You know, now that I have kinda written my introduction I feel like the entire premise of my thesis is a huge mess. Like the hypothesis makes sense to me and all but the experiments I did feels so inconsitent.
My supervisor can be such a hypocrite at times becuase he says that it's important to keep all the conditions the same but then he never actually does it? Like what??? I needed to add mast cells to my media and he said we should keep the number at 50k but when I ask him how much I should add after I've counted cells he's just like: add all of it even thought the cell number I counted was clearly above it?
Also he kept leaving out the control conditions at the beginning until I commented on it because IF YOU DON'T HAVE A NEGATIVE CONTROL HOW THE FUCK ARE YOU SUPPOSED TO SEE THE DIFFERENCE IN NORMAL AND CHANGED CONDITIONS THAT'S SCIENCE 101
Yes of course it's more work and more cells but dude you can't leave out the negative control that's so tremendously stupid. And then he adds a positive control in one experiment but not in the others? Because as far as I know I have four experimental set ups and for one of them no postive control. Maybe for two because I don't know how the models for the last experiment look like.
Also this reminds me that I need to do my results section soon. I need to figure out how exactly I should structure it, going by type of result (staining and qpcr) or experiment (2D, epidermis, dermis, whole skin). I think i might go with type of result.
