Differences:

comes from ”to become” なる;
used when we refer to future;

used to refer to present situations;
more objective;

a bit more formal;

conditional form; If we don’t do sth…
しなくては
more “delicate”;
a tad less formal;
might imply “my own duty”;
more common in speech;
ない in its て form + は = to not do something is…
 しなければなりません - If we don’t do X, sth else won’t become or we won’t be able to move forward – that would be the literal meaning.
しなくてはなりません - By not doing X, we won’t be able to move forward.
しなければいけません – if we don’t do X, “we will be in trouble…” kind of vibe (We will be in trouble because of the law, rules, my parents will scold me, Levi hechou is going to kick my ass…)

In the original Eren says:
なきゃ is more casual way to say しなければ
