the cytosol, the goo that fills the open space of the cell. Here, it can finally do its damage.

Exactly what genes underpin that series of steps, however, is not entirely clear. That’s where Bassik and Tsui hoped their screen would come in. Indeed, their experiment revealed a handful of previously unknown genes critical to the trafficking of the drug. In particular, they found that new genes involved in the first stage of endosome-mediated transportation were critical to the success of both varieties of ADCs.

Bassik and Tsui also found evidence to suggest that not all ADCs are digested through the lysosome.

“But the screen revealed genes that implied that the digestion processes happened much earlier, presumably, before the ADC ever even makes it to the lysosome,” said Bassik. “Since ADCs can be metabolized in different ways, we see these types of new insights into their mechanism as crucial for future ADC design and development.”

1. Man who had open-heart surgery wins a $1 million lottery prize after the ticket came in a get-well card

A Massachusetts man recovering from open-heart surgery won a $1 million lottery prize on a scratch-off ticket given to him by a friend, according to lottery officials.

Alexander McLeish received three scratch-off tickets in a get-well card from a friend earlier this month, according to a news release from the Massachusetts State Lottery Commission

2. Student who sews sanitary pads for refugees now leads 1,000 volunteers

A student who began sewing reusable sanitary products for refugees during lockdown is now running a global network of 1,000 volunteers. University of Bristol student Ella Lambert, 21, learned to stitch using YouTube videos during the first national lockdown, and has since launched the Pachamama Project, which aims to end period poverty.

The languages student, from Chelmsford in Essex set up the not-for-profit group with university friend Oliwia Geisler in August last year.

“I’ve always really struggled with period pain, like absolutely atrocious period pain which would mean that I’d have to miss out on school and cancel plans last minute,” she said.

“So although I’ve been really lucky, and I’ve never had to experience period poverty as such, I do know what it’s like to have to miss out on really important things and appointments because of my period.

3. Parliament of Canada unanimously passes Bill C-4 banning conversion therapy for adults and youth

The federal government's updated bill to ban the practice of conversion therapy has been adopted unanimously by the House of Commons.

MPs voted to fast track Bill C-4 on Wednesday afternoon. The Commons erupted in applause after the vote and several Liberal cabinet ministers walked across the floor to shake hands with and — in some cases — hug their Conservative counterparts.

4. UK; 13-year-old girl uncovered a hoard of 1300 BCE Bronze Age axes on her third metal-detecting trip

Milly was scouring a field near Royston, Hertfordshire, when she made the find - the first of 65 artefacts dating from about 1300BC. When her father dug out the first one, Milly said they joked it might be an axe - and it was.

Archaeologists were called in to excavate the entire hoard, which will be sent to the British Museum, London.

5. Making aviation history: Madison plants fuel United Airlines aircraft with passengers aboard

For the first time, a jet flown by United Airlines entirely powered by 100% plant based fuel from corn stalks flew from Madison WI to Chicago Illinois. The fuel is made from farm waste corn stalks and cobs, not corn seed based ethanol and is 100% renewable with 50% less green house gasses than regular jet fuel.

6. 30 white rhinoceros have been successfully transported from South Africa to Akagera national park in Rwanda, in the largest translocation of rhinoceros in history. A fantastic measure to protect this amazing species.

The 30 white rhinos arrived at their new home, Akagera national park in eastern Rwanda, yesterday. It is hoped Akagera will become a new breeding stronghold to support the long-term survival of the species. Down to an estimated 18,000 animals across Africa, white rhinos are classified by the IUCN as near threatened, with numbers in decline largely due to poaching, driven by demand for their horns.

7. Thousands of bees make it out alive after being buried by La Palma volcano ash for 50 days

They had sealed themselves in by creating propolis, a resinous material with which they plugged any gaps, and were able to survive on their food reserves.

ON1 360 plans start out with 10x the storage amount for 20% less cost compared to competitive plans available today. ON1 360 plans include the Software and Service option as well as a Service Add-on option in both monthly and annual payment options. The Software and Service plan includes a subscription license to ON1 Photo RAW 2020, any and all software upgrades and updates, the ON1 Photo Mobile connectivity, and the 360 syncing service. The starting plan includes 200GB of storage at the low cost of $7.99 per month or $89.99 for the year. ON1 still offers Photo RAW as a perpetual-license product separate from ON1 360, for those who prefer to own their software. Customers who own Photo RAW can choose the ON1 360 Add-on and those plans start at $5.99 per month or $59.99 for the year and come with 200GB of storage. What’s Ahead in the ON1 Roadmap

ON1 will continue developing ON1 Photo RAW for the desktop and ON1 Photo Mobile for devices. There is a detailed product roadmap for ON1 Photo Mobile already available from the ON1 website. Future plans include ON1 getting back into developing products that work within other photo editing workflows as well.
